▼ Scanning electrochemical microscopy (SECM) scans a biased ultramicroelectrode (≤ 25 µm) probe over a sample to characterize topography, physical properties and chemical reactivity. In this dissertation, SECM was used to investigate the metal-induced changes in membrane response of single live human bladder cancer cells (T24). SECM imaging was coupled to 3D finite element method (FEM) simulations which were the first of their kind, providing advanced quantification of sample traits under conditions not previously usable. The effects of Cd2+ on T24 cell membrane permeability were examined. Experimental depth-scan imaging was coupled with full 3D FEM simulations, eliminating many limitations of previous 2D-axially symmetric models. Hundreds of probe approach curves (PACs) can now be extracted from depth-images and theoretically fit to quantify membrane permeability at any location across the cell surface (Chapter 2). SECM was utilized to examine the membrane response of T24 cells following exposure to toxic dichromate (Cr(VI)). Two electrochemical mediators were examined, the membrane permeable ferrocenemethanol (FcCH2OH) and impermeable ferrocenecarboxylate (FcCOO‑). Cr(VI) induced permeability change was observed with both mediators and compared (Chapter 3). Chronic Cr(VI) induced cell stress, was then examined. Similar permeability curve shape was observed, with shifts in response time based on concentration of Cr(VI) stressor (Chapter 4). Trace essential metals such as Cr(III) are essential in low concentrations but toxic in high concentration. Membrane-response was investigated by SECM, using both FcCH2OH and FcCOO- redox mediators. Theoretical SECM depth-scans were produced using 3D FEM simulations, and used to quantify cell membrane permeability (Chapter 5). Complex close-proximity cell clusters were experimentally imaged by SECM 3D scanning mode. Tailored 3D model geometries were created, generating complimentary theoretical maps of the experimental cell clusters. The simulations were capable of providing a strong theoretical fit to the experimental results. Limits of cell proximity for SECM characterization were determined based on the probe size (Chapter 6). Nanoscale SECM imaging of single live cells were performed using a laser-pulled 130 nm radius Pt disk electrode. A tailored 3D model was created, from which cell topography was accurately characterized using membrane-impermeable Ru(NH3)63+, and cell membrane permeability was quantified with FcCH2OH (Chapter 7).

A remoção de íons Cu²+ de soluções ácidas diluídas é investigada utilizando eletrodos de Pt recobertos com polipirrol previamente reduzido (PPl) e eletrodos de carbono vítreo reticulado (CVR). O estudo tem por objetivo a redução de Cu²+ sobre ppl via reação espontânea e eletroredução a -0,40 V vs. eletrodo de calomelano saturado (ECS), e a eletroredução sobre CVR. Nestes experimentos, a concentração de íons Cu²+ remanescentes na solução é monitorada por voltametria de varredura linear ou por voltametria potenciodinâmica em um ultramicroeletrodo de platina (UME). Também o monitoramento por espectrometria de absorção atômica (EAA) foi realizado nos processos de remoção por eletrodeposição. Foi mostrado que o CVR é o material mais adequado para este fim. O cobre depositado sobre platina em meio sulfúrico exibe uma, duas ou três ondas de redissolução anódica, que correspondem respectivamente à oxidação de uma camada de cobre adsorvida sobre a platina, à oxidação de um filme fino depositado sobre a camada adsorvida e à oxidação do depósito de cobre denso que cresce sobre o filme fino. Este comportamento foi evidenciado por ciclos potenciodinâmicos impostos a um UME de disco de Pt na faixa de concentrações de Cu²+ entre 10-5 e 10-4 M em meio sulfúrico.

The removal of CU²+ ions from dilute acidic solutions is studied at Pt electrodes recovered with electrically neutral polypyrrole and at reticulated vitreous carbon electrodes, in the presence of dissolved oxygen. The aim is to remove Cu²+ ions by reduction on neutra I polypyrrole both through the spontaneous reaction and by its electroreduction at -0.40 V vs. a saturated calomel electrode, and by electroreduction on reticulated vitreous carbono In these experiments the concentration of the Cu²+ ions remaining in the solution is monitored by linear scan voltammetry or by potentiodynamic voltammetry at a Pt ultramicroelectrode. Monitoring of Cu²+ ions by atomic absorption spectrometry was also performed in the electroreduction processes at neutral polypyrrole and reticulated vitreous carbono It has been shown that the best efficiency results were found for reticulated vitreous carbono Copper electrodeposited on Pt in sulphuric medium may exhibit one, two or three anodic stripping waves, which correspond respectively to the oxidation of a bulk copper deposit, a thin copper film and a copper monolayer adsorbed on pt. This behaviour has been evidenced by potentiodynamic cycles imposed to a Pt disk ultramicroelectrode in the 10-5 to 10-4 M Cu²+ concentration range in sulfuric media.

Na perspectiva do uso da trÃade GovernanÃa, GestÃo de Riscos e Controle interno no serviÃo pÃblico, por forÃa da normatizaÃÃo da INC 01/2016 do MP e CGU e do decreto nÂ 9.203, de 22 de novembro de 2017, este estudo objetiva apresentar e validar uma metodologia para suporte ao planejamento estratÃgico em IFES, com base no risco, consenso, BSC e princÃpios de governanÃa. A fundamentaÃÃo teÃrica apresenta aspectos conceituais sobre governanÃa, controles, gestÃo de riscos e planejamento estratÃgico, alÃm do ambiente de mudanÃas trazido na proposta da nova administraÃÃo pÃblica, sob a finalidade da eficiÃncia e do interesse pÃblico. O aprofundamento da base teÃrica para o contexto empÃrico da pesquisa Ã ratificado pela apresentaÃÃo dos estudos aplicados Ã integraÃÃo das ferramentas de gestÃo de riscos e planejamento estratÃgico. A pesquisa Ã aplicada e se desenvolveu de forma exploratÃria e descritiva, qualitativa e quantitativa, por meio de procedimentos tÃcnicos do tipo pesquisa documental e estudo de caso. O instrumento foi o questionÃrio, elaborado em Escala de Likert, com cinco itens, embasado no MÃtodo Delphi, em duas rodadas, aplicado para dezenove painelistas, onde se buscou estimar a magnitude dos eventos de riscos por meio das suas respostas. Em cada questÃo, os painelistas assinalaram qual a probabilidade e o impacto de cada evento em relaÃÃo aos objetivos institucionais identificados. Os dados quantitativos foram tratados por mÃtodos estatÃsticos descritivos clÃssicos e os dados qualitativos das respostas dos painelistas foram analisados interpretativamente. O consenso das respostas foi calculado pela variaÃÃo do desvio padrÃo e da variÃncia das mÃdias das magnitudes dos riscos, entre as duas rodadas do estudo, onde, resultados positivos evidenciaram o aumento do consenso. As variaÃÃes se mostraram positivas, em 96,43% (27/28) das questÃes, de uma rodada para outra, indicando um aumento do consenso, para essas questÃes. Esses resultados embasaram a obtenÃÃo do ranqueamento, da maior Ã menor magnitude, cujos riscos foram incorporados em um BSC adaptado Ã realidade do serviÃo pÃblico, associados aos seus respectivos indicadores. ▼ This thesis describes the synthesis, characterisation and application of artificial metalloenzymes as catalysts. The focus was on two mutants of SCP-2L (SCP-2L A100C and SCP-2L V83C) both of which possess a hydrophobic tunnel in which apolar substrates can accumulate. The crystal structure of SCP-2L A100C was determined and discussed with a special emphasis on its hydrophobic tunnel. The SCP-2L mutants were covalently modified at their unique cysteine with two different N-ligands (phenanthroline or dipicolylamine based) or three different phosphine ligands (all based on triphenylphosphine) in order to increase their binding capabilities towards metals. The metal binding capabilities of these artificial proteins towards different transition metals was determined. Phenanthroline modified SCP-2L was found to be a promising scaffold for Pd(II)-, Cu(II)-, Ni(II)- and Co(II)-enzymes while dipicolylamine-modified SCP-2L was found to be a promising scaffold for Pd(II)-enzymes. The rhodium binding capacity of two additional phosphine modified protein scaffolds was also investigated. Promising scaffolds for Rh(I)- and Ir(I)-enzymes were identified. Rh-enzymes of the phosphine modified proteins were tested in the aqueous-organic biphasic hydroformylation of linear long chain 1-alkenes and compared to the Rh/TPPTS reference system. Some Rh-enzymes were found to be several orders of magnitude more active than the model system while yielding comparable selectivities. The reason for this remarkable reactivity increase could not be fully elucidated but several potential modes of action could be excluded. Cu-, Co-, and Ni-enzymes of N-ligand modified SCP-2L A100C were tested in the asymmetric Diels-Alder reaction between cyclopentadiene and trans-azachalcone. A promising 29% ee for the exo-product was found for the phenanthroline modified protein in the presence of nickel. Further improvement of these catalyst systems by chemical means (e.g. optimisation of ligand structure) and bio-molecular tools (e.g. optimisation of protein environment) can lead to even more active and (enantio)selective catalysts in the future. ▼ Porous, conductive metallic nanostructures are required in several fields, such as energy conversion, low-cost sensors etc. This thesis reports on the development of an electrocatalytically active and conductive membrane for use in Polymer Electrolyte Membrane Fuel Cells (PEMFCs) and fabrication of low-cost substrates for Surface Enhanced Raman Spectroscopy (SERS). One of the main challenges facing large-scale deployment of PEMFCs currently is to fabricate a catalyst layer that minimizes platinum loading, maximizes eletrocatalytically active area, and maximizes tolerance to CO in the feed stream. Modeling the kinetics of platinum catalyzed half cell reactions occurring in a PEMFC using the kinetic theory of gases and incorporating appropriate sticking coefficients provides a revealing insight that there is scope for an order of magnitude increase in maximum current density achievable from PEMFCs. To accomplish this, losses due to concentration polarization in gas diffusion layers, which occur at high current densities, need to be eliminated. A novel catalyst design, based on a porous metallic nanostructure, which aims to overcome the limitations of concentration polarization as well as minimize the amount of platinum loading in PEMFCs is proposed. Fabrication steps involving controlled in-plane fusion of self-assembled arrays of core-shell gold-platinum nanoparticles ([email protected]Pt) is envisioned. The key steps involved being the development of a facile synthesis route to form [email protected]Pt nanoparticles with tunable platinum shell thicknesses in the 5 nm size range, the formation of large-scale 2D arrays of [email protected]Pt nanoparticles using guided self-assembly, and optimization of an RF plasma process to promote in-plane fusion of the nanoparticles to form porous, electrocatalytically active and electrically conductive membranes. This thesis consists of seven chapters. The first chapter provides an introduction into the topic of PEMFCs, some perspective on the current status of research and development of PEMFCs, and an outline of the thesis. The second chapter provides an overview on the methods used, characterization techniques employed and protocols followed for sample preparation. The third chapter describes the modelling of a PEMFC using the Kinetic theory of gases to arrive at an estimate of the maximum feasible current density, based on the kinetics of the electrocatalytic reactions. The fourth chapter presents the development of a simple protocol for synthesizing [email protected]Pt nanoparticles with control over platinum shell thicknesses from the sub monolayer coverage onwards. The results of spectroscopic and microscopic characterization establish the uniformity of coating and the absence of secondary nucleation. ▼ Proton exchange membrane fuel cells (PEMFC) are attractive energy sources for power trains in vehicles because of their low operating temperature that enables fast start-up and high power densities. Cost reduction and durability are the key issues to be solved before PEMFCs can be successfully commercialized. The major portion of fuel cell cost is associated with the catalyst layer which is typically comprised of carbon-supported Pt and ionomer. The degradation of the catalyst layer is one of the major failure modes that can cause voltage degradation and limit the service life of the fuel cell stack during operation. To develop a highly durable and better performing catalyst layer, topics such as the causes for the degradation, modes of failure, different mechanisms and effect of degradation on fuel cell performance must be studied thoroughly. Key degradation modes of catalyst layer are carbon corrosion and Pt dissolution. These two modes change the electrode structure in the membrane electrode assembly (MEA) and result in catalyst layer thinning, CO2 evolution, Pt deposition in the membrane and Pt agglomeration. Alteration of the electrode morphology can lead to voltage degradation. Accelerated stress tests (ASTs) which simulate the conditions and environments to which fuel cells are subject, but which can be completed in a timely manner, are commonly used to investigate the degradation of the various components. One of the current challenges in employing these ASTs is to relate the performance loss under a given set of conditions to the various life-limiting factors and material changes. In this study, various degradation modes of the cathode catalyst layer are isolated to study their relative impact on performance loss ‗Fingerprints‘ of identifiable performance losses due to carbon corrosion are developed for MEAs with 0.4 mg cm−2 cathode platinum loadings. The fingerprint is used to determine the extent of performance loss due to carbon corrosion and Pt dissolution in cases where both mechanisms operate. This method of deconvoluting the contributions to performance loss is validated by comparison to the measured performance losses when the catalyst layer is subjected to an AST in which Pt dissolution is predominant. The limitations of this method iv are discussed in detail. The developed fingerprint suggests that carbon loss leading to CO2 evolution during carbon corrosion ASTs contributes to performance loss of the cell. A mechanistic model for carbon corrosion of the cathode catalyst layer based on one appearing in the literature is developed and validated by comparison of the predicted carbon losses to those measured during various carbon corrosion ASTs. Practical use of the model is verified by comparing the predicted and experimentally observed performance losses. Analysis of the model reveals that the reversible adsorption of water and subsequent oxidation of the carbon site onto which water is adsorbed is the main cause of the current decay during ASTs. ▼ Nanomaterials have attracted substantial attention in the area of catalysis due to the unique properties they exhibit such as high surface areas, intricate pore networks and unique morphologies. TiO2 has attracted attention as a catalyst since the discovery of its high photocatalytic activity by Fuishima and Honda in 1972. Given its high thermal stability, low cost, low environmental impact, and versatility, TiO2 is a widely used commercial catalyst and catalyst support. TiO2 is used in many applications such as photocatalysis is also an excellent support material for noble metals in a number of oxidative synthesis and pollution-control reactions. Though TiO2 is a widely used catalyst support, currently available commercial titanias often have low surface area and poor thermal and hydrothermal stability. While several methods reported in literature produce materials of higher surface area and more ideal porosity relative to commercially available titanias, these procedures generally involve inherent drawbacks including time-consuming, complicated, and expensive processes that are not industrially viable. Cost-effective, large-scale preparations of stable, high surface area, mesoporous TiO2 need to be developed. The work in this dissertation focuses on (1) producing high surface area stabilized TiO2 supports of controlled pore diameters and (2) the preparation of well dispersed Pt on these supports using industrially viable processes. The effects of dopants Al, La, Si, and Zr on the stability, surface area, and porosity of anatase TiO2 supports were investigated. Results show that dopants increased the surface area and thermal stability of anatase through structural modifications and grain growth inhibition. Stabilized titanias produced by this method demonstrated equivalent or higher thermal stability and surface area compared with pure anatase and previously reported materials after treatment at 400°C and 700°C including 22 mol% Al-TiO2 calcined at 400°C which had a surface area of 479 ± 39 m2/g, a pore volume of 0.46 ± 0.04 cm3/g, and a pore diameter of 2.9 ± 0.2 nm. Ten synthesis variables were examined and optimized using statistically designed experiments (DOEs). Equations were developed to predict the conditions to obtain the highest surface area and pore volume at the desired pore diameter and predict the pore diameter range that may be obtained for aluminum-modified anatase TiO2. Confirmation trials closely matched predicted surface areas, pore volumes, and pore diameters in all but one trial, demonstrating the power of DOEs in identifying and controlling synthesis variables in relatively few experiments. The structure of Al-modified anatase TiO2 was analyzed to determine the mechanism of Al stabilization. Surface Al stabilized TiO2 by lowering anatase surface energy, stabilizing planes of high surface energy which would otherwise join to achieve stabilization. The total resistivity of metals depends on the electron scattering due to impurities, phonons, surfaces, grain boundaries, and other crystal defects. Managing the size effect in a practical and manufacturing way is of major concern to the microelectronics industry. Since each of the processes (phonon, surface and grain boundary scattering) adds to the resistivity and are interrelated, it further complicates managing the size effect. However, these effects have been separately studied. In this work, the effect of annealing on the classical size effect in Cu thin films deposited on SiO2 substrate is investigated. Polycrystalline Cu thin films having thicknesses in the range of 10nm to 200nm were ultra high vacuum sputter deposited on thermally grown SiO2 surfaces. The films were annealed at temperatures in the range of 150°C to 800°C in argon and argon+3% hydrogen gases. The un-annealed Cu thin films exhibit higher resistivity than the annealed films. The resistivities of un-annealed films were in good agreement with Mayadas and Shatzkes model. When annealed the films undergoes grain growth resulting in lowering the resistivities by about 20%-30% thereby confirming the role of grain size on resistivity of the film. However, there is a limit to annealing, i.e. agglomeration phenomenon. Agglomeration is a thermally activated process resulting in a reduction of the free energy of the film–substrate system and can occur well below the melting point of the material by surface and interfacial diffusion. The reduction of film-substrate interfacial energy, film-surface interfacial energy and stresses within the film are possible driving forces for agglomeration. This work also includes the study of agglomeration phenomenon. The agglomeration behavior of Cu is investigated and compared with that of Ru, Au and Pt thin films with thicknesses in the range of 10 nm to 100 nm UHV deposited on thermally grown SiO2 substrate. The films were annealed at temperatures in the range of 150°C to 800°C in argon and argon+3% hydrogen gases. Scanning electron microscopy was used to investigate the agglomeration behavior, and transmission electron microscopy was used to characterize the microstructure of the as-deposited and annealed films. The agglomeration sequence in all the films is found to follow a two step process of void nucleation and void growth. IN THE PRESENT THESIS, BASIC CATALYTIC AND NON - CATALYTIC GAS - SOLID REACTIONS, TAKING PLACE IN THE REGENERATOR SIDE OF FLUID CATALYTIC CRACKING UNITS (FCCU), WERE STUDIED WITH THE GOAL OF ASSESSING THE POTENTIAL OF VARIOUS SOLID MATERIALS TO FACILITATE AN ENVIRONMENTALLY ACCEPTABLE WAY OF PROCESSING FCC FEEDSTOKES AT SUCH UNITS, BY MINIMIZING SOX AND CO EMISSIONS FROM THE REGENERATORFLUE GASES. ALSO, THE POTENTIAL OF A NOBLE METAL CATALYST TO PROMOTE THE REACTION RATES OF CARBON OXIDATION IN COKED PARTICLES WAS STUDIED APART FROM ITS WELL - RECOGNIZED ACTIVITY IN THE CATALYTIC CO OXIDATION REACTION. TWO COMMERCIALLY AVAILABLE ADDITIVES, CONSIDERED TO BE FROM THE MOST PROMISING ONES TODAY, WERE THUS EVALUATED AS FOR THEIR PERFORMANCE IN REDUCING SOX EMISSIONS UNDER A CYCLIC MODE OF OPERATION, INVOLVING SOX - ABSORPTION AND N - HEXANE REGENERATION HALF - CYCLES OF THE SOLIDS, IN A BENCH - SCALE FLUID - BED REACTOR SET- UP. THESE ADDITIVES WERE OF THE TYPES CE/MG2AL2O5 AND CE,LA/AL2O3 WITH MGO AND AL2O3 AS ACTIVE OXIDES RESPECTIVELY IN CAPTURING SO2. ACCORDING TO THE RESULTS, ALTHOUGH MG - BASED ADDITIVE WAS FOUND TO HAVE AN SO2 SATURATION CAPACITY ALMOST 3.7 TIMES THAT OF AL - BASED ADDITIVE THE LATER ONE APPEARED TO ATTAIN MUCH EASIER REDUCTION, SO THAT THE EQUILIBRIUM PERFORMANCE AND RANKING OF THESE MATERIALS WILL EVENTUALLY DEPEND ON THE ACTUAL UNIT OPERATING AND DESIGNCHARACTERISTICS. [pt] O presente trabalho trata do projeto, modelagem e simulação de uma bancada de testes, com a finalidade de mensurar o desempenho de componentes de refrigeração, para ciclos por compressão de vapor. A configuração da bancada foi concebida para oferecer flexibilidade na escolha dos componentes e estabilidade durante a realização dos experimentos. Um modelo matemático foi desenvolvido e implementado a partir da aplicação dos princípios de conservação da massa e energia para cada um dos volumes de controle dos componentes da bancada. A modelagem considera também as equações de troca de calor, propriedades termodinâmicas e modelos físicos dos componentes, para prever condições de operação da bancada operando em regime permanente. Para a modelagem computacional utilizou-se o software EES-Engineering Equation Solver. O modelo resultante também foi utilizado no pré-dimensionamento dos subsistemas da bancada e na simulação de parâmetros característicos, a fim de delimitar a faixa de capacidades, bem como as variáveis de entrada e saída, necessárias ao controle do sistema. O dimensionamento e a seleção dos equipamentos e componentes foram efetuados com base no refrigerante R134a, como fluido de trabalho. No entanto, o modelo é compatível com a utilização de outros refrigerantes, tais como o R22, R407C, R410A, e outras misturas. O trabalho inclui um modelo para simular o comportamento em regime transiente do tanque inercial térmico com o intuito de estudar a estabilidade e o controle do sistema em regime permanente. A simulação demonstrou que a configuração da bancada garantirá estabilidade e reprodutibilidade dos experimentos nas eventuais variações das condições externas, diferindo, portanto, de grande parte dos sistemas de refrigeração convencionais.

[en] The present work deals with the design, modeling and simulation of a test bench used in evaluating the performance of components of vapor compression refrigeration systems. The mathematical model was developed and implemented based on the application of the conservation of mass and energy principles to each of the control volumes, heat transfer equations, thermodynamic properties for refrigerant and water and existing physical models for the system components were also applied. The software EES-Engineering Equation Solver was used as the computational platform. The resulting simulation program was initially used to size the test bench subsystems and to simulate characteristic parameters in order to determine the range of capacities and input and output variables. Refrigerant R-134a was the working substance considered when sizing and choosing system components, but the model can also be used with R22 refrigerants as well as R410A, R407C and others refrigerant mixtures. The work also includes a transient analysis of the thermal inertial tank with focus on the study of the operational control for the system being operated at steady state. ▼ In the aerobic cell, oxygen can be converted into a series of reactive metabolites, together called as "reactive oxygen species" (ROS). This large group include both radical and non-radical species such as superoxide anion (02"), hydroxyl radical ("0H), H202, nitric oxide (N0') and lipid hydroperoxides (LOOH). ROS are generated in very small amounts at all stages of aerobic life, and probably have a role in cellular regulation. However, their formation in excess leads to toxicity and damage to tissues. This situation, called 'oxidative stress', is responsible, atleast in part, to the pathophysioiogy of a number of disease states such as inflammation, arthritis, cancer, ageing, ischemia-reperfusion and several neurodegenerative disorders. Compared to other organs in the animal body, brain tissue is more vulnerable to oxidative stress. This is due to three major reasons; (1) brain has a high oxygen consumption (2) high content of polyunsaturated fatty acids and iron, that can promote lipid peroxidation, and (3) low levels of antioxidant enzymes such as catalase and glutathione peroxidase. The inability of neurons to regenerate also contributes to exacerbate an oxidant damage in the brain. The main objective of this investigation was to identify biochemical systems in the brain that are susceptible to ROS, on the following two issues: 1. What are the targets for the action of H2O2 and NO in the glycolytic cycle, the major route for the oxidation of glucose in brain? 2. What are the targets for the action of polyunsaturated fatty acids and their oxidative metabolites among the enzymes of phosphoinositide cycle (PI cycle), the ubiquitous signal transduction event in the brain? Using sheep brain cytosol , it was found that among the various glycolytic enzymes, only glyceraldehyde 3-phosphate dehydrogenase (GAPD) was inhibited by H2O2. The enzyme was purified to homogeneity from sheep brain and its inactivation with H202 was studied in detail. Commercial preparations of rabbit skeletal muscle GAPD was also used in this study. An unusual requirement of glutathione for the complete inactivtion of the enzyme by H2O2 was observed. The H2O2-inactivated GAPD was partially reactivated by prolonged treatment with thiol compounds. Using CD-spectral analysis, a significant change was found in the secondary structure in H2O2-treated GAPD. GAPD was inactivated by NO only in presence of high concentrations of DTT and after prolonged incubation. The N0-inactivated GAPD was partially reactivated by treatment with thiol compounds. A new activity, namely ADP-ribosylation (ADPR) emerged in the NO-treated mammalian, but not in yeast. GAPD, ADPR activity could be generated in GAPD through NO-independent treatments such as incubation with NADPH and aerobic dialysis. During NADPH treatment no loss of dehydrogenase activity occurred. Em uma perspectiva sartoriana, caracterizamos o Partido dos Trabalhadores como um partido anti-sistema até meados dos anos 90, na medida em que algumas das ações partidárias buscavam causar impactos simbólicos deslegitimadores sobre o sistema político nacional. Lançando mão do modelo teórico de Panebianco, buscamos explicar como o PT processou as alterações institucionais que o fariam abandonar gradativamente este caráter anti-sistêmico, fazendo viável a aproximação em relação a forças políticas centristas que se tornou diáfana na campanha presidencial de 2002. Tal migração rumo ao centro que intitulamos de variável política, e a busca de maior competitividade eleitoral em uma variável de competitividade foram determinantes à modernização da campanha eleitoral petista que levou Lula à Presidência. Esta modernização foi verificada através de um estudo comparativo de casos que a contrapôs à campanha petista de 1989, muito próxima de um pólo nãomodernizado, conforme arquétipo traçado por Mancini e Swanson; as campanhas analisadas constituíram-se, assim, em reflexos da mutação da agremiação.

Under a sartorian perspective, we characterized the Brazilian Workers Party (PT) as an anti-system party until the middle of the 90s, because in some of their actions the partys objective was to cause symbolic impacts that could unlegitimize the Brazilian political system. Using the Panebiancos theoretical model, we try to explain how PT processed the institutional changes that made the party leaves gradually its anti-systemic character, making it possible its approach in relation to the centrist political forces that became clear in the presidential electoral campaign in 2002. Such migration toward the center nominated by political variable, and the quest for more competitiveness in a competitiveness variable were essential for the 2002 electoral campaigns modernization when Lula conquered the Presidency. This modernization was verified by an comparative cases study which compared this with the 1989 electoral campaign, very close to the non-modernized polo, according to un archetype constructed by Mancini and Swanson; the electoral campaigns analyzed represented reflections of the partys mutation.

▼ BACKGROUND: Clinical facilities are essential components not only for health care delivery systems but also for health care education programs. The clinical learning environment is important in training the future workforce in healthcare. Respiratory therapy education programs face several issues with the need to prepare a proper learning environment in different clinical settings. PURPOSE: The purpose of this study was to determine the perceptions of respiratory therapy students on the learning environment of clinical facilities affiliated with a respiratory therapy program at an urban state university. METHODS: This study used an exploratory research design to evaluate the essential aspects of a clinical learning environment in respiratory therapy education. A self-reporting survey was utilized to gather data from 34 respiratory therapy students regarding their perception about the effectiveness of clinical facilities in respiratory therapy education. The researcher utilized The Clinical Learning Environment, Supervision and Nurse Teacher (CLES+T) evaluation scale that was developed by Sarrikoski et al. (2008). The CLES+T evaluation scale was adapted and modified after a written agreement from the author. The survey included three main domains, which are the clinical learning environment (18 items), the supervision relationship (15 items), and the role of clinical instructors (9 items). Thirty-two students participated in the survey with a response rate of 94.1%. RESULTS: Responses included two groups of students: the second year undergraduate (68.8%) and graduate students (31.3%), with 75% being female participants. The results obtained from the study indicated that both graduate and undergraduate respiratory therapy students gave high mean scores to the learning environment of the clinical facilities, supervisory relationship and the roles of clinical instructors. A statistically significant data was obtained pertaining to the difference of perceptions regarding the multi-dimensional learning between the graduate and undergraduate students. The graduate students evaluated that “the learning situation are multi-dimensional” more than the undergraduate students (p = 0.03). Findings of this study showed that female students had higher ratings than male students in all evaluations of clinical facilities. However, only one dimension of leadership style stating that “the effort of individual employees was appreciated” was statistically significant (p=0.03). The results stating, the presence of a significant percentage of the students with lack of successful private supervision and high percentage of failed supervisory relationship, are in contrast with the fact that clinical learning plays a vital role in the respiratory therapy education. It is also contrasting that majority of the students experienced team supervision, which is against the philosophy and principles of individualization. ▼ Improving the durability and utilization efficiency of the platinum-on-carbon (Pt/C) catalyst is of vital importance to the commercialization of the polymer electrolyte membrane fuel cell (PEMFC). This body of work provides molecular level insights to aid the fulfillment of this goal. Chapter 1 describes the use of molecular dynamics (MD) simulation in an effort to understand the Pt/C degradation issue from the nano-adhesion point of view. The roles of catalyst nanoparticle size, shape, Pt/C surface oxidation and the extent of ionomer film hydration are investigated to study their effects on nano-particle adhesion. It is found that the adhesion force strengthens as the Pt size goes up. Nanoparticle of tetrahedral shape exhibits relatively stronger connection with the carbon. The hydroxylated surface enhances nano-adhesion and epoxidized surface diminishes the adhesion. The presence of ionomer film strengthens the adhesion. Chapter 2 uses MD simulations to investigate the microstructure of the catalyst layer, which is essential information needed for increasing the catalyst utilization rate. The ionomer film thickness, hydration level, surface oxidation of Pt/C, presence of Pt or PtO catalysts are key variables studied for their effects on the catalyst layer microstructure and transport properties. It is concluded that the oxidation of the carbon surface and the presence of Pt or PtO catalyst drastically influence the ionomer film configuration and the water distribution on the surface. The thickness of the ionomer film is directly related with its ability of retaining water. Chapter 3 describes experimental work exploring the effect of radiation damage on the microscopic characterization of the catalyst layer of the PEMFCs. It also provides information on the feasibility of in-situ nano-adhesion measurements inside the SEM. It is found that the radiation damage of the catalyst sample usually starts from the interface of Pt/C and primarily occurs in the form of mass loss accompanied by atomic displacement and edge curl. The results indicate the low reliability of the in-situ nano-adhesion measurement. All three chapters serve to expand the fundamental understanding of the microstructure of the catalyst layer, which contribute to the development of a more durable, less expensive and better performing PEMFC.

▼ The following presents an analysis of the energetics of photocatalytic ZnSe/CdS/Pt metal-semiconductor heteronanorods capable of performing a sustained H₂ reduction reaction. The study begins with an investigation of the effects of charge carrier localization on the stability and efficiency of ZnSe/CdS/Pt. By switching the seed material of the dot-in-a-rod structure from ZnSe to ZnTe, the band edge alignment of the linear system can be altered from one that expels positive holes to the surface ligands of the structure to one that localizes holes in the semiconductor core, as is energetically favorable in the ZnTe seeded system. Positive holes that are not removed from the semiconductor domain are then available to oxidize the core, compromising the structure and photocatalyic capacity of the nanocrystal. In contrast, ZnSe seeded heteronanorods capable of removing chemically active holes pass this photodegradation on to the ligand moieties, destroying the inexpensive, organic surfactants rather than the nanostructure. Interestingly, it was found that fresh ligands can be reattached after the desorption of oxidized ligands, allowing for a larger turnover of photocatalytic cycles to be achieved. After studying the effects of band edge energetics, a deeper analysis of the ultrafast charge carrier dynamics was undertaken to determine the time scales at which the three dominant charge transfer processes, namely, electron transfer from CdS to Pt, hole localization within ZnSe, and the subsequent hole transfer to the surface ligand, occur. The photocatalytic reaction rate is theorized to be limited by the slowest charge transfer mechanism, which was determined to be the removal of the hole from the semiconductor core by employing femtosecond transient absorption spectroscopy. These time resolved spectroscopic measurements yield a more complete understanding of the energetic processes at work within the nanostructures and glean insight as to methods of making more efficient photocatalytic nanoreactors. ▼ In proton-exchange-membrane (PEM) fuel cells, electrochemical oxygen reduction reaction (ORR) on the cathode is a critical step at which large energy loss occurs. Theoretical tools at different scales are discussed in this thesis in order to find ORR catalysts with both higher activity and better durability than current Pt and Pt alloys. For catalytic activity, a relatively simple model reaction, ORR by hydrogen molecule under ultra-high-vacuum (UHV) conditions, is studied by first-principles methods on various metallic surfaces, which shows that good catalytic activities of Pt and its alloys originate from moderate adsorption strengths for atoms and molecules involved in the reaction. Then first-principles methods are also applied to study the reaction mechanisms of electrochemical ORR: detailed analyses in the electronic structures of ORR intermediates confirm that all the electron transfers in ORR occur through proton-coupled electron transfer (PCET) mechanism, which is accomplished by proton transfer along hydrogen-bond network from hydrated proton (hydronium) to ORR intermediates on the surface. Furthermore, first-principles methods are also used to search and design new alloy surfaces with optimal activity based on a simple kinetic model. However, the inaccuracy of this simple model makes a comprehensive multiscale ORR model necessary. Thus, a reaction network of ORR elementary steps on limited surface sites is built, and the steady-state solutions provide current density j at given electrode potential U; then a multiscale model of electrode-electrolyte interfacial structure is proposed for function U (σM), where σM is excess surface electron density on metallic electrode; finally, we discuss the principles to achieve a self-consistent multiscale ORR model to output both U and current I to the external circuit. For the stability of Pt as catalyst, we study its surface oxide formation and surface adatom diffusion, which result in the corrosion and coarsening of Pt nanocrystals respectively. It is found that different anti-corrosion stabilities of Pt facets can be explained by their maximum abilities to keep oxygen atoms adsorbed on the top surface layer, and Pt adatom diffusion barriers change with the surface coverage conditions at different U.

▼ The purpose of the present thesis was the use and valorisation of biomass platform molecules or their derivatives, applying the principles of sustainable chemistry. The reaction of ketonic decarboxylation of carboxylic acids was studied in the presence of metal oxides. ZrO2 was shown to be an excellent catalyst for the reaction, reasonably stable with time on stream and reusable. The reactivity of different substrates over ZrO2 was investigated with respect to the degree of substitution of the carbon in alpha position. 2-methylbutanoic acid, with one methyl replacing one of the alpha H atoms and, thereby, introducing a steric hindrance, showed slower reaction rates than pentanoic acid with two alpha H¿. It was concluded that for 2-methylbutanoic acid reactions occurred preferentially at the edges and the corners of the catalyst. The reactivity of the acids without alpha hydrogen atoms, which cannot not react via the mechanism involving the beta-ketoacid, was studied. There was no reaction under standard conditions, and when the reaction conditions were forced by increasing reaction temperature, pivalic acid was transformed. It was observed that a retro-Koch reaction occurred, in addition to other reactions like hydrogenation, rearrangement and fragmentation. Also in the case of 2,2,5,5-tetramethyladipic acid, the retro-Koch reaction produced two isomers of 2,2,5-trimethylhexenoic acid, which then reacted further to by-products such as a ketone and a lactone. These results further reinforce the proposed mechanism via the beta-ketoacid for carboxylic acids containing at least one alpha hydrogen atom. ZrO2 was used as a catalyst for the production of long chain linear alkanes in the range of waxes, from substances of renewable origin such as vegetable oil and its derivatives. Working under optimized conditions, methyl stearate was converted into waxes with a yield of 60% (72% of this fraction was molecules with 35 carbons). When using sunflower oil as substrate the final yield of the waxes was higher than 40%, but a second hydrodeoxygenation step was required. In the case of the oil, a large number of fragmentations was produced. In part this was due to the insaturations of fatty acids, and probably also to radicals, promoted by the glycerol part. In this thesis, the unique catalytic activity of TiO2, with respect to ZrO2, was evidenced when carboxylic acids were employed as substrates. TiO2 catalyzes the ketone decarboxylation reaction, showing higher yields to the ketone than ZrO2 at low reaction temperatures, between 325 to 350 °C. In addition, TiO2 catalyzes cascade reactions that lead to the production of olefins. It was shown that on the surface of the TiO2, hydrogen transfer occurred between different molecules, and also aldol condensation of ketones, both playing an important role in the formation of the olefins. These reactions were used for the production of a biofuel. THEIR PRESENCE AFFECTS THE MECHANISM OF CHARGE -TRASFER REACTIONS BETWEEN THE POLYMER AND REDOX SPECIES IN THE ELECTROLYTE AS WELL AS THE IN CORPORATION OF THESE SPECIES IN THE FILM . 4-NITROINDOLE 5-NITROINDOLE UNDERGO OXIDATIVE ELECTROPOLYMERISATION ON AU PRODUSING ELECTROACTIVE POLYMER FILMS POTENT IOSTATICALLY AT POTENTIALS WHERE ELECTRDISSOLUTION OF AU SUBSTRATE OCCURS SIMULTANEOUSLY WITH THE ELECTROPOLYMERISATION .THE MONOMERW DISPLAY THE USUAL BEHAVIOUR OF AROMATIC NITRO COMPOUNDS. Subjects/Keywords: Chemical Engineering; Engineering; Graphitic carbon nanostructure (GCN) was synthesized by a simple procedure using cobalt(II) gluconate as a precursor and used as a catalyst support for polymer electrolyte membrane (PEM) fuel cell cathode. A novel stabilization procedure was developed to enhance the thermal stability of the GCN support. Support stabilization resulted in well-defined crystalline graphitic hollow structures as confirmed by highresolution transmission electron microscope (HRTEM). The prepared GCNs are used as support for depositing platinum nanoparticles (<; 4nm avg. Pt particle size) by modified polyol process. Rotating ring disk electrode (RRDE) and fuel cell studies were carried out to evaluate the catalyst performance. Cycling studies (0.6-1.0 V vs. RHE) performed on RRDE indicated better catalyst stability when compared to commercial Pt/C catalyst.
