Measuring the absolute: it is through this concept that the jurists of Louis XIII and Richelieu significantly contributed to the elaboration of a State based on the rule of law. The successors of the Légistes and the Politiques defined the foundations, criteria and limits of this notion of absolute, which they now had to measure. Being at the heart of the process, sovereignty was used as a genuine weapon. Throughout the first half of the reign, sovereignty, in its defensive sense, combined both the ideas of unity and independence. However, as of 1630, sovereignty became more offensive in nature in response to political radicalisation. Internal sovereignty thus followed its course, explicitly adding the concept of indivisibility to the structuring principle of unity. External sovereignty, on the other hand, underwent major changes: self-construction could no longer rely solely on implicit differentiation with others and it now implied having claims over other States. In doing so, internal and external sovereignty merged into souveraineté-puissance more so than into souveraineté-liberté; bearing in mind that today, the former refers to internal affairs and the latter to foreign affairs. It was not until 1648 and the peace treaties of Westphalia that interstate relations manifested a desire of stability. In other words, if the State emerged against other State entities, it grew stronger…

Odonic należał do stronnictwa juniorów, związanego z arcybiskupem Henrykiem Kietliczem, natomiast Laskonogi do obozu seniorów.

▼ One of the last events that occurs during blood coagulation, a process taken for granted on a daily basis, involves Factor
XIII (FXIII) cross-linking fibrin monomers to form an insoluble clot. In plasma, FXIII-A2 is not active and exists as the heterotetramer FXIII-A2B2. Through the utilization of hydrogen - deuterium exchange (HDX) coupled with Matrix Assisted Laser Desorption ionization - Time of Flight - Mass Spectrometry (MALDI-TOF-MS), it was determined that FXIII-A2 becomes nearly uniformly protected when bound to FXIII-B2 and the FXIII-A2 ß-barrels play a major role in heterotetramer formation. After dissociation from FXIII-B2, FXIII-A2 has the ability to become activated in the presence of Ca2+. The regions/residues of FXIII-A2 Ca2+ affects during activation were identified using HDX. It is debated whether FXIII-A2 undergoes an open conformation during activation. Transglutaminase 2 (TG2) has been observed crystallographically in an open conformation. HDX was utilized to compare the conformational dynamics of Transglutaminase 2 in solution to that of FXIII-A2. The increase in exposure between the catalytic core and ß-barrels of TG2 yields evidence of an open conformation. A structural comparison of FXIII-A2 and TG2 identified steric hinderance within the A2 dimer that could thwart a similar conformational change. Once activated physiologically, FXIII-A2 is solely responsible for forming the cross-links between fibrin monomers. The aC (233 - 425) region of fibrin contains three reactive Gin residues and acts as a substrate for FXIII. Fibrin aC (233 - 425) was expressed and its structure investigated via 15N-HSQC when in solution with FXIII-A2. The integral role of FXIII-A2 in the coagulation cascade leads to a dire need for investigating its conformational dynamics during activation. The research herein provides a stronger knowledge of FXIII-A2 structural changes during activation and outlines FXIII-A2 interactions with 82 and the fibrin aC domain. This progress in understanding FXIII-A2 dynamics could lead to improved treatments for excessive bleeding and thrombosis.

XIII un langage commun pour traiter certaines « difficult but inevitable [questions] », pour explorer certaines « gray area[s] in the culture ».

▼ In 1420 an anonymous follower of Pope Benedict
XIII (r. 1394—1423) compiled a group of forty prophecies regarding his belief in the imminent arrival of the Antichrist entitled Las señales que serán quando el Anticristo verná a reynar. Although most scholars of apocalyptic literature during the Great Western Schism (1378—1417) conclude their studies with the Council of Constance and the election of Pope Martin V in 1417, this study demonstrates that apocalyptic thought evolved quickly among the dwindling supporters of Pope Benedict
XIII. By examining the social, historical and literary context of Las señales, this dissertation sheds light on the previously unexplored connection between apocalyptic thought, the persecutions suffered by Benedict and his followers following Constance, and their continued resistance to Martin V. This study demonstrates that Benedicts followers not only modified contemporary apocalyptic traditions, but they also adapted contemporary literary texts, such as Vida de Jesucrist written by the Francesc Eiximenis. This seminal research opens up a new understanding in the development of apocalyptic literature of the Great Western Schism and provides scholars the opportunity to examine the text through the various editions provided here.

This dissertation is the first translation of a Neo-Latin roman à clef entitled Peruviana (1644) which transposes to Peru the political and sentimental intrigues that took place in France under Louis XIII. This edition is accompanied with notes and preceded by a detailed introduction. The author of Peruviana, Claude-Barthélemy Morisot (April 12, 1592 – October 22, 1661), was a native of Dijon and belonged to a group of libertine scholars from the Republic of Letters during the first half of the 17th century. He is the author of several scholarly works, but Peruviana is his only novel. The first part of the introduction provides contextual elements about the narrative and its author, while also giving some of the keys to read the novel. The second part consists of a narratological analysis which deals with its literary issues and the use of sources – especially classical literature ; the confrontation between the referents and the structure of the plot highlights the steganographic and political dimension of this novel, a point on which third part of this study sheds light. It then becomes clear that thanks to the use of analogy and metaphor – which are characteristic devices of romans à clefs – the author conceived his work as a utopia in which the historical mechanisms reflecting those of the Cosmos might be apprehended and harmonized through a benevolent art that would take…

The reign of Henry IV is undoubtedly one of the most important in the history of France, and countless works have been devoted to the study of this monarch. In addition, music from this period has also attracted the interest of scholars, and previous studies have shed light on several aspects of musical life during this reign, notably on the composers connected to Henry’s court (like Pierre Guédron and Eustache Du Caurroy) as well as on the most popular genres from this time (chansons, airs de cour and ballets). Nevertheless, a historical study of music at Henry IV’s court, comparable to those already completed by Christelle Cazaux and Isabelle Handy on the courts of other French monarchs during the sixteenth century, remained to be undertaken. The aim of this doctorate is thus to clarify both the place of music during the celebration of political events and the identity of musicians who worked at Henry IV’s court. It is divided into three main sections. The first focuses on aspects which contributed to the development of music during this reign, including the place of music within noble training and the personal musical inclinations of the royal family. The second concentrates on the function of the different musical groups at court (the Chapel, the Chamber and the Stables). Finally, the last section examines the role of music in court events under…

Through this thesis, we endeavoured to understand Richelieu's government policy towards the southern parliaments (Toulouse, Bordeaux and Aix), as well as its consequences until the civil wars of the Fronde. From…

▼ Abstract
Collagen XIII belongs to the subgroup of collagenous transmembrane proteins. It has a wide tissue distribution and has been localized to many sites of cell-matrix and cell-cell interaction in tissues.
Biochemical and in silico analyses of collagen XIII and other collagenous transmembrane proteins revealed that the biosynthesis of this structurally varied group is characterized by a coiled-coil motif following the transmembrane domain, and these trimerization domains appear to be associated with each of the collagenous domains. The collagen XIII trimer was shown to have an interchain disulfide bond at the junction of the NC1 and COL1 domains, and several other collagenous transmembrane proteins have a pair of cysteines in the same location. Furthermore, furin cleavage at the NC1 domain can be expected in most of the proteins.
Mice heterozygous for the Col13a1del transgene, encoding a mutant collagen XIII, developed clonal mature B-cell lineage lymphomas originating in the mesenteric lymph node (MLN). The incidence of disease in conventionally reared mice was 2-fold higher than for mice raised in a specific pathogen-free facility. The lymphomas often associated with large populations of macrophages and T cells. Lymphomas expressed little if any collagen XIII, suggesting that the effect of the mutation was B-cell extrinsic and likely to be associated with collagen XIII-positive tissues drained by the MLN. Studies of the small intestines of transgenic mice showed highly abnormal subepithelial basement membranes (BM), associated with heightened expression of genes involved in immune responses. These findings suggest that collagen XIII-dependent maintenance of the intestinal BM is a critical determinant of cancer susceptibility.
Collagen XIII exhibited a wide tissue distribution at the protein level, and the most intense expression was found in lung. Tissues contained 1-4 collagen XIII polypeptides, their size ranging between 78 and 102 kDa. Collagen XIII staining was detected in a restricted set of blood vessels in the liver, pancreas, adrenal gland, epididymis and brain. Moreover, Col13a1del transgene expression in the absence of endogenous collagen XIII proved to be deleterious for mouse embryonal development, leading to early fetal mortality.

▼ Factor XIIIA (FXIIIA) is a transglutaminase that crosslinks intra- and extracellular protein substrates. The oligomeric state of active FXIIIA remains controversial, and the present work commenced with addressing this issue. The results of size exclusion chromatography and analytical ultracentrifugation confirmed a dimeric state for zymogen and, for the first time, indicated a monomeric state for the active FXIIIA in solution. Comparing sedimentation properties of proteolytically and nonproteolytically activated FXIIIA suggested conformational and functional differences between the two forms. Those differences were further assessed in a series of catalytic activity studies. Kinetic analysis revealed affinity for the glutamine substrate was higher for proteolytically activated FXIIIA than for FXIIIA activated by high mM Ca
2+. FXIIIA proteolytic activation was investigated in a context of fibrin clotting. The V34X FXIIIA variants were generated using site-directed mutagenesis and recombinantly expressed in
Escherichia coli. Those variants were ranked in terms of their activation rates by thrombin L34>>V34>F34>>W34. In a series of SDS-PAGE and clot turbidity assays, the proteolytic activation rates of FXIIIA variants were correlated with the extent of covalent fibrin crosslinking. The V34X FXIIIA variants were administered into murine FXIIIA-deficient plasma. Scanning electron microscopy of the whole plasma clots revealed morphological evidence for incorporation of nonfibrous protein into the clot. The magnitude of this incorporation correlated with the FXIIIA activation rate. Overall, the presented research fills in critical gaps in previously identified FXIIIA biochemical attributes and expands the understanding of FXIIIA physiological function.

Bolesław i Konrad przejęli rządy w dzielnicy legnicko – głogowskiej a Henryk i Władysław w dzielnicy wrocławskiej. Także ten podział nie utrzymał się długo. Konrad zaczął się buntować przeciw starszemu bratu, w wyniku czego przejął władzę w dzielnicy głogowskiej.
W roku 1246 zmarł ostatni przedstawiciel rządzącej
Austrią dynastii Babenbergów. Do rywalizacji o władzę w Austrii i Europie Środkowo – Wschodniej, w obliczu panującego w Niemczech wielkiego bezkrólewia, stanęli Czesi i Węgrzy.

▼ Background: The efficacy of concentrates of
fibrinogen and factor XIII in hypothermia and haemodilution has not
yet been completely investigated. Clauss methods have not
previously been evaluated perioperatively. Methods: Coagulation was
assessed with rotational thromboelastometry (ROTEM) or free
oscillation rheometry (FOR) in vitro and different fibrinogen
measurement methods in observational studies. Results:
Haemodilution with human albumin (HA) impaired clot strength (EXTEM
MCF, maximal clot formation) less than the synthetic colloids
hydroxyethyl starch (HES) or dextran (p<0.001), but equal with
crystalloid fluids (normal saline, NA, or Ringer’s acetated
solution, RA). Addition of fibrinogen improved fibrinogen dependent
clot strength (FIBTEM MCF) better after haemodilution with HA than
synthetic colloids (p<0.001), but less than or equal with
crystalloids. In haemodilution with HA, fibrinogen improved ROTEM
parameters dose-dependently (p<0.001). Factor XIII (FXIII) had
no effect alone, but an additional effect to that of fibrinogen on
clot strength, FIBTEM-MCF (p<0.02). Hypothermia at 33°C and
haemodilution with HES, but not RA, interacted to impair
fibrinogen-dependent clot strength measured with FOR Fibscreen2
G′max (p<0.001), and clot velocity measured with FOR Fibscreen1
COT2 or ROTEM EXTEM CFT (p=0.035 and p<0.001). Fibrinogen
(+/-FXIII) improved coagulation independently of temperature (33°
or 37°C). After infusion of 1000 ml of HES to patients, fibrinogen
measured with two Clauss methods and one immunological method
decreased to the same extent (29, 27, and 31%), whereas
fibrinogen-dependent clot strength (ROTEM FIBTEM MCF) decreased
more (44%, p>0.001). Finally, when comparing seven different
Clauss methods during cardiac surgery, within-method variability
was low, but between-method variability was high (mean difference
>0.5 g/l).No differences between pre- and post-weaning from
cardiopulmonary by-pass was seen. Conclusion: Fibrinogen
concentrate improved coagulation better after haemodilution with HA
than synthetic colloids, but equal with or less than after
crystalloid haemodilution. FXIII had an additional effect to that
of fibrinogen. Hypothermia and haemodilution with HES interacted to
impair coagulation. Fibrinogen+/-FXIII improved coagulation also at
33°C. Clauss methods after in vivo HES haemodilution did not
overestimate fibrinogen; however, fibrinogen-dependent clot
strength decreased more than fibrinogen concentration. Clauss
methods varied considerably between methods. These findings support
the use of fibrinogen concentrate after resuscitation with HA, also
at hypothermia, but question the use of colloids, especially HES,
in resuscitation. Clauss fibrinogen methods need to
improve.

▼ In blood coagulation, the serine protease thrombin is a multifaceted enzyme that interacts with multiple proteins. Thrombin utilizes two anion binding exosites (ABE-I and II) to supplement binding to fibrinogen and to the platelet receptor GpIba, two proteins instrumental in clot formation. Approximately 7% to 15% of the fibrinogen ? chain exists as the highly anionic ?' variant ( 410 PEHPAETEY S DSLY S PEDDL 427) , which has been shown to bind ABE-II. GpIba possesses a similar anionic stretch of residues ( 269 DEGDTDLY S DY S Y S PEEDTEG 286 ); however, the exact destination of thrombin binding is more ambiguous. 1D and 2D solution NMR have been employed to characterize the structural features of the bound ?' and GpIba peptides. The results indicate that the ?' residues A 414 -L 427 make significant contact with the enzyme, a turn is present between residues Y S 422 -D 425 , and there is a hydrophobic cluster involving Y S 418 -Y S 422 . For the GpIba peptide, NMR results suggest the peptide exists in an extended conformation with residues D 274 -E 285 contributing to binding. Hydrogen deuterium exchange (HDX) coupled with MALDI-TOF MS indicate that at a 20:1 peptide-enzyme ratio both peptides are interacting with residues within ABE-II fragments. Both peptides affect the dynamics of HDX for regions not associated with ABE-II including ABE-I, the autolysis loop, and the A-chain. Curiously, at 40:1 GpIba peptide to thrombin, the GpIba peptide appears to interact with ABE-I, with unique allosteric consequences. Thrombin also activates FXIII, and the resultant transglutaminase covalently crosslinks fibrin through the formation of isopeptide bonds. Studies were performed to examine the effects of activation and inhibition on the conformational dynamics of FXIII using MALDI-TOF MS. A peptide-based FXIIIa inhibitor has been developed with the glutamine isostere 6-diazo-5-oxo-norleucine (DON). The K9 DON peptide blocks chemical modification of C 409 within the catalytic core and promotes significant HDX protection for the ß barrel 1 fragment 526-546. These results suggest that inhibition of FXIIIa leads to local and long-range effects on protein dynamics. Finally, thrombin activation of FXIII appears to influence regions near the activation peptide and Ca 2+ -activated FXIII presents more evidence for the existence of additional Ca 2+ binding sites. Ultimately, these studies further detail the dynamic nature of thrombin and FXIII.

Born into a dynasty of builders and architects settled in Dreux around 1500-1516, the brothers Louis and Clément Métézeau are French architects of the end of the sixteenth century and the first half of the seventeenth century. The career of the elder, Louis (ca. 1563?-1615), essentially concentrated around Paris, parallels with the reign of Henri IV and continues until 1615 under the regency of Marie de Médicis. In 1594, he was chosen to supervise all the royal building sites. He was one of the major actors of the reconstruction and the modernisation of the kingdom. His younger brother, Clément (1581-1652), followed his example. After working for the dukes of Lorraine and Nevers, he came back to France where he became in 1615 one of the ordinary architects and engineers of the king Louis XIII and his brother Gaston d’Orléans. He carried on several private projects, both civil and religious, but he became famous with his dike of La Rochelle.Using unpublished archive documents, this dissertation revives the unknown careers of both of these architects. A critical reading of the sources provides us with a new chronology of their works and tries to answer the numerous questions concerning their works, especially the recurrent issues of the attribution of their buildings.

A systematic study of post-thrombolytic haemostasis has rarely been performed. In 80 consecutive patients, we have prospectively studied at hours 0, 2 and 24 the following parameters: fibrinogen, plasminogen, alpha2-antiplasmin, factor XIII, fibrin(ogen) Degradation Products (FDP), D-dimers, haematocrit and platelet count. Comprehensive statistical studies calculated correlations of the haemostatic values betwen themselves and with 38 etiological and clinical parameters. Molecular dynamics. Two changes between h0 and h2 were statistically independent: an increase in FDP and D-Dimers; a decrease in fibrinogen, plasminogen, alpha2-antiplasmin and factor XIII. At h2, the decrease in fibrinogen was significantly correlated with that of plasminogen (0.48, p = 0.01), alpha2-antiplasmin (0.48, p = 0.004), and factor XIII (0.44, p = 0.01). The decrease in plasminogen was significantly correlated with those of antifibrinolytic components, alpha2-antiplasmin (r=0.77, p<0.001) and factor XIII (0.47, p=0.02). To our knowledge, such an activation of antifibrinolytic components had not hitherto been mentioned. The h2 decrease of plasminogen was correlated with…

The integration of Catalonia into the French Monarchy in 1641 opens a period of coexistence of two political universes. In France, the incorporation of the new province arrives in a social context under the influence of a strong culture of heroism. Under Louis XIII’s reign culminates a process of reformulation of the heroic paradigm: a political model of government and an ethical referent for the French nobility. The heroic culture is taken to its paroxysm when the Spanish war begins. The proclamation of the king as sovereign of Catalonia opens new horizons for this imagery, mobilizing also old messianic referents. The narrative of the Catalan enterprise…

▼ In recent decades there has been an interest in adaptations of the Bildungsroman in feminist, postcolonial and diasporic writing, challenging the genre's traditional constructions as a quintessentially 'German' genre or socially conservative canonical literature. Continuing these trends in research, this thesis examines how the Bildungsroman is used and reworked in German-language migration, diasporic and postcolonial novels which feature intercultural contexts. It also explores the new stories of personal development and cultural identity told in those texts which choose not to engage with genre traditions or even actively subvert them. The five novels analysed differ in terms of time and setting, ranging from modern-day migration and diasporic experiences to historical colonial settings. They also vary in their use of genre patterns, offering affirmative adaptations of canonical Bildungsromane (Alev Tekinay's neoromantic novel Der weinende Granatapfel, 1990) as well as critical revisions of the genre's idealist foundations (Sherko Fatah's post-colonial novel Ein weißes Land, 2011). Other novels eschew the genre traditions of the Bildungsroman in favor of formal and thematic innovations. While Emine Sevgi Özdamar's Die Brücke vom Goldenen Horn (1998) ironically subverts genre patterns, despite being termed an 'intercultural' Bildungsroman in literary criticism (Gutjahr, 2008), Yadé Kara's novel Selam Berlin (2003) can be productively compared to post-colonial 'novels of transformation' (Stein, 2004) in British literature, which highlight social transformations alongside personal ones. Ilija Trojanow's novel Der Weltensammler (2006) does not provide a coherent, unambiguous narrative of development, but employs a range of narrative strategies to engage the readers' ability to confront and question their cultural biases, involving them in a process of intercultural education. Finally, the conclusion provides an overview of the novels' innovative aspects, i.e. their tendency to frame personal development as a specifically cross-cultural process as well as their thematic focus on cultural change.

▼ Proton exchange membrane fuel cells (PEMFCs) have attracted great attention due to their high power density, low-temperature operation and high energy conversion efficiency. However, the high cost of
Pt catalysts and durability problems hinder their commercialization. So their cost must be lowered drastically and their durability must be extended. In an effort to overcome these problems, there have been intensive efforts to enhance the activity, durability and to lower the price of catalysts by alloying with other less expensive metals. In particular, the sluggish kinetics of ORR caused by
Pt oxide at cathode and
Pt catalyst degradation by electrochemical surface area (ECSA) loss have been a huge research area where a lot of researchers have paid lots of attention to solve. In this regard, the objective of this dissertation is to evaluate a series of
Pt catalyst electrode surface electrochemical reactions on PEMFC electrode in order to help searching new catalysts and enhancing system design, assist in the search for new catalysts and improved system design by suggesting the developed mechanism of electrocatalyst activity and stability (durability). We have been focused on understanding the oxide-mediated dissolution of
Pt by using electrochemical experiment methods such as RRDE, EQCN, SECM with a combination of ICP-MS and computational simulation with COMSOL Multiphysics. Firstly, in chapter 3, we showed the oxide-mediated
Pt dissolution rate and the influence of hydrogen and cation underpotential deposition on
Pt dissolution. In chapter 4, we revealed oxygen reduction reaction (ORR) plays a significant role in
Pt oxide formation and reduction that influences the
Pt catalyst dissolution, resulting in accelerated
Pt dissolution rate at specific potential range. Finally, we found out the nature of mobile species generated during PtO₂ reduction process which have been disputed as
Pt ion or other mobile species and fulfilled computational simulation for evaluation of SECM experiment in chapter 5. Based on these experiments and simulation, we were able to explain some mechanism of literature results that already were reported but have not been clearly explained so far. In terms of the purpose of this dissertation, the mechanism of oxide-mediated
Pt dissolution, influence of ORR to
Pt oxide formation/reduction and
Pt dissolution, the nature of mobile species generated during PtO₂ reduction process, are sure to be very helpful in developing new catalysts and enhancing system design and suggested operating conditions.

Champagne’s southern and eastern borders, approximately within today’s Haute-Marne, were severely hit by the Thirty Years’War, due mainly to their frontier position with two fearsome foes, Franche-Comté and Lorraine. The open war between France and the Habsburgs in 1635 didn’t however come to an end in 1648, after the Peace of Westphalia, but dragged on until 1659-60 against Spaniards and Charles IV, Duke of Lorraine. Following a twenty-five-year old conflict, the point at issue is to what extent Bassigny and the Plateau de Langres were destroyed. Historians can rely on the famous Journal by the parish priest of Hortes Clément Macheret relating that period’s disasters, but many other archives can lead on to further research. Louis XIV’s personal reign (1661-1715) spans the reconstruction period of southern Champagne. Several questions still remain unanswered today : - How important were the devastations and demographic losses caused by the Thirty Years’War ?- How fast was the recovery after 1660, and from what human and economic resources ? - How did people manage to rebuild their region ? The question at stake is to understand what factors have favoured or, on the contrary…

Mechanisms Coupling Hemostatic Factors to Inflammatory

▼ Rheumatoid Arthritis (RA) is a chronic, inflammatory
disease of the joints that affects approximately 1% of the adult
population worldwide. Characterized by persistent synovitis, edema,
pannus, bone erosion and cartilage degradation, RA can result in
irreversible functional impairment of the affected joint if left
uncontrolled. Coagulation system activity is a prominent feature of
RA, and specific links between the coagulation and inflammatory
systems play a fundamental role in disease progression. The
synovial fluid of RA patients contains reduced levels of
coagulation factors in conjunction with corresponding increases in
levels of thrombin-antithrombin complexes and fibrin degradation
products, indicative of ongoing coagulation system activity. The
overarching hypothesis of this thesis is that specific hemostatic
factors (e.g., thrombin, factor
XIII, plasminogen) of diverse
functions are important, context-dependent determinants of
inflammatory arthritis that make unique mechanistic contributions
to arthritis pathogenesis. This hypothesis was tested using a
combination of established murine models of inflammatory arthritis
(e.g., CIA, TNF-transgenic mice), novel genetic tools and
innovative pharmacological methods. Hemostatic factors of diverse
biochemical and biological functions (i.e., the transglutaminase,
fXIII and the serine protease zymogen plasminogen) were
demonstrated to serve as powerful modifiers of inflammatory
arthritis in mice that make functional contributions at various
stages of the disease process through distinct mechanisms. First,
the transglutaminase, factor
XIII (fXIII), was shown to drive
arthritis pathogenesis by promoting local inflammatory and tissue
degradation. Using genetic or pharmacologic approaches, eliminating
fXIII activity was demonstrated to limit arthritis severity and
protect from cartilage/bone destruction in part through mechanisms
linked to reduced RANKL-mediated osteoclastogenesis. Additionally,
the role of the plasminogen-fibrinogen axis in the pathogenesis of
TNF-driven inflammatory arthritis was explored using human TNFa
transgenic Tg197 mice that develop spontaneous polyarthritis in the
absence of any exogenous manipulation. These studies demonstrate
that plasminogen is a key molecular determinant of inflammatory
joint disease capable of simultaneously driving or ameliorating
arthritis pathogenesis in distinct anatomical locations of the same
subject. Plasminogen can either drive or ameliorate arthritis
within the same animal as a function of anatomical location of the
joint, the availability of fibrin(ogen), and the activation status
of matrix metalloproteinase (MMP)-9. In conclusion, the work
presented in this dissertation demonstrates that fibrin
stabilization and fibrin dissolution are integral to the arthritic
process and make unique mechanistic contributions to arthritis
pathogenesis. This work strongly suggests that immunological as
well as hematological factors determine the precise incidence and
modulate the outcome of arthritis and suggests that targeting of…

▼ Fibrinogen is the most abundant protein involved in blood coagulation and has been associated with many pathological implications in cardiovascular disease. At the final stages of blood clot formation, the transglutaminase Factor XIIIa introduces γ-glutamyl-ε-lysinyl covalent bonds between reactive glutamines and lysines in fibrin, which results in a tighter clot network that is resistant to fibrinolysis. Factor XIIIa crosslinks specific reactive glutamines on fibrinogen, selecting more reactive glutamines in the αC region of fibrinogen than any other chain. Although crosslinking pairs in the αC region have been identified, little is known about the extent of crosslinking and the role played by each reactive glutamine. For the first time, we have ranked three reactive glutamines crosslinked under physiological conditions. A combination of MALDI-TOF mass spectrometry, LC-MS, and 2D
15N-
1H HSQC NMR studies was used on Fibrinogen aC(233-425) to understand Factor XIIIa’s substrate specificity. Factor XIIIa’s specificity for each reactive glutamine in aC(233-425) was monitored by the enzyme's ability to cross-link a lysine mimic, glycine ethyl ester (GEE), to the reactive glutamines (Q237, Q328, Q366). We showed that Factor XIIIa crosslinks these three glutamines to GEE in the order Q237 >> Q366 ≈ Q328. Fibrinogen aC(233-425) variants were generated in which each reactive glutamine (Q) was replaced with an inactive asparagine (N). We demonstrated that Factor XIIIa still crosslinks each reactive glutamine independently and is more selective in the absence of the most reactive glutamine Q237. To examine the role of the putative Factor
XIII binding site on αC(233-425), we performed experiments in which an important amino acid E396 was replaced with an alanine (Fbg αCE396A). Our results showed that both plasma Factor
XIII A
2B
2 and recombinant Factor
XIII A
2 can crosslink all three reactive glutamines in Fbg αCE396A to a similar extent as with the WT. For structural characterization, aC(233-425) was expressed in
15N or
15N/
13C-enriched media. 2D HSQC NMR experiments confirmed that the
15N-labeled αC(233-425) is mostly intrinsically disordered. Residues surrounding Q366 and Factor
XIII's binding site αC(389-402) showed a higher propensity for order. 2D NMR experiments suggest the possibility of pre-structured motifs (PreSMOS) within Fibrinogen aC(233-425).

In 1631, Mary de’ Médici (1573-1642), queen mother of the French king, opposed to Richelieu’s government, precipitately leaves the kingdom to find shelter at the court of Brussels. Until her death in 1642, she never succeeds, in spite of many attempts, to be allowed to come back to France. She lives in the Spanish Netherlands from 1631 to 1639. Then, after a short passage through the United Provinces, she stays in London until 1641, to move out again to Cologne, where she finally dies. Whereas the Thirty Years War tears Europe to pieces, Mary de’ Médici weaves bonds not only with foreign princes, but also with other French banned emigrants, such as Gaston of Orleans or the duchess of Chevreuse. The study of this outstanding situation reveals official or secret networks, which are built up and undone around those emigrants. The making and good working of those networks are ensured by a pool of ambassadors, courtiers, servants, who have various profiles and follow different purposes. By rebuilding the individual stories of these men, we can disclose the organic logic of this complex and moving party, in which the news arise and flow. We try to provide an analysis from the point of view of the material support and routing of information, but also according to its content, which may vary following the peculiar circumstances and political issues. The circulation and changes of the news affect the diplomatic deeds in a way…

A considerable amount of legends conveys hackneyed images of Henri IV and Louis XIII. The first one was then evoked as a good-natured, warlike figure and a Green Gallant who preferred the popular enjoyments of the boiled chicken and the bawdy songs of the Pont Neuf to the celebrations of the Valois court. His son was, as for him, considered as a taciturn monarch, dominated by his mother or by the almighty Cardinal Richelieu. Curiosity is…

The study on the Mo-Pt-Si system contributes to the knowledge of complex metal alloy, which are intended for applications at high temperatures in the aerospace or glass industries. Its thermodynamic description is difficult because its refractory character related to the presence of the molybdenum and to its complexity due to the high number of the constituents (3 elements). Only an approach of problem by a modelling thermodynamics of CALPHAD type gives to a significant result. As imposes the establishment of a modelling of a ternary system by the study of three binary borders, the first part of this work was devoted to the study of Pt-Si, Mo-Pt and Mo-Si systems. Along with the optimization, an experimental measurements used in this study (X-ray, thermal analysis, microprobe,….) allowed to determine the equilibria phases, the enthalpies of formation of the various intermetallic compounds as well as the structural mechanisms responsible for large domains of stability observed for some phases. The first approach of the Mo-Pt-Si ternary system obtained from the database including the binary optimisations, allowed to interpret the experiments of controls of equilibria phases in isotherm and anisotherm conditions. A new ternary silicide was discovered and took into account in calculation. Finally, through many examples we show the agreement of the modelling with all experimental…
