▼ SAS-6 is the structural core of the forming centriole - a cylindrical protein complex, which is an essential component of the centrosome. Oligomerisation of SAS-6 is crucial for successful centriole duplication and is achieved through two dimerisation domains in the SAS-6 protein; a long C-terminal coiled-coil domain and a globular N-terminal dimerisation domain. As core components of the centrosome, centrioles help facilitate various cellular functions. They are involved in the anchoring of flagella and cilia to the membrane and in coordinating the spindle apparatus during chromosome segregation. A deeper insight into the molecular mechanisms at play in the centriole duplication process would have implications on our understanding of fundamental cell division processes and a number of related diseases. Here the involvement of an unstudied loop region in the C. elegans SAS-6 N-terminal domain dimerisation is described. Combining structural biology, biophysical and computational techniques, the molecular interactions of this loop were explored, contributing to the oligomerisation of SAS-6 at the N-terminal dimer interface. Furthermore, the screening and testing of small molecule inhibitors of the SAS-6 N-terminal domain dimerisation is described, targeting a hydrophobic pocket in the domain. Two candidate compounds are presented as a result of the screens and next steps towards structure based compound design are suggested, based on computational analysis. The search for inhibitory compounds includes a set-up of an in-house virtual screening pipeline, as well as in vitro screening efforts and a new crystallographic structure of the H. sapiens SAS-6 N-terminal domain. By investigating the making and breaking of the SAS-6 N-terminal domain dimerisation, light is shed on so far neglected details of this essential protein-protein interaction and advancements towards a SAS-6 oligomerisation inhibitor described, which could ultimately be used for new approaches in cell cycle research and might open up new avenues for medical research by binding a disease relevant target.

▼ Fluorescent imaging technologies that offer new ways to visualise and quantify fluorescently labelled molecules are increasing, necessitating the development of fluorescent molecules that can efficiently and specifically label targets in vitro and in vivo. The first aim of this thesis was the study of human neutrophil elastase. Human neutrophil elastase is an important enzyme in the regulation of inflammation but if over expressed can become part of the cause of inflammation itself. To elucidate this dual function and have a greater understanding of this enzyme, an imaging probe for neutrophil elastase was designed. Firstly, the syntheses of fluorescently labelled three branched dendron core structures were optimised, and studied in neutrophils. The selected core structure was functionalised with an elastase specific peptide sequence and fluorescently labelled. The probe was specifically cleaved by neutrophil elastase in an enzymatic assay and in the presence of activated neutrophils (Chapter 1). Fluorescein and rhodamine are dyes that are readily available, are affordable and have convenient wavelengths for microscopy and flow cytometry. Carboxyfluorescein diacetate N-succinimidyl ester (CFDA-SE) is a commonly used fluorescein derivative, widely used in cell proliferation assay. It is mainly used as a mixture of isomers and its synthesis is not reported. Herein a short and simple synthesis of the two individual isomers of carboxyfluorescein diacetate N-succinimidyl ester as well as the equivalent rhodamine variation (carboxytetraethylrhodamine N-succinimidyl ester) is reported (Chapter 2). The labelling properties of these probes were studied in proliferation assays on mouse and human T lymphocytes. Finally, the nuclear penetration of the dendron structure combined with nuclear localisation sequences (NLS) was investigated. Attachment of nuclear localisation sequences to the probe in the presence of fluorescein demonstrated successful entry into the nucleus in human alveolar adenocarcinoma cell line (A549) (Chapter 3).

▼ The glucocorticoid receptor (GR) is an important regulator of insulin sensitivity and glucose tolerance. In view of the fact that in vitro the histone deacetylase 6 (HDAC6) has been shown to be a major deacetylase of heat shock protein 90 (HSP90) which in turn is mediating correct folding of the GR and subsequent ligand interaction depending on HSP90 acetylation status the effect of HDAC6 on GR function in a metabolic context in vivo and in vitro was investigated. For this study primary hepatocytes isolated from wildtype and HDAC6-deficient mice as well as chronically dexamethasone- and vehicle-treated wildtype and HDAC6 knockout mice (3 weeks i.p. injection; 1mg/kg body weight) were used for experiments. Additionally experiments in H4IIE rat hepatoma cells with different HDAC inhibitors were performed. Primary hepatocytes isolated from wildtype and HDAC6 knockout mice were transfected with a GR-eGFP plasmid before stimulation with dexamethasone revealing attenuated glucocorticoid-induced nuclear translocation of the GR in HDAC6-deficient hepatocytes by analysis with fluorescence microscopy. This effect was confirmed by evaluating GR protein content in cytoplasmic and nuclear fractions of livers of the corresponding chronically dexamethasone- and vehicle-treated mice. Isolated mRNA from the livers of the animals was used to conduct a whole genome microarray (Affymetrix®) confirming that HDAC6 does not act as a transcriptional regulator but attenuates GR-mediated gene regulation. The expression of major hepatic gluconeogenic genes such as phosphoenol pyruvatecarboxykinase (PEPCK), glucose-6-phosphatase (G6P), pyruvate carboxylase (PCX) and fructose-1,6-bisphosphatase (FBP) was evaluated by qPCR: all genes were significantly upregulated in livers of wildtype mice after dexamethasone treatment. In contrast, dex-induced G6P and FBP upregulation was absent in HDAC6-deficient mice and PEPCK and PCX induction by dexamethasone was significantly reduced compared to dexamethasone treated wildtype mice although significantly induced compared to vehicle treated HDAC6 knockout mice. In addition this effect of HDAC6 was confirmed by the use of tubacin as selective HDAC6-inhibitor in H4IIE hepatoma cells. Here tubacin potently blocked dexamethasone-induced gene expression of G6P and PEPCK. Reduced gene expression of some of the analyzed genes is evidently sufficient to reduce glucose output in primary hepatocytes as dexamethasone stimulation in wildtype and HDAC6-deficient hepatocytes revealed. In wildtype mice chronic dexamethasone injection resulted in a marked glucose intolerance, insulin resistance and hyperinsulinemia. In HDAC6 knockout mice glucocorticoid-induced glucose intolerance was completely abolished. Furthermore, dexamethasone- mediated hyperinsulinemia in wildtype mice did not occur in HDAC-deficient mice and GR-mediated insulin resistance in these mice was ameliorated ascertained by an insulin tolerance test. ▼ Proteolytic enzymes have important functions in physiological and pathophysiological processes, and are frequently used in industrial processes. The search for new proteases with different specificities is always on. In the present work 311 plants from 91 genera from 11 plant families were examined for the presence of proteolytic activity in the latex. 75% of the studied plants were tested positive for the presence of proteolytic activity. Regarding the strength of the activity the families of Euphorbiaceae, Moraceae, and Caricaceae showed the highest fluorescence values. The families of Asteraceae in contrast, had relatively low proteolytic activity. Furthermore, it was found that in the genus Euphorbia, the proteolytic activity is constitutive, and could be found throughout the year. In contrast, some Asteraceae and Campanulaceae showed that the observed activity is dependent on the developmental state of the plant. The investigations showed the different functions of latex proteases. In Euphorbia, they represent defence proteins and are associated with Diterpene derivatives in the defence system of the plant. In Asteraceae and Campanulaceae they appear to play a role in development processes. The subsequent screening of the genus Euphorbia for determining the type of protease showed that serine protease activity was detected in all tested species. The residual activity after treatment with AEBSF ranged from 1-80%. Other inhibitors had also an influence on the proteolytic activity. The assumption that the proteolytic activity in the genus Euphorbia is based exclusively on serine proteases was not confirmed. In some species all tested inhibitors had an effect on the activity. Latices serine proteases have been isolated from also indicate the presence of other protease types. From the latex of Euphorbia mauritanica L. a protease fraction was isolated after the removal of gum and subsequent DEAE- ion exchange and size exclusion chromatography. The unknown protease Mauritanicain has a molecular mass of approximately 95 kDa, an optimum temperature of 55-65°C and a pH optimum of pH 6.5-7.5. It shows activity over a pH range from pH 4.5 to 12 and is stable up to 80°C. The protease has a preference to cut behind the amino acids leucine, lysine and alanine, but also behind non polar amino acids like methionine, valine, phenylalanine, isoleucine, proline and glycine. The testing with specific inhibitors showed serine protease activity. The contact with latices of genus Euphorbia sometimes leads to severe inflammatory reactions. For this reaction primarily diterpene esters are responsible. It could also be shown that proteases lead to an increased secretion of the proinflammatory cytokine Il-6 in acute inflammation in differentiated U-937 cells. The serine protease thrombin, the cysteine protease papain and the diterpene ester PMA affect the secretion of Il-6 in a concentration and time dependent manner. Des substances toxiques générées par les activités portuaires, urbaines et industrielles sont déversées à certains niveaux du milieu marin côtier marocain. Les mollusques peuvent concentrer les polluants et avoir des effets néfastes sur la santé humaine par l’intermédiaire de la chaîne alimentaire. Malgré le renforcement des mesures de sécurité alimentaire, l’implication de la pollution chimique des aliments dans les troubles métaboliques n’est pas à exclure. Pour prédire l’impact des polluants sur l’écosystème aquatique et sur la santé humaine, le développement d’outils de biosurveillance est nécessaire.Nous avons quantifié les métaux lourds (Cd, Cr et Pb) chez les moules (Mytilus galloprovincialis) issues de la côte atlantique marocaine (site industriel Jorf Lasfar (JL) et site touristique d’Oualidia (OL)) en raison de la proximité d’une plateforme d’extraction de phosphate puis caractérisé leurs profils lipidiques (acides gras, cholestérol, oxystérols, phytostérols et phospholipides). Les extraits lipidiques totaux de moules ont été testés in vivo sur des rats pour déterminer leurs effets sur les paramètres biochimiques plasmatiques et in vitro sur une lignée de cellules β pancréatiques murine (MIN-6) en conditions normo et hyperglycémique. Les effets des extraits de moules JL et OL ont été comparés par rapport à ceux de moules d’origine espagnole (ES) destinées à la consommation en France.Les métaux lourds dans les moules JL dépassent les normes internationales. Les concentrations métalliques dans tous les extraits lipidiques sont à l’état de trace. Les moules JL et OL sont moins riches en acides gras insaturés, plus riches en oxystérols et en phospholipides par rapport aux moules ES, suggérant un stress environnemental. Les extraits lipidiques des moules JL et OL administrés à des rats, ont provoqué une perturbation des paramètres plasmatiques, notamment des taux de glucose, créatinine, triglycérides et transaminases avec une augmentation de cholestérol-HDL. In vitro seuls les extraits lipidiques JL et OL induisent la mort des cellules MIN-6 par un processus non apoptotique.
