▼ Chitosan (CH) is a biopolymer obtained from the deacetylation of chitin, a polysaccharide present in the exoskeleton of crustaceans and cell walls of fungi. Application of CH may be limited by its water-insolubility. It can be dissolved in acids and forms relatively high viscosity solutions. CH solutions have antioxidant and antimicrobial properties. CH penetration into shrimp may be facilitated by vacuum tumbling with low viscosity chitosan-sodium tripolyphosphate (CH-TPP) nanoparticle or water-soluble chitosan (WSC) solutions. It is expected that this would reduce lipid oxidation and microbial loads. In this study, CH-TPP and WSC solutions were developed and applied to shrimp, and the quality characteristics during frozen storage were evaluated. This research was conducted in two separate studies. In the first study, four treatment solutions were prepared: (1) a 1% acetic acid (AA) solution, (2) a CH in AA solution, (3) a sodium tripolyphosphate (TPP) in AA solution, and (4) a CH-TPP in AA solution. The solutions were sonicated and then sheared in an ultra-homogenizer to reduce particle size. In the second study, WSC was produced by enzymatic hydrolysis. Two treatment solutions were prepared: (1) a 0.1 M AA solution and (2) a WSC in distilled water (DW) solution. In both studies, fresh shrimp meat was separately vacuum tumbled with the solutions, cryogenically frozen, and evaluated for quality characteristics under frozen storage. Fresh shrimp meat tumbled with DW and fresh shrimp meat without tumbling (NT) were used as controls. Shrimp treated with CH, CH-TPP, and WSC had lower aerobic plate counts (APC) compared to other treatments after 120 days of storage at -20 °C. However, only the WSC treatment decreased yeast and mold counts (YMC) in the shrimp. CH, CH-TPP, and WSC treatments could aid in retention of color, texture, and moisture content of shrimp. Additionally, CH, CH-TPP, and WSC treatments produced the highest reduction in lipid oxidation compared to other treatments. This research indicated that CH, CH-TPP, and WSC solutions, combined with vacuum tumbling, can be effective at reducing APC and lipid oxidation in shrimp during frozen storage. As WSC can be dissolved in water, it may have greater application potential in seafood than CH or CH-TPP.

▼ The objective of this study was to evaluate the effects of non-meat ingredients, blade tenderization and vacuum-tumbling on the textural, visual and sensory characteristics of steaks from Biceps femoris, Supraspinatus, Triceps brachii long head, and Longissimus dorsi muscles packaged in high oxygen, modified atomosphere (MAP) system. United Department of Agriculture (USDA) Select muscles (n=72) from each cut were obtained from a commercial processor over three processing days. Muscles were aged for five days at 4ºC. Denuded muscles within a processing day were randomly assigned to one of 24 treatments. This study was a 2 x 4 x 3 factorial arrangement where treatments were control, injection (injected or non-injected), blade tenderization (0, 1, or 2 passes) and vacuum-tumbling (0, 5, 10 or 20 minutes). Injected muscles contained up to 10% of a brine containing 1.55% potassium lactate, 0.1% sodium diacetate, 0.3% sodium tripolyphosphate blend and 0.4% salt in the final product. Muscles were vacuumtumbled and blade tenderized sequentially after injection. Steaks from the muscles were stored in a high oxygen (80% O2, 20% CO2) MAP system for 0, 3, 7, 10 and 14 days at 2ºC. Steaks were evaluated for package purge (%), Warner-Bratzler shear force (kg), cook loss (%), cook time (min), pH, CIE L* a* b* color space values and trained color panel scores on each storage day. A trained descriptive attribute sensory panel evaluated steaks on day 1 only. Warner-Bratzler shear force (P<0.01) and trained sensory panel results (P<0.05) showed that the addition of non-meat ingredients improved tenderness in all four muscles. Sensory detectable connective tissue was lower (P<0.01) in injected steaks for all muscles except in Biceps femoris steaks. Injected steaks had higher pH (P<0.01) measurements than non-injected treatments in all muscles except the Triceps brachii long head. Neither blade tenderization nor vacuum-tumbling had consistent effects in all four muscles. Vacuum-tumbled Biceps femoris steaks had lower bitter flavor aromatics (P<0.05). In conclusion, enhancing beef with non-meat ingredients had the greatest impact on the quality attributes of high connective tissue cuts and Longissimus dorsi steaks. ▼ This thesis documents the successful development of a novel form of robotic motion, inspired partially from bio-mimicry and partially from a search for unknown forms of legged robotic motion. The novel motion developed from the research in this thesis, was created by implementing a tilting and falling action on the robot as a result of its leg movements to form a unique robotic gait. The tumbling gait was developed on a six legged robot that featured a tri-pedal stance, with the robot driving its legs in such a way as to unbalance the robot and force it to fall onto an opposing leg, creating a tumbling motion. To create the desired tumbling motion, several robotic concepts were investigated. The final robotic version used a series of leg actuations that induced a tumbling action on the robot. This was achieved by moving one of the three grounded legs towards the centre of the robot, causing the robot to tumble in that direction, giving a triangular footprint to the motion. After determining the method most suitable for motion and reviewing the current literature in this field, the robotic chassis design was created based around the manufacturing abilities of a plastics three dimensional (3D) rapid-prototyping printer. This design was cube-shaped with a single leg protruding from each face of the cube, giving a total of six legs. This cube design included three axes of symmetry, featuring a collinear pair of legs mounted on each of the robot’s axes. The cube design incorporated leg mounting, electronic board mounts, battery compartments and internal strengthening. Due to the complexity of the design and the high level of integration between the electronics and mechanics, ‘off-the-shelf’ components could not be used, and a complete electronics design was created, prototyped and built, comprising multiple circuit boards for the required power, processors and input-output devices. Considerable electronic development permitted a wireless controlled semi-autonomous robot mobilised by the combined use of twelve servo motors for evaluation of its gait. The electronic design also facilitated interchangeable electrical systems for expansion and continued development of its control and motion. An initial direction for the robot to follow was provided by a wireless controller to the onboard navigation system. The navigation system then performed mathematical calculations based on the robot’s triangular footprint, to determine which leg to move, in order to proceed in the required direction. The tri-pedal stance inherent in the robot’s footprint, allowed each of the robot’s steps to be directed toward one of three directions. It was found that the accuracy of following a given heading over time was dependant on the number of steps taken by the robot. The manually programmed leg movements for Spike’s gait were generated from a lookup table, and programmed into the robot’s processors, relating servo motor angular displacement to leg destination position for each orientation of the robot. ▼ Mobile robots that are able to move about and effectively negotiate their environment are attractive for a wide variety of applications. Such applications include surveillance, inspection, and mobile sensing where robots often present cost-effective alternatives to human labor. Other applications include those that are potentially hazardous to humans; examples of these include search and rescue, monitoring and maintenance of toxic environments, and planetary exploration. A vast majority of research into mobile robots has been limited to structured environments such as research labs, indoor office environments, industrial settings, maintained roads, etc. As the number of mobile robot applications grows, so does the need for such systems to be able to operate in unstructured (general) environments. Such environments often exhibit a wide variety of terrain including uneven surfaces and significant terrain irregularities. In some cases hazardous areas can be tactically avoided with careful path planning, but in general this is not always possible and obstacles must be negotiated directly. For these applications it is imperative that the robot exhibits a sufficient level of mobility to be able to perform required tasks. In addition to mobility requirements, many mobile robot applications are further constrained by limitations on physical size and/or cost. It is often the case that small (inexpensive) robots are preferable if not required. In general, however, it is the case that miniature mobile robots sacrifice mobility in exchange for their small size. Additionally, the increased design complexity of miniature systems often increases both design and manufacturing cost. In this thesis we present a relatively new and unexplored form of robotic locomotion called tumbling which addresses many of the aforementioned existing limitations of miniature mobile robots; the thesis is comprised of three main parts. In the first, tumbling and tumbling robots are defined and discussed in detail as well as other useful notation. Additionally, we present a classification of tumbling robots along with a catalog of existing designs to establish the state of the art. This treatment marks the first of its kind and establishes the first formal definitions with respect to tumbling locomotion for mobile robots. In the second, we examine terrainability of the class of serial multiply actuated tumbling robots by looking at the underlying principles of tumbling interactions with several idealized obstacles. Specifically, we derive configuration equations that relate terrainability to the parameters of an idealized tumbling robot. The results are supported through experimentation using the Adelopod, a physical tumbling robot developed as part of this thesis, over a variety of repeatable terrains. Finally, we conclude by examining the maneuverability for the class of serial multiply actuated tumbling robots and begin to address motion planning for such devices. ▼ Comminution or size reduction such as crushing and grinding is an important unit operationwidely used in many industries. However, current grinding technology is low-efficient andpower-intensive (Wills, 1992). Therefore, it is important that the grinding process is properlydesigned and grinding devices are operated at optimum operating conditions. To achieve thisaim, reasonably accurate models based on the basic physics and mechanics are required tounderstand the dynamic characteristics of grinding. Both mathematical method and numericalsimulations were conducted in tumbling ball mills to investigate the effects of grinding mediaand material, and operation conditions.For mathematical method, back-calculation method was used to reasonably predict thegrinding performance. It shows that large grinding ball (25mm) is preferred for particle beingbroken. However, the physical batch grinding test is time consuming.For numerical simulation, this work investigates the grinding process in tumbling ball millsusing a discrete element method (DEM) based model. Binary particles are used to representgrinding balls and ground powders, respectively. Three forms of energy inside mills, collisionenergy, dissipated energy and maximum impact energy, are investigated and linked toparticle breakage. By linking the energy information with population balance models, theevolution of product size with grinding time is predicted and the results are compared withthe experimental data from the literature. The results indicate that the collision energy is moredirectly linked with particle breakage and can predict the product size reasonably wellwithout adjustment. Other two energies can also be used for size prediction but the selectionfunction needs to be carefully calibrated. The effects of operation conditions such as rotationspeed and loadings of grinding balls and ground material are also investigated.The size and size distribution of grinding medium are two important variables in the grindingprocess. While some models have been developed to predict the optimum grinding mediumsize for improved grinding performance, these methods often require parameters determinedfrom experiments, which is time and capital consuming and also limits their applicability forgeneral use. This work performs a discrete element method (DEM) based study of grindingprocess in a tumbling ball mill. The effects of grinding medium/ore particle size ratio from4:1 to 10:1 are investigated. The micro-dynamic properties of particle flow, the maximumimpact energy among particles are analysed, in which it is found the average of the maximumimpact energy reaches the largest at the size ratio of 4:1. Two methods of mixture of grindingballs, same number and same mass of four sizes grinding balls, were selected to investigatetheir effect. According to recent findings, some species modify their environment in such a way that they facilitate the creation of new niches for other species to evolve to fill. Given the vast speciesdiversity of insects, the occurrence of such sequential radiation of species is likely common among herbivorous insects and the species that depend on them, many of them being insects as well. Herbivorous insects often have close associations with specific host plants and their preferences for mating and ovipositing on a specific host-plant species can reproductively isolate host-specific populations, facilitating speciation. Previous research by our laboratory has established that there are two distinct populations of thegall fly, Eurosta solidaginis (Tephritidae), which attack different species of goldenrods, Solidago altissima (Asteraceae) and S. gigantea. The gall fly’s host-associated differentiation is facilitating the divergence and potential speciation of twosubpopulations of the gall-boring beetle Mordellistena convicta (Mordellidae) by providing new resources (galls on stems of the galdenrods) for the gall-boring beetles. These beetles exist as two host-plant associated populations of inquilines that inhabit the galls induced by the gall fly. While our previous research has provided genetic and behavioral evidence for host-race formation, little is known about the role of their host plants in assortative mating and oviposition-site selection of the gall-boring beetles’ hostassociated populations. Volatile emissions from host plants can play a major role in assisting herbivores to locate their natal host plants and thus facilitate assortative mating and host-specific oviposition. The present study investigated the role of host-plant volatiles in host fidelity (mating on the host plant) and oviposition preference of M. convicta by measuring its behavioral responses to the host-plant volatile emissions using Y-tube olfactometers. In total, we tested behavioral responses of 615 beetles. Our resultsshow that M. convicta adults are attracted to their natal host galls (67% of S. altissima-emerging beetles and 70% of S. gigantea-emerging beetles) and avoid the alternate host galls (75% of S. altissima-emerging beetles and 66% of S. gigantea-emerging beetles),while showing no preference for, or avoidance of, ungalled plants from either species. This suggests that the gall beetles can orient to the volatile chemicals emitted by the galls and can potentially use them to identify suitable sites for mating and/or oviposition. Thus, host-associated mating and oviposition may play a role in the sequential speciation of the gall-boring beetle.

▼ Many living organisms have evolved to anticipate daily circadian cycles and changing seasons of their environment. In mammals, the suprachiasmatic nucleus (SCN) of the hypothalamus, a brain region of about 20,000 neurons, serves as the master circadian clock coordinating timing throughout the body and entraining to daily external light cycles. The remarkable precision of the SCN clock relies on intercellular signaling. In its absence, each SCN neuron and the SCN as a whole have significantly less stable oscillations. Though there are candidate signaling neuropeptides and anatomical surveys of the SCN, it is still unknown how the SCN as a whole responds to changes in the environment and regulates function in the body. We model the unstable oscillations in individual cells by developing a stochastic model based on the cell clock's gene regulatory network, then investigate the intercellular signaling properties of the SCN to understand its behavior as a whole. Though many existing deterministic models contain details of the gene regulation in the cell, their output has been compared to the behavior of the SCN as a whole, rather than to individual cells. Characterizing properties of individual cells such as period, phase, and synchronization is challenging due to their non-linear and unstable oscillations. We developed a wavelet analysis method to characterize cell behavior in biological experiments and compare with stochastic cell models. This analysis led to an examination of how period distributions could be influenced by stochastic fluctuations in a nonlinear cell oscillator model, and a hypothesis that the poor or strong oscillators observed in biological experiments could be a stochastic oscillator operating near a bifurcation point, between non-oscillatory and oscillatory conditions.It was observed in SCN tissue and in the SCN stochastic model that the oscillator is less likely to shift phase in response to a vasoactive intestinal polypeptide (VIP) dose at circadian time (CT4) than at other times. A reexamination of the behavior of the SCN as a whole, when modeled as linked stochastic oscillators, led to the hypothesis that the cells of the SCN synchronize to each other using a ``phase tumbling'' process. Our hypothesis is that the SCN synchronizes by its cells shifting with a wide phase distribution when they are perturbed at phases not near CT4. Rather than shifting in a deterministic manner, where all the cells stay synchronized and shift together to a new light schedule, they instead temporarily desynchronize then reorganize aligned to the new light/dark cycle. Within a few cycles the system can rapidly shift to a new light schedule. This rapid re-entrainment to both new light/dark and temperature schedules was confirmed in mice by first desynchronizing the SCN using a neuropeptide that has been considered a synchronizing agent, vasoactive intestinal polypeptide, or by a brief bright light exposure before exposing the animals to a new shifted schedule.Finally, since the behavior of the SCN as a whole may…

▼ The delivery of fusion material to the center of a laser inertial confinement engine happens by injecting a frozen hydrogen isotope mixture that resides inside a carefully crafted capsule called hohlraum. Because of the injection mechanism, the capsule necessarily has the shape of a modified short cylinder traveling along its axis of symmetry. The flow details and stability of this geometry are of significant importance to the reliable operation of the device. The gas inside the fusion chamber is very hot and the viscosity is relatively high, which for the typical injection velocities of the capsule results in a low Reynolds number flow. In this connection, a low Reynolds number compressible flow past a dynamically moving rigid short cylinder has been investigated using three-dimensional direct numerical simulation (DNS) as a model of the real capsule. A Cartesian-based novel embedded geometry method for compressible fluid-structure interaction problems is developed and coupled with a low-numerical-dissipation compressible flow solver to perform simulations in a parallel multiprocessor environment. In this method, the surface of the cylinder is uniquely identified by a zero level set separating the fluid and solid region. A PDE-based global extension technique inspired by analytical continuation is used to smoothly propagate the surface boundary conditions inside the fictitious solid domain. This approach ensures smooth and noise-free reconstruction of pressure and viscous stresses on the surface of the solid body and utilization of a global high-order spatial discretization scheme due to the smooth nature of the flow quantities at each time step stage. The translational and rotational dynamics of the cylinder by modeling the 6-degree of freedom (DoF) motion employ a non-singular quaternion-based framework. The longitudinal stability of the cylinder is affected by the variation in angle of attack, aspect ratio and Reynolds number. The present work focuses on Reynolds number, Re = 60 and a Mach number, M = 0.25 to closely mimic the flow conditions inside an inertial confinement fusion chamber. A critical angle of attack has been identified beyond which the short cylinder will start to tumble based on a set of stationary simulations. ▼ Dielectrophoresis (DEP) has been a subject of active research in the past decades and has shown promising applications in Lab-on-Chip devices. Currently researchers use the point dipole method to predict the movement of particles under DEP and guide their experimental designs. For studying the interaction between particles, the Maxwell Stress Tensor (MST) method has been widely used and treated as providing the most robust and accurate solution. By examining the derivation processes, it became clear that both methods have inherent limitations and will yield incorrect results in certain occasions. To overcome these limitations and advance the theory of DEP, a new numerical approach based on volumetric-integration has been established. The new method has been proved to be valid in quantifying the DEP forces with both homogeneous and non-homogeneous particles as well as particle-particle interaction through comparison with the other two methods. Based on the new method, a new model characterizing the structure of electric double layer (EDL) was developed to explain the crossover behavior of nanoparticles in medium. For bioengineering applications, this new method has been further expanded to construct a complete cell model. The cell model not only captures the common crossover behavior exhibited by cells, it also explains why cells would initiate self-rotation under DEP, a phenomenon we first observed in our experiments. To take a step further, the new method has also been applied to investigate the interaction between multiple particles. In particular, this new method has been proved to be powerful in elucidating the underlying mechanism of the tumbling motion of pearl chains in a flow condition as we observed in our experiments. Moreover, it also helps shed some new insight into the formation of different alignments and configurations of ellipsoidal particles. Finally, with the consideration of the Faradic current from water electrolysis and effect of pH, a new model has been developed to explain the causes for the intriguing flow reversal phenomenon commonly observed (but not at all understood) in AC-electroosmosis (ACEO) with reasonable outcomes. ▼ Marination by vacuum tumbling is commonly practiced in the meat industry; however, the optimal temperature of the process to maximize marinade absorption and retention is yet to be defined. Furthermore, the role of marinade functional ingredients and pH of the marinade in alleviating problems with pink color in cooked meat and water retention during cooking is yet to be elucidated. For denaturation model study, myoglobin pigments from chicken gizzards were extracted. Observed results from the model study were validated in whole muscle and comminuted chicken meat. In addition, temperature of vacuum tumbling operation was optimized with respect to marinade pick- up, cook yield and expressible moisture. Also, trivalent Eu+3 was used to trace the penetration of marinade in chicken breast meat. Results showed that increasing concentration of salt and pH in marinade increased the persistence of the pink color in cooked meat. Furthermore, degree of denaturation of myoglobin pigments was not a determining factor for cooked meat color under the condition of higher ORP values in meat products. On the other hand, temperature of marination was found to be a significant factor in marinade penetration and retention in a vacuum tumbling process. Higher marination temperature promoted deeper penetration of marinade in the meat as traced by trivalent Eu+3. Consequently, marinade pick-up was found at higher temperature. However, cook yield was found highest when marination process was initially at a higher temperature followed by lowering to near refrigeration temperature. Amount of salt soluble protein extracted at lower temperature was higher compared to higher temperature of marination that may be a factor in higher marinade retention on cooking In conclusion, pink color incidence in cooked meat is dependent to the ORP value of meat during and after cooking. Pink color can be induced in a product that has the cooked meat color by altering the ORP by addition of a reducing agent. In addition, Eu+3 tracing of marinade penetration in meat showed marinade to be only near the surface of the chicken breast meat even at the elevated temperature of vacuum tumbling.

Le marinage et le saumurage améliorent la durée de conservation, la tendreté et la jutosité de la viande et du poisson. En complément, un malaxage ou « barattage » est souvent appliqué aux pièces de viande préalablement à la cuisson. Le but principal de cette étude était d’évaluer l’impact des conditions de traitement sur les modifications biochimiques et structurales du tissu musculaire ; une grande partie concerne le malaxage. Les essais relatifs au marinage ont permis de fixer le pH et la teneur en NaCl de petits échantillons de Semitendinosus de bœuf respectivement à 6,5, 5,4 ou 4,3 et à 0,9 ou 2 % en masse ; une ANOVA a révélé les effets de ces 2 paramètres sur la taille des cellules et des espaces extracellulaires, l’oxydation des lipides et des protéines et l’hydrophobie de surface de ces dernières. Un simulateur de saumurage-malaxage conçu par le laboratoire a permis d’imposer des successions de déformations contrôlées (nombre de 350 à 2500, taux de compression de 10 à 30%) à des muscles Semimembranosus (SM) ou Rectus femoris (RF) de porc. Des traitements mécaniques semblables à ceux existant dans des barattes industrielles de tailles différentes ont ainsi été reproduits. Les principales conclusions sont : (1) le malaxage augmente nettement la diffusivité apparente du NaCl, d’une part, en altérant la structure tissulaire (+ 20 %), et d’autre part, en induisant une convection s’ajoutant à la diffusion (+200 %) ; (2) le traitement mécanique entraine en lui-même une augmentation de la solubilité des protéines, connue pour moduler les qualités des viandes transformées, de 20 à 50% par rapport à des échantillons non malaxés, qu’ils soient salés ou non ; (3) il entraine aussi une augmentation modérée de l’hydrophobie des protéines ; (4) la dégradation de l’endomysium, servant d’indicateur des modifications structurales, est plus marquée au milieu qu’en périphérie du muscle malaxé et d’autant plus importante que le traitement mécanique est fort et long.

Marinating and brining improve shelf-life, tenderness and juiciness of meat and fish. As a supplement massaging or tumbling is often applied to meat pieces before cooking. The main aim of this study was to assess the impacts of the processing conditions on the biochemical and structural changes in the meat tissue; a great part is focused on massaging. In the marinating trials the pH and NaCl content of thin samples of beef Semimembranosus muscle were set at 6.5, 5.4 or 4.3 and at 0,9 or 2 % (w/w), respectively; an ANOVA have shown the effect of these two parameters on the cells and extra cellular space sizes, the oxidation of lipids and proteins and the protein surface hydrophobicity. A brining-massaging simulator built by the laboratory was used to apply controlled successions of deformations (number from 350 to 2500, compression ratio from 10 to 30 %) to Semimembranosus (SM) ou Rectus femoris (RF) pork muscles. ▼ The purpose of this study was to compare the effects of the command and reciprocal teaching style on the tumbling skill acquisition of college students. Fifty-one undergraduate students from Virginia State University in Petersburg, Virginia served as subjects. Subjects were pretested to assess their performance on twelve tumbling skills. Two raters scored videotaped performances utilizing specific criterion established for each skill. A twelve item tumbling skill test devised by the investigator was used for all testing. Group I subjects (N_c = 25) were instructed utilizing the command style of teaching and Group II subjects (N_r = 26) the reciprocal style. The same instructor taught both groups the tumbling skills for twelve class periods of fifty minutes each. The instructional period lasted six weeks. At the end of the experimental period, posttest performances were once again videotaped and evaluated. Mean scores for each groups' performances were analyzed to determine the degree of skill improvement and to test for any difference that might have been attributed to treatment effects. One way Analyses of Variance were applied to the data using the .05 level of confidence for significance. As a result of the study the following conclusions were reached: (1) both command and reciprocal teaching styles promote tumbling skill acquisition of college age students in beginning tumbling; (2) neither teaching style used in the study is more effective than the other in promoting tumbling skill acquisition for college students over a six week instructional period. On the basis of the data found in this study, it is apparent that college students are capable of teaching each other beginning tumbling skills. The non-significant differences between treatment group performances tends to suggest that practitioners could utilize the reciprocal style equally effectively as the command style while at the same time furthering each student's ability to learn in alternative modes.
