▼ [pt] As tecnologias da informação e comunicação (TICs)
vêm avançando de forma continuada e se amalgamando à cultura de uma
grande parcela da juventude mundial, inclusive a brasileira. Nessa
pesquisa, citamos o processo pelo qual está passando os espaços de
ensino-aprendizagem, do qual, aos poucos, estão acolhendo a demanda
desse público-alvo, de jovens estudantes, que desejam estar
conectados durante suas atividades escolares. Contudo, muitos games
com suas habilidades e competências têm contribuído para o
aprendizado e podem cumprir muitas tarefas, inclusive a de
reaproximar estudantes que estejam desestimulados com o ensino do
dia-a-dia. A sociedade, por sua vez, vem acompanhando esse processo
e, nesse documento, apresentamos um histórico de como evoluímos com
os games desde os arcades até chegarem aos celulares. Como
consequência dessa evolução tecnológica, há uma profusão de títulos
de games para os dispositivos móveis, muitos deles educativos, o
que vem de encontro com a necessidade de crianças e jovens em idade
escolar. Em seguida, apresentamos um estudo dos artifícios que
fazem desses games tão atrativos a esse público. Com todo esse
subsídio, a pesquisa propõe a formatação de uma taxonomia para
games educativos, a fim de oferecer subsídios para que alunos, seus
pais e professores tenham facilidade em identificar games que
possam contribuir para o seu aprendizado.
[en] Information and communication technologies have
been consistently moving forward and impacting a segment of the
youth population across cultures, including Brazil. In this
research paper we examine the transformations occurring in the
field of education, both from a teaching and a learning
perspective. These two arenas have gradually been taking on current
demands from their target audience, the youth, who desire to stay
connected while performing their typical school work. As a result,
many educational games emphasizing particular skills contribute to
learning and can end up playing a number of roles, one of wich is
to reengage demotivated students in their day to day learning
activities. Meanwhile, society as a whole has been paralleling this
transformation. This paper presents a history of the evolution of
games, from the arcades to the latest cell phone platforms, and how
the myriad of mobile educational games has been meeting the
academic needs of school age children. We present a closer look at
the tools and devices that make them so attractive to this
particular age segment of the population. Our research proposes the
implamentation of taxonomy exclusively of educational games, aiming
to facilitate their identification of games that contribute to
learning for students and their parents and teachers.

▼ L'objet de cette recherche consiste à capturer et analyser les relations entre l'architecture et les technologies récentes sous l'angle de la perception. Loin de la subjectivité supposée, l‘analyse guidée par la notion d'expérience perceptive s'appuie à la fois sur des concepts esthétiques et phénoménologiques. Cette double réflexivité porte également sur une étude de cas conceptuels et « concrets » qui opèrent la vision de la spatialité architecturale dans des tensions spéculatives entre le monde « numérique » et physique. La problématique porte sur la réception de l'architecture comme valeur culturelle et production intellectuelle, coordonnée par des moyens technologiques qui l'amplifient. S'inscrivant dans l'hypothèse qu'aujourd'hui l'architecture est loin de « servir » uniquement de réponse aux questions formelles, structurelles et fonctionnelles, la recherche démontre que l'architecture s'approche de plus en plus du champ artistique en phase d'expérimentations permanentes. Analysant les facteurs d'esthétisation de concepts variés, l'étude des cas vise à appréhender l'aspect cognitif et la valeur esthétique de la réception technologique — notion nouvelle, introduite comme un outil opératoire permettant d'aborder les aspects sensibles, hybrides et variables de la production architecturale. L'originalité de cette recherche consiste donc à analyser les connexions instables qui caractérisent les processus actuels de transitions diverses dans le domaine de l'architecture. À ce titre, cette recherche propose d'ouvrir le champ de la discussion autour du « digital » selon les principes de la singularité architecturale dans la pluralité des réceptions possibles
The purpose of this research is to capture and analyze the relationships between architecture and recent technologies in terms of perception. Far from the supposed subjectivity, this analysis guided by the notion of perceptual experience, is based on both phenomenological and aesthetic concepts. This double reflexivity also relates to the conceptual and "concrete" case studies operating the architectural vision of spatiality in speculative tensions between the "digital" and the physical word. The problematic concerns the reception of architecture as a cultural value and intellectual production, coordinated by technological means that amplify it. In keeping with the assumption that today's architecture is far away to "serve" only as a response to formal, structural and functional questions, this research shows that architecture is increasingly approaching the artistic field in phase of permanent experimentations. By analyzing the factors of aestheticization of various concepts, case study seeks to understand the cognitive and the aesthetic value of the technological reception — new notion, introduced as an operational concept to approach the sensitive, hybrid and variables issues of architectural production. uma pesquisa sobre a linguagem da Animação e seu potencial
expressivo para a formação educacional e profissional de indivíduos
portadores de síndromes cognitivas. Analisamos os aspectos
envolvidos na produção da animação – tais como concentração, noção
espacial e noção de tempo – e suas relações com indivíduos com
síndromes, classificados como especiais. O processo legitimador
dessa experiência foi o estudo da narrativa e suas práxis nas
Oficinas de Animação ministradas no Instituto Helena Antipoff (IHA)
e no Centro de Desenvolvimento do Humaitá (CDH). Nossa expectativa
com essa pesquisa é provocar e ampliar a discussão da inclusão dos
portadores de síndromes no universo da Animação, tanto do ponto de
vista educacional quanto expressivo e profissional, através dos
filmes desenvolvidos nas oficinas de Animação
Especial.
[en] This thesis presents the progressive process and
results of a research on language of Animation and its expressive
potential for educational and professional accomplishment of people
with cognition syndromes. We have analyzed the aspects involving
the production of Animation – such as concentration, space and time
notion – and its relations towards the individuals with syndromes,
classified as persons with special needs. The process that ratifies
this experiment as a legitimate one was the thorough study of
narrative and its praxis during the Animation workshops, which took
place at the Helena Antipoff Institute (IHA) and at the Humaitá
Development Center (CDH). Our expectation with the present research
is to provoke and to expand the discussion concerning the inclusion
of individuals with syndromes in the universe of Animation,
considering both the educational as well as the expressive and
emotional point of view, through the films produced during the
Special Animation Workshops.

(TIC’s) no processo de ensino, ou e-learning, como estão se
tornando conhecidas, seja para uso no apoio ao ensino presencial,
seja para a educação a distância (EAD), estão, cada vez mais, se
tornando uma parte integrante, tanto do ambiente cultural, quanto
do ambiente tecnológico da universidade moderna. Quando se observa
a introdução e implementação do e-learning nas escolas de gestão
brasileiras, tem-se percebido nas recentes experiências que o
processo exacerba certas tensões internas. Por outro lado, os
estudos na literatura que investigam o fenômeno são, na maioria dos
casos, estudos descritivos ou normativos, e que não explicam o
processo. Entretanto, sustenta-se que a implementação de tecnologia
é um fenômeno complexo e que envolve a interrelação de diversos
fatores e que deve ser considerado não somente os aspectos
funcionais, mas também o significado que a tecnologia tem para os
envolvidos. Com o objetivo de preencher essa lacuna, este estudo
buscou compreender como as escolas de gestão gerenciam o processo
de implementação do e-learning em seus ambientes a partir do
significado que seus gestores atribuem às suas experiências. A
partir da análise da experiência de sete escolas com a
implementação do e-learning, esta pesquisa propõe uma teoria
substantiva do processo, utilizando os procedimentos metodológicos
da Grounded Theory (Glaser & Strauss, 1967; Corbin &
Strauss, 2008). Foram entrevistados 14 gestores dessas escolas e os
resultados da análise revelaram cinco categorias conceituais: 1) a
incongruência; 2) a estrutura; 3) a forma de entrada; 4)
aculturando o e-learning; e 5) disseminando o elearning que
integradas em torno da categoria central – a incongruência –
serviram de base para construir um modelo relacional que explica
como ocorreu o processo de implementação do e-learning nessas
escolas. A conclusão principal que se chegou é que para o processo
de implementação do e-learning ser efetivo deve ter claramente em
suas premissas que, para que possa ser aceito e disseminado dentro
do ambiente interno, deve vir acompanhado de ações que consigam
transmitir aos stakeholders uma clara perspectiva de valor
relacionada ao uso de tecnologia no processo de ensinoaprendizagem.
Somente dessa forma os envolvidos podem perceber os benefícios de
seu uso. Assim, a implementação do e-learning, independente do
objetivo pela qual a escola introduz em seu ambiente, deve
privilegiar a questão pedagógica, definindo estratégias que
evidenciem o e-learning como uma ferramenta capaz de melhorar as
práticas existentes de ensino, estimulando que se desenvolva
internamente uma percepção de valor em relação a ele, caso
contrário, não surge a motivação para que o uso da tecnologia possa
se disseminar no ambiente. Espera-se que os resultados encontrados
contribuam para aumentar o conhecimento sobre o processo de
implementação do elearning, principalmente nas escolas de gestão, e
para gerar reflexões acerca do fenômeno no contexto
brasileiro.

▼ [pt] Os projetos envolvendo tecnologias digitais,
design e educação têm uma presença significativa no Brasil e no
mundo. Por meio deles, são criados e modificados serviços, produtos
e modelos. No âmbito da educação formal, as atividades lúdicas
digitais (ALDs) são recursos usados por professores e alunos
durante situações de ensino-aprendizagem em instituições de ensino
como escolas, universidades e cursos. Esta pesquisa tem como
objetivo caracterizar o desenvolvimento das ALDs para o
ensino-aprendizagem de crianças, visando compreender aspectos
relativos ao contexto, à experiência de uso e aos processos de
projeto. No universo dos projetos, é dado enfoque aos que são
idealizados em empresas com estrutura editorial. A investigação se
dá sob uma perspectiva multidisciplinar e utiliza como base os
achados provenientes de revisão de literatura e de análise de
conteúdo de entrevistas realizadas com gestores, professores,
editores, designers e programadores. Pretende-se contribuir com os
campos do Design e da Educação ao investigar, analisar e discutir
métodos, processos, ferramentas e técnicas presentes em projetos de
ALDs, bem como empreender reflexões sobre a atuação do designer em
projetos desta natureza.
[en] Projects that involve digital technologies,
design and education have a significant presence in Brazil and in
the world. Through them, services, products and models are created
and modified. In the context of formal education, digital playful
activities (DPAs) are resources used by teachers and students
during teaching-learning situations in educational institutions
such as schools, universities and courses. This research aims to
characterize the development of DPAs for teaching-learning of
children, to understand aspects related to the context and the user
experience and design processes. In the universe of projects, the
focus is on those that are idealized in companies with an editorial
structure. The research takes place from a multidisciplinary
perspective and uses as a base the findings from literature review
and content analysis of interviews conducted with managers,
teachers, editors, designers and programmers. It is intended to

▼ [pt] O objetivo desta tese é refletir sobre o
documentário animado. Um produto audiovisual que mistura
documentário e animação e está redefinindo o papel do design na
produção das novas mídias. Mostramos também que o documentário
animado reacende uma série de debates e reflexões relativas à
teoria do documentário e da animação em relação às concepções de
realismo. A nossa principal premissa é que o documentário sempre se
apropriou da tecnologia de forma a favorecer a experimentação e, o
documentário animado, em 3D, é a sua forma de se apropriar das
tecnologias digitais. Para comprovar esta hipótese mostramos como a
experimentação da tecnologia se deu no documentário em diferentes
épocas, mais precisamente nos períodos do primeiro cinema, das
vanguardas e da escola britânica – de 1900 a 1930, os anos de 1960,
e os anos de 1990 (novas mídias). O que nos interessa é investigar
esta relação entre os meios de produção utilizados na realização do
documentário – quer tenham esta denominação ou não – e os modos de
representação e estilos resultantes da tecnologia de cada época,
que pretendemos delinear brevemente. O Design está presente nos
dois aspectos: nos meios de produção, por sua tradição ligada ao
desenvolvimento de tecnologias e instrumentos para viabilizar a
produção, e nos modos de representação, porque as referências
estéticas da animação são oriundas das artes gráficas (quadrinhos,
ilustrações, etc). Isto também explica, em parte, a ausência da
animação na teoria do cinema, aspecto que questionamos, mostrando
como a animação está contemplada na teoria Deleuziana. Para
concluir, fazemos um estudo de caso de um documentário animado
emblemático, por sua abordagem da tecnologia 3D. Ryan (2004), de
Chris Landreth, resume o potencial do documentário animado enquanto
experimentação da tecnologia, intensidade narrativa e estética não
figurativa, em singular referência ao pintor Francis
Bacon.
[en] The objective of this thesis is to reflect on the
animated documentary. An audiovisual product that mixes both
documentary and animation and is redefining the role of the design
in the production of new media. We also show that the animated
documentary rekindled a series of debates and reflections related
to documentary s theory and realism concepts in animation. Our main
premise is that the documentary has always appropriated of
technology in order to allow experimentation, and the animated
documentary, in 3D, is the way to appropriate itself of digital
technologies. To prove this hypothesis, we show how the trial of
the technology occurred in documentary s history at different
times, more precisely during the periods of the early cinema, the
vanguards and the British School – from 1900 to 1930, the 1960 s
and the 1990 s (new media). What concern us is to investigate this
relationship between the means of production used in the making of
documentary movies – having this denomination or not – and ways of
representation and styles resulting from the technology of each

s ohledem na konstrukci mobilního navařovacího systému, které nepočítá
s možností předehřátí základního materiálu. Jako nejvhodnější přídavný materiál, který vyhověl všem požadavkům, se ukázal STEIN-MEGAFIL A 740 M.; Diploma thesis examines the most appropriate solution of filler materials, suitable
technology and welding parameters for the renovation of the crane wheel. Five different filler materials were suggested and have been
subject of macroscopic examination and measurement of quality - primary the hardness of the weld deposit. Additional materials have been chosen to design a mobile weld system, which does not count with the possibility of preheating the base material. As the most suitable filler material, which satisfy all the requirements that emerged STEIN-MEGAFIL A 740 M.

▼ Tématem diplomové práce jsou strukturní a základní mechanické změny, které probíhají v litině
s lupínkovým grafitem po přetavení a legování povrchu prostřednictvím chrom-niklové oceli pomocí EB technologie. Dále je sledován vliv nastavených parametrů na vlastnosti a hloubku natavené oblasti. Experimentální část diplomové práce se zabývá mikrostrukturami, mikrotvrdostí a rozložením prvků, které byly analyzovány po natavení a legování povrchu za určitých nastavených parametrů na přístroji elektronového paprsku. Na základě mikrostruktur jsou popsány strukturní změny, kterými prošly jednotlivé oblasti materiálu po natavení. V závěru práce jsou rozebrány výsledky využití techniky elektronového paprsku na LLG a vyvozeny závěry, které z nich vyplývají.; In this thesis, the structural and mechanical changes, which took place in flake graphite cast iron after electron beam surface melting and chrome-nickel surface alloying, were studied. Furthermore, the effect of set parameters on properties and depth of the melted and alloyed region is also analyzed. In the experimental part, the analyzed microstructures, micro-hardness and the distribution of elements after melting and alloying, were presented. Structural differences of material regions caused by melting were described based on microstructures. In the end of diploma thesis, the conclusions of using EB
technology for surface melting and alloying were discussed.

▼ Este estudo tem como objetivo analisar como se dá a inserção e a vivência cotidiana de um grupo de mulheres em sala de aula nos cursos de Engenharia Elétrica e Ciência da Computação em uma faculdade localizada na Região Metropolitana de São Paulo RMSP, investigando os obstáculos enfrentados por elas nesses cursos e considerando as estratégias que utilizam para se manterem neles. Foram realizadas entrevistadas semi-estruturadas com 49 alunas, sendo sete do curso de Engenharia e 42 do curso de Computação. Com base na análise das entrevistas foi possível perceber não apenas as diferentes formas de exclusão a que ficam expostas as alunas desses cursos cujo corpo discente é em maioria do sexo masculino, mas também as táticas utilizadas por elas para conseguir permanecer no curso e enfrentar a discriminação e as atitudes preconceituosas de colegas e professores, em geral evitando o embate direto e inventando formas sutis de convivência com as adversidades em sala de aula.
This study aims to examine the insertion and daily life of a group of women in the classroom in the courses of Electrical Engineering and Computer Science at a college located in the metropolitan region of Sao Paulo - RMSP. It investigates the obstacles faced by them in these courses and the strategies they use to remain in them. We performed semi-structured interviews with 49 students, seven students of Engineering and 42 students of computing. Based on the analysis of the interviews, we could comprehend not only the different forms of exclusion that the female students of these courses are exposed to, in which the majority of students are male, but also the tactics they use to remain in the course and to deal with discrimination and biased attitudes of colleagues and teachers. In general, they avoid clash and invent subtle ways to co-exist with adversities in the classroom.

de recepção das reportagens produzidas pela Caravana do Jornal
Nacional, um projeto de telejornalismo executado pela Rede Globo,

reencontrar as pessoas entrevistadas pela equipe de jornalistas, na
época, e questioná-las sobre essa experiência de participar de um
projeto de telejornalismo e de vivenciar um contato com os
profissionais do meio. Vamos avaliar como esses grupos entenderam
as representações feitas nas reportagens da Caravana JN, realizadas
nas comunidades visitadas, investigar como foi o encontro dos
profissionais do JN com o seu próprio público, de que forma as
comunidades decodificaram a passagem da Caravana e que tipo de
interações sociais o projeto foi capaz de promover. O trabalho de
campo foi realizado em duas cidades da região Sul e uma do Sudeste
do Brasil. Nossas análises se voltam para as rotinas produtivas do
telejornalismo e ao que a Caravana JN significou, em termos de
inovação, dentro da trajetória de um produto da comunicação de
massa, líder de audiência há quase quatro décadas.
[en] This research belongs to the field of
relationship between mass culture and national identity and
proposes an assessment on how the special coverage produced by the
Caravana do Jornal Nacional was received by the Brazilian
communities. Caravana JN was a significant project on broadcast
journalism executed by Rede Globo right before the national
presidential elections. As it was traveling from south to north of
Brazil, the Caravana JN went to the encounter of citizens that live
in very remote areas with the objective of investigating what would
be their wishes and dreams. One of the challenges of this study was
to find the same people that were interviewed by the journalist
team then, in order to inquire about the whole experience. More
than an evaluation about the representations illustrated on this
special coverage of the visited communities, this study also
investigates how was the encounter between the television
professionals and their audience, in what way the communities
decoded the Caravana passage and what kind of social interactions
the project was capable to promote. The field part of this research
was done in two cities of the South and one of the Southeast
regions of Brazil. Our analyses was focused on the production
routines of broadcast news in which the Caravana JN made impact in
terms of innovation, and on the trajectory of a mass communication
product that maintains the highest and record audience ratings of
the for more than four decades.

do estudante do Curso de Especialização em Tecnologias em Educação
- Lato Sensu (2006-2007), realizado na modalidade a distância, como
projeto de parceria entre a Coordenação Central de Educação a
Distância (CCEAD/PUC-Rio), o Fundo Nacional de Desenvolvimento da
Educação (FNDE), a Secretaria de Educação a Distância (SEED) e o
Ministério da Educação e Cultura (MEC). A partir da análise da
interface gráfica, dos conteúdos e dos registros documentais
hospedados no ambiente virtual, foram investigados indicadores que
denotam maior ou menor grau de autonomia dos estudantes no trato
com os conteúdos específicos dos componentes curriculares e com a
proposta metodológica em si, no que diz respeito à aquisição de
conhecimentos. Trazendo uma análise, em linhas gerais, da
otimização dos recursos da Internet e sua utilização na aquisição
de informações e conhecimentos, o trabalho pretende integrar-se ao
conjunto das discussões científicas que vêm sendo realizadas acerca
da integração das tecnologias de informação e comunicação na
formação de professores. Dessa maneira, visa contribuir para as
reflexões sobre as potencialidades telemáticas aplicadas à
educação, relacionados à autonomia e à interatividade dos
estudantes. A interpretação dos dados, pautada na análise de
conteúdo, possibilitou, ainda, a compreensão das especificidades
metodológicas das propostas educacionais para o ensino a distância
como elemento indicador de favorecimento da autonomia dos
estudantes.
[en] This paper presents an analysis of possibilities
and limitations with regard to promoting the autonomy of the
student of the Curso de Especialização em Tecnologias em Educação -
Lato Sensu (2006-2007), carried out in the distance mode, as a
partnership project between the Coordenação Central de Educação a
Distância (CCEAD/PUC-Rio), the Fundo Nacional de Desenvolvimento da
Educação (FNDE), the Secretaria de Educação a Distância (SEED) and
the Ministério da Educação e Cultura (MEC). From the analysis of
the graphical interface, contents and the documental records hosted
on the virtual environment, were investigated indicators which show
greater or lesser degree of autonomy of the students in dealing
with the specific content of the curriculum components and the
proposed methodology in itself, as regards the acquisition of
knowledge. Bringing an analysis, in general, of the optimization of
the resources of the Internet and its use in the acquisition of
information and knowledge, the work seeks to integrate itself to
all the scientific discussions being held about the integration of
information and communication technologies in teacher education.
Thus, it aims to contribute to the reflections about the telematic
potentials applied to education, regarding the autonomy and

licenciatura da Universidade Federal de Mato Grosso do Sul. O
objetivo geral da pesquisa foi identificar e analisar as
características das práticas pedagógicas com o uso de tecnologias
digitais, sob o ponto de vista dos projetos institucionais e das
percepções dos professores e alunos desses cursos, com vistas a
identificar o nível de capital tecnológico dos professores e a
relação desse capital com as práticas pedagógicas consideradas
inovadoras dentro do contexto institucional. O caminho metodológico
da pesquisa se iniciou com a revisão de bibliografia, passando pela
análise de projetos de curso, aplicação de questionário para os
coordenadores, questionário para professores e alunos, finalizando
nas entrevistas com professores e alunos. Privilegiamos, nesse
estudo, a integração de diferentes procedimentos analíticos com o
objetivo de obter um conhecimento descritivo, relacional e
compreensivo. A pesquisa indica que, embora existam, na maioria dos
projetos pedagógicos, disciplinas que abordem o uso das tecnologias
digitais e que os professores usem vários recursos digitais em suas
práticas pedagógicas, o nível de incorporação ainda não permite que
esse uso ultrapasse o instrumental. As poucas experiências
inovadoras encontradas demonstram que o capital tecnológico dos
professores é um fator preponderante para as mudanças nas práticas
pedagógicas, mas essas experiências isoladas ainda não têm um
potencial de difusão e adesão do grupo, pois não há na Instituição
um clima favorável à inovação, talvez por não existir um projeto
institucional de fomento ao uso de tecnologias digitais na prática
pedagógica ou uma política institucional de formação de
professores, que estabeleça metas para a melhoria na formação dos
professores.
[en] The present study focused on pedagogical
practices with the use of digital technologies in undergraduate
courses at the Federal University of Mato Grosso do Sul (UFMS). The
general objective of the research was to identify and analyze the
characteristics of pedagogical practices with the use of digital
technologies from the point of view of institutional projects and
the perceptions of teachers and students of these courses, in order
to identify the level of technological capital of teachers and the
relation of this capital to pedagogical practices, considered
innovative within the institutional context. The methodological
path started with a literature review, through the analysis of
course projects, a questionnaire for coordinators and a
questionnaire for teachers and students, finalizing with teachers
and students interviews. We highlighted on this study the
integration of different analytical procedures in order to obtain
descriptive, relational and comprehensive knowledge. The research

▼ L'objectif de notre thèse est d'examiner la place qu'occupe la créativité dans les technologies de l'information (TI). Nous présentons dans un premier lieu une discussion des principaux travaux qui ont traité de la créativité dans les systèmes d'information (SI). Ensuite, nous discutons de la relation entre la créativité et la technologie. Cette thèse contient trois articles, et nous présentons ici le contenu de ces articles. Dans le premier article, nous avons effectué une revue de la littérature et nous avons examiné la place de la créativité dans les systèmes d'information et positionné cette recherche à partir de la classification d'Orlikowski et Scott (2008) de la variance, des processus et de la relationalité. Nous avons situé la littérature sur la créativité dans la recherche SI dans une logique de variance et nous avons proposé d'élargir la recherche en étudiant cette relation dans une logique de processus sociaux. Dans le second article, nous nous sommes concentrés sur l'impact des technologies sur la créativité et nous avons mené une étude ethnographique sur un festival à Québec. La contribution de cette article consiste à voir cet impact comme transmissif, interactif et imaginatif. Dans le troisième article, nous nous sommes concentrés sur l'impact de la créativité sur l'utilisation de la technologie et nous avons mené une recherche qualitative dans le secteur de la radio à Montréal. La contribution de cet article est de montrer l'importance de la créativité dans l'usage des systèmes d'information. L'objectif général de cette thèse est d'étendre la connaissance scientifique et pratique en ce qui concerne les SI et la relation entre la créativité et la technologie à partir de la logique du processus social.

▼ Epidermal growth factor receptor in cancer therapy Recently, cancer research has been able to identify molecular targets that are specific for (or highly expressed by) cancer cells. These molecular targets serve as models for the development of rationally designed anticancer drugs that target important mediators of tumour development. Also, some of these molecular targets have been explored for targeted drug delivery, to ensure that anticancer drugs are specifically delivered to the tumours. The epidermal growth factor receptor (EGFR) is an extensively investigated growth factor and a recognized target for cancer therapy. Many human epithelial cancers overexpress EGFR and the dysregulated EGFR signalling is associated with uncontrolled cell proliferation, inhibition of apoptosis, stimulation of angiogenesis, tissue invasion and metastasis. Two main strategies have been employed to inhibit EGFR: tyrosine kinase inhibitors (TKIs) and monoclonal antibodies (mAbs), some of them have already been approved for clinical use. EGFR: target for silencing A different strategy to inhibit EGFR was employed in this thesis. Here, the expression of EGFR protein was interrupted by use of small interfering RNA (siRNA). For efficient gene silencing, siRNA must be delivered intact into the target cell cytoplasm, where RNA interference (RNAi) takes place. Carriers, such as Lipofectamine, are generally used to promote cellular uptake. However, when internalization occurs via endocytosis, the endosomal escape becomes one of the limiting steps for efficient knockdown of the target protein. In this thesis, two different methods were successfully employed to facilitate the endosomal escape of siRNA: an influenza-derived fusogenic peptide and photochemical internalization (PCI). PCI combines a photosensitizer with light to induce photochemical reactions that destabilize endosomal membranes and allow the release of entrapped molecules. PCI was also successfully applied in vivo for enhancing the endosomal escape of anti-EGFR siRNA-Lipofectamine, delivered by intratumoral injection. This study demonstrates that also in vivo endosomal escape is a limiting step that can be facilitated by PCI, improving silencing efficiencies. Local administration of siRNA is not always possible and, therefore, novel targeted drug delivery systems are developed for cancer-specific molecular targets, to direct siRNA and other therapeutic molecules to cancer cells. EGFR: target for delivery Liposomes are attractive delivery systems that have been employed for delivery of a wide range of therapeutic molecules. Liposomes surface has been modified with shielding polymers and targeting ligands that increase their circulation time in the bloodstream and mediate specific target-cell interactions, respectively. In this thesis, a novel targeted drug delivery system was described: anti-EGFR nanobody-liposomes. ▼ Summary Factor (F)VIII is an essential cofactor in the enzymatic coagulation cascade. A defect in the gene encoding for FVIII results in the X-linked inherited bleeding disorder hemophilia A. In the circulation, FVIII is protected from proteolytic degradation and premature clearance by forming a complex with its carrier protein von Willebrand factor (VWF). While the complex of FVIII and VWF involves one of the strongest protein-protein interactions in the human body, it has remained unclear when and where FVIII and VWF first interact. The most widely accepted view is that FVIII and VWF are secreted from different cells and form a complex in the circulation, merely by virtue of their high-affinity interaction. During the last decade however, increasing evidence has been presented that suggests that the human body should also comprise a releasable storage pool of both proteins. In this thesis, we have studied the relationship between high-affinity interaction in the circulation and intracellular co-storage of FVIII and VWF. We have taken a new approach towards this issue by creating fluorescently tagged VWF and FVIII proteins that are established as being deficient in high-affinity interaction. We have demonstrated that FVIII is stored in VWF-containing storage organelles in absence of high-affinity interaction and that the extent of FVIII storage is independent of its affinity for VWF. In addition, we studied the therapeutic benefit of intracellular co-expression of FVIII and VWF in terms of potential treatment options for hemophilia A. We have shown that blood outgrowth endothelial cells can produce high levels of functional FVIII over a prolonged period of time. In addition, we have demonstrated that a substantial amount of FVIII can be stored in VWF-containing storage organelles whereas the majority of FVIII is released in a constitutive manner. From our data, we conclude that the structural requirements that determine high-affinity interaction between FVIII and VWF in the circulation differ from those that determine intracellular co-trafficking. Our data provide a molecular explanation for the fact that hemophilia A and von Willebrand’
s disease patients that suffer from impaired complex assembly of FVIII and VWF in the circulation show concomitant increase of FVIII and VWF plasma levels following treatment with desmopressin. The mechanism that determines FVIII storage in VWF-containing granules as well as the therapeutic potential of FVIII expression in VWF producing cells now awaits further exploration.

▼ The mature epidermis is an effective barrier which prevents the body from dehydration and protects it against various environmental influences. If the natural barrier is immature or damaged, the skin barrier is impaired and desiccation occurs. Hence, the regeneration of impaired skin is an essential process for survival. In patients, the natural recovery is sometimes rather slow, in particular in the cases of large defects. To support skin regeneration, a large variety of skin creams is presently available, aiming to modify the hydration level of the skin and to protect the body against excessive water loss and thus to enhance epidermal barrier function. However, these creams clearly have a different structure than stratum corneum (SC), resulting in low water binding capacity and uncontrolled hydration of the skin. The basic mechanism of these skin creams is that they act as moisturizer and increase the hydration of SC by either blocking the TEWL (transepidermal water loss) or by delivering exogenous hygroscopic substances to the SC. Therefore, it has been suggested that the development of barrier creams with high water-holding capacity and thus controlled water transport, such as the natural cream VC (vernix caseosa), might help in the development and/or restoration of the SC barrier. The aim of this thesis was the development of a new generation of skin-surface biofilms, combining the structure and properties of VC, to protect diseased, dry and premature skin and to facilitate wound healing. These biofilms were based on synthetic cells embedded in a lipid matrix. The synthetic cells consisted of crosslinked hydrophilic polymers (hydrogels) with similar size and shape as natural corneocytes. Combined with the work of Rissmann (presented in thesis R. Rissmann), who focused on the development of a synthetic lipid mixture, we were able to prepare biofilms. The biofilms were prepared by mixing the synthetic corneocytes with the lipid matrix. These biofilms were characterized and optimized to mimic VC. Various biofilms were applied topically on disrupted mouse skin to investigate their effect on superficial wound healing. All formulations promoted a rapid formation of SC and prevented epidermal hyperproliferation. In general the synthetic VC analogues showed strong effects concerning the recovery rate, however, only one biofilm mimicked the effects of native VC the most closely. In conclusion, this thesis describes the development of a new generation of skin-surface biofilms mimicking closely the structure and properties of VC. These biofilms act as skin protectants and facilitate the restoration of the SC barrier. These unique systems have therefore great potential to protect diseased, dry and premature skin and to facilitate wound healing in humans.

▼ In this research, several block copolymers were synthesized and characterized with regard to possible pharmaceutical applications. All block copolymers were thermosensitive and self-assembled at 37 °C into structures like micelles and hydrogels, which can be used for innovative drug delivery purposes. Some of the synthesized polymers were biohybrid, in the sense that they contained peptide segments which enabled their cleavage by enzymes that are upregulated in diseased tissues. First, methods were explored for the synthesis of polymer (bio)conjugates. One suitable and popular reaction for synthesizing such conjugates is the copper-catalyzed azide-alkyne cycloaddition ‘click’ reaction. A novel method was developed to introduce a reactive azide functionality on polymers after their synthesis by Atom Transfer Radical Polymerization (ATRP). The newly developed reaction is catalyzed by the ATRP copper catalyst and can be followed by a ‘click’ conjugation reaction using again the same catalyst. Another, relatively new, method to synthesize polymer bioconjugates is to grow a polymer from a peptide or protein. In this research, a peptide containing an enzymatic cleavage site for the tumor-associated matrix metalloproteases (MMPs) 2 and 9 was functionalized at its C-terminus with an ATRP initiator, followed by the polymerization of a hydrophilic polymer block, poly(oligo(ethylene glycol) methyl ether metharcrylate).Then, another ATRP initiator was conjugated to the N-terminus of the peptide followed by the polymerization of a thermosensitive block, poly(N-isopropylacrylamide). At 37 °C in water, all polymers formed micelles with a favorable size for drug delivery purposes. It was shown that the coronas of the micelles could be cleaved off enzymatically. This system may function as a long-circulating drug delivery vehicle with enzyme-triggered destabilization in the target tissue to release e.g. a cytotoxic payload. Subsequently, a study of the physico-chemical differences between ‘star-like’ micelles and ‘flower-like micelles’ was performed. Star-like micelles are formed from amphiphilic diblock copolymers, whereas flower-like micelles are formed from triblock copolymers that consist of two hydrophobic blocks separated by a hydrophilic block. Both Static Light Scattering and 1H NMR T2 relaxation measurements indicated that hydrophilic polymer blocks in flower-like micelles are less flexible than in star-like micelles. The last part of this research focused on a particular property of flower-like micelles, namely the fact that at high concentration they are in equilibrium with a hydrogel. When a thermosensitive triblock copolymer hydrogel was brought into water, flower-like micelles were released. When the hydrogel was pre-loaded with the cytostatic drug paclitaxel, this was co-released with the micelles. ▼ Therapeutic proteins have become invaluable in treating a wide range of serious and life-threatening diseases. However, repeated administration of these drugs to patients often induces the formation of undesirable anti-drug antibodies, also known as immunogenicity. Among the factors that are known to play a role in immunogenicity of therapeutic proteins, the presence of protein aggregates has been indicated as one of the main product-related risk factors. Even though several studies have shown that protein aggregates have the propensity to trigger an antibody response against the monomeric form of the protein, little is known about the particularities that make them immunogenic. The work presented in this thesis focuses mainly on aggregate characterization and aggregate-related immunogenicity of monoclonal antibody formulations, using a monoclonal human IgG1 with k light chains as a model. The use of emerging analytical techniques for the characterization of IgG aggregates is evaluated; non-native aggregation mechanisms of IgG are studied; the fate of fluorescently labeled IgG aggregates upon contact with human biological fluids in vitro and upon injection in mice is investigated; and the correlation between type and amount of IgG aggregates with their immunogenic potential is studied in transgenic mice (containing the human genes for Ig heavy and light chains) and their nontransgenic counterparts

▼ Since its discovery in 1996, numerous functions have been proposed for Factor VII-activating protease (FSAP) including interactions with the haemostatic system. These include activation of the single chain urokinase-type plasminogen activator (scuPA) and coagulation factor VII (FVII), and interaction with the endothelium and smooth muscle cells. With its decreasing specificity, the physiological role of FSAP has become increasingly unclear. About 5-10 % of apparently healthy individuals carry a FSAP variant (known as “Marburg-1”) which contains a glycine (G) to glutamate (E) substitution located in the catalytic domain. This FSAP variant is associated with a 50-80 % impaired activation of scuPA in vitro, suggesting an effect on fibrinolysis. Epidemiological studies however, vary strongly in their conclusions concerning the implications of this polymorphism. The studies presented in this thesis were performed to assess the role of FSAP in the haemostatic system and the potential pathophysiological implications of the Marburg-1 FSAP polymorphism (G534E) for FSAP biological activity

▼ The aim of gene therapy is to treat, cure or prevent a disease by replacing defective genes, introducing new genes or changing the expression of a person’
s genes. Success of gene therapy is dependent on successful delivery of DNA from the site of administration into cell nuclei. Naturally occurring and highly efficient gene delivery systems are viruses. However, issues of safety, limited targeting possibilities and limited loading capacity justify the search for safe and flexible synthetic (non-viral) alternatives. Despite numerous efforts, success of non-viral gene delivery is hampered due to four major problems: toxicity, short-lived expression, heterogeneity of expression and low efficiency. The aim of this thesis is to rationally design DNA-Transporting Nanoparticles (DTN), focusing on three levels: (1) design of DNA to increase safety, efficiency and duration of transgene expression, (2) design of formulations to increase efficiency of transfection and (3) development of methods to allow better analysis of DTN. First it is shown how rational design of plasmid DNA can increase safety, efficiency and duration of transgene expression. Next, two strategies to improve nuclear uptake of DTN are examined. The first strategy involves modification of plasmid with DNA nuclear Targeting Sequences (DTS) which would facilitate nuclear localization by inducing coating of plasmid DNA with nuclear localization signals (NLS)-containing proteins and subsequent binding to importins. Modification of plasmids with DTS was not shown to increase gene expression, regardless of the cell line, dose and carrier system chosen, indicating that a bottleneck exists downstream of nuclear delivery. The second strategy was based on the non-covalent coupling of NLS-peptides to DNA. Peptides were shown to enhance transfection efficiency, but in an NLS-independent way. Next, the number of plasmids per DTN is addressed. The amount of active DNA could be reduced substantially while maintaining transfection activity by replacing active DNA with non-coding junk DNA. The results indicate that not the total amount of DNA, but the number of active DNA-containing particles is the critical factor in determining transfection efficiency. Size of DTN is a critical parameter, but preparation of DTN generally yields heterogeneously sized populations. Additionally, the size of DTN is affected by exposure to salts and proteins. A new method based on flow cytometry was developed to measure particle size distributions directly in biological fluids. Simultaneous detection of fluorescence and side scattering intensity (SSC ; indicative of size) provided the possibility to distinguish fluorescently labeled nanoparticles of interest from other particulate matter frequently present in biological fluid. Additionally, a proof of principle to sort heterogeneous populations into different size fractions was obtained. ▼ With about 200 new products in the pipeline, recombinant human (rh) therapeutics are becoming the most dominant class of drugs. One of the reasons to create rh therapeutics was to avoid recognition by the immune system due to foreign origin. Nevertheless, rh therapeutics induced formation of anti-drug antibodies. Aggregation of the therapeutic protein became a major concern, directing the focus on an ancient mechanism of the immune system to recognize repetitive epitopes on pathogens and activation of a non-classical T-cell – B-cell pathway leading to a fast and efficient immune response. It was hypothesized that aggregated rh therapeutics mimic such repetitive epitope carrying pathogens, named T cell independent type 2 antigens. The thesis entitles ‘Immunological aspects of antibody formation against recombinant human therapeutics’ investigated the immunological mechanisms triggered after administration of aggregated rh therapeutics in transgenic mice immune tolerant for the therapeutic of interest. A three step approach was used: (I) Memory formation after administration of aggregated rh therapeutics?, (II) Is the response T cell independent?, (III) Do Marginal Zone B cells influence the immune response against rh therapeutics?. Two model therapeutics were used: (1) rh interferon beta, and (2) recombinant human interferon alpha. Both models showed the absence of memory formation after initial challenge and boost with both aggregated and non-aggregated rh drugs. In addition, a lack of neutralizing antibodies was observed. Though memory was absent, animals had an abundant pool of isotype switched antibodies, a sign for T cell dependency. And indeed, depletion of CD4+ T cells led to a complete abolishment of the antibody formation against aggregated therapeutic. Lastly, it could be shown that the depletion of marginal zone B cells influence the response against rh therapeutics. In conclusions, it was shown that the immune response directed against aggregated rh therapeutics is neither comparable to a classical nor to a T cell independent type 2 antigen. In addition, the presence of naturally occurring antibodies in human blood samples was investigated. It is hypothesized that patients having natural occurring antibodies against a rh therapeutic may be more susceptible to formation of anti-drug antibodies when treated with the rh therapeutic. The presence of naturally occurring antibodies against interferon alpha, beta and gamma and bone morphogenetic proteins 2 and 7 could be found in around 2% of individuals (n=400). Their influence on the immunogenicity of rh therapeutics need to be further explored.

▼ Molecular imprinting is a technique to create template-shaped cavities in polymer matrices with memory of the template molecules, to be used in molecular recognition. Molecular imprinting of low molecular weight compounds is a well established technique used to create high affinity materials. On the other hand, molecular imprinting of high molecular weight compounds such as proteins is not nearly as successful for various reasons. Nevertheless, the use of hydrogels has led to some encouraging protein imprinting results. A hydrogel consists of a three dimensional polymer network that is able to retain large amounts of water with conservation of the network structure. As a result of the high water content, hydrogels are generally considered as biocompatible materials, which makes them particularly interesting for biomedical and pharmaceutical applications. In this thesis, an innovative method to prepare hydrogel nanoparticles with protein imprints on their surface is proposed. Polyacrylamide nanoparticles were prepared inside the internal aqueous compartment of liposomes, prepared by detergent removal. Macroscopic hydrogel formation due to polymerization of the monomers outside the liposomes was prevented by inhibition of free-radical polymerization of non-encapsulated monomers by addition of ascorbic acid. This method offers straightforward control over the shape and size of the particles. Moreover, the liposomal bilayer offers the possibility to incorporate membrane proteins, which can then be used as templates for surface imprinting. It is shown that SN/GpA is successfully incorporated in the liposomal bilayer. Importantly, the hydrophilic domain of SN/GpA was oriented towards the internal compartment of the liposome, and therefore would be available for the formation of imprints. However, despite many attempts towards the synthesis of PINAPLES, a proof of concept was not obtained. This is partially due to the current state of the art of protein imprinting. Protein imprinting is still a relatively new scientific field. Although the number of research papers that describe protein imprinting is increasing, the actual scientific proof of the feasibility of selective recognition of proteins by molecular imprinted polymers in aqueous environment is not convincing. Electrostatic interaction between protein and hydrogel network can cause high aspecific binding, and may therefore not be suited for molecular imprinting. However, this strong interaction may be useful as a tool for controlled release of therapeutic proteins from pharmaceutical formulations. Charged hydrogels were obtained by copolymerization of methacrylated dextran (Dex-MA) with either methacrylic acid (MA) or 2-N,N-dimethylaminoethyl methacrylate (DMAEMA). The effect of incorporation of the charged monomers on hydrogel charge, equilibrium swelling and release of model proteins was assessed both at low (10 mM HEPES) and physiological ionic strength (HEPES buffered saline, HBS). ▼ The therapeutic value of various targeted nanomedicines was evaluated in several inflammatory disorders, including cancer, while exploring which nanocarrier may be most suitable for a specific therapeutic application. First, an overview of drug targeting systems presently available for anti-inflammatory therapy was given. Then, the thesis fell apart in three sections: two sections diverging the therapeutic scope of several novel and established targeted nanomedicines to various inflammatory diseases and cancer, and a final section to bring these and other efforts together in the quest for the ‘optimal’ nanomedicine for a given application. The first section dealt with the preclinical evaluation of targeted dexamethasone-containing carriers in three different models of inflammatory disease. On the one hand, the therapeutic activity of dexamethasone encapsulated into the aqueous core of long-circulating liposomes was studied in mice with experimental autoimmune encephalomyelitis (EAE), a model for multiple sclerosis (MS), as well as in mice with dextran sodium sulfate (DSS)-induced colitis, a model for inflammatory bowel disease (IBD). Interestingly, the targeted delivery of dexamethasone using long-circulating liposomes was therapeutically efficacious in preclinical models of RA and MS, but not in IBD. On the other hand, a dexamethasone-delivery system based on core-crosslinked polymeric micelles demonstrated strong therapeutic efficacy in two preclinical models of rheumatoid arthritis (RA). The second section focused on the development of new nanomedicines derived from colchicine with vascular disrupting activity. The synthesis, liposomal encapsulation and in vitro characterization of two polymeric prodrugs of colchicine were described, and one of these prodrugs was selected for preclinical evaluation in tumor-bearing mice, to demonstrate a highly efficacious in vivo vascular disrupting activity of this new type of colchicine-derivatives. As part of the third and last section, which addressed the application-oriented search for an optimal nanocarrier system for drug targeting, three different nanocarriers of dexamethasone were evaluated in a comparative study to assess their therapeutic efficacy in two animal models of RA, providing deeper insights in the therapeutic efficacy of each of the systems. Then, to allow for the screening of different nanomedicines without requiring extensive usage of laboratory animals, an in vitro assay for predicting the in vivo circulation kinetics of liposomes was presented. Finally, the thesis concludes with a summarizing discussion regarding the findings presented in this thesis

▼ Tailorable Trimethyl Chitosans as Adjuvant for Intranasal Immunization Active vaccination has proven to be the most (cost) effective tool in the fight against infectious diseases. Nowadays, most vaccines are administered via parenteral injection. However, the risk of contaminated needles and need for trained personnel have risen interest in alternative immunization routes. As live attenuated vaccines raise considerable safety issues, (nasal) vaccine development now mainly focuses on purified, well-characterized antigenic proteins. However, these subunit vaccines are generally less immunogenic and need potent adjuvant (system)
s to elicit an adequate immune response. The use of muco-adhesive polymers like N,N,N-trimethylchitosan (TMC), a partially quaternized, water-soluble chitosan derivative, can enhance the immune responses against antigens. In this thesis novel synthetic methods are presented to obtain structural variants of TMC that allow tailorability of the degrees of quaternization (DQ) and acetylation (DAc) and exclude the introduction of other alterations such as O-methylation (DOM)and polymer chain scission. Additionally, thiol-moieties are introduced in a controllable manner to yield thiolated TMC. This tailorability of TMC provided the possibility to properly establish structure-activity-relationships in in vitro biological assays and in in vivo (nasal) vaccination studies. On Caco-2, cells O-methyl free TMCs demonstrated a larger decrease in trans-epithelial electrical resistance (TEER) than O-methylated TMCs while TMCs with a high DAc (~50%), were not able to open tight junctions. Also, with increasing DQ, an increase in cytotoxicity (MTT) and membrane permeability (LDH) was observed. On the other hand, TMCs with a high DAc (~50%) demonstrated no cell toxicity (MTT, LDH release) up to a concentration of 10 mg/ml. The extent of lysozyme-catalyzed degradation of TMC was highly dependent on the DAc; polymers with the highest DAc showed the largest decrease in molecular weight. Using Calu-3 cells, XTT and LDH cell viability tests showed a slight reduction in cytotoxicity for thiolated TMCs as compared to the non-thiolated polymers with similar DQs. The adjuvant properties of TMCs as nasal adjuvant in mice, using whole inactivated influenza virus as antigen, were strongly decreased by re-acetylation of TMC, whereas the DQ and DOM hardly affected the adjuvanticity of TMC. The inferior adjuvant effect of TMC-RA over that of TMC might be caused by a slightly lower stability of TMC-RA in the nasal cavity. In vitro experiments with human and murine dendritic cells indicated that the degree of N-acetylation is critical for the adjuvant effect of TMC on human DCs, but not on mice DCs. Finally, a covalently-stabilized polymeric nanocarrier system was introduced based upon the formation of intracellularly degradable disulfides between thiolated TMC (TMC-SH) and (oppositely charged) thiolated hyaluronic acid (HA-SH). apontada como uma estratégia eficiente para a formação inicial e
continuada de professores para a educação básica. Contudo, a
expansão dessa modalidade também vem acompanhada de críticas e
preocupações quanto à qualidade dessa formação. Uma das
problemáticas principais da EAD reside na mediação pedagógica, em
relação à díade aluno–mediador e/ou professor. Nesse sentido, esse
trabalho teve como objetivo investigar a concepção dos alunos de um
curso de especialização na modalidade a distância sobre a mediação
pedagógica, partindo das seguintes questões de estudo: Quais
atribuições os alunos delegam à mediação pedagógica? Quais as
contribuições da mediação pedagógica para a qualidade da formação
dos cursistas? Como as tecnologias da comunicação e informação
(TIC) potencializam a mediação pedagógica? A fim de compreender o
conceito de mediação, procuramos fazer uma pesquisa desse termo em
campos interdisciplinares, relacionando-o com os avanços das TICs
na educação a distância. Optamos, metodologicamente, por um estudo
de caso do Curso de Especialização Tecnologias em Educação – PUC
Rio. Para a produção de dados fizemos um recorte das respostas dos
cursistas sobre a mediação pedagógica exercida no decorrer do
curso, após aplicação de um questionário. As questões fechadas
foram analisadas com a ajuda do programa SPSS, e selecionou-se uma
questão aberta que foi trabalhada através da análise de conteúdo,
utilizando-se o programa Alceste. Os resultados provenientes das
questões fechadas mostraram uma grande aceitabilidade da atuação do
mediador pedagógico. Na análise qualitativa, o software Alceste
originou quatro classes, sendo posteriormente identificadas como:
estratégias pedagógicas de mediação e mediador assistente; mediador
tira-dúvidas; motivação ao aluno; formação do mediador pedagógico e
influências da mediação na atuação profissional do aluno. As
concepções dos cursistas revelaram o reconhecimento e a valorização
da mediação como essencial para que o aluno construa o conhecimento
esperado no processo de aprendizagem. Ficou evidenciada a
complexidade da prática profissional do mediador tendo, inclusive,

espaços profissionais e, entre suas possibilidades de uso, está seu
emprego no contexto educacional. As pesquisas para desenvolvimento
de tecnologias partem de um referencial ainda distante da realidade
educacional e com predominância de visões advindas do público
adulto. Nesse sentido, há espaço para o entendimento de como as
crianças experimentam tais tecnologias - impressão 3D (manufatura
aditiva), modelagem 3D e escaneamento 3D - no ambiente escolar da
Educação Infantil. A presente pesquisa busca investigar qual a
contribuição da experimentação das tecnologias 3D para a Educação
Infantil, objetivando abrir possibilidades para o entendimento das
formas de experimentação pelos alunos. A partir da leitura de
documentos governamentais e de teóricos da educação e do
desenvolvimento infantil, além do levantamento de outras
experiências semelhantes, foi estruturado um experimento composto
por três encontros, baseados no método de pesquisa-ação. O primeiro
encontro ocorreu no espaço não formal do museu - para apresentação
das tecnologias - e os outros dois no ambiente de sala de aula -
para a experimentação e reflexão sobre as tecnologias. Para o
experimento, foram selecionadas três turmas do último ano da
Educação Infantil da escola Curiosa Idade, além do Museu
Internacional de Arte Naif. Para a análise dos dados obtidos, foi
utilizada a abordagem da Sociolinguística Interacional. Foi
constatado que os alunos do último ano da Educação Infantil são
capazes de compreender todo o processo, do escaneamento 3D de
objetos, passando pela customização via modelagem 3D e chegando ao
objeto modificado impresso em 3D. Além disso, verificou-se que a
ludicidade e a criatividade são potencializadas pelo uso das
tecnologias 3D.
[en] The 3D technologies are ever more present in
professional spaces, and among their possibilities is their
employment in the educational context. The research for
technologies development starts from a referential still far from
the educational reality and with visions coming predominatly from
the perspective of adults. In this sense, there is room for
understanding how children experience such technologies - 3D
printing (additive manufacturing), 3D modeling and 3D scanning - in
the preschool environment of Educação Infantil (brazilian stage
similar to the Early Childhood Education). The present research
seeks to investigate the contribution of the 3D technologies
experimentation to Educação Infantil, aiming to open possibilities
for the understanding of the ways of experimentation by the
students. Having reviewed governmental documents and education and
child development theorists, besides surveying similar experiences,
an experiment was structured consisting of three meetings, based on

pedagógico das Tecnologias da Informação e Comunicação (TIC) têm
por objetivo promover a integração do uso das TIC nas práticas
pedagógicas. Com o intuito de contribuir para a análise dos
resultados alcançados por esses momentos de formação, este trabalho
estudou a relação entre a participação do professor nesses momentos
e as suas práticas com TIC. Investigamos a presença das tecnologias
nos momentos de formação e analisamos as políticas públicas de
disseminação das TIC nas escolas. Paralelamente, realizamos uma
pesquisa quantitativa com o objetivo de estudar a correlação entre
as variáveis latentes formação específica em TIC e uso pedagógico
de TIC pelos professores com os dados da TIC Educação 2014. Para
atribuirmos uma medida a essas variáveis latentes, construímos
quatro escalas utilizando a Teoria de Resposta ao Item (TRI). Duas
escalas de formação, uma que considera os momentos
institucionalizados e outra os informais dos quais os professores
participaram; e duas escalas de uso, uma considerando as atividades
com os alunos e outra as ações de preparação de aula. Para explicar
a variável dependente uso pedagógico de TIC, construímos um modelo
de regressão linear considerando como variáveis explicativas: as
variáveis de formação específica em TIC e as variáveis de controle.
Os resultados principais indicam que o uso pedagógico de TIC
(Atividade e Preparação) é influenciado positivamente pela formação
específica em TIC Institucionalizada e Informal, disciplina
específica em TIC na graduação, renda mensal familiar e titulação,
negativamente pela idade, e não é influenciado por curso específico
em TIC, sexo e cor/raça.
[en] Moments of professional training for the
pedagogical use of information and communication technologies (ICT)
aim to promote the integration of the use of ICT in pedagogical
practices. In order to contribute to the analysis of results
achieved by these training moments, this dissertation presents a
study on the relationship between teacher s participation in these
moments and his practices with ICT. We investigate the presence of
technology in formation and we analyzed public policies of
dissemination of ICT in schools. Meanwhile, we conducted a
quantitative research to study the correlation between those latent
variables: specific training in ICT and used ICT pedagogy on the
TIC Educação 2014 data basis. To assign a measure to latent
variables, we constructed four scales using the Item Response
Theory (IRT). Two of them are training scales: the first one
considers the institutionalized moments, while the second one takes
the informative information of the assistant teachers into account.
The other two are scales of use, one considering the activities
with students and the other the actions of preparing classes. In
order to explain the dependent variable use of ICT for pedagogy, we
composed a linear regression model including as explanatory
variables: the specific training variables in ICT and the control

discussões contemporâneas sobre abordagens às tecnologias da
tradução na formação universitária de tradutores profissionais.
Este estudo parte de uma análise de um afastamento percebido entre
ensino universitário e prática profissional no âmbito estrito da
formação de tradutores para investigar o papel do tratamento
dispensado às tecnologias da tradução nessa conjuntura. Para tanto,
como base teórica e metodológica, este trabalho usa postulados dos
Estudos da Tradução sobre o objeto de seus ramos descritivos e
aplicados e os estudos desenvolvidos pela pedagogia da tradução em
torno do que seriam competências tradutórias para, então, propor um
modo de inserção da tecnologia no processo de formação de
tradutores. É feita, então, uma inclusão das tecnologias da
tradução no contexto dos modernos Estudos de Ciência e Tecnologia
em seus aspectos descritivos - notadamente os trabalhos sobre
Ciência, Tecnologia e Sociedade (CTS) e a Teoria Ator-Rede (TAR) -
para verificar expectativas sobre processos e resultados da
tradução por causa da presença intensiva da tecnologia. Por fim,
com base nas inter-relações expostas pelo exame anterior, este
estudo sugere uma forma de abordagem às tecnologias da tradução na
universidade que contribua para valorizar a universidade, reduzir o
afastamento percebido entre ensino universitário e prática da
tradução e destacar as pesquisas acadêmicas da tradução como
protagonistas no âmbito das Humanidades Digitais.
[en] This work follows the contemporary discussions on
how to approach the translation technologies in the education of
professional translators. This study starts with an analysis of a
perceived distance between university education and professional
practice in the strict framework of translators training and
education, and then investigates the role that current approaches
to translation technology play in that scenario. Therefore, as a
theoretical and methodological basis, this work uses postulates
from Translation Studies on the object of their descriptive and
applied branches, and from the translation pedagogy studies on
translational competences, to propose a mode of inserting
technology in the translator education process. Then, the
translation technologies are included in the modern context of
Science and Technology Studies in their descriptive aspects -
notably the Science, Technology and Society (STS) studies, and the
Actor-Network Theory (ANT) approach - in order to check
expectations on translation processes and results associated with
the intensive presence of translation technologies. Finally, based
on the interrelations found during investigations, this study
suggests an approach to translation technology at the university
that would contribute to distinguish the university, reduce the
distance perceived between university education and translation
practice, and highlight the academic translation research as
protagonist within the Digital Humanities studies.
