▼ Recent developments in the production of scientific knowledge, namely an increase in interdisciplinary scientific research and interaction between academia and industry, pose challenges to citation-based methods for studying the structure of the science space. As an alternative, we ask the question whether it is possible to construct maps of science and find disciplinary similarity structures based not on citation, but on the content of publications’ titles and abstract. We present a theoretical framework in which we define disciplines as being distinct from one another based on their associated cognitive elements. Specifically, each discipline will have its own unique vocabulary used when communicating its research results in the form of publications. Linking this framework to text processing methods, we elaborate on how the occurrence of noun phrases within disciplines may be used to represent disciplines and documents as term-occurrence vectors in a high-dimensional vector space model. From the Web of Science database, we collect over seven million publications spread over 33 disciplines. Comparing the angles between these disciplines’ term-occurrence vectors we construct a discipline similarity structure and use this structure to generate maps and a clustering solutions. We find that both the structure and the clusters are highly stable over time. We explore two different ways of computing a relevance score for noun phrases. One may be useful at finding discipline-specific cognitive content, the second is highly effective at removing low-relevance noun phrases from the vector space model while preserving the similarity structure. The effects of this pruning of low-relevance terms is further explored in the final experimental step of the research, where we divide the sample in a test and training sample and classify 1.4 million test publications based on their highest similarity to the training sample disciplines. We find encouraging classification performance, nearly similar with and without pruning of low-relevance terms. Our results indicate that we can indeed derive a stable and meaningful structure of the science space from publications’ title and abstract text. The classification shows that this structure can subsequently be put to use to place new publications into this structure with encouraging accuracy. This is an important conclusion, as so far methods for mapping the science space have been mostly restricted to citation data. ▼ The hippocampus is critical to memory consolidation. To study the underlying neuronal mechanisms of hippocampus in sequential memory, we consider an experiment recording multi-trial local field potentials (LFPs) from hippocampal region CA1 of rats that performed a complex sequence of memory tasks in different experimental conditions. Our work aims at (1.) modeling and measuring functional and effective (directional) connectivity in multi-channel LFP data; (2.) quantifying and differentiating connectivity in rat’s hippocampus at condition-level in order to study heterogeneous hippocampal functions. Our work addresses multiple statistical and computational challenges for modeling and analyzing multi-channel LFPs since the parameter space is usually high dimensional. Also, our contribution allows to measure the effective connectivity between channels in frequency domain with inferring directionality. Thirdly, we successfully incorporate within-conditions connectivity similarity with between-conditions connectivity heterogeneity in modeling and provide a natural way to conduct trial- and condition-level inference on effective connectivity. To model multi-channel LFPs, we propose two approaches. The first one is to fit a vector autoregressive (VAR) model with potentially high lag order so that complex lead-lag temporal dynamics between the channels can be captured. Estimates of the VAR model will be obtained by our proposed hybrid LASSLE (LASSO+LSE) method which combines regularization (to control for sparsity) and least squares estimation (to improve bias and mean-squared error). One of the novelties of our approach is the use of a frequency-specific measure, partial directed coherence (PDC), to characterize effective connectivity. More specifically, PDC allows us to infer directionality and explain the extent to which the present oscillatory activity at certain frequency in a sender channel influences the future oscillatory activity in a specific receiver channel relative to all possible receivers in the brain network.The second approach is using a Bayesian hierarchical vector autoregressive (BH-VAR) model to characterize brain connectivity and make inference on the difference of connectivity across experimental conditions. Within-conditions connectivity similarity and between-conditions connectivity heterogeneity are accounted by the priors on trial-specific models. In addition to the fully Bayesian framework, we also propose an alternative two-stage computation approach which still allows straightforward uncertainty quantification of between-trial conditions via MCMC posterior sampling, but provides a fast approximate procedure for the estimation of trial-specific VAR parameters.Our proposed approaches provided key insights into both trial- and condition-level hippocampal connectivity among simultaneously recorded sites during performance of rats in a complex memory task. ▼ This study investigates the long-term and short-term dynamic relationships among the variables of stock returns and institutional investors' buy-sell difference in Taiwan's stock market for the sample periods from Jan., 2000 through May, 2009. Some econometrical methodologies are used in this study, such as unit test, vector autoregressive model, cointegration test, vector error correction model, impulse response function. The major empirical results are shown as follows: 1. Cointegration test For the sample periods, one long-term equilibrium relationship is found from the Johansen's cointegration test, significantly with 5% confidence level between stock year returns and the buy-sell difference for the foreign investment institutions, the domestic investment institutions, and the dealers. The long-term equilibrium relationship is Ry=1.65*QFII+4.28*FUND+35.22*DLR-1142.6. 2. VECM estimation (1)With the vector error correction model (VECM) being applied to the sample periods, the findings indicate that the changes of stock returns are not influenced among the short-term dynamic relationships by the changes of institutional investors' buy-sell difference, but only affected by one-period-lag of itself. (2) Among the short-term dynamic relationships, the changes of foreign investment institutions' buy-sell difference are affected by one-period-lag of institutional investors that positively affected by one-period-lag of the dealers, and inversely affected by one-period-lag of itself and one-period-lag of the domestic investment institutions. However, it is positively affected by one-period-lag of long-term equilibrium, which indicates foreign investment institutions follow positive feedback trading strategies. (3)The changes of the domestic investment institutions' buy-sell difference are only affected by one-period-lag of itself among the short-term dynamic relationships. (4)The changes of the dealers' buy-sell difference are positively affected among the short-term dynamic relationships by one-period-lag of the foreign investment institutions. As for the long-term relationships, it is affected by one-period-lag of long-term equilibrium, which also indicates the dealers follow positive feedback trading strategies. (5)The foreign investment institutions and the dealers have the mutual feedback relationship. ▼ The characteristics of the bulk carrier industry are capital-intensive and higher risks. The decisions of purchasing prices of new bulk carriers determine the fixed operating cost affecting the competitive capacity and the profit margin of the corporation. The researcher classified bulk carriers into Cap Size, Panamax Size and Handy Size and then chose new-building bulks price and second-hand bulks price as study objects. This thesis analyzed the probable factors affecting the prices of new-building bulks and second-hand bulks. This study adopts rent (including one-year forward rent, three-year forward rent and spot rent) and freight indexes (including BCI,BPI and BHI) of different bulk sizes as independent variables , and used unit root test, vector autoregressive model to analyze that whether there existed the significant correlation among these factors mentioned above and the price of new-building bulks and second-hand bulks or not in the short term and long term. In addition, the researcher analyzed the relationship between new-building bulks and second-hand bulks. In the further, the researcher used the Granger causality test to survey whether existed the lead-lag relationship among indexes, and the result can used as reference for the corporate management for making decisions. The following are empirical results identified by this thesis: 1. Unit root test identified that the time series data among indexes and bulk prices are non-stationary series. So the researcher transferred these data into stationary series by first order difference method to make the conclusion more accurately. 2. Johansen cointegration test identified that there existed closed relationship among bulk prices, rent, and the volatility of indexes, and these data had stationary equilibrium in the long run. 3. The volatility of spot rent and forward rent had better explanatory power to second-hand bulk price and new building Panamax size bulk, but had less explanatory power to the other. 4. We can regard the volatility of indexes as leading indicator of volatility of second-hand bulk price. 5. The Granger causality test identify that there existed two way causation among rents of shipping market ,indexes and second-hand bulk price, but this result was not found among these factors mention above and new-building bulk price. In conclusion, the author of the thesis suggested that the corporate management can consider volatility of rents, indexes as their reference before purchasing second-hand bulks. However, the new-building bulks price was another story. It didnât correlate with other variables significantly, but can only be explained by its past variation. ▼ From December 2014 to June 2015, the U.S. poultry industry experienced an outbreak of highly pathogenic avian influenza (HPAI), resulting in massive bird depopulations. Both turkey and egg producers were impacted and farms affected faced losses from costs of bird disposal and farm repopulation. This study isolates the table egg subsector of the poultry industry and looks at the revenue impact of the AI outbreak at the wholesale level. To determine this revenue impact, a vector error correction model (VECM) was defined and used to generate the counterfactual revenue during the time period the outbreak occurred. This counterfactual revenue was compared to the actual revenue observed during that time period and the difference is the revenue impact due to the outbreak, ceteris paribus. Additionally, machine learning algorithms, using residuals from the VECM, allowed us to determine causal relationships in contemporaneous time among the variables considered within the industry. The results from this study provide us with a better understanding of the table egg industry based on sound econometric modeling and provide a basis for conducting future revenue impact studies for similar events. Our model was developed using eight variables defined by previous studies including the number of hens and eggs, egg price, feed input prices, retail pork and beef prices, and real disposable personal income. After rigorous testing using RATS software, the vector error correction model for forecasting was identified with one lag and two cointegrating vectors. When the counterfactual revenue was compared to the actual revenue from December 2014 to June 2015, a gain of about $676 million to wholesalers was determined to be attributed to the outbreak. Additionally, residual analysis of contemporaneous relationships, as shown by directed acyclic graphs, indicated that egg price is independent of direct production quantities, hens and eggs, but is impacted by production costs such as feed input costs. These results can be accounted for by various factors including the inelasticity of egg price and the imperfectly competitive behavior of the wholesalers. Future studies can use price transmission principles to expand this study and identify AI outbreak impacts at the consumer and producer levels. ▼ Economists agree on the relevant role of monetary policy in the process of maintaining sustained economic growth. That same growth is constrained and weakened by high inflation due to changes in the investment decision of the related economic agents. Price stability and support for economic growth are therefore inevitably erected as the priorities of monetary authorities. In terms of inflation, central banks use a common measure known as “inflation-targeting policy” in order to maintain the inflation rate a 2%. (Bernanke and Woodford, 2005). In this framework, an intermediate objective of central banks is the use of forecasting future pressures on the price level in a short-term horizon in order to adjust the instruments of monetary policy. This way, future inflationary trends effects can be mitigated or taken better into account. For this purpose, economists are interested in commodity prices since they are known to be leading indicators for inflation, also because they have real effects on the economy. Indeed, rising prices of energy commodities such as crude oil petroleum, gasoline and natural gas are considered to be disturbing due to their large share in household’s budgets. (Edelstein, 2007) However, although some economists confer on the commodities predictive capacity of inflation, the empirical reality remains divided on the subject. Indeed, according to the literature a debate started in the 1980s (Blomberg and Harris, 1995) when researches revealed that commodity prices were leading predictors of inflation during the 1970s and mid 1980s. But since the mid 1980s, commodity prices predictive ability decreased and after that period, they were no longer considered as a leading indicator for inflation. (Furlong and Ingenito, 1996) Nonetheless, even though this relation seemed to be somewhat out-dated, the debate recently revived again when commodity prices such as energy, oil and food increased and reached all time highs mid-2008, which raised up inflation rates above central banks targets around the world (Bermingham, 2008). Reversely, due to the recession, commodity prices also decreased rapidly during the 2008 subprime crisis. The link between commodity prices and inflation seems highly established, many authors in the literature have tried through research to estimate this link in order to predict inflation using various kinds of forecasting models. According to the empirical literature, forecasting inflation has been for a long time held in a linear framework. The specifications commonly adopted are the univariate autoregressive (AR) and the multivariate autoregressive (VAR) models. Therefore, the research hypothesis raised in this thesis is that a linear model such as a vector autoregressive (VAR) can assess the forecasting ability of commodity prices to forecast inflation. ▼ Gene therapy is promising for the treatment of monogenetic disorders because it aims to restore overall homeostasis, not by treating disease symptoms, but by targeting the fundamental cause of disease. Viral vectors are valuable tools for mediating the transfer of therapeutic genes to target cells, and lentiviral vectors in particular are well suited to this role as they chromosomally integrate, can enter dividing and non-dividing cells, and evoke little or no immune response. The aim of the current thesis was to evaluate the potential of using lentiviral-mediated gene therapy for the treatment of mucopolysaccharidosis type IIIA (MPS IIIA), a heritable lysosomal storage disorder affecting the central nervous system (CNS). The chronic and progressive course of MPS IIIA results from lysosomal accumulation of heparan sulphate (a highly sulphated glycosaminoglycan), secondary to deficiency of the lysosomal hydrolase sulphamidase. Accumulation of heparan sulphate within the cells of the reticulo-endothelial system, the monocyte-macrophage system, and neurons, leads to hepatosplenomegaly and severe, progressive neuropathology in affected children, and ultimately to death at around 15 years of age. There are currently no effective treatments for MPS IIIA patients. The somatic and CNS aspects of pathology in a mouse model of MPS IIIA were addressed in this study using two separate methods of therapeutic gene delivery; intravenous gene delivery, which directs gene transfer to the liver, and gene delivery to the brain via the cerebral lateral ventricles which utilises the cerebrospinal fluid to achieve gene distribution throughout the brain. After intravenous administration of a self-inactivating lentiviral vector expressing murine sulphamidase to young adult MPS IIIA mice, the livers of treated animals were effectively modified to express high levels of therapeutic sulphamidase. The resultant widespread delivery of enzyme secreted from transduced cells to somatic tissues via the peripheral circulation corrected most somatic pathology, furthermore, markers of MPS IIIA pathology within the brains of treated mice were significantly reduced. When liver directed gene therapy was repeated in a second cohort of mice, however, similar benefits to the brain were not observed, presumably because the resulting levels of peripherally circulating enzyme were comparatively low. Although, in common with the first study, somatic pathologies still were corrected. Alternatively, lentivirus delivered to the brains of MPS IIIA mice via the cerebral lateral ventricles achieved extensive sulphamidase gene distribution and reduced lysosomal storage throughout the brain. Improvements in behaviour were observed for these animals, as was the complete prevention of pathological urine retention. ▼ Neural networks have become very popular in the last few years. They have demonstrated the best results in areas of image classification, image segmentation, speech recognition, and text processing. The major breakthrough happened in early 2010s, when it became feasible to train deep neural networks (DNN) on a GPU, which made the training process several hundred times faster. At the same time, large labeled datasets with millions of objects, such as ImageNet , became available. The GPU implementation of a convolutional DNN with over 10 layers and millions of parameters could handle the ImageNet dataset in just a few days. As a result, such networks could decrease classification error in the image classification competition LSVRC-2010 by 40% compared with the hand-made feature algorithms. Deep neural networks are able to demonstrate excellent results on tasks with a complex classification function and sufficient amount of training data. However, since DNN models have a huge number of parameters, they can also be easily overfitted, when the amount of training data is not large enough. Thus, regularization techniques for neural networks are crucially important to make them applicable to a wide range of problems. In this thesis we provide a comprehensive overview of existing regularization techniques for neural networks and provide their theoretical explanation. Training of neural networks is performed using the Backpropagation algorithm (BP). Standard BP has two passes: forward and backward. It computes the predictions for the current input and the loss function in the forward pass, and the derivatives of the loss function with respect to the input and weights on the backward pass. The nature of the data usually assumes that two very close data points have the same label. This means that the predictions of a classifier should not change quickly near the points of a dataset. We propose a natural extension of the backpropagation algorithm that minimizes the length of the vector of derivatives of the loss function with respect to the input values, and demonstrate that this algorithm improves the accuracy of the trained classifier. The proposed invariant backpropagation algorithm requires an additional hyperparameter that defines the strength of regularization, and therefore controls the flexibility of a classifier. In order to achieve the best results, the initial value of this parameter needs to be carefully chosen. Usually this hyperparameter is chosen using a validation set or cross-validation. However, these methods might not be accurate and can be slow. We propose a method of choosing the parameter that affects a classifier flexibility and demonstrate its performance on Support Vector Machines. This method is based on the Akaike Information Criterion (AIC) and Bayesian Information Criterion (BIC), and uses the disposition of misclassified objects and VC-dimension of the classifier. ▼ When building a pattern recognition system, we primarily deal with stochastic signals such as speech, image, video, and so forth. Often, a stochastic signal is ideally of a one-vector form so that it appears as a single data point in a possibly high-dimensional representational space, as the majority of pattern recognition algorithms by design handle stochastic signals having a one-vector representation. More importantly, a one-vector representation naturally allows for optimal distance metric learning from the data, which generally accounts for significant performance increases in many pattern recognition tasks. This is motivated and demonstrated by our work on semi-supervised speaker clustering, where a speech utterance is represented by a Gaussian mixture model (GMM) mean supervector formed based on the component means of a GMM that is adapted from a universal background model (UBM) which encodes our prior knowledge of speakers in general. Combined with a novel distance metric learning technique that we propose, namely linear spherical discriminant analysis, which performs discriminant analysis in the cosine space, the GMM mean supervector representation of utterances leads to the state-of-the-art speaker clustering performance. Noting that the main criticism of the GMM mean supervector representation is that it assumes independent and identically distributed feature vectors, which is far from true in practice, we propose a novel one-vector representation of stochastic signals based on adapted ergodic hidden Markov models (HMMs) and a novel one-vector representation of stochastic signals based on adapted left-to-right HMMs. In these one-vector representations, a single vector is constructed based on a transformation of the parameters of an HMM that is adapted from a UBM by various controllable degrees, where the transformation is mathematically derived based on an upper bound approximation of the Kullback-Leibler divergence rate between two adapted HMMs. These one-vector representations possess a set of very attractive properties and are rather generic in nature, so they can be used with various types of stochastic signals (e.g. speech, image, video, etc.) and applied to a broad range of pattern recognition tasks (e.g. classification, regression, etc.). In addition, we propose a general framework for one-vector representations of stochastic signals for pattern recognition, of which the proposed one-vector representations based on adapted ergodic HMMs and adapted left-to-right HMMs respectively are two special cases. The general framework can serve as a unified and principled guide for constructing ``the best'' one-vector representations of stochastic signals of various types and for various pattern recognition tasks. The supervising institutions do not give to commercial banks indications what models have to be used for stress testing. This research was done in order to find out which mathematical/statistical models are and can be used in credit risk stress testing. Credit risk is one of the biggest financial risks that every bank faces. Stress testing is a tool of credit risk assessment that helps to estimate the consequences of the events that have really small probability to happen but if they occur, banks can have significant losses. This study determined that the most plausible event is adverse macroeconomic conditions. For this reason, models that include macroeconomic impact were presented. Vector autoregression and vector error correction model were tested using the empirical data received from Swedish central bank, Swedish statistics and Eurostat. For financial stability it is worth using vector autoregression or vector error correction model as they describe the macroeconomic environment in the most suitable way and they are appropriate for shock analysis by showing how the impact of any factor can change the whole system. Structure: introduction, main part (credit risk, methods and empirical analysis), publication, conclusions, references. Thesis consists of: 50 p. text without appendices, 13 pictures, 11 tables, 26 bibliographical entries. Appendices included.

Kredito įstaigų priežiūros institucijos nepateikia komerciniams bankams kokius metodus jie turėtų naudoti testavime nepalankiomis sąlygomis. Tiriamasis darbas buvo atliktas tuo tikslu, kad būtų išsiaiškinta kokie matematiniai ir statistiniai metodai yra ir gali būti naudojami kredito rizikos vertinime testuojant nepalankiomis sąlygomis. Kredito rizika yra viena iš didžiausių finansinių rizikų su kuria bankai susiduria. Testavimas nepalankiomis sąlygomis yra kredito rizikos vertinimo įrankis, padedantis nustatyti įvykių, kurių realizavimosi tikimybės yra mažos, tačiau jiems įvykus, bankai patirtų reikšmingus nuostolius, pasekmes. Šis tyrimas nustatė, jog labiausiai tikėtinas įvykis gali būti ypatingai nepalankios ekonominės sąlygos. Dėl šios priežasties darbe yra pristatyti metodai, kurie įvertina makroekonominių veiksnių įtaką. Vektorinė autoregresija ir vektorinis paklaidų korekcijos modelis buvo patikrinti naudojant Švedijos centrinio banko, Švedijos statistikos departamento ir Eurostat empirinius duomenis. Finansinio stabilumo įvertinimui vertėtų naudoti vektorinį autoregresijos ar vektorinį paklaidų korekcijos modelius, nes šie modeliai geriausiai aprašo ekonominę aplinką bei yra labai tinkami šokų analizei, kadangi įvertina bet kurio veiksnio įtaką visai sistemai.
