副腎髄質におけるP2X4受容体を介した脳由来神経栄養因子(BDNF)の発現とTrkBの役割について検討した。ラット副腎髄質由来クロマフィン細胞(PC12)を用いた。抗TrkB(clone 47/TrkB)がTrkB作用抗体として作用し、60分ストレス条件下でのラット副腎髄質のTrkBがTrkB作用抗体によりカテコールアミン放出を誘起した。BDNF発現は、ラットの副腎髄質で確認し、BDNFは副腎髄質で自己分泌機能をもつ可能性が示唆された。副腎髄質では急性ストレス条件下でBDNF-TrkB相互間の自己分泌を通したポジティブフィードバックループの役割を担う可能性が示唆された。副腎髄質のATP受容体のうちP2X4受容体がBDNF産生に関して強く関係していることが示唆された。
