Увод: Биокомпатибилност се тумачи као способност материјала да делује уз адекватан одговор домаћина у одређеној ситуацији. Одговор домаћина на страно тело зависиће од његове хемијске реактивности, површинске структуре и топографије. Доказано је да наноструктурирана површина ближе подражава хијерархијску организацију коштаног ткива у односу на микроструктурирану, повећањем концентрације активних биомолекула који утичу на ћелијску миграцију, адхезију, диференцијацију и пролиферацију. Током деведесетих година, минерални триоксидни агрегат (МТА) представљен је као материјал избора за ретроградне кавитете због нижег апикалног микропропуштања, везивања у присуству влаге и високе pH вредности. Упркос повољним особинама, МТА показује суву конзистенцију, ниску вискозност и дуго времена везивања, што ограничава његову клиничку употребу. Циљ ове студије био је да се синтетишу наноматеријали на бази веома активних калцијум-силиката унапређених физичко-хемијских својстава и испитају њихова биокомпатибилност и биоактивност у односу на МТА+ (Cerkamed, Stalowa Wola- Пољска). Материјали и Метод: Поступак сол-гел методе у комбинацији са методом високо-температурне ланчане реакције сагоревања примењен је у току синтезе испитиваних материјала. Анализа фаза пре и после хидратација изведена је помоћу XRD и FTIR, док је морфологија узорака проучавана SEM и EDS. pH мерења су обављена коришћењем pH-метра, док је количина ослобођених јона одређивана коришћењем ICP-OES. Биокомпатибилност свежих материјала и њихових екстраката (24h, 7 и 21 дан) је испитана применом индиректног теста (МТТ), директног теста (LDH), теста пролиферације (3H-тимидин) и директном методом (Неутрално црвена). Интеракција директно примeњених испитиваних материјала са пулпним ткивом анализирана је на моделу културе хуманих зуба (15 и 30 дана). Осамнаест заморца служило је за интрамишићну имплементацију стерилних полиетиленских цевчица испуњених испитиваним материјалима (15, 30 и 60 дана). За интракоштани имплантациони тест употребљено је дванаест новозеландских белих зечева подељених у две групе (30 и 90 дана)...

Crvena detelina (Trifolium pratense L.) je izrazito stranooplodna biljna vrsta koja se odlikuje gametofitskim sistemom inkompatibilnosti, a populacije su heterogene i sastoje se od heterozigotnih genotipova. Da bi biljni genetički resursi mogli biti korišćeni u programima oplemenjivanja, neophodno je izvršiti prethodnu karakterizaciju i evaluaciju uzoraka, koji se nalaze u kolekcijama germplazme. Precizan i sveobuhvatan opis germplazme crvene deteline je jedan od osnovnih preduslova u ostvarivanju uspešnog oplemenjivanja. Cilj ove doktorske disertacije je obuhvatao evaluaciju fenotipske varijabilnosti 46 genotipova crvene deteline koji su deo kolekcije Instituta za ratarstvo i povrtarstvo u Novom Sadu, a na osnovu agronomski značajnih osobina i morfoloških markera po UPOV-u. Primenom odabranog seta od 15 mikrosatelitskih markera izvršena je molekularna karakterizacija 46 genotipova crvene deteline, kako bi se ustanovila genetička varijabilnost materijala i njegov značaj za oplemenjivanje na određene agronomski važne osobine. Osim toga, kod ispitivanih genotipova crvene deteline su određeni i značajni parametri kvaliteta: sadržaj sirovih proteina, vlakna nerastvorljiva u kiselom deterdžentu (ADF-acid detergent fiber) i vlakna nerastvorljiva u neutralnom deterdžentu (NDF- neutral detergent fiber). Na osnovu analize četiri morfološke osobine (forma rasta, maljavost stabljike, boja lista i intenzitet obojenosti pege na listu) i jedne fenološke osobine (vreme cvetanja), utvrđen je visok nivo diverziteta ispitivanih genotipova crvene deteline, a vreme cvetanja je određeno kao deskriptor najveće diskriminacione moći. Postignut je određeni stepen saglasnosti u grupisanju genotipova primenom analize homogenosti u odnosu na klaster analizu. Pri utvrđivanju morfološke sličnosti i određivanju osobina koje najviše doprinose diskriminaciji genotipova crvene deteline metod analize homogenosti se pokazao kao bolji u odnosu na klaster metod...

Este trabalho avaliou a bioacumulação de metais e as respostas biológicas em mexilhões Perna perna coletados na Praia da Cocanha, Ilha das Palmas e Ponta de Itaipu, e ostras Crassostrea brasiliana coletadas nos Estuários de Cananéia, Santos e Bertioga em diferentes estações do ano. Foram realizadas duas campanhas: 2008/09 e 2016/17. A Cocanha apresentou elevados níveis de metais, indução de EROD e DBF e efeitos peroxidativos e genotóxicos. Em Palmas houve ativação de EROD, DBF, GST e GSH. Itaipu evidenciou bioacumulação de metais, ativação de GST e GSH e danos no DNA. Em Cananéia houve bioacumulação de As e Cr, respostas das fases I e II e efeitos peroxidativos. Santos exibiu piores condições fisiológicas, com alterações bioquímicas em todas as estações do ano. Bertioga piorou em 2016/17 em relação a 2008/09, evidenciado por maior bioacumulação de metais e respostas de biotransformação e antioxidantes e efeitos neurotóxicos. Os dados evidenciam fontes de contaminação diferentes para cada local e variação de acordo com o período do ano em função de diversos fatores (e.g. pluviosidade, reprodução, turismo). Os locais escolhidos como referência demonstraram estar sob influência de fontes pontuais de contaminação. Deve-se considerar que Itaipu e Cananéia integram unidades de conservação.

The present study assessed the bioaccumulation of metals and the biological responses in mussels Perna perna from Cocanha Brach, Palmas Island and Ponta de Itaipu, and oysters Crassostrea brasiliana from the estuaries of Cananéia, Santos and Bertioga in different seasons. Two campaigns were carried out: 2008/09 and 2016/17. Cocanha showed high levels of metals, induction of EROD and DBF and peroxidative and genotoxic damages. Palmas exhibited induction of EROD, DBF, GST and GSH. Itaipu exhibited bioaccumulation of metals, activation of GST and GSH and DNA damage. Cananéia showed bioaccumulation of As and Cr, phases I and II responses and peroxidative damage. Santos exhibited the worst physiological conditions, with biochemical alterations in all seasons. Bertioga conditions worsened in 2016/17 compared to 2008/09, evidenced by the higher content of metals and biotransformation and antioxidant responses and neurotoxic damage. These data show different sources of contamination for each site and variation in accordance with the time of the year due to several factors (e.g. rainfall, reproduction, tourism). The sites chosen as reference may be influenced by point sources of contamination. It should be considered that Itaipu and Cananéia are within protected areas.

Dentre os fármacos, os antibióticos têm recebido atenção especial devido aos problemas que podem causar à biota aquática. O objetivo do presente estudo foi avaliar a citotoxicidade de Amoxicilina e Clavulanato de Potássio isolados e em associação para mexilhões Perna perna utilizando o ensaio do tempo de retenção do corante vermelho neutro, que avalia a estabilidade da membrana lisossômica de hemócitos dos organismos teste. A Amoxicilina causou citotoxicidade aos mexilhões nas concentrações de: CEO: 1 ng.L-1, CI25-24h: 0,44 ng.L-1, CI25-48h: 1,19 ng.L-1 e CI25-72h: 0,85 ng.L-1, o Clavulanato de Potássio foi citotóxico nas concentrações de: 10 ng.L-1 em 24h e 50 ng.L-1 e 100 ng.L-1 em 48h e 72h. Os valores de concentração inibitória foram de CI25-24h: 3,11 ng.L-1, CI25-48h: 3,45 ng.L-1 e CI25-72h: 3,43 ng.L-1. No ensaio realizado com a associação dos fármacos todas as concentrações foram citotóxicas aos mexilhões em 48h e em 72h apenas 40 ng.L-1 de Amoxicilina + 10 ng.L-1 de Clavulanato de Potássio e 200 ng.L-1 de Amoxicilina + 50 ng.L-1 de Clavulanato de Potássio. As concentrações inibitórias foram: CI25-48h: 1,67 ng.L-1 e CI25-72h: 1,36 ng.L-1 a partir dos dados de Amoxicilina e CI25-48h: 0,42 ng.L-1 e CI25-72h: 0,34 ng.L-1 a partir dos dados de Clavulanato de Potássio.

Pharmaceutical compounds are identified in environmental matrices in orders of magnitude from ng.L-1 to μg.L-1. Among the drugs, antibiotics have received special attention due to the problems that can cause to aquatic biota. The aim of this study was to evaluate the cytotoxicity of Amoxicillin and Potassium Clavulanate in isolated and associated forms to marine mussels Perna perna through the neutral red retention time assay which assesses the stability of hemocytes lisossomal membrane of test organisms. Amoxicillin caused cytotoxicity to the mussels in concentrations of: OEC: 1 ng.L-1, IC25-24h: 0.44 ng.L-1, IC25- 48h: 1.19 ng.L-1 and IC25-72h: 0.85 ng.L-1, Potassium Clavulanate was cytotoxic at concentrations of 10 ng.L-1 in 24h; 50 ng.L-1 and 100 ng.L-1 at 48h and 72h. The inhibitory concentration values were IC25-24h: 3.11 ng.L-1, IC25-48h: 3.45 ng.L-1 and IC25-72h: 3.43 ng.L- 1. The test conducted with the combination of drugs all concentrations were cytotoxic to mussels in 48h and 72h only 40 ng.L-1 Amoxicillin + 10 ng.L-1 and Potassium Clavulanate 200 ng.L-1 Amoxicillin + 50 ng.L-1 Potassium Clavulanate. The inhibitory concentrations were IC25-48h: 1.67ng.L-1 and IC25-72h: 1.36 ng.L-1 from the data of Amoxicillin and IC25- 48h: 0.42 ng.L-1 and IC25-72h: 0.34 ng.L-1 from the Potassium Clavulanate data.

▼ A aroeira (Myracrodruon urundeuva) é uma árvore cujas folhas vem sendo estudadas com fins terapêuticos na medicina e odontologia por apresentar potencial antiinflamatório e antimicrobiano, além de favorecer o processo de reparo e cicatrização. O objetivo do trabalho foi caracterizar quimicamente o extrato de aroeira e avaliar seu efeito sobre fibroblastos gengivais humanos. Foi realizado a caracterização química da aroeira por meio do processo de triagem cromatográfica que consistiu na coleta das folhas sadias, maceração em metanol (MeOH) 80%, partição entre os solventes hexano, acetato de etila (AcOEt) e n-butanol (n-(BuOH)) Em seguida, realizaram-se análises dos compostos químicos em cromatografia liquida de alta eficiência acoplado a detector de arranjo de fotodiodo HPLC-PAD, análises por espectrometria de massas MS e cromatografia liquida acoplada a espectrometria de massas (LC-MS). Posteriormente foi realizada a análise de citotoxicidade do extrato hidroalcoólico de aroeira em fibroblastos gengivais humanos (linhagem FGH). Para a realização dos experimentos foram plaqueadas 2x103 células/poço em placas de 96 poços. O meio de cultivo foi substituído por meio de cultura de Eagle modificado por Dullbecco complementado por 10% soro fetal bovino (SFB) em diferentes concentrações do extrato hidroalcoólico de aroeira (extrato bruto; 1:10; 1:100; 1:1000; 1:10000 e controle). As análises de viabilidade foram feitas nos tempos experimentais de 24, 48, 72 e 96 horas, por meio dos testes de redução do MTT (brometo de 3-(4,5-dimetiltiazol-2-yl)-2,5-difeniltetrazólio), captação do vermelho neutro e coloração pelo cristal violeta. A estatística foi realizada em análise de variância de 2 critérios seguido de uma análise de teste Tukey (p<0,05). Os compostos encontrados no extrato foram os derivados de ácido gálico, galotaninos, além de flavonoides. Para a redução do MTT os resultados mostraram uma ligeira oscilação da absorbância em todos os grupos nos períodos experimentais. Destaque para as diluições de 1:100, 1:1000 e 1:10000 que apresentaram, em geral para todos os períodos, os maiores valores; enquanto os grupos de extrato bruto e diluição 1:10 apresentaram valores inferiores aos outros grupos (p<0,05). Na análise da captação do vermelho neutro também houve uma leve oscilação dos valores de absorbância, no entanto os valores obtidos para o grupo do extrato bruto foram os menores em todos os períodos estudados (p<0,05). Enquanto os grupos controle e de diluições de 1:100, 1:1000 e 1:10000 apresentaram valores semelhantes sem diferença significante (p>0,05). Para o teste do cristal violeta, os resultados encontrados seguiram o mesmo padrão do vermelho neutro: valores de absorbância menores nos grupos do extrato bruto e 1:10, ao contrário dos outros grupos que apresentaram valores maiores (p<0,05). Em vista dos resultados, podemos concluir que o extrato metanólico de aroeira apresenta em sua composição taninos, flavonóides e derivados de ácido gálico e de galotaninos. ▼ Tutkielma tarkastelee uusmaterialistisen kirjallisuudentutkimuksen mahdollisuuksia analysoimalla kielen mekanismeja William S. Burroughsin trilogiassa The Red Night Trilogy (1981-1987). Lähestyn kohdetekstiä paitsi uusmaterialistisen teorian, myös Burroughsin oman kielifilosofian kautta. Tutkielmani pyrkii tuottamaan uutta tutkimusta kahdesta näkökulmasta, sillä uusmaterialistisen kirjallisuudentutkimuksen perinne on toistaiseksi liki olematon. Vastaavasti burroughslaista kielifilosofiaa ei ole riittävästi päivitetty kirjailijan koko tuotannon laajuuteen, mihin sen päättävän trilogian tarkastelu uusmaterialistisesta näkökulmasta tarjoaa mahdollisuuden. Muodostan tutkielmassani hypoteesin kolmesta kielellisestä strategiasta, joiden toimivuutta koettelen erillisissä analyysiluvuissa. Niistä ensimmäinen, karnevalismi, tarkastelee kerronnallistetun kielen rakennetasoa. Tarkoituksena on osoittaa, kuinka trilogiaan muodostuu keinotekoisuuttaan manifestoiva tekstuaalinen taso, joka raottuessaan avaa materialle väylän tekstiin. Toinen kielistrategia hybridismi kiinnittyy kielen aihetasoon. Sen avulla tarkastelen sitä, kuinka kieli kykenee rikkomaan itse rakentamiaan konsepteja ja sekoittamaan näin omia mekanismejaan. Kolmas kielistrategia katafaattisuus toteutuu lähimpänä kieltä, sanan tasalla. Sen kautta ehdotan trilogian kielen kykenevän tuottamaan materiaalista meditaatiota, joka johtaa materian radikalisoitumiseen tekstipinnan alla. Tutkielmani metodologiassa korostuu prosessuaalisuus, joka on uusmaterialismissa olennainen menetelmä. Prosessuaalisuus toteutuu ensinnäkin kolmen kielistrategian skaalautuvassa liikkeessä kerronnallistetuista rakenteista kohti kieltä itseään. Lisäksi käyttämäni teoriakenttä on kokeileva, sillä sovellan tutkielmassani uusmaterialistisen teorian lisäksi karnevalismitutkimusta, posthumanistista teoriaa hybrideistä ja keskiaikaisen mystisen kieliperinteen tutkimusta. Uusmaterialismi julistaa pyrkivänsä voimakkaaseen epämetodologisuuteen, mikä antaa sallivan taustan tällaiselle teoriakenttien luovalle ja innovatiiviselle käytölle. Metodiikan lisäksi prosessuaalisuus ilmenee myös asennoitumisena trilogiaan itseensä, jonka ehdotan olevan eräänlainen prosessuaalisesti operoiva tekstuaalinen kone. Tarkoituksenani on kysyä, missä määrin uusmaterialistinen materia eli materiaalinen voi päästä osaksi kielellistä esitystä. Uusmaterialismin materiakäsitys edustaa ontologista monismia asettaen dualististen ajattelumallien romahduttamisen lähtökohdakseen. Kieli ei kuitenkaan ole toistaiseksi ollut tervetullut osaksi tuota monistista materiaa, mikä johtaa monismin toteutumisen kyseenalaistumiseen. Samoin burroughslainen kielifilosofia suhtautuu kieleen hyvin pessimistisesti. Burroughsille kieli näyttäytyi kontrolloivana ja vierauttavana viruksena. Tutkielmani etsiikin näille molemmille kieleen kielteisesti suhtautuville lähtökohdille positiivisia arvoja. Syfte och frågeställningar Studien syfte var att undersöka kunskapen om RED-S bland tränare inom svensk truppgymnastik och få en översikt om tränarna kände till Svenska Gymnastikförbundets eller Internationella Olympiska Kommitténs åtgärdsplan för identifiering, behandling och återgång till idrottande. Detta undersöktes med följande frågeställningar: 1) ”Hur mycket kunskap om RED-S har dagens tränare inom truppgymnastik?” 2) ”Skiljer tränares kunskaper åt beroende på kön, division och erfarenhet i år?” och 3) ”Har tränarnas förening en åtgärdsplan för RED-S?”. Metod En digital enkät baserad på liknande studier och IOC’s rekommendationer, skickades ut till både en sluten Facebookgrupp och via mail till föreningar på subelitnivå. Enkäten bestod av 28 frågor där de första åtta frågorna riktade sig mot tränarnas karaktärsdrag och de följande tjugo frågorna riktade sig mot kunskaper kring RED-S enligt IOC’s rekommendationer. All data analyserades i Google Excel och via SPSS. För att avgöra kunskapsnivån bland tränarna skapades ett poängsystem för frågorna om RED-S medan en icke parametriskt T-test genomfördes för att upptäcka eventuella kunskapsskillnader mellan gruppfördelningarna kön, division och erfarenhet. Resultat Inga tränare följde en åtgärdsplan för RED-S och ingen kände till Gymnastikförbundets eller IOC’s åtgärdsplan för RED-S. Det fanns inga kunskapsskillnader mellan gruppfördelningarna och utifrån resultaten är tränarnas kunskaper högre i frågor om återgång och behandling, men lägre i frågor om identifiering och grundläggande kunskaper. Av 35 deltagande var det endast 20% som tidigare hört talas om RED-S. Slutsats Denna studie upptäckte att det fanns kunskaper om RED-S men inte tillräckligt för att tränare skulle kunna hjälpa gymnaster som är utsatta eller löper risk för RED-S. Däremot var intresset att lära sig mer om ämnet högt bland deltagarna. På grund av bristande deltagande är studien inte representativ för svenska trupptränare på subelitnivå och kräver fler studier för att fastställa resultaten.

▼ Bioelectrochemical systems (BESs) are bioreactors that use bacteria as catalysts to drive oxidation and/or reduction reactions at solid-state electrodes. They are considered as a promising technology for energy efficient wastewater treatment, power production, bioremediation, and the production of valuable chemicals. Currently, the practical application of this technology is limited by low current and power densities. Electrode surface modification has been proved an efficient way to enhance of the performance of BESs. However, there is still limited knowledge on the effects of surface chemistry on the biofilm formation and electron transfer. Moreover, most of the reported electrode modification methods are either complex, expensive or unscalable. There is a need to fill in some of these knowledge gaps and to develop simple, effective, and scalable surface modification methods to enhance the bioanode current output of BESs. Therefore, the objectives of this work were: (i) to clarify the effects of surface chemistry of carbon electrode on anodic biofilm formation and current generation in BESs; (ii) to develop a quick approach to achieve desirable surface chemical properties on carbon electrode for faster anodic biofilm formation; (iii) to invent a novel strategy to graft electron transfer mediator to carbon electrode surface to enhance extracellular electron transfer (EET); (iv) to establish a simple method to make stainless steel (SS) surface biocompatible for scalable electrodes of BESs. To investigate the effects of surface charge and surface hydrophobicity on anodic biofilm formation and current generation in BESs, glassy carbon electrodes were modified with –OH, –CH3, –SO3-, or –N+(CH3)3 functional groups and then tested as anode in BESs. The results demonstrated that: (i) positively charged and hydrophilic surfaces were more selective to electroactive microbes (e.g. Geobacter) and more conducive for electroactive biofilm formation; (ii) The effects of the surface hydrophilicity were more pronounced than surface charge; (iii) differences in the maximum current output between surface modifications were correlated with biomass quantity; and (iv) all biofilms were dominated by Geobacter populations, but the composition of –CH3-associated biofilms differed from those formed on surfaces with different chemical modification. Now that the mode of action of the surface charge was established with expensive aryl diazonium salts, the next step was to perform the modification in a more cost effective, realistic manner. To convert carbon the felt surface into hydrophilic and positively-charged surface, a cationic surfactant, cetyltrimethylammonium bromide (CTAB) based treatment method was developed. In an acetate-fed bioanode, the start-up time of current production and the time to reach stable current output at the CTAB-treated felt anodes were 36.1% and 49.4% shorter than the untreated anodes, respectively. O fitoplâncton é sensível às perturbações ambientais e identificar a viabilidade destes organismos é importante para o monitoramento aquático. O termo viabilidade tem sido usado neste contexto para determinar basicamente se o organismo está vivo ou morto. O uso de corantes vitais e mortais tem sido uma das técnicas aplicadas neste tipo de análise, mas ainda não se conhece a potencialidade de alguns corantes que se apliquem a espécies de fitoplâncton. Neste trabalho, foram utilizadas espécies de diferentes grupos taxonômicos mantidas em cultivo, as quais foram submetidas a uma série de corantes vitais e mortais com o objetivo de detectar a viabilidade do fitoplâcton marinho. Dentre os corantes testados, o vermelho neutro, azul de Evans e o fluorescente CMFDA tiveram os melhores resultados (observado x esperado) em testes com diferentes porcentagens de células vivas e mortas. A combinação entre o corante vital vermelho neutro e o mortal azul de Evans não foi efetiva para análise simultânea. Alexandrium tamiyavanichii foi a espécie que teve menor afinidade com os corantes. Também foram comparados diferentes métodos de observação e registro de células coradas e não coradas, provando que é possível substituir a observação direta da microscopia pela filmagem no microscópio ou pela FlowCAM. A vantagem do uso destes métodos é que além de serem mais rápidos, é possível salvar as imagens capturadas e não é necessário fazer a análise instantaneamente. O método da filmagem é vantajoso, pois é fácil de ser desenvolvido em qualquer laboratório.

Phytoplankton cells are sensitive to environmental perturbations and, therefore, identifying their viability is important for aquatic monitoring. The term viability has been used in this context to determine whether an organism is alive or dead. The use of vital and mortal stains to detect phytoplankton viability is a promising approach. In this study we investigated the efficiency of several vital and mortal dyes in detecting marine phytoplankton viability. Best results were achieved with neutral red, Evans blue and the fluorescent stain in tests with different percentages of live and dead cells. The combination of neutral red and Evans blue (vital and a mortal stains, respectively) was not effective in simultaneous analysis. Alexandrium tamiyavanichii had low affinity for any given stain. Different observational methods were compared, suggesting that direct microscopic counts can be replaced by image acquisition methods using either a microscope-mounted camera or a FlowCAM. Such imaging methods are fast, allow image archiving, and image processing can be performed on a later stage, which is useful when several experiments need to be run in a short period of time.

Syfte: Att genom en litteraturstudie jämföra Schirmers tårtest (STT) och fenolröd tråd (PRTT), för att se hur god korrelation det är mellan de två testerna vid diagnostisering av torra ögon. Metod: Analysera och jämföra studier med hjälp av olika artiklar. Dessa artiklar har hittats genom sökning i databasen Google scholar med sökfraserna schirmer´s test versus phenol red thread och tear test correlations. Fem relevanta artiklar valdes ut för närmare granskning. Resultat: Det har framkommit varierande resultat angående huruvida det finns en god korrelation mellan Schirmers tårtest och fenolröd tråd. Slutsats: Efter att ha tittat närmare på de fem studierna så kan man konstatera att det behövs ytterligare studier av testerna Schirmers tårtest och fenolröd tråd, dessutom behövs det gemensamma gränser för vad som är torra ögon. När det gäller korrelationen mellan Schirmers tårtest och fenolröd tråd så är det svårt att ta ställning till det eftersom alla studier har olika urvalskriterier.

Objective: This literature review is a comparison of the Schirmer's tear test (STT) and the Phenol Red Thread Test (PRTT). The aim of the review is to find out if there was a good correlation between the two tests regarding diagnosing dry eye disease. Method: Analyze and compare studies using different articles. These articles were found by searching the Google Scholar. The search phrases were Schirmer´s test versus phenol red thread and tear test correlations. Five relevant articles were selected for further review. Results: There have been varying results regarding whether there is a good correlation between Schirmer's tear test and Phenol Red Thread. Conclusion: After a closer look at the five studies, it can be concluded that further studies of the Schirmer's tear test and Phenol Red Thread are required, in addition, there is a need of common cutoffs for diagnosing dry eye disease. It is hard to find a correlation between Schirmer's tear test and Phenol Red Thread, since all studies use different criteria for inclusion.

▼ Le paludisme est une maladie tropicale résultant de l'infection par le parasite Plasmodium falciparum transmis par piqûre de moustique. Les symptômes du paludisme résultent du développement de P. falciparum dans les globules rouges (GR). Depuis des siècles, le paludisme a exercé des pressions sur le génome humain, sélectionnant des mutations conférant une protection contre les formes sévères de la maladie. C'est le cas de la mutation du gène de l'hémoglobine (HbA), composant principal des GRs. La forme mutée du gène produit une hémoglobine anormale (HbS). Contrairement à la drépanocytose (HbSS), le portage hétérozygote (trait drépanocytaire) de cette mutation (HbAS) est asymptomatique. Les porteurs sains HbAS développent moins de symptômes graves du paludisme. Aujourd'hui, les mécanismes responsables de cette protection restent partiellement élucidés. Lors de son développement, P. falciparum modifie la membrane et le cytosquelette du GR afin d'exposer des protéines parasitaires à la surface de l'hématie. Parmi ces protéines, l'adhésine majeure parasitaire "P. falciparum erythrocyte membrane protein 1" (PfEMP1), se lie aux récepteurs endothéliaux, entraînant la cytoadhérence et la séquestration des GRs infectés (iGRs). Cette cytoadhérence permet d'éviter le passage et la clairance splénique des iGRs. Des études ont montré que les iGRs HbAS cytoadhèreraient moins, en association avec une présentation anormale de PfEMP1. Cette thèse porte sur l'étude des mécanismes de résistance du trait drépanocytaire contre le paludisme à P. falciparum. Le premier projet de cette thèse porte sur le phosphoprotéome des membranes de GRs HbAA et HbAS infectés par P. falciparum. Les protéines parasitaires exposées à la surface du GR interagissent avec des protéines érythrocytaires impliquées dans l'ancrage de la membrane du GR au cytosquelette. Il s'agit des protéines du complexe Ankyrine-R et du complexe jonctionnel. Le stress oxydant généré par le trait drépanocytaire et par l'invasion parasitaire perturbe l'équilibre kinase/phosphatase dans la cellule, pouvant entraîner des modulations de la phosphorylation des protéines, interférer dans les interactions protéiques et par conséquent dans la présentation des antigènes parasitaires. Des extraits membranaires de GRs HbAA et HbAS infectés ont été produits et analysés en spectrométrie de masse et en Western-Blot. Cette étude a montré que le trait drépanocytaire modulait la phosphorylation des protéines érythrocytaires de la membrane du iGR (transporteurs membranaires et protéines du cytosquelette majoritairement), mais aussi celle de protéines parasitaires. Le deuxième projet porte sur la réponse anticorps anti-VAR2CSA dans le cadre du paludisme gestationnel selon le portage de l'HbS. Le paludisme gestationnel est une des formes sévères du paludisme, due à la cytoadhérence des iGRs dans le placenta. A utilização de resíduos perigosos em produtos construtivos, técnica denominada estabilização por solidificação (E/S), requer um conhecimento aprofundado de sua composição, viabilidade técnica e econômica, além de uma avaliação ambiental ao longo do uso e pós-uso do produto. Quando se trata da avaliação de um produto da E/S durante a fase de uso, o Brasil não dispõe de um procedimento adequado que considere a integridade física da amostra e as características do cenário onde o resíduo encapsulado se encontra. O objetivo geral desta pesquisa é avaliar a lixiviação de cromo total em monólitos de cerâmica vermelha durante a fase de uso, por imersão e irrigação. Foram confeccionados corpos de prova com adição de 0,05% de óxido de cromo III (Cr2O3), além de corpos de provas referência (sem adição Cr2O3). Com base no regime pluviométrico da região metropolitana de Porto Alegre/RS desenvolveram-se dois testes de lixiviação para a avaliação da efetividade da E/S: imersão e irrigação. O primeiro com o propósito de simular um ambiente hostil em situação de prolongado contato da cerâmica com o lixiviante e o segundo para uma condição de exposição à chuva ácida ou ciclos de molhagem e secagem, possibilitando a avaliação do desempenho ambiental do material. A E/S do Cr na cerâmica vermelha foi acima de 99,90%. Nos corpos de prova com Cr2O3 a concentração de Cr lixiviado foi de 0,1243 ppm no teste de imersão, 0,01183ppm no teste de irrigação e 0,023 ppm no teste de lixiviação da norma ambiental brasileira para resíduos sólidos. As duas formas de avaliação da lixiviação de cromo total em monólitos de cerâmica vermelha, durante a fase de uso, propostas nesta pesquisa, apresentaram baixo coeficiente de variação e resposta técnica em 28 dias, representaram o fenômeno da difusão e se diferenciaram em seus cenários.

▼ Shade stress on creeping bentgrass (Agrostis stolonifera) causes elongation of leaves, reduced energy availability for growth and lower levels of disease resistance resulting in poor quality putting surfaces. Decreased quantity or photosynthetic photon flux density (PPF) and quality or the red:far red light ratio (R:FR) of light affects the morphology of turfgrass plants by increasing production of giberellic acid and reducing available non-structural carbohydrates. The object of the study was to determine which source of nitrogen (N) is best suited to retaining turfgrass quality in shade and to see if the plant growth regulator trinexapac-ethyl [4-(cyclopropyl-α-hydroxy-methylene)-3,5-dioxo-cyclohexane carboxylic acid ethyl ester] (TE) (Syngenta AG, Switzerland) can improve density and combat the loss of surface quality of bentgrass under tree shade. Field studies were conducted on ‘Penncross’ creeping bentgrass (Agrostis stolonifera) in Columbus, in 2006, and 2007. Under three light environments: full sun (FS), reduced R:FR (DS), and reduced PPF (NS). Light reduction was >90% in both shade areas and the R:FR ratio was .38 in the DS area, 1.28 in NS, and 1.29 in FS. The three N treatments included (NH₂)₂CO, Ca(NO₃)₂ and (NH₄)₂SO₄ foliarly applied weekly at a rate of 0.43g m^-2 with or without TE applied bi-weekly at 0.3975 L ha^-1. Chlorophyll content, along with non-structural carbohydrates, flavonoids, turf canopy color, dry matter and canopy densities were measured. There were no differences in soluble carbohydrate levels between treatments. Chlorophyll levels varied between TE treated and non-treated areas. Applications of (NH2)2CO consistently resulted in higher content of N15 in turfgrass plants also but there was no effect in nitrogen uptake kinetics due to light environment differences. TE had a negative impact on quantities of flavonoids. There was variation between the N treatments in relation to dry matter and carbohydrate content. (NH2)2CO in combination with TE provided the best visual quality surface as the season progressed. TE was effective in reduced PPF conditions and R:FR conditions which would indicate it as a possible product for use in shade management of creeping bentgrass Agrostis stolonifera. It was equally efficient in both light environments suggesting that prior shade research using NS is relevant to shade management in private industry. There was not a strong indication as to a preferred source of N.(NH₂)₂CO had positive impacts on total chlorophyll content. Advisors/Committee Members: Gardner, David (Advisor).

▼ Radicals are potential building blocks to prepare conductive and magnetic materials. In order to achieve high conductivity, materials displaying a large bandwidth W and a low on-site Coulomb repulsion energy U must be generated. Semiquinone-bridged bis-1,2,3-dithiazolyl radicals (R = Cl, Ph, Me and the MeCN adduct of R = Cl) represent a new family of resonance stabilized neutral radical for use in the design of single-component conductive materials were prepared and fully characterized. In solid state these radicals remain as unassociated (monomers) in the solid state and typically form superimposed alternating π-stacks or slipped π-stacks, arranged in several different space groups. The predominate intermolecular interactions are S•••N′ and / or S•••O′ contacts, which increase the dimensionality from one dimensional π-stacked systems (i.e., poor lateral overlap) to two dimensional systems in the solid state. Thus the semiquinone-bridged bis-dithiazolyl radicals exhibit a significant decrease in activation energy (ca. 0.1 – 0.2 eV) and the conductivity is two to three orders of magnitude (ca. σ ≈ 1E-5 – 1E-2 S / cm) higher in comparison to the previously reported pyridine based systems. This high conductivity is attributed to the low on-site Coulomb repulsion energies (U) which were estimated from the solution cell potentials (EPC) obtained from CV measurements and improved bandwidth (W) from the S•••N′ and / or S•••O′ interactions. Furthermore, the all sulphur containing semiquinone-bridged bis-dithiazolyls have the lowest activation energies and the highest conductivity under ambient conditions compared with other all sulfur nitrogen based radicals known to date. The semiquinone-bridged bis-dithiazolyl (R = Cl) orderes as spin-canted antiferromagnets, TN = 8 K, and displayed large coercivity (80 Oe). The ZFC-FC measurement at low field (i.e., H = 100 Oe) established the phase transition temperatures and the spontaneous magnetization was used to estimate the spin canting angles (~ 0.14°). In the case of R = Ph, the antiparallel alignment of the ferromagnetic coupled chains leads to a spin-canted antiferromagnet (TN = 4.5 K), which undergo a unique field induced spin flop transition. The MeCN solvated of R = Cl behaves as a simple paramagnet at room temperature with bulk antiferromagnetic interactions, but no observed magnetic ordering from 2-300 K.

▼ HIV/AIDS is the most threatening and challenging infectious diseases of our time, with the highest increase of newly infected cases reported. This infectious disease was discovered in the early eighties under homosexual men and was later to be discovered in heterosexuals. HIV is a systemic immunosuppressive disorder which causes a depletion of CD4+ T cells and develops into the acquired immunodeficiency syndrome - AIDS. Africa is the continent most affected by HIV/AIDS with the southern parts of Africa having the highest prevalence rates compared to the rest of Africa. Statistics indicate that AIDS is responsible for 3% of deaths in children worldwide - one in seven people dying of an HIV-related illness is a child under the age of 15 years. It was stated by the WHO that countries should develop improved antiretrovirals regimes for the prevention of mother-to-child transmission. Difficulties in administering antiretrovirals (ARVs) to patients (especially children) are the strict dosage regimes and the severe adverse reactions. These factors complicate patient adherence. The list of problems in treating patients is endless and includes the distribution, stability as well as the low efficacy of these drugs. Most of the above mentioned problems and obstacles related to ARVs and ARV treatment could be minimized or eliminated by the use of a stable and effective drug delivery system. Enhancing ARV treatment may be accomplished by the use of the Pheroid™ drug delivery system. Pheroids™ consists mainly of fatty acids and sterile nitrous oxide gassed water. Pharmacological active substances are entrapped into submicron and micron sized structures called Pheroids™. Research showed promising results and advantages in delivering drugs through oral and transdermal routes using Pheroid™ technology. The focus of this study was to test the possible enhancement of the efficacy of antiretrovirals using Pheroid™ technology. The assays used to study this possible enhancement were a modified neutral red and a modified 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium (MTT) assay. These assays confirmed and illustrated the toxic and protective properties of the tested ARVs (stavudine, lamivudine and nevirapine). An MT-2 cell line was used and infected with an HIV-1 strain, SW7-TCL. Applying Pheroid™ technology in these assays resulted in massive cell death, due to increased ARV toxic levels within the cells. Viability tests proved that Pheroids™ had no effect on the viability of cells at the concentration typically used. This confirmed the enhancing properties of Pheroids™ in the delivery of drugs into the cells. The MTT assay was further adapted from a seven day incubation period to a three day incubation period. By using a low concentration series and a three day incubation period the loss of cells through toxicity was partially overcome.
