Este estudo teve por objetivo avaliar a influência de sessões de treinamento hipóxico intermitente (THI) sobre parâmetros hematológicos, capacidade aeróbica e o desempenho de corrida em condições de hipoxia e normoxia. Fizeram parte do grupo de estudo 9 atletas voluntários, sexo masculino (idade média 23 anos - DP 3,6), praticantes de pentatlo militar. Os indivíduos foram submetidos a pré e pós-testes de esforço máximo em normoxia e hipoxia 13% O2 (3.800 m de altitude). Analisou-se hemograma, freqüência cardíaca, concentração sangüínea de lactato [lact], saturação de oxigênio na hemoglobina, Índice de Percepção de Esforço e variáveis ergoespirométricas. O tratamento foi composto por 29 sessões de THI, concentração de 13% de oxigênio (1 hora, 5 vezes por semana). As análises estatísticas, feitas através do teste de Wilcoxon, apresentaram diminuição significativa no número de eritrócitos e CHCM (p <0,05). Não foi constatada alteração da [lact], FC e VO2 máx. em normoxia. Porém, houve diminuição significativa da [lact] em diferentes velocidades de corrida em hipoxia (5,4, 7,2, 9,0, 10,8, 12,6 km/h / p<0,05). Observou-se também aumento significativo na distância de corrida em normoxia e hipoxia (p<0,05) e diminuição do tempo de corrida em hipoxia. Conclui-se que o THI, na dosagem empregada neste estudo, foi eficiente para proporcionar aumento de performance em ambas as situações, apesar da significativa redução no número de eritrócitos e da pequena queda do hematócrito. O resultado em hipoxia demonstra ainda, que o método pode ser usado, ao nível do mar, como alternativa de preparação para competições em altitude.

The goal of the present study aimed at the assessment of the influences of intermittent hypoxic training (IHT) sessions on haematological parameters, aerobic capacity (AC), and running performance (RP), under hypoxic and normoxic conditions. The group of study was constituted by nine (09) volunteers of Military Pentathlon Athletes, male sex, with mean ages of 23 years old (23 3,6). These athletes underwent (pre-) and post-testing measurements of maximum effort in normoxyc and hypoxic conditions - 13% O2 (simulated altitude 3.800 meters). In the course of the research work hemograms, cardiac frequency, blood lactate concentration [lact], haemoglobin oxygen saturation, the index of perception of effort and ergospirometry variables were fully measured and analysed. The assessments were done in twenty-nine (29) sessions of IHT, with 13% of oxygen concentration (one hour, five times a week). The statistics analyses using the Wilcoxon test showed a significant decrease in the number of erythrocytes and MCHC (p<0,05). No changes were noticed in the blood lactate concentration [lact], cardiac frequency (FC) and in the maximum VO2 in normoxyc conditions. However, there was an expressive reduction of the [lact] in different running velocities in hypoxic (5, 4, 7, 2, 9, 0, 10, 8, 12,6 km/h / p<0,05). ▼ The aim of this study was to evaluate the effects of heat stress on the metabolic and endocrine status in cows. The experiment was performed on 20 Holstein Friesian cows from lactation day 18 to 45 during the summer period for a duration of 37 days. During the experiment the „hourly“ heat indices (THI) and thereon the values of the average morning (from 10PM the previous day to 9AM of the current day), afternoon (10AM to 9PM of the current day) and overall daily THI were determined. Blood sampling and trias readings were recorded on the 1st, 2nd, 8th, 11th, 14th, 18th, 25th, 29th and 37th day of the experiment, in the morning and afternoon periods. According to the values for hourly THI the entire experimental period was divided into three periods: period A in which the cows were exposed to a severe heat stress (THI ≥ 78) at least for 7 hours during a 24h period (1st, 8th, 14th and 37th day of the experiment); period B during which the cows were exposed to moderate heat stress (72 ≥ THI ≤ 78) at least for 7 hours during a 24h period (2nd, 18th, and 29th day of the experiment), period C during which the cows were exposed to heat stress (THI ≤ 72) during 24h (11th and 25th experimental day).The average daily THI for period A (73,25 ± 0,89) was significantly higher (p<0.01, individually) compared with period B (71.45 ± 0.96) and period C (65.41±2.09), during period B THI was significantly higher compared with period C (p<0.01). The daily feed intake was by 2.9 and 1.74 kg DM feed less in cows exposed to heat stress compared with the period in which the cows were kept under optimal ambient conditions. In the period with optimal environmental temperature conditions the average milk yield was significantly higher (43.08 ± 5.15 l), than in conditions of moderate (41.96 ± 5.51) and severe heat stress (39.47 ± 5.15). However, this decrease was not in unison with the quantity of not eaten feed. Hyperthermia (39.72 ± 0.47°C) and tahipnoa (86.60±6.39 n/min) were recorded only during the afternoon hours in the cows exposed to severe heat stress and can be used as reliable clinical indicators of heat stress. Indicators of arising heat stress are the changes in blood electrochemical reaction (alkalosis), also. Cortisol concentration was increased in conditions of moderate (10,44±3,07 nmol/L) and severe heat stress (10,44±3,07 nmol/L) compared with the period with optimal ambient temperature (6,41±2,11 nmol/L). The differences in the concentrations of cortisol were significant between all tested periods (p<0,01, individually). The intensive effect of heat stress had an inhibitory effect on the activity of the thyroid as the concentration of T4 measured during severe heat stress (48,44±12,38 nmol/L) was significantly lower compared to the other two periods (59,20±17,51 nmol/L for period B and 57,62±12,48 nmol/L for period C). Patologija i terapija životinja - Bolesti papkara / Clinical pathology and therapy of animals - Farm animal diseases

Cilj ovog rada bijo je da se ispita uticaj toplotnog stresa na metabolički i endokrini status krava. Ogled je izveden na 20 krava Holštajn – frizijske rase od 18. do 45. dana laktacije tokom letnjeg perioda u trajanju od 37 dana. Tokom perioda izvođenja ogleda određivana je vrednost „satnih“ toplotnih indeksa (THI) a zatim izračunata vrednost prosečnih jutarnjih (od 22 h prethodnog do 9 h tekućeg dana), popodnevnih (od 10 h do 21 h tekućeg dana) i celodnevnih THI. Uzorkovanje krvi i određivanje trijasa izvršeno je 1., 2., 8., 11., 14., 18., 25., 29. i 37. dana ogleda, u jutarnjem i popodnevnom periodu. Na osnovu vrednosti satnih THI ceo ogledni period je podeljen na tri perioda: period A u kome su krave bile izložene izrazitom toplotnom stresu (THI ≥ 78) najmanje 7 sati u toku 24 sata (1., 8., 14., i 37. dan ogleda); period B u kome su krave bile izložene umerenom toplotnom stresu (72 ≥ THI ≤ 78) najmanje 7 sati u toku 24 sata (2., 18., i 29. dan ogleda); period C u kome krave nisu bile izložene toplotnom stresu (THI ≤ 72) u toku 24 sata (11. i 25. dana ogleda). Prosečni dnevni THI u periodu A (73,25 ± 0,89) je bio značajno veći (p<0,01, pojedinačno) u odnosu na period B (71,45 ± 0,96) i period C (65,41±2,09). THI u periodu B je bio značajno viši nego u periodu C (p<0,01). Dnevni unos hrane je bio manji kod krava tokom perioda izloženosti toplotnom stresu za 2,9 odnosno 1,74 kg SM obroka u odnosu na period kada su bile u optimalnim ambijentalnim uslovima držanja. U periodu optimalnih ambijentalnih uslova temperature prosečna dnevna mlečnost je bila značajno viša (43,08 ± 5,15 L), nego u uslovima umerenog (41,96 ± 5,51 L) odnosno izrazitog toplotnog stresa (39,47 ± 5,15 L) ali ovo smanjenje mlečnosti nije bilo u skladu sa količinom nepojedene hrane. Hipertermija (39,72 ± 0,47 °C) i tahipnoja (86,60±6,39 n/minut) kod krava su ustanovljene samo u popodnevnim časovima i mogu da posluže kao pouzdani klinički poklazatelj toplotnog stresa. Pokazatelji razvoja toplotnog stresa su i promene vrednosti elektrohemijske reakcije krvi (alkaloza). Koncentracija koritzola je bila povišena u uslovima umerenog (10,44±3,07 nmol/L) i izrazitog topolotnog stresa (8,74±2,49 nmol/L) u odnosu na period sa optimalnim ambijentalnim temperaturnim uslovima (6,41±2,11 nmol/L), pri čemu su razlike u koncentracijama ovog hormona bile značajne između svih ispitivanih perioda (p<0,01, pojedinačno)...

▼ Abstract: Trinh T. Minh-ha is a filmmaker and Vietnamese intellectual, rooted in the United States since the 1970s has multidisciplinary academic education with studies in musical composition, ethnomusicology and comparative literature. In recent decades it has worked prominently in various fields of artistic production and also the academic production. Held eight films, including medium and feature films, he has set up five multimedia installations and published fourteen books, including works of art, collections of scripts and interviews and theoretical essays. His films provoke the conventions of different filmic cornfields, moving in ranges between documentary, ethnographic film, experimental cinema and narrative cinema. In its production, both intellectual and filmic, we find issues that cross from one field to another, subverting the boundaries and creating intersections between theory and practice in order to question the places determined between knowledge, relating the arts and innovative way science . His topics of interest revolve around the forms of representation of cultural difference, the female condition and identity issues. Our research focuses on his films proposing a dual relationship of movement: a first that comes from theory toward the movies, to investigate how innovative strategies used in your movie dialogue with social theory, to think other milestones in the relationship between cinema and anthropology. The second part of the film toward the film theory, specifically the theory dedicated to documentary cinema, to analyze how his films can help to expand the debate on the cinematic domain by offering unique and challenging contributions from the essential pillars of this kind of cinema, which are those linked to filming strategies and the relationship between who films and who is filmed. We understand his films as exemplary cases that allow us to think the documentary film us in his poetic fullness and contribute in an original way in relation to what may be the great documentary tradition of contribution to the cinema, the ethical question. ▼ Vietnamese consider Trinh Hoai Duc as the nineteenth century's most outstanding founding minister of Nguyen Dynasty . His road to fame went very well. He possessed a perfect character and was loved by the Emperor of Nguyen Dynasty and admired by the monarch. Aside from being grateful and proud of Trinh Hoai Duc, people in the southeast and Dong Nai regions regarded Trinh Hoai Duc as a famous Vietnamese Chinese official. At that time, many neighboring countries and regions in the southeast Asia had initially entered the colonial era. This led to the strong economical as well as social changes and threats in the local areas of Vietnam. During that period, two countries, China and Vietnam, had similar situations at home and abroad. Trinh Hoai Duc was sent to China as an ambassador. In the âan Trai Thi Tap-Quan Quang Tap,â he described the figures he made contact with in the seven provinces, lyrics, scenery and religion, thus showed his diplomatic skills. In the âCan Trai Thi Tap-Kha Di Tap,â the topics of scenery and making friends are also covered. Being an ambassador to China, Trinh Hoai Duc recorded the social situation at that time, his homesickness and his ambassadorship in China in the forms of poetry, paintings and songs. He not only provided precious information to the descendants in future generations, but expanded their knowledge and visions. He also offered useful references in the field of literature. From the above reasons, this paper carried out the data collection, archiving, processing, and elaborating outline followed by discussion, and finally determining the direction and the motivation to study writing. Trinh Hoai Ducâs âCan Trai Thi Tapâ poetry as based on ancient Chinese language, can hardly be understood by Vietnamese scholars who are proficient in the Chinese language, let alone the ordinary Vietnamese. Some scholars of ancient Chinese, can only make Chinese and Vietnamese phonetic research but cannot publish book officially publicly offering its content for public readers. Very few Vietnamese scholars studied such poetry, and did not do in-depth research, the contents of which had only a few issues initially proposed, and lacked depth of academic discussion. To allow more readers to read the content of Trinh Hoai Ducâs work , and with more perspective to improve the content, this paper was created. The scope of this paper focuses mainly on researching the content of âCan Trai Thi Tapâ. First, besides the authorâs relevant introduction, this paper starts from âCan Trai Thi Tapâ by understanding Trinh Hoai Ducâs literatureâs status and influence in the south of his hometown. Second, from "Can Trai Thi Tap" sequentially ordered as âThoi Thuc Truy Bienâ, âQuan Quang Tapâ, âKha Di Tapâ, Trinh Hoai Ducâs main topic on poetry and song are progressively analyzed. ▼ Production loss due to heat stress is a major concern in the livestock industry. Dairy cattle are especially susceptible to heat stress. Milk production loss due to heat stress accounted for about 1.2 billion dollars in 2010 (Lundeen, 2014). Heat stress occurs when combinations of environmental parameters (temperature, airspeed, relative humidity, etc) reach levels where the animal struggles to release internally produced heat. Implementing mitigation strategies to reduce heat stress has been a crucial need as dairy housing has transitioned from pasture to indoor housing systems. Currently, limited recommendations exist directing producers towards implementing one cooling system over another, specific to the climate in their region. In order to maximize production, producers need the most optimal cooling system in their operation in order to reduce heat stress. To assist producers in heat stress mitigation decision making, a thermal interaction model was developed to quantify heat dissipation from a dairy cow’s core to her surrounding environment using procedures and parameters from published thermal balance models. Environmental input parameters used for the model were taken from typical meteorological year (TMY3) data sets. The objectives of this research were to: (i) analyze the thermal environment’s ability to reduce heat stress in dairy cattle in selected regions using TMY3 data, (ii) model Holstein cattle subjected to evaporative cooling + airspeed, and direct water sprinkling + airspeed cooling systems by region, (iii) create a universal barn/cooling system model to apply to selected regions with given TMY3 data inputs, and (iv) develop contour maps with optimal cooling system recommendations throughout the United States. A thermoregulation model was developed combining equations from previous models to analyze a cow in her ambient environment in a barn to determine if she is heat stressed using various cooling strategies. The model was tested in two stations in California (SN:723890) and Wisconsin (SN:726435). The model’s predictions were within one standard deviation of field data. Once the model was validated, the model was ran for all 215 TMY3 Class 1 stations and contour maps of the U.S. were created for producers to determine which cooling strategy is the most economical in their region. The results from this work show that CCI is a better thermal index than using THI and that the MVCE and LVCE cooling strategies are the most economical cooling strategy for dairy cattle housing when the milk price is fixed at 0.363 kg-1, water price is fixed at 0.0015 gallon-1 and energy cost is fixed at 0.12 kW-hr-1. Multiple maps were made for each of the eight cooling strategies that producers can use with a developed equation to determine which cooling strategy is the most economical for their region.

▼ Two strategies to reduce impact of heat stress on high producing dairy cows were examined. The first was to recalculate the temperature-humidity index (THI) using high producing dairy cows under diurnal summer conditions. This re-evaluation confirmed that current THI values underestimate the severity of heat stress levels. Therefore, cooling of dairy cattle during warm summer months should begin at a THI of 68. Previously, a THI equal to or greater than 72 has been used to define onset of heat stress. This study demonstrated that a THI greater than or equal to 68 is sufficient to increase body heat storage, respiration rate, skin evaporative heat loss, declines in feed intake and milk yield. A second objective involved three studies carried out to evaluate use of niacin in dairy cow rations to improve evaporative heat loss and resistance to heat stress. Niacin is known to cause intense vasodilation in human and lab species. We hypothesized that increasing vasodilation would improve evaporative heat loss in dairy cows. In the first niacin study, supplementation of lactating dairy cows with an encapsulated rumen by-pass form of niacin (NIASHURE™; Balchem Corporation, New Hampton, NY) at a dose of 12 g/d proved effective in alleviating some affects of heat stress during mild thermal stress. We hypothesized that encapsulated niacin would induce vasodilation effects documented in humans and lab animals increasing evaporative heat loss. Past research demonstrated that the possible mechanism for vasodilation affects seen by niacin were most likely due to prostaglandin D secretions. Niacin may act through increased prostaglandin D and E production and secretion by Langerhans cells which then act upon vascular endothelial prostaglandin D receptors to increase vasodilation. No studies have evaluated impact of encapsulated niacin on milk yield and composition during periods of thermal stress under commercial dairy conditions. The objective of the last study was to examine the effects of encapsulated niacin during heat stress on milk production and composition as well as core body temperatures under commercial conditions. Les cellules du système immunitaire sont capables de reconnaître et d'éliminer les cellules cancéreuses prévenant ainsi l'apparition de cancers. Parmi celles-ci, l'activité antitumorale est principalement attribuée aux lymphocytes T CD4 helper de type 1 (Thl). Les lymphocytes CD4 sont activés lors de la reconnaissance d'un antigène (Ag) de tumeur présenté par le complexe majeur d'histocompatibilité de classe II (CMH-II). Ils possèdent des propriétés cytotoxiques propres et activent les autres cellules immunitaires. Dans un premier temps, nous nous somme intéressés au mécanisme de présentation sur le CMH-II de la télomérase (hTERT). La protéine hTERT est capable d'interagir avec les HSPGs facilitant ainsi son internalisation par les DC. Elle emprunte ensuite les voies endolysosomale et cytosolique pour générer des peptides nommés UCP présentés dans le contexte HLA-DR. Cette découverte soutient son utilisation en immunothérapie associée aux chimiothérapies. Nous avons ensuite identifié quatre peptides dérivés de hTERT restreints HLA-DP4 puis comparé leur immunogénicité avec les UCP. Cette analyse a mis en évidence la supériorité des UCP en termes d'immunoprévalence et d'immunodominance. Enfin, nous avons étudié l'impact de l'acquisition d'une résistance à l'oxaliplatine sur le profil antigénique de lignées tumorales de cancers colorectaux (CCR). L'évaluation des réponses immunitaires de patients atteints de CCR nous a permis d'identifier des peptides immunogènes dérivés d'Ag surexprimés après une exposition à l'oxaliplatine. En conclusion, ces travaux pourront participer à l'amélioration des stratégies d'immunothérapie et d'immunomonitoring ciblant les lymphocytes CD4 Thl.

Immune cells are able to recognize and eliminate cancer cells to prevent from cancer development. Among them, antitumoral activity is mainly attributed to CD4 T helper 1 (Thl) cells. CD4 Thl cells are activated upon recognition of tumor antigen presented on the Major Histocompatibility Complex class II (MHC-II) molecules. These cells possess their own cytotoxic capacities and activate other immune cells. Firstly, we were interested in the mechanism of presentation of the catalytic subunit of telomerase (hTERT) which is an attractive tumor antigen target for immunotherapies. hTERT protein can interact with cell surface HSPGs facilitating its internalization by DC. Then, hTERT uses thé endo-lysosomal and cytosolic proteolysis pathways to generale immunogenic peptides named UCP (Universal Cancer Peptide) presented in HLA-DR context. This discovery is an additional argument in favor of using hTERT as a target for cancer immunotherapies. Then, we identified four novel hTERT-derived peptides presented by HLA-DP4 and compared their immunogenicity with UCP. This analysis highlighted the superiority of UCP in term of immunoprevalence and immunodominance. O estresse calórico em gado leiteiro especializado torna-se um fator limitante para a produção de leite no estado do Rio Grande do Sul durante os meses de maior índice temperatura-umidade. O impacto fisiológico sobre o animal tem repercussão não somente sobre o metabolismo animal, mas também na quantidade e na qualidade do leite produzido. Não existem estudos prévios sobre o grau de impacto que o estresse por calor possa causar em gado da raça Holandesa no Rio Grande do Sul com enfoque no metabolismo e na qualidade do leite. O presente projeto estudou as alterações que o estresse calórico ocasiona em vacas leiteiras especializadas sobre indicadores clínicos, produtivos e metabólicos. Um total de 450 observações foram realizadas durante dois anos consecutivos em uma fazenda leiteira de alta produção da região do Planalto Médio do Rio Grande do Sul. O grau de estresse calórico foi considerado quando o índice temperatura-umidade (THI) ficou acima de 80. O estresse calórico foi evidente sobre os indicadores clínicos (temperatura corporal e frequência respiratória) e causou redução de 17% da produção de leite, bem como diminuição na quantidade de lactose e aumento na CCS, porém sem afetar o teor de gordura nem de proteína. Os indicadores metabólicos mais afetados foram proteínas totais, albumina e ureia, com respostas compatíveis com grau de desidratação leve, glicose com aumento atribuído à maior secreção de cortisol, e colesterol com aumento atribuído à menor secreção de tiroxina, em animais em estresse por calor. Lactato foi menor em animais estressados, sendo seu mecanismo objeto de futuros estudos. Ficou evidente uma condição de alcalose respiratória e a resposta compensatória de acidose metabólica. Conclui-se que o estresse calórico nos animais estudados repercute em mudanças clínicas, produtivas, metabólicas e ácido-básicas.

Heat stress is a limiting factor for milk production in high yielding dairy cows in Rio Grande do Sul (Brazil) during the months with the highest temperature-humidity index (THI). The physiological impact caused by high THI has not only consequences in the animal’s metabolism, but also in the animal’s milk yield and milk composition (quality). To our knowledge, there are no previous studies that related the impact of heat stress on metabolism and milk composition of high yielding Holstein cows in RG. The objective of the present study was to evaluate the effects of heat stress on clinical, productive, and metabolic parameters in high yielding dairy cows. A total of 450 observations were taken during 2 consecutive years in a commercial dairy farm in the RG’s plateau region. Heat stress was characterized as a THI greater than 80. Sono implicati nel controllo di proliferazione, differenziazione e sopravvivenza cellulare ma anche di fenomeni legati alla morte, tra cui l’apoptosi. Ceramide (Cer), alla base degli sfingolipidi complessi, può andare incontro a deacilazione generando sfingosina e sfingosina-1-fosfato (S1P). Quest’ultimo è considerato un mediatore di sopravvivenza e proliferazione cellulare, in opposizione a Cer, che promuove arresto del ciclo ed apoptosi. La degenerazione retinica ed, in particolare la Retinite Pigmentosa (RP) sono associate all’accumulo di Cer ed all’induzione di morte cellulare. L’inibizione della sintesi e dell’accumulo di Cer, inattivando la serina palmitoiltransferasi (SPT) attraverso l’utilizzo di Miriocina, è stata dimostrata essere coinvolta nel recupero morfologico e funzionale dei fotorecettori retinici, in particolare dei coni, in un modello murino di RP (mutante rd10). Scopo: Il nostro scopo è di intervenire sul metabolismo degli sfingolipidi per ridurre il danno di fotorecettori retinici, attarveso l’utilizzo di una linea cellulare di fotorecettori coni. In particolare, promuovendo l’aumento di S1P intracellulare, tramite l’inibizione dell’enzima S1P liasi, e la riduzione di Cer attraverso l’inattivazione di SPT. Metodi: le cellule murine 661W, una linea di fotorecettori (coni), sono state trattate con 2-acetil-4(5)-(1(R),2(S),3(R),4-tetraidrobutil)-imidazolo (THI) 75µM, un inibitore dell’enzima S1Pliasi, per 2 ore; quindi, le cellule sono state sottoposte a deprivazione di siero e trattate con H2O2 1mM per differenti lassi di tempo. La curva di crescita di queste cellule è stata determinata attraverso il saggio MTT e la vitalità con Typan blue. Il saggio TUNEL e il FRAP test sono stati impiegati per verificare, rispettivamente, il grado di apoptosi e il potere antiossidante intrinseco dei fotorecettori. I contenuti intracellulari di sfingolipidi sono stati misurati attraverso l’analisi LC-MS. La tecnica del Western blotting, attraverso l’impiego di anticorpi specifici, è stata utilizzata per valutare la fosforilazione di ERK1/2 e Akt/PKB, il rapporto tra Bcl-2/Bax e l’espressione proteica di Nrf2. Attraverso Real time-PCR, le modulazioni nell’espressione dei trascritti di HO-1 e dei recettori di S1P sono stati valutati in seguito a stress ossidativo e trattamento con THI. S1P esogeno (100nM) e Miriocina (10µM) sono stati impiegati, rispettivamente 1 ora e 5 ore prima H2O2, per antagonizzare l’effetto dello stimolo ossidativo sui fotorecettori. In fine, abbiamo esaminato 30 composti di nuova sintesi per determinare la loro capacità inibitoria su SPT, attraverso un saggio di attività enzimatica. ▼ Organisms have evolved a variety of mechanisms for regulating gene expression. Expression of individual genes is carefully modulated during different stages of cell development and in response to changing environmental conditions. A number of regulatory mechanisms involve structural elements within messenger RNAs (mRNAs) that, in response to an environmental signal, undergo a conformational change that affects expression of a gene encoded on that mRNA. RNA elements of this type that operate independently of proteins or translating ribosomes are termed riboswitches. In this work, the THI box and S_MK box riboswitches were investigated in order to gain insight into the structural basis for ligand recognition and the mechanism of regulation employed by each of these RNAs. Both riboswitches are predicted to regulate at the level of translation initiation using a mechanism in which the Shine-Dalgarno (SD) sequence is occluded in response to ligand binding. For the THI box riboswitch, the studies presented here demonstrated that 30S ribosomal subunit binding at the SD region decreases in the presence of thiamin pyrophosphate (TPP). Mutation of conserved residues in the ligand binding domain resulted in loss of TPP-dependent repression <i>in vivo</i>. Based on these experiments two classes of mutant phenotypes were identified. Class I mutations resulted in increased accessibility of the SD region and binding of 30S ribosomal subunits regardless of the presence or absence of TPP. In contrast, Class II mutations resulted in constitutive occlusion of the SD region even in the absence of ligand. The latter class represents the first example of riboswitch mutations that result in stabilization of the ligand-bound conformation when no ligand is present. For the S_MK box riboswitch, mutational analysis verified the importance of conserved residues for binding to <i>S</i>-adenosylmethionine (SAM). A minimal S_MK box element that retains the ability to bind SAM was used to determine the high resolution structure of the riboswitch in complex with ligand using x-ray crystallography. The SD sequence was shown to be sequestered in the ligand-bound conformation and is essential for SAM binding, providing one of the first examples of a riboswitch in which the regulatory domain is intrinsic to the ligand binding domain. Specific binding to SAM is dictated through direct and indirect contacts with the adenosine and sulfur moieties of the ligand. These results were verified using a fluorescence assay with a variety of SAM analogs. The half-life of the SAM-RNA complex was shown to be much shorter than the half-life of the mRNA inside the cell, suggesting that the RNA may undergo multiple regulatory events in its lifetime. Finally, nuclear magnetic resonance (NMR) was used to investigate the solution structure of the S_MK box riboswitch in the presence and absence of ligand. ▼ Dairy farms confront unique risks from weather conditions. Hot and humid weather induces heat stress, which brings a series of risks to dairy farm operations including reductions in milk production and pregnancy rate and increases in cull rate and death rate. Traditional heat abatement technologies control the environment through ventilation, misting or evaporative cooling. Adoption of abatement equipment, however, is hindered by its high initial cost and possibly long payback period, especially for small- and medium-scale firms. Weather derivatives provide an alternative method of risk management for dairy producers. Instead of reducing production losses, weather derivatives make payments based upon observed weather conditions over a period of time so that they offer the potential to offset profit losses caused by adverse weather events. Chapter 2 tests the risk management value of weather derivatives acting as a substitute for traditional abatement technologies within a utility maximization framework. Previous research has identified the problem of basis risk in weather derivatives. Little theoretical or empirical work has been done to examine the effect of basis risk on weather derivatives. Chapter 3 examines the effect of basis risk in weather derivatives, and whether the existence of basis risk mitigates the usefulness of weather derivatives for dairy risk management. A decision that must regularly be made by a dairy farmer is when to maintain his abatement equipment and when to replace it. This decision affects both current and expected future revenues. Considering that weather derivatives can be purchased periodically, Chapter 4 tests the risk management value of weather derivatives for dairy producers and examines how weather derivatives can affect dairy producers’ abatement equipment decisions. In this chapter, I employ a dynamic programming framework to study the case that a representative dairy farmer maximizes his long-run utility using weather derivatives and abatement equipment. ▼ Ce travail doctoral traite de Hà-Nội, de son paysage construit et de son identité patrimoniale. Plus précisément, il porte sur l'analyse des processus de formation et de transformation des architectures résidentielles d'un quartier central et historique de Hà-Nội - le quartier Bùi Thị Xuân dans l'arrondissement Hai Bà Trung. Par l'analyse du panorama de la production architecturale des maisons du quartier, l'étude cherche à définir et à articuler le lien entre le paysage urbain tel qu'il se présente nos yeux aujourd'hui et son caractère patrimonial. Par ces termes, la thèse cherche à expliquer comment les interventions architecturales contemporaines s'inscrivent dans une continuité historique des modèles d'habitat et des modes d'habiter hanoiens et comment elles sont aujourd'hui porteuses de sens pour leurs habitants. L'étude utilise une approche systémique qui propose une vision holistique et culturelle du phénomène de transformation et d'appropriation de l'espace bâti. La méthodologie choisie suit les modèles historico-interprétatifs de recherche scientifique qui cherchent à formuler un discours interprétatif à partir de l'étude des relations entre les données récoltées et le phénomène observé. Afin d'identifier l'inscription historique des constructions récentes à travers l'analyse des processus de ses transformations vernaculaires, l'étude vise à reconnaître et à caractériser les modèles originels et générateurs, les ruptures qui s'y sont produites et les nouveaux prototypes qu'ils ont engendrés. Elle dégage des éléments formels susceptibles de définir une identité paysagère et architecturale propre au quartier. L'objectif général de la thèse est la compréhension de l'émergence de formes urbaines particulières et originales dans une société donnée. La ville est considérée ici comme un objet, comme un ensemble d'artefact. La thèse s'intéresse au bâti physique, à sa formalisation et à sa spatialisation. Elle prend pour postulats de base que l'analyse de la forme urbaine est une des portes d'entrée qui permet de comprendre les sociétés qui les ont produites et que le paysage urbain représente la spatialisation des identités collectives. Étudier la ville comme paysage, c'est-à-dire comme réalité physique et comme objet de représentation dans un même système dual, permet de comprendre et d'identifier comment les habitants d'un quartier mettent en scène leur culture d'habiter et la formalise dans un « patrimoine » bâti. Les résultats montrent que la morphogénèse du quartier Bùi Thị Xuân a connu des processus particuliers dans l'histoire de la ville de Hà- Nội, qui ont favorisé le développement de formes d'habitat originales et qui lui permettent aujourd'hui de revendiquer une identité patrimoniale. Connu à son origine en 1902 sous la dénomination de « Nouveau Quartier Indigène », le quartier Bùi Thị Xuân est la première extension périphérique planifiée par l'autorité coloniale française à l'intention exclusive de la population vietnamienne. L’objectif de cette étude est de comprendre les relations entre les organisations constructives, formelles et spatiales et les modes d'appropriation de la façade des maisons dans un quartier résidentiel à Hanoi, au Vietnam. Elle présente les analyses et les caractéristiques des murs de la façade des maisons du point de vue morphologique et de l'appropriation des espaces. La recherche comporte quatre chapitres. Le premier chapitre présente d’abord une introduction générale de la ville de Hanoi et du quartier Bui Thi Xuan. Ce chapitre présente également la pertinence de l’étude des murs de façade des maisons du quartier Bui Thi Xuan, ainsi que les modes d’appropriation de la façade. Le deuxième et le troisième chapitre présentent le cadre conceptuel, théorique et méthodologique de la recherche. Le quatrième chapitre fait état des résultats de la recherche, décrivant et interprétant les caractéristiques de la façade des maisons et les relations entre les organisations constructives, formelles et spatiales et les modes d’appropriation de la façade du quartier Bui Thi Xuan. La recherche permet de comprendre les caractéristiques principales de la façade des points de vue constructif, formel et spatial et de montrer l’influence de ces organisations sur les modes d’appropriation de la façade par le contrôle que les habitants ont sur les espaces, à l'intérieur et à l'extérieur, d'un bâtiment et les usages de ces espaces. Ce mémoire décrit également la relation entre les fonctions des espaces intérieurs et les organisations spatiales et formelles de la façade de maisons dans le quartier Bui Thi Xuan.

The researching purpose of the thesis is to understand relationships between structure, form, space organization and ways of using the housing's façade in the quarter Bui Thi Xuan, Hanoi. The thesis will make analysis and describe main characters of the housing's façade from the angle of morphology and space use. Researching contents include 4 chapters. Chapter I will state an overview on the situation of Hanoi and Bui Thi Xuan ward, as well as the necessity of the research on the wall of housing's facades in the quarter Bui Thi Xuan and ways of using the housing's façade. Chapters II and III will indicate scientific bases and methodologies of the research in the thesis. These two chapters will focus on clarifying basic concepts of the housing wall, housing's façade as well as its structure, ways of using the façade and researching methodologies. Chapter IV will mention researching results of the thesis by describing and explaining main characters of the housing's façade, the relationship among structure, forms, space organization and ways of using the housing's façade in the quarter Bui Thi Xuan. La soufflante de la nouvelle génération de moteurs Snecma est caractérisée par des pales de rotor aux bords de fuite épais, ainsi que par un OGV hétérogène intégrant les bras supports.L’objectif de cette thèse est de caractériser l’influence de ces innovations technologiques sur la génération des sources et le rayonnement sonore dans la manche secondaire liés à l’interaction du sillage du rotor avec l’OGV. Ce mécanisme est principalement responsable des émissions tonales et à large bande qui sont traitées indépendamment. D’une part, une simulation URANS de l’étage rotor-stator fournit les sources déterministes sur le stator hétérogène ainsi que le champ sonore dans la manche traversée par les bras supports. L’extension de modèles acoustiques permet d’accéder au spectre de raies obtenu soit en propageant les sources par un modèle fondé sur le principe de l’analogie acoustique, soit directement à partir du champ sonore issu de la CFD. Il est alors montré que l’hétérogénéité du stator induit l’émergence de la raie du fondamental, et que les bras supports engendrent une onde stationnaire dans la manche secondaire.D’autre part, la simulation d’un canal complet du rotor avec une approche ZDES permet d’accéder aux perturbations turbulentes du sillage. Ces dernières alimentent un code acoustique fondé sur la théorie d’Amiet permettant de prévoir le bruit à large bande rayonné en conduit. Bien que plus coûteuse, la ZDES permet de s’affranchir des hypothèses restrictives et du calibrage délicat des modèles de turbulence homogène isotrope en fournissant des données directement exploitables, conduisant ainsi à des estimations plus précises.L’ensemble des simulations numériques et des prévisions acoustiques est validé grâce aux mesures issues d’une campagne d’essais au banc RACE.

Modern turbofans are characterized by rotor blades with large chords, as well as heterogeneous OGV with mechanical struts integrated.The aim of this thesis is to characterize the impact of these technological innovations on sources generation and sound radiation in the bypass duct due to the interaction of the rotor wakes with the OGV. This mechanism is mainly responsible in tonal and broadband noise emissions which are dealt separately.On one hand, a URANS computation of the rotor-stator row provides the periodic sources on the heterogeneous stator as well as the sound field in the bypass duct crossed by the struts. The expansion of sound models allows to access to the tonal spectra obtained either by propagating sources thanks to a model based on the acoustic analogy principle, or by the sound field directly issued from the CFD. Then, it is shown that the stator heterogeneity induces the appearance of the fundamental tone, and that the struts lead to standing waves in the bypass duct.On the other hand, the simulation of a complete rotor channel with a ZDES approach provides the turbulent wakes.
