▼ El estudio de las córneas candidatas a cirugía refractiva Lasik ó sospechosas de queratocono es de suma importancia en la práctica clínica diaria. Hasta hoy se realizaba analizando la curvatura de la cara anterior corneal, la elevación de las caras anterior y posterior de la córnea, además de un exámen médico el cual incluye la biomicroscopía, entre otros. En la actualidad existe un nuevo módulo para el topógrafo Pentacam, llamado Belin/Ambrósio Enhanced Ectasia, el cual es el primero que realiza un estudio 100% computacional de los datos de la elevación corneal, estudiando el aspecto más importante en las ectasias: La elevación de la cara posterior de la córnea. El objetivo de esta investigación es describir y analizar este nuevo módulo, para llevarlo a cabo se estudiaron 100 ojos, divididos en 5 grupos: 20 ojos emétropes, 20 ojos miopes, 20 ojos hipermétropes, 20 ojos astigmatas y 20 ojos con queratocono. El estudio se realizó de forma retrospectiva, revisando las fichas clínicas de cada paciente, se estudió específicamente el Módulo Belin/Ambrósio Enhanced Ectasia de cada uno de ellos, analizando cada datos que entrega. El estudio demostró que los ojos sin queratocono, entregan resultados normales en todos los puntos analizados, excepto en 2 de ellos: la diferencia de elevación de la cara anterior en el grupo astigmata y la diferencia de elevación de la cara posterior en todos los grupos. En el grupo con queratocono, se encontraron valores menores a los esperados, pero aún así patológicos, excepto en el estudio paquimétrico, en el cual los valores de la distancia ápex-punto más delgado de la córnea resultaron ser normales y el índice de progresión sólo estuvo en el intervalo sospechoso. El exámen demostró ser útil para detectar ectasias precoces y córneas riesgosas para la cirugía refractiva con láser, sin embargo el estudio de la cara posterior de la córnea demostró ser muy sensible.; The study of the corneas for refractive surgery with laser candidates or keratoconus suspect is of great importance in daily clinical practice. ▼ This dissertation comprises experimental studies on the synthesis and applications of metallic nanoshells. These are a class of nanoparticles composed of a dielectric core and a thin metallic shell. Metallic nanoshells play an important role in nanotechnology, particularly in nanomedicine, due to their peculiar optical properties. The overall objectives of the dissertation were to improve the fabrication of these nanoparticles, and to demonstrate new applications of these materials in cancer research and spectroscopy. The fabrication of nanoshells is a multi-step process. Previously to our work, the procedures for the synthesis of nanoshells reported in the literature lacked systematic characterization of the various steps. The procedure was extremely time-consuming and the results demonstrated a high degree of size variation. In Chapter 3, we have developed characterization tools that provide checkpoints for each step of the synthesis. We demonstrated that it is possible to control the degree of coverage on the shell for a fixed amount of reagents, and also showed important differences on the shell growth phase for gold and silver. The synthetic optimization presented in Chapter 3 led to an overall faster protocol than those previously reported. Although the improvements presented in Chapter 3 led to a higher degree of control on the synthesis of nanoshells, the variations in the resulting particle population were still too large for applications in single particle spectroscopy and imaging. In Chapter 4, the synthesis was completely reformulated, aiming to narrow the size distribution of the nanoshell colloids. Through the use of a reverse microemulsion, we were able to fabricate ultramonodisperse silica (SiO2) cores, which translate into nanoshell colloids with narrow extinction bands that are comparable to those of a single nanoshell. We then fabricate a library of colloids with different core sizes, shell thicknesses and composition (gold or silver). The localized surface plasmon resonance (LSPR) of these colloids span across the visible range. From this library, two nanoshells (18nm silver on a 50nm SiO2 core, and 18nm gold on a 72nm SiO2 core) were selected for a proof of principle cell imaging experiment. The silver nanoshells were coated with a nuclear localization signal, allowing it to target the nuclear membrane. The gold nanoshells were coated with an antibody that binds to a receptor on the plasma membrane of MCF-7 human breast cancer cells. The nanoshells were easily distinguishable by eye in a dark field microscope and successful targeting was demonstrated by hyperspectral dark field microscopy. A comparison was made between fluorescent phalloidin and nanoshells, showing the superior photostability of the nanoparticles for long-term cell imaging. The results from Chapter 4 suggest that the nanoshells obtained by our new synthetic route present acceptable particle-to-particle variations in their optical properties that enables single particle extinction spectroscopy for cell imaging. Em conformidade com o Fundo Mundial para a Natureza (WWF) (2019), o Brasil ? o quarto maior produtor de lixo pl?stico no mundo, com cerca de 11,3 milh?es de toneladas, posi??o inferior relativo aos Estados Unidos, China e ?ndia. Desse montante, coletaram-se mais de 10,3 milh?es de toneladas (91%), e somente 145 mil toneladas (1,28%) foram efetivamente recicladas, ficando bem abaixo da m?dia global de 9% concernente a reciclagem pl?stica. Em outra vertente, tem-se os dados provenientes do Anu?rio Estat?stico Consolidado da Confedera??o Nacional de Transporte (CNT), em 2019, que classificou 1.046 km de rodovias pavimentadas no Estado do Amazonas, em regular (65,59%), ruim (7,55%) e p?ssimo (26,86%). Tal resultado aliado ? preserva??o ambiental motivam o emprego de res?duos na constru??o de pavimentos asf?lticos, objeto do presente trabalho. Nesse contexto, avaliou-se o comportamento mec?nico de misturas asf?lticas alternativas com res?duo de Polietileno de Alta Densidade (PEAD), nos teores de 1% e 2%, em substitui??o ao agregado mi?do, p? de pedra. Tal material originou-se do descarte p?s-consumo de tubos de PEAD,procedentes da instala??o de ramais prediais na cidade de Manaus. A mistura asf?ltica refer?ncia, do tipo Concreto Asf?ltico (CA), seguiu uma das dosagens aplicadas em servi?os de pavimenta??o da Prefeitura Municipal de Manaus (PMM). Determinou-se o comportamento mec?nico desses comp?sitos de acordo com os ensaios de Resist?ncia ? Tra??o por Compress?o Diametral (RT) e M?dulo de Resili?ncia (MR), submetidos ?s temperaturas de 25?C e 40?C. ▼ El queratocono (QTC) es la patología ectásica corneal por excelencia. Es una enfermedad corneal progresiva, donde la córnea adopta forma cónica, por adelgazamiento del estroma, con el consiguiente astigmatismo irregular y miopía, que conducen a un compromiso visual. Aparece en la pubertad y progresa hasta la tercera o cuarta década de la vida cuando se paraliza, por ello se considera única entre las enfermedades oculares. La aparición de una técnica aditiva de materiales sintéticos intraestromales llena un espacio en el tratamiento de la ectasia, retrasando el trasplante. Entre ellos están los anillos de sección triangular Keraring ® , que son los que ocupan este estudio. La redistribución de la curvatura corneal que inducen, es responsable de la detención de la enfermedad por una “nueva biomecánica”. El mecanismo de acción por el que actúan no está totalmente establecido. Se desconoce si existe una correlación entre los parámetros biomecánicos, clínicos y morfológicos de estos pacientes, o si se ven influenciados por las características físicas de los anillos, objetivos secundarios de la presente tesis. El diámetro, espesor y su forma triangular, son las diferencias físicas que los caracterizan, pudiendo ser posibles explicaciones al mayor efecto de aplanamiento corneal en comparación con el resto de anillos disponibles. Se evalúan cambios biomecánicos, refractivos, topográficos y morfológicos que acontecen en córneas queratocónicas tras el implante de estos anillos en sus opciones de diseño. Se evalúan también correlaciones entre estos resultados y la agudeza visual (AV) alcanzada. Como objetivos secundarios, correlaciones entre modificación de dichos parámetros y grado de ectasia, número y grosor de anillos. Se ha realizado un análisis retrospectivo. El seguimiento se prolonga hasta los 6 meses postoperatorios, con evolución al mes y 3 meses. Los resultados biomecánicos, en concreto el factor de histéresis corneal (CH), es distinto en función del diámetro utilizado. La mejora del equivalente esférico (EE) es independiente del diámetro. La mejora de la AV es más evidente con 5mm. La curvatura K1 disminuye con intensidad con 5mm, con 6mm el impacto sobre K2 es mayor. La profundidad de cámara anterior (CA) desciende progresivamente, más con 6mm. La paquimetría mínima aumenta, fundamentalmente en ojos con 5mm. El volumen de CA es más estable. La relación entre variaciones de parámetros biomecánicos y mejora de AV, se manifiesta para el grupo de 6mm. La asociación entre mejora del EE y la AV, es específica en ojos implantados con 6mm. Se observa que los ojos con mayor descenso de curvatura, no necesariamente son los de mayor mejoría visual, para ningún diámetro. Los ojos con grado de ectasia superior a I y de 6mm, experimentan mayor recuperación de la CH. La influencia del grado de ectasia sobre el factor de resistencia corneal (CRF) es indefinida. Le lien entre les éléments volatils CO₂-H₂O et la fusion mantellique a depuis maintenant longtemps été illuminé par l’expérimentation. Une large base de données expérimentales existe et souligne l’effet primordial de ces éléments sur l’abaissement des températures de fusion de la péridotite ainsi que sur la composition des liquides magmatiques produits comme une fonction des conditions P – T – fo₂ – composition du système. Néanmoins, la diversité et la complexité de cette base de données peuvent compliquer sa compréhension globale. Dans cette étude, une analyse détaillée de la composition des liquides magmatiques riches en CO₂ et H₂O est réalisée, soulignant notamment une transition non-linéaire et plus ou moins abrupte entre des liquides carbonatitiques et des liquides silicatés. Un modèle thermodynamique est élaboré afin de calculer l’activité de SiO₂ dans les liquides magmatiques riches en CO₂-H₂O (aSiO₂(l)) et coexistant avec un assemblage péridotitique, depuis des termes carbonatitiques jusqu’à des termes basaltiques. L’application de ce modèle dans des conditions de ride océanique prédit la stabilisation des liquides carbonatitiques au démarrage de la fusion redox (liée à la transition graphite/diamant- carbonates) jusqu’à environ 100 km de profondeur, avant d’évoluer plus ou moins abruptement vers des liquides silicatés riche en CO₂. Au niveau des cratons, les kimberlites de Groupe I sont stabilisés en base de lithosphère (~250 km de profondeur), et peuvent être formés à partir d’un plume mantellique. L’épaisseur de cette lithosphère empêche la remontée du plume et la formation des OIB. Afin de décrire plus pleinement les propriétés thermodynamiques du liquide magmatique, un modèle plus complexe (système CMAS-CO₂) est en construction, dont la méthodologie est modifiée par rapport au précédent modèle calculant aSiO₂(l) ; un effort tout particulier est ici mené afin de mieux considérer les incertitudes expérimentales et thermodynamiques.

The link between volatiles (CO₂-H₂O) and mantle melting has so far been illuminated by experiments. A large experimental database exists and emphasizes the importance of volatiles on lowering solidus temperatures of peridotite and modifying the melt composition as a function of P – T – fo₂ – bulk composition. Nevertheless, the diversity and the complexity of this experimental database may complicate its global understanding. In this study, an analysis of CO₂-H₂O-rich melt composition is done, emphasizing the non-linear and more or less abrupt character of the transition between carbonate-rich melts and silicate-rich melts. A thermodynamic model is accomplished to calculate the silica activity in CO₂-H₂O-rich melts coexisting with peridotite assemblage and covering carbonatitic to basaltic terms. Along an oceanic ridge adiabat, the model predicts that carbonatitic melts can be stabilized at the onset of “redox melting” (transition between graphite/diamond-carbonates) to about 100 km depth, before abruptly evolving towards carbonated silicate melts. T-helper (Th) lymphocytes are present in significant numbers in the atherosclerotic plaque. Even though coronary artery ectasia (CAE), a variant of coronary atherosclerosis, has been associated with increased levels of inflammatory markers, the underlying mechanism that determines the role of inflammation in CAE remains controversial.We evaluated the hypothesis that CAE might be associated with a specific pattern of Th-lymphocyte activation by measuring the Th-1 cytokine, interleukin-2 (IL-2) and the Th-2 cytokines, interleukin-4 (IL-4) and interleukin-6 (IL-6) in patients with CAE, obstructive coronary artery disease (CAD) and normal individuals.Plasma levels of IL-2, IL-4 and IL-6 were measured in 74 patients undergoing an elective cardiac catheterization due to symptoms of chest pain and positive or equivocal results during non-invasive screening for cardiac ischemia: 34 patients had CAE and non-obstructive CAD (group A), 22 had obstructive CAD without CAE (group B) and 18 had normal coronary arteries (group C). Group A had significantly higher levels of IL-4 compared to group B and group C (p<0,001 and p=0,006, respectively). In contrast, Group A had markedly lower levels of IL-2 compared to group B and group C (p<0,001 for both comparisons). Serum IL-4 was higher in group C compared to group B, whereas group B had higher IL-2 levels than group C (p<0,001 for both comparisons). Serum levels of IL-6 were significantly higher in group A and B compared to group C (p<0,001 for both comparisons), whilst they were comparable between group A and B. Multivariate logistic regression analysis showed that higher levels of IL-4 and lower levels of IL-2 were the strongest independent predictors associated with the incidence of CAE whilst IL-6 depicted a weaker positive relation (OR: 3,846, CI: 1,677-8,822, p=0,001, OR: 0,567, CI: 0,387-0,831, p=0,004 and OR: 1,481 CI: 1,036-2,033, p=0,03, respectively).Our data demonstrates for the first time that Th-2 inflammatory response exhibited through increased serum levels of IL-4 and low serum IL-2 constitutes a fundamental feature of CAE phenotype. The above findings may be interpreted, apart from their pathophysiologic significance, towards the effort of discovering a novel approach in the diagnosis, prognosis and therapy of patients with coronary artery ectasia.

▼ Se ha escogido la técnica PECVD para obtener depósitos de subóxido de silicio, SiOx, de espesores del orden de la micra, sobre substratos de Si de dos pulgadas. Hemos logrado depósitos con contenidos de oxígeno entre 1.3 y 2.0, y un intervalo de índices de refracción comprendidos entre 1.96 (próximo al Si3N4) y 1.46 (SiO2). La estructura (en particular, la porosidad), uniformidad y tensión de las capas son mejores cuanto menor es el contenido de oxígeno. Además, las capas crecidas contienen impurezas en forma de enlaces Si-H, N-H, Si-OH y H2O. El contenido de impurezas varia de forma gradual desde capas pobres en oxígeno con una gran cantidad de enlaces Si-H y N-H, y nulo contenido de Si-OH y H2O, y capas de estructura próxima al óxido estequiométrico, con nulo contenido de enlaces Si-H y N-H, y alto contenido de Si-OH y H2O. Se ha investigado las condiciones de temperatura, potencia y presión de los procesos de depósito, susceptibles de mejora de la calidad de las capas crecidas. El aumento de la temperatura de depósito (hasta 350 °C) y de la potencia (hasta 50 W, límite impuesto por la disminución de uniformidad), disminuye el contenido de impurezas de las capas; sin embargo, el aumento gradual de la tensión con el aumento del contenido de oxígeno de las capas, aconseja que los depósitos se crezcan a menores valores de temperatura y potencia en la obtención de capas ricas en oxígeno. Por lo que se refiere a la presión, valores bajos (100 mTorr), favorecen la disminución de impurezas; sin embargo, el aumento del orden estructural que comporta el aumento de la presión (hasta 300 mTorr), inhibe el acomodo de impurezas en capas ricas en oxígeno, por lo que la calidad de los depósitos se verá favorecida procediendo a un aumento de la presión a medida que se crezcan capas con un previsible mayor contenido de Si-OH y H2O. La mejora de las condiciones de depósito no soluciona dos problemas fundamentales; por un lado, no se han podido suprimir las impurezas ni las tensiones de las capas y, por otro, las capas ricas en oxígeno son inestables frente a la humedad. El estudio de la acción de la humedad y los esfuerzos posteriores para estabilizar las capas, al proceder a un recocido térmico a una temperatura inferior a 350 °C, dan resultados parciales, siendo los efectos reversibles. Como solución se ha propuesto proceder a un recocido térmico rápido a alta temperatura (950 °C) de las capas. Las capas tratadas son estructuralmente compactas e insensibles a la acción exterior de la humedad, no contienen cantidades de impurezas apreciables, excepto un bajo contenido de enlaces Si-H en entornos O3-SiH; además, el nivel de tensiones desaparece en capas ricas en Si y aumenta gradualmente con el contenido de oxígeno, llegando a ser un 50% inferior al de las capas depositadas sin un tratamiento posterior. Objetivo: Investigar as alterações biomecânicas da córnea de suínos induzidas pela confecção de lamelas pediculadas de diferentes espessuras pelo laser de femtossegundo. Métodos: Para a formação dos dois grupos, 12 olhos de porcos foram usados: lamelas pediculadas de 100 e de 300 micrômetros confeccionadas pelo laser de femtossegundo. Cada olho foi submetido aos seguintes exames, antes da criação das lamelas: topografia por rasterstereography, Ocular Response Analyzer (ORA), tomografia do segmento anterior por coerência óptica para a avaliação paquimétrica corneal e das lamelas criadas e sistema de velocidade de onda (SVO), que mede a velocidade de propagação de ondas acústicas entre dois transdutores posicionados na superfície corneal antes e imediatamente, após a feitura da lamela. O primerio passo foi desenhado para o estudo das diferenças em relação à histerese corneal, fator de resistência corneal, mudanças na curvatura e velocidade de propagação de onda acústica entre córneas com lamelas finas e espessas. Posteriormente, as lamelas foram amputadas, e as medidas do sistema de velocidade de onda foram repetidas. Resultados: A média de espessura das lamelas ± desviopadrão (DP) foi de 108,5±6,9 (8,5% da espessura total) e 307,8±11,5 m (22,9% da espessura total), para os grupos de lamelas finas e espessas, respectivamente (p< 0,001). Histerese corneal e o fator de resistência corneal não apresentaram diferença estatística, após a criação de lamelas finas (p = 0,81 e p = 0,62, respectivamente). Histerese corneal foi significantemente mais baixa, depois da confecção de lamelas mais espessas (8,0±1,0 para 5,1±1,5 mmHg para medidas pré e pós-operatórias, respectivamente, p = 0,003, diminuição de 36,25%) e fator de resistência corneal também mostrou significante diminuição nesse grupo, após o procedimento cirúrgico; valores médios pré e pós-operatórios de 8,2±1,6 e 4,1±2,5 mmHg respectivamente (p= 0,007), diminuição de 50%. A ceratometria média simulada apresentou maiores valores, após a confecção das lamelas mais espessas em relação ao pré-operatório (ceratometria pré e pós-operatória de 39,5±1 D e 45,9±1,2 D, respectivamente, p= 0,003). Para o grupo de lamelas finas, não houve diferença estatisticamente significante (ceratometria pré e pós-operatória de 40,6±0,6 D e 41,4±1,0 D, respectivamente, p=0,55). Em relação ao Sistema de Velocidade de Onda, após a criação das lamelas e sua amputação, houve diminuição da velocidade de propagação acústica, embora na maior parte das posições não fosse estatisticamente significante. Conclusão: Nas condições experimentais estabelecidas por este estudo, a criação de lamelas de maior espessura pareceu exercer efeito mais relevante sobre a biomecânica da córnea de suínos

▼ Surgical small intestinal diseases occur commonly in canine patients seen in veterinary practices. Failure to identify ischemic tissue intra-operatively can result in surgical dehiscence, require surgical revision, and lead to sepsis or death. Currently contrast-enhanced computed tomography (CT) is the standard of care for evaluating ischemic lesions of the bowel in humans. Contrast-enhanced ultrasound (CEUS) is an alternative method for evaluating tissue perfusion whose utility for evaluation of canine small intestinal lesions remains to be fully tested. Thirteen client-owned dogs were prospectively recruited after presenting with surgical diseases within the small intestine. Presenting conditions included discrete foreign bodies, linear foreign bodies, intestinal abscess, and neoplasia. Patients underwent pre-operative CEUS, triple-phase contrast-enhanced CT, and intra-operative CEUS. Surgical specimens obtained from each patient included resected intestinal segments or biopsy. Both CT and CEUS data were compared to histopathology findings. All patients that required a resection (n=4) received an ischemia score of 4 or 5. A higher ischemia score was correlated with an increased chance of necrosis (p=0.003) and thrombosis (p=0.024). The presence of a visual perfusion deficit on CT was correlated with a higher ischemia grade (p=0.048) and higher chance of necrosis (p=0.024); however, observers only detected a perfusion deficit in 3/4 (75%) of patients requiring a resection. Ischemia was correlated with lower Hounsfield (HU) values aborad to the lesion in the pre-contrast CT (p=0.049). Necrosis was correlated with increased HU values at normal intestine in the arterial phase (p=0.017) and orad to the lesion in the venous phase of CT (p=0.014). Thrombosis was correlated with lower HU values aborad to the lesion in the pre-contrast CT (p=0.025) and higher HU values at normal intestine in the arterial phase (p=0.037) and orad to the lesion in the venous phase CT (p=0.041). Edema was correlated with lower HU values aborad to the lesion in the arterial phase (p=0.03) and higher HU values at normal intestine in the venous phase CT (p=0.038). Ulceration and erosion were correlated with higher HU values orad to the lesion in both the arterial (p=0.028) and venous phases of CT (p=0.044). No significant correlations were found at the site of the lesion in any phase of CT. For pre-operative CEUS, edema was correlated with higher peak intensity (p=0.014) while necrosis (p=0.046) and thrombosis (p=0.04) were correlated with a faster time to peak. For intra-operative CEUS, ulceration and erosion were correlated with a greater inflow slope (p=0.045), hemorrhage was correlated with a longer time to initial rise (p=0.028), thrombosis was correlated with a faster time to peak (p=0.014), and necrosis was correlated with increased baseline pixel intensity (p=0.046). Observers were able to detect a perfusion deficit on CEUS exam in 4/4 (100%) of patients requiring a resection. ▼ The purpose of this study is to explore the nature of complex adaptive systems (CAS) and network dynamics in international education (IE) programs in a U.S. higher education institution. I analyze the IE programs through a lens of complexity and network theories and ask how measures of engagement in complex networks affect performance in the IE system. This study presumes that today's international education programs in the U.S. higher education institutions are complex adaptive systems and that traditional leadership is no longer adequate to address the overwhelming opportunities and challenges posed by global education. A two-stage quantitative research design is adopted to investigate network structures and interactions within the IE system and to describe how such network measures impact organizational performance. In Stage 1, Dynamic Network Analysis (DNA) is used to calculate agent-level network measures for each participant within the university IE system's bounded networks and to produce optimized simulations of the IE system for use in Stage 2. In Stage 2, Response Surface Methodology (RSM) is used to examine the relationship between independent and dependent measures. In this study, the independent network measures include (a) informal leadership, which is operationalized as betweenness centrality, (b) clique engagement, which is operationalized as clustering coefficient, and (c) social capital, which is operationalized as hub centrality. These independent measures are used to analyze the dependent measure, organizational performance, which is operationalized as task accuracy. A dynamic network framework of international education is proposed as a useful network model and leadership framework for enabling international education functions, senior international officers (SIOs), and their institutions to achieve excellence and succeed in a new era of global education and knowledge producing. ▼ While Africa is confronted with the urgency to provide its citizens with the basic requirements of life, the rest of the world is heading towards a ‘globally-networked’ information economy. Many communication scholars believe that without access to information resources and telecommunication services, an understanding of its significance, and the ability to use it for social and economic growth, Africa is facing an unavoidable predicament. This dissertation presents a case study of the National Council of Churches of Kenya (NCCK) Huruma Community Telecentre as an arena where governmental, non-governmental and other private organizations are collaborating to test the contribution that a Community Telecentre can make towards providing universal access to telephony and other telecommunications and information services to a disadvantaged community. While the theoretical starting point for this dissertation is grounded within communication and development theories, it employs Bijker, Hughes, and Trevor’s (1987) Social Construction of Technology concept of ‘interpretive flexibility.’ This constructivist approach offers the possibility of looking at the technological process and empowerment as a dynamic process where the ICT users are actively involved in its integration within their existing environment. The study reveals that, while a number of factors have acted as barriers to communication access to information technology, the biggest hindrances are the lack of sound telecommunication regulations and clear government policies as well as the absence of an environment conducive to ICT development due to an inadequate telecommunication infrastructure. This dissertation employs the case study as an overall strategy and also draws upon multiple data sources to develop a triangulation of methods ranging from in-depth interviewing, participant observation, historical and document analysis as well as analysis of telecommunication debates in local newspapers over the last 4 years. The study concludes that, although the telecommunication is beginning to show some impact due to the partial privatization of the sector, it is too early to assess the overall impact of new media technologies on Africa’s development. This calls for a more realistic approach that incorporates the need to harness the potential of ICTs for purposes of addressing locally relevant problems in innovative and cost-effective ways. Depuis 2005, l’augmentation exponentielle de la prévalence d’Entérobactéries productrices de BLSE est liée à la diffusion mondiale des enzymes de type CTX-M et à leur prévalence au sein de l’espèce Escherichia coli. Le rôle de certains plasmides conjugatifs, en particulier de groupe IncF, A/C, ou encore IncI, est fondamental dans la diffusion des CTX-M et ce, malgré le coût biologique qu’ils confèrent à la bactérie hôte. L’étude de ce paradoxe montre que le couple hôte/plasmide peut s’adapter et co-évoluer afin de réduire ce coût, notamment par des mutations compensatoires. Ceci pourrait expliquer le succès évolutif de certains clones pandémiques, tel que le clone E. coli O25b:H4 ST131 H30. Dans ce travail, nous avons étudié l’impact de plasmides conjugatifs, codant pour ces enzymes CTX-M, sur E. coli : (i) coût du plasmide IncF de type F2:A1:B- décrit dans le clone pandémique E. coli O25b:H4 ST131 H30, et de plasmides A/C (un plasmide IncC et un plasmide IncC-IncR) dans la souche E. coli J53-2 par une approche d’évolution expérimentale in vitro (ii) impact d’un plasmide conjugatif IncI CTX-M-1 dans des souches d’E. coli commensales du porc, ayant évolué ou acquis le plasmide au cours de travaux précédents (Mourand et al. 2016, 2017). Malgré un coût important imposé à la bactérie hôte immédiatement après conjugaison, nous avons pu montrer qu’après 1 120 générations d’évolution in vitro, le coût du plasmide F2:A1:B- IncF devenait non significatif alors qu’il se maintenait pour le plasmide IncC-IncR. Aucune modification phénotypique des transconjugants évolués n’a été observée, mais nous avons décrit de nombreux réarrangements chromosomiques et plasmidiques lors de la conjugaison du plasmide IncC-IncR. L’étude de l’impact du plasmide IncI évolué in vivo a montré, pour les souches ayant acquis le plasmide, que le coût était variable, indépendamment de leur phylogroupe. En revanche, nous avons montré une absence de coût initial du plasmide sur la bactérie hôte mais nous avons mis en évidence que les souches évoluées ayant perdu le plasmide amélioraient significativement leur fitness comparativement aux souches évoluées ayant conservé le plasmide et également aux transconjugants ancestraux. De plus, bien que non significatif, les souches évoluées ayant perdu le plasmide ont en moyenne un taux de croissance supérieur à celui de la souche ancestrale dépourvue de plasmide. Au final, nos travaux montrent que la diffusion mondiale du gène blaCTX-M au sein de clones E. coli épidémiques pourrait être le résultat d’une mobilisation de la cassette de transposition du gène dans des plasmides résidents et particulièrement bien adaptés à la bactérie hôte

Les travaux de cette thèse concernent l'étude de l'influence des conditions de dépôt sur la microstructure et les propriétés optiques de monocouches dans le but de réaliser des filtres optiques particuliers commercialisables. La méthode de dépôt utilisée est l'évaporation par canon à électrons avec assistance ionique. L'un des deux projets a été de vérifier la faisabilité de filtres optiques infrarouges dans la gamme [2-15] µm avec des matériaux non toxiques. Les monocouches des trois fluorures étudiées ont montré des comportements différents suivant les conditions de dépôt, notamment suivant la température du substrat qui influe fortement sur leur microstructure. L'association du BaF₂ au germanium, avec des réglages optimisés, montre pour différents filtres réalisés des résultats proches de la théorie. Ces filtres sont donc commercialisables. Le second projet a été de développer une machine de dépôt dans le but de pouvoir réaliser des filtres complexes d'une centaine de couches d'épaisseur variable (de 15 à 300 nm) associant des monocouches de TiO₂ à des monocouches de SiO₂. L'étude de l'influence des paramètres de dépôts (pression, assistance ionique, préchauffage du matériau) au travers différentes caractérisations (MEB, AFM, ellipsométrie) a permis une optimisation des réglages de l'enceinte de dépôt plus efficace et adaptée en fonction des spécifications des filtres optiques à réaliser. Puis, une étude ellipsométrique poussée de l'indice de réfraction des monocouches de TiO₂ en fonction de leur épaisseur a conduit à une nette évolution des résultats pour la réalisation du filtre complexe.

This thesis deals the study of deposition conditions 'effect on the microstructure and optical properties of monolayers in order to achieve marketable optical filters, with particular specifications. All filters and monolayers were deposited by ion-assisted electron-beam evaporation technique. One of the two projects was to test the feasibility of infrared optical filters in the range [2-15] µm with non-toxic materials. The study of monolayers of three fluorides showed different behaviors depending on the deposition conditions especially as a function of the substrate temperature that affects their microstructure. For different filters realized, the association of BaF₂ with germanium, settings of deposition optimized, shows results close to the theory and these filters are marketable. The second project was to develop a deposition chamber in order to realize complex filters built with one hundred layers of variable thickness (15 to 300 nm) associating monolayers of TiO₂ with monolayers of SiO₂. The study of the influence of deposition parameters (pressure, ionic assistance, preheating of material) through different characterizations (SEM, AFM, ellipsometry) allowed the optimization of deposition's parameter settings more effective, and adapted according to the specifications of the optical filters to realize. ▼ We have developed continuous random network (CRN) model based Metropolis Monte Carlo simulation tools which are capable of predicting the structural properties of amorphous semiconductor and oxide materials as well as their interface. To bolster the reliability of the CRN model, we have developed force fields based on gradient corrected density functional theory (DFT) calculations. Our in-house CRN-MMC tools have been massively parallelized, which allows us to create fairly large model structures within a reasonable computational time. Using the integrated CRN-MMC tools, we have elucidated the complex growth and structure of self-interstitial and vacancy clusters in silicon and the effect of strain on the structure and stability of the defect clusters. Our work for vacancy clusters suggests that small vacancy defects exclusively favor fourfold-coordination thermodynamically with no significant kinetic limitation rather than void-like structure formation, which has widely been adapted to explain the behavior and properties of vacancy defects. Our results also highlight the identification of stable high-symmetry fourfold-coordinated V₁₂ and V₃₂ clusters that could be expected to exist to a large extent in a vacancy rich region although its direct characterization appears impractical at present. Our work for self-interstitial clusters provides the first theoretical support for earlier experiments which suggest a shape transition from compact to elongated structures around n = 10. When the cluster size is smaller than 10, the stable I₄ and I₈ compact clusters are found to inhibit the formation of elongated defects, whereas the newly discovered fourfold-coordinated I₁₂ state is found to serve as an effective nucleation center for large extended defects. Our CRN-MMC approach also enabled us to elucidate the underlying mechanisms of synthesis and manipulation of Si rich insulators as well as the fundamental understanding of the relationship between the atomic structure and properties. We developed a valence force field based on a modified Keating model for the structure and energetics of amorphous Si rich oxide materials. In particular, our work emphasizes the importance of correctly describing the wide Si-O-Si angle distribution. Our work also suggests that the relative rigidity between Si and SiO₂ matrices is critical in determination of the Si/SiO₂ interface structure. The present potential model coupled with the CRN-MMC method can be used to create structural models (free of coordination defects) for complex a-SiO[subscript x]-based materials, which will further allow thorough studies of the properties of these materials.
