▼ Les protéines 14-3-3 sont des protéines adaptatrices qui exercent leurs fonctions biologiques en modulant l’activité de centaines d’autres protéines. De part leur impressionnant interactome, les protéines 14-3-3 sont des acteurs qui influencent de nombreux événements cellulaires et, par conséquent, de maladies associées. La stabilisation ou l’inhibition sélective d’interactions protéine-protéine (IPP) de 14-3-3 sont considérées comme des approches prometteuses pour trouver des thérapies innovantes contre des maladies comme la maladie d’Alzheimer, certains cancers ou la maladie de Parkinson.Notre premier but afin de trouver des petites molécules capables de moduler ces cibles a été d’étudier au niveau moléculaire des IPP de 14-3-3. Dans ce but, nous avons utilisé la Résonance Magnétique Nucléaire (RMN) pour attribuer les déplacements chimiques des atomes du squelette de 14-3-3σ. Nous avons ensuite étudié l’interaction entre 14-3-3 et la protéine Tau phosphorylée. Nous avons découvert que Tau se lie strictement dans la cavité amphipathique de 14-3-3 et peut s’ancrer aux deux monomères du dimère de 14-3-3. Nous avons aussi étudié l’interaction 14-3-3/p53 et avons découvert, en utilisant la RMN, que l’affinité du peptide p53 envers 14-3-3 est liée à des interactions intramoléculaires au niveau du peptide. Nous nous sommes enfin focalisés sur l’optimisation d’expériences RMN visant le criblage et la caractérisation de l’activité des petites molécules qui se lient à 14-3-3 ou à des complexes de 14-3-3 avec des peptides phosphorylés. Nous avons aussi utilisé des peptides phospho-mimétiques pour inhiber l’interaction 14-3-3/Tau. D’autre part, nous avons criblé une bibliothèque de fragments contre 14-3-3σ et trouvé trois hits qui se lient à des régions différentes de la protéine. Des expériences RMN ont ensuite permis de caractériser l’activité de certaines petites molécules actives sur des complexes de 14-3-3 avec, par exemple des peptides de p53 ou p65, et nous avons aussi démontré la capacité de certains de ces composés à stabiliser les complexes.
14-3-3 proteins are adapter proteins that exert their biological functions by modulating the activity of hundreds of proteins. This remarkable interactome makes 14-3-3 proteins influent actors in many cellular events and, by consequence, in several pathologies. The selective stabilization or inhibition of 14-3-3 protein-protein interactions (PPIs) are therefore seen as promising approaches for finding innovative therapies for a number of conditions like Alzheimer’s, cancer or Parkinson. Our first objective towards finding small molecule modulators of these targets was to obtain the molecular detail of 14-3-3 PPIs. To this end, using Nuclear Magnetic Resonance (NMR), we assigned the backbone chemical shifts of 14-3-3σ. 3-3 family members are a group of highly conserved scaffolding proteins that are present in all eukaryotes. Despite having limited endogenous activity, the 14-
3-3s bind to numerous client proteins and directly modulate their activities by a variety of mechanisms. In mammals, there are seven isoforms (beta, gamma, epsilon, eta, sigma, theta, and zeta), and between them over 200 known binding partners, which allow them to function in nearly every aspect of cellular biology. The 14-
3-3 family has long been studied in regards to cancer, as aberrant changes in their expression patterns have been associated with numerous human cancers. It is, therefore, a shared goal, among many, to elucidate the role of 14-
3-3 proteins in tumorigenesis. For the last decade, our laboratory has been keenly focused on the tumor-promoting roles of 14-
3-3 proteins in the context of lung cancer, as their expression patterns are highly dysregulated in this cancer setting. Herein, we expand on this area of research and demonstrate that four of the seven 14-
3-3 isoforms are significantly elevated in both adenocarcinomas and squamous cell carcinomas of the lung. We then investigated the consequences of these isoforms being increased in malignant lung tissues and showed that when upregulated, 14-
3-3 sigma, gamma, and zeta correlate with poorer prognosis in patients with lung adenocarcinoma (LUAD). Interestingly, these associations with survival were not observed in patients with lung squamous cell carcinoma (LUSC), suggesting that the upregulation of these 14-
3-3 isoforms may influence patient survival and serve as suitable prognostic biomarkers.
To explore the cellular consequences of 14-
3-3 upregulation, our laboratory previously overexpressed 14-
3-3 gamma, the isoform that demonstrated the strongest prognostic capacity in the LUAD-TCGA dataset, in human lung adenocarcinoma cells. Overexpression of this isoform caused a fraction of cells to become polyploid, meaning they contained more than two sets of chromosomes. Amassing data have demonstrated that polyploid cells are uniquely resistant to chemo- and radiotherapy, making polyploid cells integral components in driving the ongoing evolution of the patient disease and recurrence. We, therefore, became interested in illuminating the molecular mechanism(s) driving 14-
3-3 gamma-induced polyploidy, the net effect these cells had on genomic integrity, and whether this phenomenon occurred in vivo. By utilizing the fluorescence ubiquitin cell cycle indicator (FUCCI) system, we were able to show that overexpression of 14-
3-3 gamma resulted in inhibition of mitotic entry, forcing some cells to bypass mitosis entirely, thereby facilitating the polyploid phenotype. In pursuit of investigating whether these polyploid cells could re-enter the cell cycle and undergo cell division, we developed a widely-applicable, nontoxic procedure for measuring DNA content in live cells by fluorescence microscopy. The importance of the organizational culture like key factor for the efficiency of the organization, like comparative advantage and
also like process of change is demonstrated.
The organization can through performances and policies generate behaviors that support the existence of values within the
organization.
The equality of opportunities between women and men constitutes a value that must join the organizations, understanding the
equality of opportunities of kind as the absence of obstacles or barriers that make difficult to the women the participation in the
economic area, cultural and social politician in equality of conditions that the masculine group.
The equality of sort opportunities reports to the organizations a series of advantages and benefits that have their base in the
satisfaction of the personnel, the labor climate, the commitment with the organization, the enrichment and the corporative image.
The perception of an unjust treatment or discriminatory has organizational and personal, such consequences as the decrease of
the productivity, the increase of the labor absenteeism and the increase of complaints and legal actions.
The nonexistence of information and investigations on the perception of equality of opportunities of kind and his possible relation
with the organizational culture, has motivated the exposition of this thesis. The general aim of the thesis consists of determining if
relation exists between the organizational culture and the equality of opportunities of kind. To obtain this general aim it has
considered to identify the areas or dimensions more relevant that they affect the perception of equality of opportunities of kind. To
identify if there exists a typology of organizational culture that favors or impedes the perception of equality of opportunities of kind
and to analyze the relation between the dimensions of equality of kind and the type of organizational culture.
To reach the proposed aims has followed a concrete methodology characterized by

is designed and validated a Scale of measurement of the perception of equality of opportunities of kind (EIOG), the second phase,

▼ The questions raised that are the basis of the research and the source that feeds the objectives are:
1. Is it possible to define a data model to undertake sustainability studies for territories with different types of organization, initially unknown?
2. Can data evolve incorporating new requirements and techniques, so it is re-usable for subsequent follow up studies?
3. Can the system incorporate views of researchers that come from different research areas, possibly with conflicting interests?
Three knowledge areas are involved: sustainability; data, metadata and ontologies, and information systems.
The objectives of the research are:
- Conceptual definition of variables and values in the area of sustainability.
- Establish a variables calculation and agregation method.
- Definition of an integrated data structure.
- Integration of conceptual models in the data structure.
- Validation of the proposed methodology through prototyping.
These objectives fit into the following main areas of concern: (a) sustainability measurement, calculation of indicators and indexes, territorial organization and free software. The methodology used is to collect and analyze the current status of each area, the vision as post-normal science, systems thinking and prototype development.
The thesis shows the relationships between territory, data and conceptual models, and also proposes methods that allow them to work together. It defines the concepts of variables and values, and their relationship with the values ¿¿of conceptual models (reference values, thresholds ...). Pointed out contributing methods:
- Define and integrate wide variety of organizational structures.
- Define and calculate any type of indicator, index or variable.
- Use conceptual frameworks concurrently with different sustainability criteria.
- Prototypes are developed using free software

3-diaza-Claisen rearrangement and the design and development of cationic 1,
3-diaza-Claisen rearrangement.
Our previous work has shown that isocyanates react with azanorbornenes and azabicyclo[2.2.2]octenes under thermal conditions to afford zwitterionic intermediates that undergo a thermal 1,
3-diaza-Claisen rearrangement to give both ureas and isoureas. However, some azanorbornenes and azabicyclooctenes failed to rearrange or proceeded in low yields. To address these challenging substrates for the thermal 1,
3-diaza-Claisen rearrangement, we have developed a Pd
0-catalyzed formal 1,
3-diaza-Claisen rearrangement. Interestingly, under Pd
0-catalyzed condition, both isocyanates with electron-withdrawing groups and isocyanates without electron-withdrawing groups react with azanorbornenes and azabicyclo[2.2.2]octenes to provide ureas as the only products in high yields. More importantly, the reactions that failed under thermal conditions were all successful under Pd
0-catalysis. In addition to azanorbornenes and azabicyclo[2.2.2]octenes, other ring systems were also investigated. Pd
0 catalysis has broadened the scope of tertiary allylic amines that react with isocyanates to afford 1,
3-diaza-Claisen rearrangement products.
In the presence of p-TsCl and NEt
3, allylaminopropyl benzyl ureas were initially dehydrated to form protonated carbodiimides whose presence was confirmed by the infrared absorption frequency at 2100 cm
-1 which is the characteristic band of -N=C=N-; then the in situ generated protonated carbodiimides were poised for further cationic 1,
3-diaza-Claisen rearrangement to afford synthetically challenging guanidines. The effect of acid on the rearrangement was ascertained by the fact that no rearrangement product was observed by simply heating free base carbodiimide
3.10 in benzene at reflux. Other dehydration reagents, such as Tf
2O, Ts
2O, MsCl were also investigated, and none of them provide satisfactory results. A selection of allyamino benzyl ureas with different tether length, substituents, or in varied ring systems, were synthesized to explore the scope of this methodology. This methodology works best at allylaminopropyl benzyl ureas, and the substituents on the benzyl group does not seem to affect the reaction rate in a significant way.

PPI-Stabilisatoren müssen PPIs von Interesse zunächst umfassend
charakterisiert werden. Es gilt auÿerdem zu überprüfen, ob eine
Stabilisierung der untersuchten PPI pharmakologisch sinnvoll ist.
In der vorliegenden Arbeit wurden die Wechselwirkungen von
14-
3-3-Proteinen mit MLF1 und C-RAF mit biochemischen und
strukturbiologischen Methoden charakterisiert. Um zu überprüfen, ob
eine Stabilisierung dieser Wechselwirkungen pharmakologisch
sinnvoll ist, wurde überdies mit zellbiologischen Methoden der
physiologische Effekt dieser Wechselwirkungen untersucht. Basierend
auf der im Vorfeld dieser Arbeit gelösten Röntgenkristallstruktur
des Komplexes aus 14-
3-3e und MLF1(29-42)pS34 wurde die
MLF1/14-
3-3-Wechselwirkung mittels ITC untersucht. Der erhaltene
KD-Wert für das WT-MLF1-Peptid lag erwartungsgemäÿ im niedrigen
mikromolaren Bereich. Die Untersuchung verschiedener MLF1-Mutanten
zeigte, dass neben dem pS34 nur das F33 einen signifikanten Beitrag
zur Bindung an 14-
3-3-Proteine leistet. Weiterhin wurde durch den
Einsatz von GFP-MLF1-Fusionsproteinen die subzelluläre Lokalisation
des humanen MLF1-Proteins in HEK293T-Zellen auf eine direkte
14-
3-3-Abhängigkeit untersucht, wie sie von Winteringham et al.
(2006) für murines MLF1 in Cos-7-Zellen beobachtet wurde. Es zeigte
sich jedoch, dass die subzelluläre Lokalisation von humanem MLF1 in
HEK293T-Zellen nicht direkt 14-
3-3- abhängig ist und der von
Winteringham et al. (2006) vorgeschlagene Mechanismus zur
Regulation von murinem MLF1 durch 14-
3-3-Proteine nicht direkt auf
das humane Homolog übertragbar ist. Die S259-vermittelte
C-RAF/14-
3-3-Wechselwirkung wurde im Vorfeld dieser Arbeit bereits
intensiv mit strukturbiologischen, biochemischen und
zellbiologischen Methoden charakterisiert. Daher wurde in der
vorliegenden Arbeit der physiologische Effekt von Mutationen der
14-
3-3-Bindestelle um S259, wie sie in Noonan- und LEOPARDSyndromen
gefunden wurden, untersucht. Es konnte nachgewiesen werden, dass
diese Mutationen aktivierend auf den RAS-RAF-MEK-ERK-Signalweg
wirken. Diese Auswirkung konnte bereits für die anderen, in
Noonan-und LEOPARD mutierten Gene, PTPN11, SOS1 und KRAS, gezeigt
werden (Tartaglia et al., 2001; Carta et al., 2006; Schubbert et
al., 2006; Roberts et al., 2007; Tartaglia et al., 2007). Es zeigte
sich weiterhin, dass die eingesetzten, C-terminal verkürzten
C-RAF-Mutanten keinen Einfluss auf die subzelluläre Lokalisation
der endogenen C-RAF-Proteine hatten. Dieses Ergebnis bekr äftigt
die These, dass C-RAF-Proteine über ihren C-terminalen Bereich
dimerisieren (Weber et al., 2001; Rushworth et al., 2006). Die
biochemische Untersuchung der S233-vermittelten
C-RAF/14-
3-3-Wechselwirkung zeigte, dass 14-
3-3-Proteine sowohl
über S233 als auch gleichzeitig über S233 und S259 an C-RAF binden

als die S259-vermittelte Bindung. Eine gleichzeitige Bindung über
S233 und S259 wirkt synergistisch und erhöht die Affinität

This work describes the design and synthesis of peptidomimetics for the inhibition of the protein-protein interactions (PPI) with 14-
3-3. This protein is a promiscuous binder of other proteins with a variety of key functions such as the regulation of proapoptotic proteins (e.g. FOXO, BAD), suppression of tumor growth, and sensitizing cells to other drugs. The promiscuous binding is the result of PPIs being shallow binding sites which accept many proteins and modify their activity. Compounds which inhibit 14-
3-3 include peptides which induce cellular apoptosis. As a result, this interaction has been identified as a potential drug target for future cancer therapeutics, however the 7 isoforms are not perfectly redundant adding additional selectivity development as an eventual direction.
The Borch lab has a long standing interest in phosphoramidate prodrugs, here incorporated into peptidomimetics, to improve the cellular permeability of charged phosphate compounds. The previous work by Kim et al utilized these prodrugs to develop a modified version of the peptidomimetic produced by Shao Q. Yao.
2 The original compound 1 had an IC
50 of 2.6±0.4 µM with molecular interactions, while the prodrug analogue 2, which included the difluoromethylene phosphonate analogue of the phosphate, had an IC50 near 5 µM in cells. The exact results were unable to be determined as they resulted in cytotoxicity presumably from 14-
3-3 activation.

The peptidomimetics designed herein were modeled in comparison with the compounds produced by Kim et al. In this process two prospective binding modes were identified. One involved a linear binding method while another has a bent orientation. The compounds previously produced bind in a bent method while those found in the linear method more closely resembled the orientation of several peptides designated consensus sequences. Unsurprisingly those which more closely resemble the crystallized consensus sequences produced higher G-scores. The goal of this work was then to produce these compounds for initial confirmation of increased activity by these trial runs.

▼ Neuronal connectivity is precisely determined by axonal pathfinding during development. The navigating axons detect attractive and repulsive environmental cues by axon guidance receptors. However, the biochemical means through which multiple signaling pathways are integrated in navigating axons is poorly understood. Semaphorins are the largest family of axon guidance cues and utilize Plexin receptors to exert repulsive effects on axon extension. The intracellular region of Plexins contains a Ras GTPase activating protein (GAP) domain, which is necessary for repulsive guidance effects. Previous studies suggest that activation of Plexin RasGAP requires interactions with both Semaphorin at the extracellular region and a Rho-family GTPase at the Rho family GTPase-binding domain (RBD). Interestingly, Semaphorin repulsion can be rapidly "turned-off" by other distinct cues and signaling cascades. However, the molecular mechanisms to activate or modulate Plexin RasGAP remain unclear. First, to further understand how the Plexin RasGAP is activated, I collaborated with the Zhang lab, and following determination of the crystal structure of the intracellular region of Plexin, I examined the roles of residues interfacing with the RasGAP domain using functional mutagenesis in the Drosophila model system. Our results demonstrate that Plexin exhibits an auto-inhibited conformation, and suggest that interaction among the previously uncharacterized juxtamembrane segment, the RBD, and the RasGAP domain is critical for Plexin RasGAP activation. Second, to better understand how Semaphorin/Plexin signaling is modulated, I characterized the results of a large-scale screen to look for proteins interacting with the cytoplasmic portion of Plexin and identified the phosphoserine binding protein 14-
3-3epsilon as a specific Plexin-interacting protein. My results reveal that 14-
3-3epsilon is specifically required for axon guidance during development. Moreover, Protein kinase A is found to phosphorylate Plexin in the RasGAP domain and mediates the 14-
3-3epsilon interaction. Plexin-14-
3-3epsilon interactions prevent Plexin from interacting with its Ras-family GTPase substrate, which effectively switches Plexin-mediated axonal repulsion to Integrin-mediated adhesion. These findings uncover both a new molecular integration point between important axon guidance signaling pathways and a biochemical logic by which this guidance information is coalesced to steer the growing axon. Therefore, these new observations on activating and silencing specific signals that are repulsive to axon growth also illuminate new approaches to neutralize axonal growth inhibition and encourage axon regeneration.

▼ Glioblastoma is the most malignant form of brain cancer. Due to its aggressive nature, extensive research has been performed, but little progress has been made in identifying effective treatment options. Glycogen synthase kinase-3 (GSK-3) is a ubiquitous, multifaceted protein kinase. Previous studies have shown that small molecule inhibitors of GSK-3 block the migration of glioblastoma cells and may prevent spread of tumor in the brain. However, these studies were performed using non-selective GSK-3 inhibitors (LiCl and an indirubin derivative, BIO); thus, it was unclear whether GSK-3 was the most important target. In this study, we used recently generated highly selective GSK-3 inhibitors (CHIR99021, AZD1080, and AZD2858, as well as BIO) to investigate these questions. These were applied to four glioblastoma cell lines: G30, G9, U251, and U1242, in three migration assays: transwell, spheroid, and wound healing (scratch) assay to further assess the suitability of GSK-3 as a target in glioblastoma. We also utilized the ATP Luciferase reporter assay for cell viability to assess the influence of our panel of drugs on cell migration versus viability. In addition, the TOPFlash Luciferase reporter assay was performed as an indicator of the level of GSK-3 inhibition.
The TOPFlash assay showed that all GSK-3 inhibitors were able to increase luciferase levels. This indicates that GSK-3 was inhibited in our cells after drug treatment. The transwell assays showed us that the GSK-3 inhibitors were able to block migration significantly in all cell lines tested in a dose-dependent manner. The effectiveness of GSK-3 inhibition in the three-dimensional collagen spheroid assays was cell line-dependent, with the non-selective GSK-3 inhibitor BIO showing the most potent effects. Cell migration was not blocked by any of the three selective GSK-3 inhibitors in the wound healing scratch assay. Thus we have found that the three distinct highly selective inhibitors of GSK-3 block glioblastoma cell migration, but only work consistently in the transwell assay. Therefore, we conclude that GSK-3 might be important in the contraction and morphological changes necessary for glioblastoma cells to migrate through the 8 micron pores in the transwell. Further investigation into this observation is necessary. Though results were variable between assays, we conclude that the inhibition of GSK-3 is a promising potential therapeutic strategy for glioblastoma treatment.

▼ The contribution of interleukin-3 (IL-3), a hematopoietic growth factor and immunoregulatory cytokine, to resistance to blood-stage malaria was investigated by infecting IL-3-deficient (knockout [KO]) mice with Plasmodium berghei NK65. The results show that endogenous IL-3 significantly alters the susceptibility of BALB/c mice to P. berghei NK65 infection as reflected by measures of survival, anemia, parasitemia, and splenomegaly. Male IL-3 KO mice, but not female mice, were more resistant to infection than wild-type (WT) mice, as evidenced by lower peak parasitemia and prolonged survival. Both male and female IL-3 KO mice had increased splenomegaly and were more anemic than corresponding WT mice. Anemia was compensated for by an increase in bone marrow and splenic erythropoiesis in male IL-3 KO mice, as evidenced by higher levels of erythroid progenitors. P. berghei NK65 infection was also shown to induce IL-3-dependent changes in plasma cytokine levels: plasma levels of gamma interferon (IFN-γ) and chemokine (C-X-C motif) ligand 9 (CXCL9 or monokine induced by IFN-γ [MIG]) were found to be significantly reduced in male IL-3 KO mice during early stages of infection. In contrast, granulocyte colony-stimulating factor (G-CSF) levels were significantly higher, and the percentage of peripheral blood neutrophils lower, in P. berghei NK65-infected male IL-3 KO mice than in WT counterparts. P. berghei NK65 infection induced degranulation of splenic mast cells in both male IL-3 KO and WT mice; however, there was no difference in mast cell numbers between genotypes. Overall, our results indicate that IL-3 plays a critical role in suppressing protective immunity to P. berghei NK65 infection and that it is involved in inhibiting the development of splenomegaly, anemia, and erythropoiesis. IL-3 also influences IFN-γ, CXCL9, and G-CSF production in response to infection. The abnormal responses seen in infected IL-3 KO mice may be due to the lack of IL-3 during development, to the lack of IL-3 in the infected mature mice, or to both.

▼ This thesis is concerned with estimating the
time-varying
3-d surface geometry and surface appearance of dynamic
scenes from images. An accurate reconstruction of a 4-d
(space+time) representation of the scene yields an encoding of the
4-d events in the scene and facilitates inference tasks such as
detecting and recognizing objects, and tracking their movements.
Reconstructing the full 4-d world from images is a very difficult
problem because much information is lost in the perspective
projection from
3-d to 2-d image space. Most existing works require
prior shape and motion models for objects in the scene. This thesis
presents a framework capable of reconstructing accurate 4-d models
of general scenes without knowledge of the shapes in the scene or
their motion. The thesis introduces a volumetric 4-d representation
that is able to encode complex
3-d shapes that deform in arbitrary
ways. The representation encodes probabilistic estimates of
3-d
surface geometry and appearance, which describes the ambiguities of
reconstruction. A novel 4-d data structure is defined that adapts
the resolution of 4-d cells according to the spatio-temporal
variations in the scene. Experiments demonstrate that this adaptive
scheme requires less storage compared to the common approach of
storing separate
3-d models for each time step. The thesis also
presents two reconstruction algorithms to estimate this 4-d
representation from images. The first algorithm is suitable for
synchronized multi-view video streams captured in indoor studios
and achieves high quality 4-d models from standard datasets. A
novel appearance model for non-Lambertian surfaces enables accurate
reconstruction of reflective materials in these datasets, which has
not been previously demonstrated. The second reconstruction
algorithm is suitable for realistic outdoor environments that are
imaged periodically and change slowly over time. The algorithm
proceeds by detecting and reconstructing changes in the scene to
continuously update an evolving
3-d model. Experiments show that
the algorithm requires less imagery and computation time, and leads
to higher quality 4-d models, compared to full scene reconstruction
at each time step.

3 (CR3), a β2 integrin found on
neutrophils, plays an important role in fungal recognition and
immune response. Co-ligation of CR3 with fibronectin (Fn), an
ubiquitous extracellular matrix component, at the I-domain and
β-glucan (B), a glucose polymer found in fungal cell walls, at the
lectin-like domain is possible due to spatially distinct locations.
Dual ligation of CR3 with Fn+B induces homotypic aggregation of
primed neutrophils, a response representative of the immune cells
encounter with fungi within tissues and that can be blocked by
anti-CR3 antibody. Previous findings have shown that CR3-mediated
DNA Neutrophillic Extracellular Traps (NETS) coincide with PMN
aggregates. To understand the molecular mechanisms of how CR3
coordinates these neutrophil responses, this thesis explores the
role of the integrins CR3 as well as the β1 integrin family members
VLA3 and VLA5. Using antibody blockade and receptor-FRET, we
unexpectedly discovered a CR3-mediated required role for VLA3, the
canonical laminin receptor, in aggregate formation. The role of
VLA3 in aggregate formation was specific as blockade of other β1
integrin family members such as VLA5, the canonical Fn receptor,
failed to prevent aggregation. Exploring the role of VLA5, we were
able to show a decrease in overall VLA5 activity on Fn+B and by
using an activating anti-VLA5 antibody, that active inhibition of
VLA5 activity is necessary for aggregate formation. In assessing
NETs formation, we discovered that unlike aggregate formation, VLA5
and CR3 are necessary for NET formation, where VLA3 is not.
Furthermore, activation of VLA5 with the activating VLA5 antibody
does not inhibit NET formation. Finally, a correlative signaling
role for p-ERK and p-GSK-3β was determined in the neutrophil
responses to β-glucan and Fn but not to either alone. Within this
thesis, a novel, complex and temporally regulated cross-talk
pathway was uncovered in which dual ligation of CR3 signals the
differential regulation of individual β1 integrins not often
associated with anti-fungal activity. Furthermore, it was
discovered that NETosis and aggregation are controlled by the
activation state of distinct β1 integrins, VLA5 and VLA3,
respectively.

▼ Given a set of images of a scene, it is possible to
estimate both the
3-d position of points in the scene and images
from viewpoints not present in the original data. These estimates
cannot in general be exact, however, due to uncertainties and
ambiguities present in the data. For this reason, probabilistic
scene models and reconstruction algorithms are ideal due to their
inherent ability to represent such uncertainties and ambiguities.
Unfortunately, existing probabilistic reconstruction algorithms do
not scale well to large and complex scenes, primarily due to their
dependence on large three-dimensional voxel arrays. The work
presented in this thesis generalizes previous probabilistic models
in such a way that multiple orders of magnitude savings in storage
are possible, making accurate
3-d point localization and high
resolution novel view generation of large-scale outdoor scenes
possible. Specifically, the inherent dependence on a discrete array
of uniformly sized voxels is removed through the derivation of a
continuous probabilistic model which represents uncertain geometry
as a density field, allowing implementations to efficiently sample
the volume in a non-uniform fashion. In addition, multiple
reconstruction algorithms are presented to accommodate differing
modes of operation in which imagery may be captured and used. The
proposed model combined with the reconstruction and novel view
generation algorithms comprise the first system capable of
automatically generating photo-realistic renderings of large and
complex scenes from arbitrary viewpoints based on aerial image data
alone.

▼ Nous présentons dans ce travail l’analyse des données polarimétriques obtenues depuis le sol permettant de déterminer les propriétés des aérosols. Un travail préalable de préparation des données a été mené de manière à automatiser l’ensemble des traitements. Nous avons ensuite appliqué aux données l’algorithme d’inversion en usage au sein du réseau international de photomètre solaire AERONET (Aerosol RObotic Network) en y intégrant la polarisation. Une étude de sensibilité s’appuyant sur cinq modèles représentatifs de l’aérosol désertique, industriel et urbain, jusqu’aux aérosols marins a été réalisée. Les effets du bruit, de biais sur les mesures d'épaisseur optique aérosols et des erreurs de pointage ont été évalués. Les résultats montrent l’avantage la polarisation dans le cas de concentrations importantes en particules fines. A l’inverse, la polarisation s’est révélée moins sensible à la présence d’aérosols dominés par les grosses particules. Les données acquises sur trois sites d’observation (Beijing, Dakar, Washington), ont été traitées. Les résultats montrent l’intérêt de la polarisation dans le cas de la pollution industrielle pour améliorer la restitution de la distribution de taille des particules. La prise en compte de la polarisation améliore la qualité de la restitution de partie réelle de l’indice de réfraction et de la fraction de particules sphériques, et cela en raison de la haute sensibilité de la polarisation à la forme des particules. Cette approche est particulièrement intéressante pour mieux caractériser les aérosols dans les régions affectées par la présence d’aérosols d’origine anthropique (fraction fine prononcée) ou les aérosols de mélange.
The thesis presents the efforts on including the polarimetric data to the routine inversion of the ground-based observations and analysis of the advantages obtained. To process large amount of observations a data preparation tool was developed. It is based on AERONET inversion code. To assess the importance of polarization for aerosol retrieval results, numerical test has been performed and the real measurements were processed. First, the sensitivity study has been carried out using simulated data based on five aerosol models: desert dust, urban industrial, urban clean, biomass burning and maritime. The test investigated the effects of various errors (random noise, bias in optical thickness data, pointing errors) on aerosol retrieval using polarization data. The results demonstrate the advantage of using polarization for characterization of aerosols with pronounced concentration of fine particles. In case of the coarse mode dominated aerosols, the use of polarimetric data led to minor improvements of the retrieval. Second, the extended set of AERONET observations were processed. ▼ Ce travail a pour objectif de développer un outil numérique de modélisation du comportement des aimants permanents tout en tenant compte de la démagnétisation induite par divers facteurs comme la géométrie, le champ induit et la température. Le modèle de démagnétisation est basé sur une expression analytique (l’équation de Marrocco). Il nécessite l’identification de quatre paramètres et l’implémentation des lois polynomiales de variation pour certains d’entre eux afin de prendre en compte la démagnétisation. Ce modèle a été implémenté dans le code de calcul par éléments finis code_Carmel. Pour prendre en compte l’influence de la température, un chaînage a été effectué avec un code thermique, SYRTHES. Afin de valider cet outil numérique, une maquette expérimentale a été développée. Dans un premier temps, le modèle de calcul des pertes par courants induits dans les aimants permanents a été validé en utilisant une approche basée sur une procédure de séparation des pertes qui combine les résultats expérimentaux à ceux calculés. Ensuite, différentes grandeurs globales et locales issues du modèle de démagnétisation des aimants permanents ont été comparées aux mesures expérimentales avec une bonne concordance montrant ainsi l’aptitude du modèle proposé à représenter le comportement d’un aimant permanent soumis à une élévation de la température et à un champ démagnétisant.
This work aims at developing a numerical tool able to simulate the behavior of permanent magnets while taking into account the demagnetization induced by various factors such as geometry, induced field and temperature. The demagnetization model is based on an analytical expression (the Marrocco equation). It requires the identification of four parameters and the implementation of polynomial variation laws for some of them in order to be able to take into account the demagnetization. This model has been implemented in the finite element calculation code code_Carmel. In order to also take into account the influence of temperature the model was coupled to a thermal calculation code, SYRTHES. To validate this tool, an experimental device has been developed. Initially the permanent magnet eddy current losses model has been validated using a loss balance procedure that combines both experimental and numerical approaches. This validation allows then to provide a good estimation of the temperature rise that appears inside the magnets. Finally the permanent magnet demagnetization model has been validated by comparing different global and local calculated values with measured ones. These results showed a good agreement and demonstrated a good ability of the proposed method to take into account the behavior of demagnetization of permanent magnets under the influence of induced magnetic field and temperature.

▼ Au delà de leur impact sur la qualité de l'air que nous respirons et sur la santé, les aérosols atmosphériques constituent une des pièces maîtresses de la machine climatique. De fortes incertitudes demeurent aujourd'hui encore (IPCC, 2013) du fait de la grande diversité de leurs propriétés, de leurs multiples interactions avec le rayonnement et les nuages, ainsi que de leur grande variabilité spatio-temporelle. Dans ce contexte, la mise en place d'un suivi sur le long terme est fondamentale. En 2006, le LOA a enrichi les observations photométriques qu'il conduisait, avec la mise en place de micro-lidars sur les sites de Lille et Dakar. Notre travail porte sur l'analyse à multi-échelle temporelle de ces mesures complémentaires pour détecter d'éventuelles tendances et évaluer la variabilité des aérosols. Ces observations incluent aussi des évènements extrêmes (éruption de l'Eyjafjöll, 2010). L'exploitation de cette base de données riche et originale a nécessité le développement et la mise en oeuvre d'un algorithme opérationnel (BASIC), combinant profils verticaux et mesures photométriques. Ces travaux présentent un intérêt pour la communauté française, en particulier dans le cadre du système d'observation ORAURE et du réseau de surveillance de Météo France. Nos travaux nous ont permis de préciser l'impact radiatif des aérosols. Nos résultats ont également été utilisés pour une étude de validation des profils de CALIOP et pour une première évaluation du modèle NMMB/BSC-Dust, en Afrique. Enfin, nous présentons les premiers résultats d'un système mobile permettant l'étude de la variabilité spatiale aérosol à haute résolution, déployé pendant la campagne DRAGON (USA, 2011).
Beyond their impact on air quality affecting health, atmospheric aerosols are a key parameter of the climate. Large uncertainties remain (IPCC, 2013) due to their type versatility, radiative and clouds interactions and their strong variability in space and time. In light of this, a long-term observation is highly required. In 2006, the LOA reinforced sunphotometer observations with two micro-lidars in two contrasted sites : Lille and Dakar. In this work, we analyse these complementary observations at multiple time scale in order to highlight trends and provide a variability estimation. These observations include detection of extreme events (volcanic ash from Eyjafjöll, 2010). The analysis of the unique and large database is based on the development of an automatic algorithm (BASIC) combining vertical profiles and sunphotometer measurements. This work is particularly relevant and of interest in the current and future context of observation systems (ORAURE, Météo France). Our results are used to provide a better determination of aerosols radiative effect. ▼ Afin de produire de l’électricité, une éolienne doit s’orienter de manière à être face au vent. Cette rotation est assurée par un système de giration placé entre le mât et la nacelle appelé « yaw ». C’est lors de la phase de yawing que peut apparaître le phénomène de « crissement » à cause du contact frottant entre les garnitures des freins et le disque, l’effort de freinage n’étant pas relâché afin de contrôler la giration de la nacelle. Dans un objectif de réduction voire de suppression du bruit, une démarche scientifique de compréhension du phénomène comprenant un volet structure et un volet matériau a été mise en place, aucune étude sur le sujet n’ayant jamais été réalisée dans le domaine éolien. La première étape de l’étude a consisté en une caractérisation du problème en conditions réelles sur éolienne. Les données collectées complétées par une étude bibliographique élargie ont permis de définir une stratégie d’étude : de manière globale, cette recherche est basée à la fois sur une approche expérimentale et sur une approche numérique. L’accent est mis sur l’influence du matériau de friction : plusieurs matériaux ont été investigués dont l’un correspond à une « amélioration » d’une formulation industrielle initialement relativement « bruyante ». Ces matériaux sont également décrits et caractérisés en termes de propriétés mécaniques et les surfaces frottées sont observées après essai. Au final, cette étude a permis de maitriser et d’identifier certains paramètres influents tels que le rôle des garnitures, amenant de nombreux éléments de compréhension des phénomènes mis en jeu afin d’appréhender des voies d’amélioration pour la réduction du bruit.
In order to produce electricity, a wind turbine needs to be face to the wind. The system allowing its orientation is placed between the tower and the nacelle and is called ‘yaw’. Due to the contact between friction pads and disc during the rotation phases, squeal noise can occur, the braking forces being used to control the nacelle rotation. In the aim of reducing and even suppressing the noise, a scientific approach has been set up to understand the phenomena involved, including the structure and the friction material aspects, no such study has ever been carried on in the wind turbine domain. The first step has consisted in characterizing the problem in real conditions on wind turbines. The collected data and an extended bibliographic study have been used to define a strategy: globally, the study is based both on experimental investigations and numerical simulations. The influence of the friction material has been highlighted: several materials have been investigated, one of them being a “improvement” of a commercial material initially “noisy”. Those friction materials are also described and characterized regarding their mechanical properties and the rubbing surfaces have been observed after tests. ▼ La robotique souple s’inspire de la nature, de la manière dont les organismes vivants se déplacent et adaptent leur forme à leur environnement. Contrairement aux robots traditionnels, les robots souples accomplissent des tâches avec plus de flexibilité. Les matériaux souples avec lesquels ils sont construits les rendent plus sûrs pour des environnements fragiles.Cependant, le domaine de la robotique souple pose de nouveaux défis, en particulier pour la modélisation et le contrôle. Dans cette thèse, nous visons à fournir des méthodes génériques pour leur modélisation. Les méthodes sont basées sur la méthode des éléments finis pour capturer les déformations de la structure du robot, et de son environnement, quand il est déformable. Nous formulons le problème de leur cinématique inverse et dynamique inverse comme un programme d’optimisation, permettant de gérer facilement des contraintes aux actionneurs et des problèmes de singularité. Nous sommes en mesure de contrôler plusieurs types d’actionnement, tels que les actionnements par câbles, pneumatiques et hydrauliques.De plus, la plupart des applications impliquent une interaction du robot avec des obstacles. Or, la cinématique des robots souples dépend fortement des facteurs environnementaux. Nous proposons ainsi de nouvelles méthodes qui prennent en compte les contacts dans le processus d’optimisation. Enfin, nous proposons de contrôler certaines tâches de locomotion et de préhension nécessitant l’utilisation de contacts frottants (statique). Nous accordons une attention particulière à fournir des solutions avec des performances temps réel, permettant un contrôle en ligne des robots dans des environnements changeant.
Soft robotics draws its inspiration from nature, from the way living organisms move and adapt their shape to their environment. In opposition to traditional rigid robots, soft robots are built from highly compliant materials, allowing them to accomplish tasks with more flexibility. They are safer when working in fragile environment, which allows for potential use of soft robotics in the fields of manufacturing and medicine.Yet, the field of soft robotics brings new challenges, in particular for modeling and control. Within this thesis we aim at providing generic methods for soft robot modeling, without assumptions on the geometry. The methods are based on the finite element method to capture the deformations of the robot’s structure and of its environment when deformable. We formulate the problem of their inverse kinematics and dynamics as optimization programs, allowing easy handling of constraints on actuation and singularity problems. We are able to control several types of actuation, such as cable, pneumatic and hydraulic actuations.Moreover, most of the applications involve interaction of the robot with obstacles. ▼ The thesis starts with a first study about the family business as a corporation, with its peculiarities and its engagement with existing legal forms. Considering that one of its most important elements is its vocation of continuity, and considering also that the problems with the generational change are the leading cause of death of such companies, we analyse how the Family Constitution can solve that problems.
Within the Family Constitution analysis, we review its historical background, the experience in other jurisdictions and, above all, its efficacy problems in Spain. These are the problems of all shareholders' agreements, but with an additional element: not all its content can be binding, because it conflicts with mandatory norms. Therefore, we propose to improve the effectiveness through specific legal solutions (penalty clauses, additional provisions), some relatively recent options (access to the Registry of Commerce) and, above all, the full conviction of its signatories, consequence of their consensus.

▼ In the United States, as demonstrated by the multiple cases of exonerated in recent years through DNA testing, many innocent people are convicted from misidentifications. Photography has a decisive role in this, since photo lineups are the most common method of identification today.
The main objectives of this thesis are: first, to demonstrate that photography is not an infallible tool, or even effective, as a proof of identification. Second, to understand why the US police and judicial system —cognizant of the flaws incurred by this procedure— allow the use of photo lineups. And, finally, to offer arguments from the Psychology of facial recognition and Psychology of person recognition so that the eyewitness psychologists contemplate the necessity to undertake new comparative studies between photo and video lineups.

▼ Face à l’épuisement du pétrole, l’Europe s’oriente vers la production de bioéthanol 2ème génération à partir de biomasse végétale. L’espèce Miscanthus x giganteus est prometteuse de par son fort rendement et ses faibles impacts environnementaux. Cependant, sa diversité génétique est faible et sa composition de biomasse pas forcément optimale pour produire du bioéthanol 2ème génération. Cette thèse explore donc différentes espèces de Miscanthus pour définir des idéotypes et des pistes en sélection afin d’améliorer le miscanthus pour ce débouché. Trois approches ont été menées : une étude bibliographique, une étude expérimentale au champ et des tests en pilote industriel. L’étude bibliographique montre une forte variabilité environnementale et génotypique pour les caractères associés à la production et à la composition de biomasse. L’analyse des données expérimentales révèle des caractères antagonistes : une forte production de biomasse est associée à de fortes teneurs en cellulose et lignines, mais à de faibles teneurs en hémicelluloses, solubles et cendres. De fortes teneurs en lignines contribuent également à réduire l’efficacité de la transformation de la biomasse en bioéthanol 2ème génération. Enfin, des outils de phénotypage ont été définis : la hauteur de végétation et le volume aérien de la plante permettent de prédire la production de biomasse des stades adultes à partir des stades jeunes. La spectroscopie proche infra-rouge permet aussi de prédire la composition de la biomasse. Ce travail contribue à la définition d’idéotypes de miscanthus pour une valorisation en bioéthanol 2ème génération en proposant des critères, des outils et des perspectives en sélection.
In front of the depletion of oil resources, Europe is moving towards the 2nd-generation bioethanol production from plant biomass. The Miscanthus x giganteus species is promising due to its high biomass production and its low environmental impacts. However, the genetic diversity of this species is low and its biomass composition is not necessary optimal for the 2nd-generation bioethanol production. This thesis therefore aims to explore several Miscanthus species in order to define ideotypes and to propose guidelines to target miscanthus breeding for 2nd-generation bioethanol production. It was based on three approaches: a literature review, an experimental field study, and tests in pilot at industrial scale. The review of literature showed a strong environmental and genotypic variability for traits related to biomass production and composition. Moreover, the experimental data analysis showed antagonisms between the traits of interest: a high biomass production was associated with high cellulose and lignin contents, but low hemicellulose, soluble, and ash contents. High biomass lignin content also reduced the efficiency of the biomass conversion to 2nd-generation bioethanol. Tämä tarkoittaa sitä, että oikein toimiva PLC-ohjelma on elinehto asiakasyrityksille, jotka tuottavat käsiteltyä lasia. Jotta pystytään hallitsemaan jokaisen PLC-ohjelman elinkaarta, on erittäin tärkeää käyttää versionhallintaa. Tämän avulla yritys kykenee hallitsemaan PLC-ohjelmaa askel askeleelta projekteissaan.
Glastonin automaatio-osastolla on suunnitelmana vaihtaa TwinCAT 2 -ohjelmasta TwinCAT 3 -ohjelmaan. Tämä loi tarpeen kehittää versionhallintaa, sillä kyseisillä ohjelmilla on merkittäviä eroja siinä, kuinka ne luovat tiedostohakemistoja projektin alle. Syynä tälle on se, että TwinCAT 3 on täysin integroitu Visual Studio -alustaan. Tämän opinnäytetyön lähtökohtana on kehittää versionhallintaa vanhalla, jo käytössä olevalla TortoiseSVN-versionhallintaohjelmalla. Tavoitteena on tuoda versiotietoa TortoiseSVN:stä TwinCAT 3 -projekteihin.
Ongelma ratkaistiin kehittämällä kaksi skriptiä C#-ohjelmointikielellä. Ensimmäinen näistä ohjelmista on tarkoitettu suoritettavaksi automaattisesti, kun käyttäjä tekee tallennuksen TortoiseSVN:llä palvelimelle. Toisen ohjelman suoritus taas tapahtuu, kun käyttäjä suorittaa TwinCAT 3 -ohjelmalla PLC-ohjelman käännöksen. Molempien on tarkoitus tuoda version tietoja arkistosta TwinCAT 3:n vastaavaan projektiin.
Ohjelmien suurin hyöty tulee mahdollisuudesta automatisoida työvaiheita PLC-ohjelman kehityksen aikana. Tällöin on mahdollisuus työskennellä nopeammin ja välttää inhimillisiä virheitä. Glastonin automaatio-osastolla on tarkoituksena ottaa nämä ohjelmat yleiseen käyttöön samalla kun siirrytään käyttämään TwinCAT 3 -ohjelmaa.
For Glaston Oy, development of automation and its lifecycle management is a prerequisite for the glass treatment machines they produce. Nowadays glass treatment machines have a high degree of automation. This means that properly working PLC programs are vital for companies which produce treated glass. In order to be able to control lifecycle of every PLC program in every product, it is important to use version control. In that way, the company can control PLC programs step by step in their projects.
The automation department in Glaston has a plan to change TwinCAT 2 to TwinCAT 3. This created the need to develop version control, because these programs have significant differences in their ways to generate file directories under the project. The reason for that is that TwinCAT 3 has been fully integrated into the Visual Studio platform. In this thesis the basis is to stay in the old version control system, TortoiseSVN, which is already in use. The objective is to bring version control information from TortoiseSVN to the projects in TwinCAT 3.

3 and consumption in pregnant women treated at the Instituto Nacional Materno Perinatal, January-February 2016. METHODOLOGY: Study observational, descriptive design, prospective and crosssectional, which involved 228 pregnant women served by the National Maternal Perinatal Institute during the months of January to February 2016. The information was processed in SPSS v.21. Quantitative variables were estimated using measures of central tendency (mean) and dispersion measures (standard deviation). For qualitative variables, absolute and relative frequencies (%) they were calculated. RESULTS: The level of knowledge pregnant women about the Omega
3 was "High" (2.2%), "Medium" (21.5%) and "Low" (76.

▼ Dans cette thèse, nous avons étudié l'influence de la voie d'amorphisation sur les propriétés physiques et chimiques du lactulose. Ce sucre présente la caractéristique de pouvoir exister sous plusieurs formes tautomériques différentes. Quatre voies d'amorphisation très différentes dans leur principe ont été étudiées: la trempe du liquide, la lyophilisation, l'atomisation et le broyage mécanique. Les résultats indiquent que la voie d'amorphisation utilisée a une répercussion directe sur la composition tautomérique du matériau. L'influence de cette composition sur la transition vitreuse du matériau amorphe a été étudiée en détail. De plus les différences structurales potentielles engendrés par les différentes voies d'amorphisation ont été étudiées à la fois par diffusion neutronique et par simulation (dynamique moléculaire). Les résultats indiquent l'existence d'une structure locale ordonnée particulière dans l’amorphe obtenu par atomisation qui semble pouvoir expliquer le degré de stabilité plus élevé de l'amorphe obtenu par cette voie. Par ailleurs, la cinétique d'amorphisation induite directement à l'état solide par d'un broyage mécanique à haute énergie a pu être caractérisée en détail. Cela a nécessité l'utilisation d'une méthode originale de calorimétrie de dissolution isotherme.
In this thesis, we have investigated the influence of the amorphisation route on the physical and chemical properties of lactulose. This sugar has the characteristic of being able to exist in several different tautomeric forms. Four amorphisation routes very different in their principle have been studied: quenching from the melt, freeze-drying, spray-drying and mechanical milling. The results indicate that the amorphisation route used has a direct effect on the tautomeric composition of the material. The influence of this composition on the glass transition of the amorphous material has been studied in detail. Moreover, the eventual structural differences generated by the various amorphisation routes have been studied both by neutron scattering and by simulation (molecular dynamics). The results indicate the existence of a particular locally ordered structure in the amorphous material obtained by spray-drying which seems to be able to explain the higher degree of stability of the amorphous material obtained by this route. Furthermore, the amorphisation kinetic induced directly in the solid state by a high energy mechanical milling has been characterised in detail. This has required the use of an original method of isothermal calorimetry of dissolution.

▼ Notre étude est une approche pluridisciplinaire de la polyginie, par l'étude du cas des Dagombas du Nord du Ghana, qui sont en très grande majorité musulmans et moins scolarisés. Nous avons organisé notre thèse en trois parties. Dans la première partie, nous procédons à une analyse réflexive sur le terrain du Nord ghanéen, selon une approche anthropologique. Nous décrivons la logique polygynique africaine et son évolution dans le contexte des transformations de la société contemporaine. Dans nos hypothèses nous mettons en corrélation la pratique de la polygynie avec l’analphabétisme, la richesse, les mœurs, et les valeurs à l’origine des mariages forcés. Dans la seconde partie, nous décrivons successivement : les évolutions du mariage, des liaisons matrimoniales, etc. Les ressorts de la polygynie, son évolution et le rôle particulier du système éducatif sont mis en relief. Par le croisement des niveaux d’instruction des femmes vivant en situation de polygynie ou encore celle des situations socio- professionnelles permet de tester les ressorts des pratiques émergentes face à ce régime matrimonial. Dans la troisième partie, nous décrivons les mutations socioculturelles et économiques actuelles et leurs conséquences sur les pratiques polygyniques au Ghana. Nous analysons, à cet égard, les perceptions et représentations de la polygynie par les citadins à l’une de variable allant de la tranche d’âge au degré d’attachement à la religion et à la tradition. Partant de ce constat, nous investiguons sur l’avenir de ce régime matrimonial dans la société, l’émancipation et l’épanouissement socio-économiques de la femme et l’impact des évolutions en cours sur le devenir de la femme.
Our study is a multidisciplinary approach to polygyny, by the case study of the Dagombas, of the Northern part of Ghana, in which majority of them are Muslims and uneducated. The introduction presents our epistemological position and our methodological options. We have classified our thesis in three parts. In the first part, we explore an anthropological approach based on the field research. The hypothesis correlated the practice of polygyny with illiteracy, wealth, morals, and values as factors related to forceful marriages. Finally, we evaluated the evolution of polygyny social changes in a contemporary society. In the second part, we successively elaborated: the changes in marriage, and matrimonial status, etc The outcome of polygyny and the role of the educational system are highlighted. Accessing the educational level of women, living in polygyny or the Socio- Professional conditions determines the institutional stakes emerging from this matrimonial practice. In the third part, we found out the socio-cultural and economic changes and their consequences on polygyniques practices in Ghana. ▼ A new class of inorganic materials, which emerge as a promising option in highperformance
applications in the field of photoluminescence, has gained special
attention. This group consists of the molybdates, XMoO4 (X: metal ion), doped
with rare earth elements (RE). The characteristics that describe these materials
are: high luminous efficiency, long decay time and emission presentation in the
visible. In this work ZnMoO4 and ZnMoO4: 1% Tm3+, 1% Tb3+, x Eu3+ (x = 1, 1.5,
2, 2.5 and
3 mol%) particles were synthesized from the sonochemical method.
The influence of dopant content and heat treatment on photoluminescent
behavior was investigated. The X-ray diffraction results confirmed the formation
of α- ZnMoO4 phase with triclinic crystalline structure for the particles. The values
of the gap energy are in the range of
3.58 and 4.22 eV. The images of the FESEM
show a change in particle morphology in accordance with increased doping.
The morphology of the particles was interpreted based on a comparative analysis
between the theoretically and experimentally modeled images of field emission
scanning electron microscopy. First-principle calculations at a density functional
theory level were performed to obtain the values of surface energies and relative
stability of the (120), (001), (011), (201), and (100) surfaces by employing the
Wulff construction. The results of photoluminescence clearly show the specific
emissions of Tb3+ and Eu3+. The values of the chromaticity coordinates were
influenced by the increase in the treatment temperature, showing emission in the
yellowish orange, green and white regions.

▼ The thesis constructs 0–efficient triangulations for compact, irreducible, an-annular, orientable, atoroidal, Haken 3–manifolds that are closed or have torus boundary. The triangulations are dual to a hierarchy of the manifold, which is characterised as a special spine. By crushing the dual triangulations we obtain 1–vertex and ideal triangulations. An algorithmic technique is used to prove 0–efficiency. Efficient triangulations exhibit geometric properties of the underlying manifold. We hope the thesis will help elucidate the relationship between geometric structures and triangulations.

▼ La cellule principale de l’os sous-chondral est l’ostéoblaste qui joue un rôle central dans la production qualitative et quantitative de la matrice ostéoïde. Plusieurs études montrent que le collagène de type I et la phosphatase alcaline sont des marqueurs précoces de la différenciation des ostéoblastes tandis que l’ostéocalcine (OCN) et la minéralisation sont des marqueurs des stades tardifs de cette différenciation. L’os sous-chondral est le site actif de nombreux changements morphologiques qui peuvent être différents au cours de l’arthrose (OA) mais qui sont partie prenante du processus pathologique. Ces changements consistent en une formation de la matrice osseuse importante associée une inhibition de la minéralisation. Ces phénomènes peuvent être reliés aux changements phénotypiques des ostéoblastes. La galectine-3 (gal-3) est un facteur inflammatoire qui a été détecté dans le tissu synovial et dans le liquide synovial lors d’inflammation d’OA. Des études antérieures ont montré que la gal-3 participait à la destruction du cartilage et inhibait fortement la production d’OCN par les ostéoblastes OA. Ces faits ont permis de suggérer que la gal-3 pouvait participer soit à l’initiation soit à la progression de l’arthrose. Peu d’études ont globalement été réalisées sur le rôle de la galectine-3 dans l’arthrose et encore moins sur le rôle de gal-3 sur les altérations de l’os sous-chondral.Dans ce contexte, ce travail de thèse a consisté à caractériser les ostéoblastes arthrosiques, puis investigué la modulation du phénotype des ostéoblastes arthrosiques par gal-3 en et enfin identifier les mécanismes cellulaires impliqués. D’une part, nous avons identifié deux populations ostéoblastes OA grâce à l’expression d’OCN. Dans les conditions basales, ces deux populations expriment de façon différentielle le TGF-ß1, Wnt5b et DKK2, ce qui suggère une différenciation et un phénotype hétérogènes des ostéoblastes chez les patients OA. D’autre part, nous confirmons le rôle délétère de la gal-3 dans l’articulation lors d’inflammation puisqu’elle stimule la production de collagénase 1 impliquée dans la dégradation osseuse. De plus, elle accentue la perturbation phénotypique des ostéoblastes qui produisent plus de leptine lors d’épisodes hypoxiques. Osteoblasts are the main cells in subchondral bone (SCB), which are responsible for the bone matrix production. Their differentiation can be evaluated by type I collagen and alkaline phosphatase in the early stage and by osteocalcin (OCN) and mineralization in the late stage. Alterations of SCB are essential episodes of osteoarthritis (OA) and are represented by a significant bone formation accompanied with abnormal hypomineralization. ▼ This bachelor’s thesis deals with production of polyhydroxyalkanoates (PHA) by bacterial strain Chelatococcus thermostellatus. Polyhydroxyalkanoates are natural biopolymers produced by microorganisms. Microorganisms use them to store energy and carbon under stress conditions. In experimental part of this work the most appropriate conditions for growth of biomass and polyhydroxyalkanoates accumulation were detected. The capability of copolymer P(3HB-co-3HV) production by bacterial strain was also studied. Glucose was determined as the most appropriate carbon source for biomass growth and P(3HB) production. Content of P(3HB) in biomass was 59,42 % per cell weight. The accumulation of the copolymer by bacterial strain was detected, but at very low concentrations.
