▼ The active synthetic constituent of oral contraceptives 17α- ethinylestradiol (EE2)
and its natural steroidal estrogen analogues are being released into the aquatic
environment mainly via wastewater treatment plants (WWTPs). Although steroidal
estrogens have been frequently reported in very low concentrations in the environment,
they have been placed at the top of the list of endocrine disrupting chemicals (EDCs) for
their high estrogenic activity in non-target aquatic species like fish and frogs. Almost 30
years worth of intensive research has moved the problem of endocrine disruption in the
aquatic environment from being a hypothesis to a well-known cause-and-effect story.
However, the fate and effects of many pharmaceuticals, including EE2, remain poorly
understood. Thus, the main objective of this thesis was to investigate EE2
bioaccumulation in fish by field and laboratory studies.
An optimized sample preparation and analytical method protocol was achieved to
detect EE2 in its biologically active form in whole fish tissue at trace levels (ng/g).
Shorthead redhorse suckers (ShRHS) (Moxostoma macrolepidotum) collected in
proximity to a WWTP in the St. Clair River have significantly accumulated EE2 with an
average of 1.5 ng/g. EE2 bioaccumulation was limited to fish exhibiting intersex and with
induced vitellogenin (VTG). EE2 was absent in fish from a reference site. Positive
correlations between EE2 and lipid content as well as "15N supported the hypothesis of
EE2 bioaccumulation in wild ShRHSs. A wider survey was carried out to explore
patterns of EE2 bioaccumulation in the pelagic and benthic food-wed of the St. Clair
IV
River, Ontario. In the WWTP effluents and nearby surface waters, EE2 levels were
extremely low and below our method detection limits. All of the seven sampled species
from different trophic levels and the sediment samples collected from the impacted sites
and the reference sites did not have any measurable EE2 concentrations. A laboratory
controlled study where male goldfish (Carassius auratus) were exposed to EE2 via water
versus food demonstrated that fish can rapidly accumulate EE2 by both routes of
exposure. The uptake constant for water was 45 h-1 and the bioconcentration factor for
EE2 in fish was 377. The fast uptake rate of EE2 via water coincided with a much slower
elimination rate constant of 0.0786 h-1. The assimilation efficiency of 0.106 for EE2 by
goldfish was determined by EE2 dietary exposure, which was used to predict EE2
accumulation under different exposure scenarios. The work presented here was the first
demonstration of EE2 bioaccumulation in wild fish and the first to model EE2
bioaccumulation from water and dietary exposure in laboratory-exposed fish.

▼ The objective of this thesis was to investigate the reproductive effects and mechanisms of action of three common MWWE constituents, 17α-
ethinylestradiol (EE2), nitrate and ammonia, using the zebrafish (Danio rerio) as the model. Fecundity assays indicated that ammonia and EE2, but not nitrate, inhibited spawning. Further mechanistic investigations into reproductive processes such as steroidogenesis, maturation, and ovulation indicate that ammonia and EE2 affect reproduction through different mechanisms. EE2 acts by inhibiting steroidogenesis as shown by the reduction of sex steroid levels and expression of aromatase. Ammonia blocked the stimulatory effects of the gonadotropin analog human chorionic gonadotropin (hCG) on ovulation and spawning. Molecular analysis from these experiments indicated a potential role of prostaglandins in this response through the actions of the enzyme COX-2, but no effects on prostaglandin levels or receptor expression were found. Though the mechanism through which ammonia inhibits reproduction remains unknown, these findings help narrow the likely sites of action.

▼ In rivers, association of organic contaminants with dissolved organic carbon may limit freely dissolved or bioavailable fractions and toxicity of organic contaminants. Consequently, assessment of toxicity of organic contaminants on the basis of their total chemical concentrations may lead to overestimation of risks to organic contaminants. Therefore, to achieve reliable and accurate risks assessment for organic contaminants, determination of bioavailability is important. The influence of humic acid on the bioavailability of organic contaminants in rivers was studied, using three chemicals with different properties as model contaminants, which at the start of the study were detected in wastewater effluents. It was hypothesized that in the presence of dissolved organic carbon, a fraction of the total concentration of an organic contaminant would not be bioavailable in river water. Therefore, the aim of the study was to determine bioavailability and its impact on toxicity. Bioavailability in the presence of humic acid was determined chemically and using a yeast estrogen screen assay. The chemical method comprised solid-phase extraction and liquid chromatography-mass spectrometry to determine freely dissolved and the fraction of the chemicals associated with dissolved organic carbon. The results indicated increased binding to dissolved organic carbon with the hydrophobicity of the test compounds except for perfluorooctane sulfonate. The dissolved organic carbon-water partition coefficient for ethinylestradiol was determined to be Log KDOC 2.36. Log KDOC values of 4.15 and 4.41 at 10 and 100 mg/L humic acid, respectively, were derived for hexabromocyclododecane indicating greater binding than ethinylestradiol due to the more hydrophobic character. The yeast estrogen screen was used as a biological method to measure the effect of humic acid on the bioavailability of ethinylestradiol and a more hydrophobic compound, dichlorodiphenyltrichloroethane. Results of the yeast estrogen screen indicated that the presence of humic acid had no effect on bioavailability of either of the chemicals.

▼ Testicular oocytes (TOs), a form of gonadal intersex, have been found in black basses (Micropterus spp.) from many locations in North America. The presence of TOs is often assumed to imply exposure to estrogenic endocrine disrupting compounds (EDCs), however, a definitive causal relationship has yet to be established, and TO prevalence is not consistently low in fish from areas lacking evident EDC sources. This could suggest unknown or unidentified EDC sources, or that TOs can be stimulated by other stressors, or that they arise spontaneously during normal development. The goal of this dissertation research was to improve our ability to interpret observations of TOs in smallmouth bass (M. dolomieu) in particular, with approaches that are applicable to any other species. These approaches included statistical analysis of TO occurrence in wild populations where estrogenic exposure is more and less likely, and laboratory studies to characterize the early gonadal development in smallmouth bass, both in the presence and absence of exogenous estrogen. TO occurrence in smallmouth bass was assessed in eight watersheds in Northeastern Minnesota watersheds with differing degrees of human development, and hence, likelihood of EDC exposure. Testicular tissues from mature fish were evaluated using a semi-quantitative method that estimated TO density, normalized by cross-sectional area. TO prevalence and density among populations from more developed watersheds were higher than in populations from less developed watersheds. However, TO prevalence was unexpectedly high and variable (7-43%) in some populations from less developed watersheds, and only weak evidence was found for a relationship between TO density and watershed development, suggesting alternative or more complex explanations for TO presence in smallmouth bass. To characterize early gonadal development in smallmouth bass, both in the presence and absence of estrogen, swim-up fry were exposed to control water or the potent model estrogen, 17α-ethinylestradiol (EE2), for 90 days, and reared for another 90 days in clean water. Development of gonadal tissue was assessed at 11 time points using standard histology methods. Ovarian differentiation was observed in 50% of control fish by test day 49, and testicular differentiation occurred by day 92. Among EE2-exposed groups, ovarian phenotypes were observed in 83% of fish by day 49, and in 98% of fish between days 92 and 180, indicating that EE2 can cause complete, and likely permanent, sex reversal in smallmouth bass. Among the EE2-exposed fish that developed ovaries between days 92 and 180, 38% had abnormal characteristics, suggesting that ovaries in sex reversed males can be distinguished from ovaries in genetic females. In a second laboratory study, smallmouth bass gonadal development was assessed during early life stage exposure across a range of concentrations of EE2, including environmentally realistic concentrations. 97% of fish exposed to 4.3 ng/L EE2 (mean measured concentration) had ovaries on test days…

Resumo: Defeito do tubo neural (DTN) trata-se de uma malformação congênita resultante de um mal fechamento do tubo neural. Ocorre quando a placa neural embrionária falha em completar seu desenvolvimento e não se fecha em algum ponto ao longo do seu crescimento, por volta da quarta semana do desenvolvimento embrionário. Trata-se de uma má formação fetal que pode ser evitada com a utilização do ácido fólico desde o período pré concepcional até os primeiros meses de vida da criança. O folato é essencial e sua deficiência pode ser causada por dieta inadequada, por fatores genéticos ou pela interação de fatores genéticos e
ambientais. A suplementação com ácido fólico previne 50-75% dos casos de defeitos do tubo neural. Este estudo objetivou avaliar a viabilidade da realização de suplementação do ácido fólico juntamente com anticoncepcional com a administração de etinilestradiol 0,02 mg + levonorgestrel 0,10 mg + ácido fólico 0,4 mg em comprimidos de administração oral, com base em comparações estatísticas dos níveis de folato no sangue. As concentrações foram medidas através de método analítico apropriado e validado, bem como avaliado sua segurança e tolerabilidade em voluntárias sadias do sexo feminino. Trata-se de um estudo de Fase II, monocêntrico, aberto, com um tratamento em três ciclos, com administração em doses múltiplas, nos quais as voluntárias recebem o produto teste e posteriormente o placebo. As trinta e seis voluntárias saudáveis receberam um comprimido revestido do produto teste - etinilestradiol 0,02 mg + levonorgestrel 0,10 mg + ácido fólico 0,4 mg por 21 dias e um placebo revestido
contendo apenas 0,4 mg de ácido fólico somente nos últimos 7 dias de cada ciclo de tratamento, que foram realizados em 3 ciclos. Amostras de sangue foram coletadas para quantificar o folato por cromatografia líquida acoplada a espectrometria de massa (LC-MS / MS), para mensuração do níveis de folato no sangue antes da primeira administração do primeiro, segundo e terceiro ciclo, após 21 dias do primeiro, segundo e terceiro ciclo e após 28 dias do primeiro, segundo e terceiro ciclo. A segurança foi avaliada pelo registro de eventos adversos, monitoramento de sinais vitais e avaliação de exames laboratoriais e ECG. A média do nível de ácido fólico no sangue no início (1º dia do 1º ciclo) foi de 42,7 ± 22,2 ng/mL enquanto no 28º dia do 3º ciclo foi de 47,6 ± 20,1 ng/mL, com uma variação de 11,32%. As voluntárias toleraram bem o protocolo clínico e não relataram efeitos adversos clinicamente significativos, apenas uma voluntária, por motivos particulares, abandonou o estudo durante a

▼ Les réseaux neuroendocrines qui contrôlent la reproduction sont mis en place pendant le développement sous l’action des stéroïdes sexuels endogènes. Toute perturbation de l’équilibre hormonal pendant ces phases critiques pourrait être à l’origine de troubles de la fonction de reproduction chez l’adulte. Ce travail vise à identifier l’impact d’une exposition développementale à un oestrogène de synthèse, l’éthinyloestradiol (EE2), sur les réseaux neuroendocrines et les conséquences physiologiques et comportementales chez l’adulte et sa descendance. Nos résultats ont montré que l’EE2 induit des perturbations des comportements sexuels chez les mâles et chez les femelles et une modification des réseaux hypothalamiques à GnRH et des neurones à calbindine qui contrôlent la physiologie et les comportements reproducteurs. Nous avons également montré que certains effets de l’EE2 sont transmis jusqu’à la quatrième génération, mettant ainsi en évidence le caractère transgénérationnel de ces perturbations. Ces résultats mettent en évidence la sensibilité des réseaux neuroendocrines aux perturbateurs endocriniens et la nécessité de prendre en compte ces paramètres dans l’évaluation de leurs effets sur la santé et la reproduction.
Neuroendocrine networks controlling reproductive function are established during development by the action of endogenous sex steroids. Any disturbance in the hormone balance during these critical phases may cause several disorders in reproductive function in adulthood. This work aims at identifing the consequences of a developmental exposure to the synthetic pharmaceutical estrogen ethinylestradiol (EE2) on the neuroendocrine and behavioral outcomes of the reproductive function in adult individuals and their offspring. Our findings showed that EE2 induced disturbances of sexual behaviors in males and females and modified the GnRH and the calbindin hypothalamic networks of exposed animals. We also showed that some effects of EE2 were transmitted up to the fourth generation, pointing out the transgenerational character of certain effects. All these results highlight the sensitivity of neuroendocrine networks to endocrine disruptors and the need to consider these parameters in assessing their effects on health and reproduction.

▼ Actuellement, les systèmes d’épuration des eaux n’ont pas été conçus pour éliminer les substances médicamenteuses notamment les hormones (estrogènes). Ces molécules sont ainsi déversées dans le milieu naturel et contaminent les rivières. Le 17 α-éthinylestradiol a été choisie comme molécule modèle pour illustrer la présence d’un perturbateur endocrinien dans une eau résiduaire. Cette molécule présente un réel risque pour la faune et la flore (Féminisation des poissons mâles, diminution de la reproduction…). L’objectif de ce travail est d’évaluer l’efficacité de la photocatalyse et de l’ozonation pour éliminer l’Ethinylestradiol dans les eaux. Le procédé d’oxydation utilisant l’ozone a été appliqué dans un premier temps sur des solutions concentrées d’éthinylestradiol (EE2 = 10 mg/L) préparées dans une eau ultrapure tamponnée afin de mieux comprendre les mécanismes d’élimination de ce composé et d’évaluer l’efficacité en terme de taux de conversion de EE2 et de la minéralisation du substrat. Les expériences réalisées ont permis de montrer que l’ozone est un oxydant très efficace pour oxyder l’éthinylestradiol. L’oxydation se fait essentiellement par réaction directe de l’ozone sur la molécule d’EE2 alors que sa minéralisation nécessite la conversion de l’ozone en radicaux hydroxyles qui vont ensuite réagir sur la structure saturée de la molécule organique. L’étude de l’ozonation de solutions EE2 à différentes concentrations a permis d’établir une relation entre la concentration initiale en polluant dans l’eau et la dose d’ozone à introduire pour éliminer ce composé à un pH proche de la neutralité. Cette relation a été utilisée pour estimer la dose d’ozone à appliquer pour une eau contaminée par EE2 à une concentration proche de celle rencontrée dans les eaux résiduaires et comparée à celle obtenue sur une eau provenant de la station d’épuration du Bourget du Lac. L’efficacité du procédé photocatalytique a été évaluée en travaillant sur les vitesses de disparition et de minéralisation de EE2 en présence de TiO2 «Degussa P25» et TiO2 PC500. Les cinétiques d’adsorption et de dégradation photocatalytiques ont été évaluées dans une gamme de concentration allant de 1 à 10 mg/L en travaillant à différentes masses de photocatalyseur. Les constantes d’adsorption et de vitesse, modélisés avec les modèles de Langmuir et Langmuir-Hinshelwood respectivement révèlent, (1) des propriétés d’adsorption et de photocatalyse plus importante pour le photocatalyseur TiO2 «Degussa P25», (2) des constantes d’adsorption différentes en présence ou en absence d’irradiation et aucune proportionnalité entre quantité de EE2 adsorbée et concentration en photocatalyseur. Cette absence de corrélation est expliquée en considérant la taille des particules. La formation d’agrégats de plus en plus important est observée en augmentant la masse de TiO2. La minéralisation de EE2 est totale après moins d’une heure d’irradiation en travaillant avec une puissance lumineuse de 5,5 mW/cm2 et 10 mg/L de EE2 indiquant l’efficacité de ce procédé.

▼ Les perturbateurs endocriniens (PE) sont une préoccupation majeure de par leurs effets sur les grandes fonctions physiologiques des organismes etparticulièrement sur la fonction de reproduction. Chez les vertébrés, la reproduction est sous le contrôle d’un axe neuroendocrinien nommé axegonadotrope. Celui-ci comprend différents acteurs cérébraux (gonadolibérines et kisspeptines), hypophysaires (gonadotropines) et gonadiques(stéroïdes sexuels) dont la mise en place et le fonctionnement sont régulés finement. Parmi les mécanismes de régulation, les stéroïdes vont jouer unrôle important en effectuant des boucles de rétrocontrôle. Les PE mimant ces stéroïdes sont donc potentiellement capables d’impacter ces régulations.L’objectif de ce travail était d’évaluer les effets d’un xénooestrogène sur les mises en place des acteurs cérébraux impliqués dans l’axe gonadotrope chezle bar européen (Dicentrarchus labrax). Nous avons d’abord décrit l’ontogénèse des systèmes à kisspeptine, à gonadolibérine et à gonadotropine aucours du développement larvaire. Nous avons mis en évidence une régulation différentielle de ces acteurs suggérant un rôle durant ce processusdéveloppemental. De plus, nous montrons que le 17α-éthinylestradiol (EE2) est capable de perturber l'ontogenèse des systèmes à GnRH2 et à GnRH3au cours des stades précoces du développement larvaire.Ces données constituent une base permettant l’évaluation des effets des PE sur l’ontogénèse des systèmes neuroendocriniens.
Endocrine disruptors (EDs) are a major concern because of their effects on the main physiological functions, particularly on the reproductive function.In vertebrates, reproduction is under the control of a neuroendocrine axis called hypothalamic-pituitary-gonadal (HPG) axis. It includes various actors atthe brain (gonadotropin releasing hormones (GnRH) and kisspeptins), pituitary (gonadotropic hormones) and gonadal (sex steroids) levels. Theirdévelopment and functioning are finely regulated. Among the regulators, steroids play an important role by performing feedback loops. EDs mimickingsteroids are therefore potentially able to impact these regulations. The objective of this work was to evaluate the effects of xenoestrogens on theimplementation of cerebral actors involved in the HPG axis in the European sea bass (Dicentrarchus labrax). We first described the ontogeny of thekisspeptin, GnRH and gonadotropin systems during larval development. We have highlighted a differential regulation of these actors suggesting a roleduring this developmental process. In addition, we show that 17α-ethinylestradiol (EE2) is able to disrupt the ontogeny of GnRH2 and GnRH3 systemsduring the early stages of larval development.These data provide a basis for evaluation of the EDs effects on the ontogeny of neuroendocrine systems.

Resumo: O estrógeno natural 17ß-estradiol (E2), o estrógeno sintético 17"alfa"-etinilestradiol (EE2) e o herbicida atrazina (ATZ) foram selecionados para se investigar, em escala de laboratório (jar-test), a remoção de compostos orgânicos em matriz aquosa por adsorção em carvão ativado em pó (CAP) e seu correspondente superfino (CAP-SF). Cromatografia líquida de alta eficiência com detector de arranjo de diodos (HPLC-DAD) foi empregada para a identificação e quantificação desses analitos, em duplicata. Amostras de água desionizada e de água bruta foram fortificadas
para obtenção de concentrações iniciais de 6,70, 21,64 e 26,47µg/L para ATZ, E2 e EE2, respectivamente. Isotermas de adsorção foram desenvolvidas experimentalmente em água desionizada (temperatura: 20°C; pH: 8,5; dosagens de carvão: 1, 2,5, 5, 10 e 25mg/L; tempo de contato: 2h) sendo obtidos os parâmetros dos modelos de Freundlich e Langmuir. A capacidade adsortiva máxima do CAP seguiu a ordem crescente para ATZ (o mais hidrofílico), EE2 e E2; no caso do CAP-SF, esta ordem foi ATZ, E2 e EE2. Natural estrogen 17ß-estradiol (E2), synthetic estrogen 17"alfa"-ethinylestradiol (EE2) and herbicide atrazine (ATZ) were selected in order to investigate the adsorption
removal of organic compounds by powdered activated carbon (PAC) and its superfine correspondent (SF-PAC) in aqueous matrix, at laboratory scale (jar-test). High pressure liquid chromatography plus diode array detector (HPLC-DAD) were applied for analytes detection and quantification, in duplicate. Samples of deionized and raw water were fortified in order to obtain initial concentrations of 6.70, 21.64 and 26.47µg/L for ATZ, E2 and EE2, respectively. Resumo: Constructed wetlands são sistemas naturais de tratamento de águas residuárias e de abastecimento, que removem poluentes por macrófitas aquáticas e pelo biofilme formado nas raízes dessas plantas fixadas a um meio suporte. Esse método de tratamento tem como vantagem o baixo custo de implantação e manutenção, economia de energia elétrica durante seu funcionamento, além de serem eficientes na remoção de poluentes. Os poluentes que tem gerado grande preocupação devido aos riscos à saúde humana e ao meio ambiente são os interferentes endócrinos. Essas substâncias estão cada vez mais
presentes no meio ambiente, especialmente nos corpos hídricos, e podem causar câncer em seres humanos e animais, mutagenicidade nos órgãos reprodutores das espécies animais presentes na biota aquática, esterilidade em homens, entre outros efeitos adversos à saúde. Portanto a presença dessas substâncias em águas residuárias tem despertado grande preocupação e o uso de métodos para a remoção das mesmas é de extrema relevância e importância. Neste contexto, o presente projeto visou avaliar a eficiência de constructed wetlands de fluxo subsuperficial horizontal, em escala de bancada, na retenção/remoção dos interferentes endócrinos: bisfenol A (BPA), etinilestradiol (EE2) e levonorgestrel (LNG) adicionados à esgoto sintético previamente preparado. Foram utilizados quatro diferentes tipos de wetlands: três contendo brita como meio suporte, uma cultivada com Cyperus isocladus (WL1), uma sem macrófita (WL2) e outra cultivada com Eichhornia crassipes (WL3); a quarta wetland (WLC) possuía
além da brita, carvão de bambu (Dendrocalamus asper) como meio suporte, e foi cultivada com a mesma macrófita de WL1. Foram avaliados também a toxicidade do esgoto sintético, a adsorção no meio suporte utilizado, e a absorção dos interferentes endócrinos pelas macrófitas. Parâmetros físico-químicos como pH, demanda química de oxigênio, carbono orgânico dissolvido, série de sólidos, série de nitrogênio, e fósforo total, assim como a quantidade de biomassa, e concentração de macro e micronutrientes nas macrófitas também foram avaliados. As retenções médias de BPA foram de 80,7 (± 6%) em WLC, para WL1 42,3 (± 16,8%), em WL2 30,2 (± 19,5%) e para WL3 20,0 (± 21,1%). Para EE2 as retenções médias foram 85,2 (± 3,4%) para WLC, 51,4 (± 9,7%) em WL1, 23,3 (± 17,6%) em WL2 e 55,4 (± 4,3%) em WL3. Para o hormônio LNG a WLC obteve retenção de 82,7 (± 5,0%), 74,2 (± 9,3%) em WL1, 81,2 (± 5,0%) em WL2 e por fim, 82,8 (± 6,8) em WL3. Resumo: Leitos cultivados tem sido citados como um método alternativo de redução da concentração de alguns agrotóxicos e hormônios levemente hidrofóbicos, como o 17-?-etinilestradiol, que é um dos principais responsáveis pela causa da alteração no sistema endócrino de humanos e animais. Neste contexto, o presente projeto visou avaliar a eficiência da redução de pH, 17-?-etinilestradiol, oxigênio dissolvido (OD), cor aparente, turbidez, nitrogênio total Kjeldahl (NTK), nitrogênio amoniacal (NH3), nitrito (NO2-), nitrato (NO3-), demanda química de oxigênio (DQO) e fósforo total (PT), utilizando-se três leitos tendo brita
como meio suporte ¿ dois deles cultivados com a macrófita Eichhornia crassipes (aguapé), com 43 dias de diferença de cultivo entre eles, e outro leito sem cultivo apenas com a brita para controle e comparação dos resultados. A vazão de entrada nos leitos cultivados variou entre 312 e 1.059 L.dia-1, o TDH médio permaneceu entre 2 e 3 dias, as médias máximas de retenção da concentração de DQO, cor, turbidez, NTK, NH3 e PT pelos leitos foram 94,2%, 76,7%, 90,0%, 42,7%, 39,6% e 51,0%, respectivamente. A biomassa dos dois leitos cultivados com aguapé absorveram juntos, 163 g.kg-1 de nitrogênio e 47,2 g.kg-1 de fósforo, durante o período monitorado. Dentre os parâmetros de eficiência analisados o leito cultivado com Eichhornia crassipes com maior grau de desenvolvimento foi o mais eficiente na retenção da maioria dos parâmetros avaliados com nível 5% de significância. A metodologia adaptada para análise do hormônio 17-?-etinilestradiol por Cromatografia Líquida de Alta Eficiência acoplada
à Detector de Arranjo de Diodos obteve limite de detecção de 1,26 µg.L-1, e limite de quantificação de 2,52 µg.L-1, porém as amostras analisadas ficaram abaixo do nível de detecção, não sendo possível a determinação das concentrações das amostras. Conclui-se que a macrófita Eichhornia crassipes fixada em meio suporte obteve retenções de nutrientes consideradas satisfatórias em tratamento de esgoto doméstico, e as informações obtidas por este trabalho poderão contribuir com a melhoria e desenvolvimento de novas tecnologias para retenção e métodos analíticos para quantificação de 17-?-etinilestradiol em águas residuárias, diminuindo o lançamento destes interferentes endócrinos nos corpos hídricos

Contaminantes emergentes são substâncias químicas de ocorrência natural ou antrópica que não tem controle no ambiente e têm condições latentes de ecotoxicidade e efeitos deletérios sobre a saúde humana. Produtos de higiene pessoal, repelentes, medicamentos, inseticidas, entre outros, que chegam aos corpos hídricos de maneira frequente, fazem parte destes contaminantes. O N,N-Dietil-m-toluamida (DEET) e o 3-fenoxibenzil(1RS)-cis,trans-3-(2,2-diclorovinil)-2,2-dimetilciclopropanocarboxilato (permetrina), utilizados como repelente contra insetos e no tratamento da infestação por piolhos, respectivamente, têm sido encontrados em ambientes aquáticos. Ao lado destes, os estrógenos
também têm despertado a atenção das agências ambientais, em especial o 17-α-etinilestradiol, que é um hormônio sintético empregado em pílulas anticoncepcionais e considerado um desregulador endócrino. Devido à baixa concentração destes compostos encontrada em água, sua análise tem sido conduzida por técnicas cromatográficas, empregando a detecção por espectrometria de massas, o que gera análises com alto custo e mão de obra especializada. O desenvolvimento de métodos de análise com baixo custo, que propicie a determinação de diferentes classes de contaminantes simultaneamente, com limites de quantificação aceitável, devem ser investigados. Adicionalmente vale destacar que efluentes apresentam alta atividade estrogênica, sendo observada a presença de elevadas concentrações de cafeína, tornando este composto um importante marcador da presença de contaminantes emergentes. Neste sentido, este trabalho teve por finalidade avaliar métodos de pré-concentração on-line e off-line para análise
concomitante de DEET, permetrina, 17-α-etinilestradiol e cafeína, empregando como técnicas de analise a eletroforese capilar (CE) e a cromatografia liquida (HPLC) empregando os detectores ultravioleta e de fluorescência. As condições otimizadas para o HPLC foram: coluna C18, fase móvel ACN/H2O com um gradiente na proporção 45:55 (v/v) até 8 min, seguida de 95:5 (v/v) até 18 min, vazão de 1 mL min-1 e temperatura de 25 ± 2ºC em 220 nm por UV e ex= 280 nm e em= 310 nm por fluorescência. ▼ Osmoregulation is an essential process to maintain water and ionic balance and when euryhaline fish move between freshwater and seawater environments as part of their life cycle this presents additional osmoregulatory challenges. Migrating fish can be exposed in both environments to pollutants such as endocrine disrupting chemicals (EDCs) that include natural hormones (e.g. 17β-estradiol; E2), synthetic hormones (e.g. 17α-ethinylestradiol; EE2), and industrial chemicals (e.g. nonylphenol). The focus of this thesis was to study the effects of different categories of EDCs on the osmoregulatory functions of euryhaline fish such as three-spined sticklebacks (Gasterosteus aculeatus) and rainbow trout (Oncorhynchus mykiss). Osmoregulatory variables (such as osmolality, water and ionic content) were compared in plasma and tissues (white muscle and carcass) of rainbow trout. This validated the use of specific tissue parameters as a surrogate of plasma responses to various osmoregulatory challenges. Waterborne exposure to 17α-ethinylestradiol revealed differential sensitivity of vitellogenesis in the three-spined sticklebacks (no induction) and rainbow trout, but had a significant effect on calcium homeostasis in both species. Intraperitoneal implants of 17β-estradiol reduced CaCO3 production and apparent water absorption in the intestine and increased in tissue calcium stores of seawater-acclimated trout, but fish were able to compensate and showed no overall osmoregulatory disturbance. Waterborne exposure to nonylphenol in freshwater trout was also investigated, but no effects on osmoregulation were found up to 2 ng/l. Overall, estrogens can affect osmoregulation differentially in euryhaline fish species, and sometimes at EDC levels lower than the threshold for reproductive effects (i.e. vitellogenin induction).

▼ Communities are composed of organisms that interact
with each other and with the non-biological environment. There are
also anthropogenic pollutants in the environment that also interact
with the organisms. Estrogenic endocrine disrupting compounds, for
example 17α-ethinylestradiol (EE2, the active compound in
contraceptive pills), released into surface waters can have
negative effects on the organisms living there. Fish are sensitive
to estrogenic EDCs and exposed fish can be less fertile and have
inhibited reproductive behaviors that may lead to reduced
population sizes while the majority of invertebrates are not
sensitive, with the exception of mollusks. With the difference in
sensitivity is likely that there may be consequences for aquatic
communities, such as changes in the structure and dynamics. We have
studied the effects of EE2 on different organizational levels in
the aquatic community and with the emphasis on end-points not
connected to reproduction. We have shown that fish exposed to EE2
had inhibited somatic growth, increased size of liver, decreased
size of gonads and also reduced foraging performance that
indirectly affected prey communities by releasing them from
predation. We have also shown that EE2 can inhibit or enhance
somatic growth rates in gastropods depending on specie, but
population growth rates was not affected, and that EE2 pollution
history in gastropods may also influence the effects of novel
stressors. Furthermore, we showed that EE2 can interfere with
detoxification and apoptosis in copepods (crustacea) and this can
make them more sensitive to novel stressors and affect molting and
population demography. The effects of EE2 we have observed on the
organisms in our studies, together with the known negative effect
on fish reproduction, may have consequences for community
structures and ecosystem functioning in surface waters that receive
EE2 or other estrogenic EDCs and.

The 17α-ethinylestradiol (EE2) is a synthetic estrogen used in contraceptive and hormone replacement therapies; is an environmental contaminant potential, it is not completely removed in sewage treatment plants, reaching the aquatic environment, with the aggravating factor interfere the endocrine system of humans and other animals. For this reason there is great interest in the removal of EE2 present in wastewater, surface water and groundwater. Thus the use of laccase for this purpose may be promising because of low specificity of this enzyme to the substrate. Therefore, the aim of this study was to evaluate the effectiveness of laccase produced by the fungus Pleurotus ostreatus immobilized in Luffa cylindrica (loofah) fibers in EE2 degradation. Therefore the best means for production of laccase and the best conditions of pH and temperature were investigated. The immobilization process was realized and removal of the EE2
assay was conducted in flasks and in a batch reactor. Thus, the best means for producing laccase were Potato Dextrose and Potato Dextrose Modified with guariroba straw. Readings taken with 2.2-Azino-bis (3-ethylbenzothializone-6-sulfonic acid) - ABTS revealed best pH between 3.6 and 4.6 and optimal band temperature between 30 and 50 °C for free and immobilized enzymes. The laccase produced in both media had an apparent molecular mass of 40 kDa. The best conditions found for the immobilization process were glutaraldehyde concentration of 2% and reaction with the support 1 hour under stirring (120 rpm); drying the loofah prior to the reaction with glutaraldehyde; time of 24 hours of immobilization. The immobilized enzyme showed best enzymatic activity in alkaline pH, and at higher temperatures, compared to the free enzyme. The operational stability, storage and thermostability were not significant. Best removals for tests in flasks were 79.22% and 75.0% for free and immobilized enzyme,

▼ A poluição da água doce no estado de São Paulo ocorre por diversas causas. Entre elas está a ineficiência do serviço de tratamento de esgoto sanitário. Alguns componentes do esgoto sanitário são desreguladores endócrinos, entre eles estão os hormônios naturais e sintéticos excretados pelos seres humanos. Estas substâncias causam modificações no sistema reprodutivo como, por exemplo, câncer, feminização, alterações na transcrição genética, alteração nas gônadas, indução à síntese de vitelogenina, entre outros. O objetivo desse trabalho foi testar a atividade endócrina das águas do rio Paraíba do Sul, em Pindamonhangaba, SP. Para tanto, amostras de água do rio foram utilizadas para a quantificação dos hormônios, 17-etinilestradiol, 17-estradiol e levonorgestrel por meio da técnica de cromatografia líquida de alta eficiência (HPLC). Além disso, foram realizados testes crônicos com peixes (Danio rerio) no interior do rio e em laboratório, e uma
posterior quantificação de vitelogenina. Os resultados não indicaram concentrações detectáveis dos hormônios pelo método empregado. Entretanto, foi detectada a indução de vitelogenina nos machos de Danio rerio. Esse fato indica a atividade estrogênica da água utilizada. São necessários outros estudos para a avaliação de quais substâncias presentes na água, estão causando as alterações endócrinas nos peixes, entre eles a utilização de índices para priorizar o local inicial dessas pesquisas. Freshwater pollution in São Paulo state, Brazil, occurs due a several reasons. Among them there is the inefficiency of the sewage treatment service. Some sewage components are endocrine disrupters, including natural and synthetic hormones excreted by humans. These substances cause modification on reproductive system such as cancer, feminization, gene transcription and gonad alterations,
vitellogenin synthesis induction, among others. The aim of this work was to evaluate the endocrine activity of the river Paraíba do Sul, at Pindamonhangaba city, Brazil. For this, river water samples were collected for the quantification of the hormones, 17- etinylestradiol, 17-estradiol and levonorgestrel by high performance liquid chromatography (HPLC). Furthermore, chronic assays were performed with fish (Danio rerio) into the river and in the laboratory, following quantification of biomarker vitellogenin. The studied hormones were not detected by employed methods. However, induction of vitellogenin in male of Danio rerio was detected indicating estrogenic activity of water. Further studies are necessary for the assessment of which substances into the water are causing the endocrine disruption in fish. These studies are important for the preservation of aquatic biodiversity of the Paraiba do Sul River

▼ O controle da fertilidade é obtido principalmente pela inibição da ovulação por meio da atividade combinada de dois componentes principais: estrogênio e progestina. Desta forma, novas formulações vêm sendo desenvolvidas com baixa dose de etinilestradiol associado a um novo agente progestógeno, a drospirenona, com o objetivo de garantir equilíbrio entre eficácia, segurança e controle de ciclo menstrual. Este trabalho tem como objetivo desenvolver, validar e comparar métodos analíticos para a identificação e quantificação de etinilestardiol e drospirenona presentes em uma mesma formulação por cromatografia em fase líquida de alta eficiência (CLAE) e a eletroforese capilar (EC). O método por CLAE, foi realizado empregando-se uma coluna LiChoCART® 100RP - C18, (125 x 4 mm) 5 µm, fase móvel constituída por MeCN/H2O 50:50 (v/v) e vazão de 1.0 mL min. Utilizou-se um detector UV 200 nm para drospirenona e um de fluorescência λex = 280 nm
e λem = 310 nm, para o etinilestradiol. O método por EC foi validado utilizando-se como eletrólito, tampão de tetraborato de sódio 30 mM, dodecil sulfato de sódio 20 mM e acetonitrila 30% a pH 9,2, capilar de sílica fundida de 31,2 cm sendo 21 cm efetivos, com 75 µm de diâmetro interno. O método por CLAE apresentou um coeficiente de correlação (r2) de 0,9989 para o etinilestradiol e 0,999 para a drospirenona. No método por EC, o coeficiente de correlação (r2) encontrado foi de 0,9988 para o etinilestradiol e 0,998 para a drospirenona. Os métodos podem ser considerados eficientes e confiáveis para serem empregados em análise de rotina para controle de qualidade destes produtos farmacêuticos.
The fertility control is achieved mainly by inhibiting ovulation through the combined activity of two main components: estrogen and progestin. Thus, new formulations have been developed associating appropriately low dose of ethinylestradiol to a new drug, the drospirenone,
in order to ensure the balance between efficacy, safety and cycle control. The objective of this research was to develop, to validate and to compare two analytical method for separation and quantification of ethinylestardiol and drospirenone using high performance liquid chromatography and capillary electrophoresis (CE). The HPLC method, was performed using a LiChoCART® 100RP - C18, (125 x 4 mm) 5 µm column, a mobile phase constituted of acetonitrile:water (50:50 v/v) and flow rate of 1.0 mL min. UV detection was made for drospirenone at 200 nm coupled with a fluorescence detector at λex= 280 nm and λem = 310 nm for ethinylestradiol. The CE method was validated using a solution of sodium tetraborate buffer 30 mM, sodium dodecil sulphate 20 mM and acetonitrile 30%, pH 9.2 and a fused silica capillary of 31,2 cm with 21 cm effective and 75 µm of diameter. The HPLC method showed a correlation coefficient (r2) of 0.9989 for ethinylestradiol and 0,999

▼ Nos últimos anos, a pesquisa ambiental tem se defrontado com sérios problemas em relação a algumas substâncias presentes no meio aquático em pequenas concentrações. Os desruptores endócrinos tais como produtos farmacêuticos, hormônios naturais e sintéticos, pesticidas, substancias tensosativas, polímeros de baixa massa molar e diversos outros contaminantes orgânicos presentes em efluentes municipais e industriais, são exemplos dessas substancias que contribuem para alterar o funcionamento do sistema endócrino. Varias técnicas já foram empregadas na degradação dessas substancias contida no meio aquático, mas não se demonstraram eficazes.Este trabalho tem por objetivo estudar a degradação do Hormônio etinilestradiol que é um agente terapêutico e um método contraceptivo muito usado. É preocupante o crescente uso desta substância, pois é encontrada no meio aquático de várias partes do mundo.Para a degradação do hormônio etinilestadiol testou!se quatro
técnicas de degradação, (a) eletrólise, (b) fotólise, (c) fotocatálise e (d) eletrofotocatálise.O eletrodo de trabalho utilizado foi de carbono vítreo em meio de KCl para a eletrólise. Como catalisador utilizou!se o Ti/TiO2 na fotocatálise e foteletrocatálise. Os processos degradativos foram acompanhados por espectroscopia UV-vis e cromatografia HPLC.Foram feitos testes de identificação da formação de clorato e hipoclorito nas amostras finais, pois estas espécies podem auxiliar na degradação de substâncias orgânica nos processos eletroquímicos e fotoquímicos.
In recent years, the environmental research has been facing serious problems in relation to some substances in the aquatic environment in small concentrations, such as the endocrine desruptores pharmaceuticals, natural and synthetic hormones, pesticides, tensosativas substances, polymers of low molar mass and different other organic contaminants present in municipal and industrial effluents, are examples of substances that
contribute to change and the functioning of the endocrine system. Several techniques have been used for the degradation of these substances in the aquatic environment, but none demonstrated effectiveness.This work aimed to study the degradation of the hormone ethinyl estradiol which is a therapeutic agent and is widely used in contraceptive method. It is worrying the increasing use of this substance because it is found in various parts of the world.It was tested four techniques of degradation of the hormone, (a) electrolysis, (b) photolysis, (c) photocatalysis and (d) photoelectrocatalysis. The working electrode of glassy carbon was used in KCl medium for electrolysis. As catalyst, was used Ti/TiO2 in the photocatalysis and photoelectrocatalysis. The degradation process was followed by UV-vis spectroscopy and HPLC chromatography.It was carried out tests to identify the formation of chlorate and hypochlorite in the samples once these species can help the degradation in electrochemical
and photochemical process.

A presença no meio hídrico de
contaminantes orgânicos com atividade estrogênica é um tema que tem gerado crescente preocupação na comunidade científica, uma vez que tais contaminantes são capazes de desencadear efeitos adversos no sistema endócrino de várias espécies. O estrogênio sintético 17α-etinilestradiol (principal componente utilizado em formulações de contraceptivos orais) tem sido apontado como um dos principais compostos responsáveis por provocar tais alterações. O principal objetivo deste estudo foi investigar, através de um questionário, o padrão de consumo do 17α-etinilestradiol presente em contraceptivos orais e medicamentos utilizados na TRH feminina, e estimar o aporte deste estrogênio nos corpos hídricos da área urbana do 1° distrito do município de Santa Maria Madalena. Para o desenvolvimento deste trabalho utilizou-se como metodologia um estudo do tipo inquérito realizado com 325 mulheres de 15 a 59 anos de idade, residentes na área de estudo, no período de setembro a dezembro do ano de 2013.
Das 325 mulheres entrevistadas, constatou-se que 95 fazem o uso de contraceptivo oral, sendo que destas, 91 fazem uso de formulações que contêm o 17α-etinilestradiol. Tomando a amostra (N=325) como estatisticamente representativa da área de estudo, pôde-se extrapolar o valor de consumo do 17 α-etinilestradiol para a população total de mulheres (N=2054) na faixa etária estabelecida. Com base nas informações obtidas a partir da elaboração do presente estudo foi possível estimar que há um considerável aporte desta substância nos corpos hídricos da área urbana do 1° distrito do município de Santa Maria Madalena (16 mg/dia), o que sugere a possibilidade de efeitos adversos à biota aquática local. Uma vez que o presente estudo não encerra a discussão sobre o assunto, ao fim desta dissertação há uma proposição para um trabalho futuro no sentido de se avançar neste tema.

▼ Os desreguladores endócrinos são compostos micropoluentes de ação
deletéria que causam efeitos aos animais e aos seres humanos. Estes vêm trazendo
a várias décadas, problemas relacionados à má formação ou ainda infertilidade de
várias espécies. A presença destes compostos na ordem de ng L-1 já é capaz de
gerar efeitos nocivos ao sistema endócrino. Este trabalho visou à determinação da
presença e quantidade de alguns destes compostos como 17α-etinilestradiol,
bisfenol A e estrona entre outros, nas águas do rio Arroio Fundo, localizado em
Jacarepaguá no RJ que possui um índice de qualidade de água ruim. A primeira
etapa deste trabalho foi a implementação e validação de metodologia para
determinação do 17α-etinilestradiol por cromatografia líquida acoplada a detector de
fluorescência onde foram estabelecido limite de detecção, de quantificação e
linearidade conforme orientação do Inmetro (2010). Além da avaliação das amostras
ambientais através da metodologia estabelecida. Na etapa subsequente foram
realizados os experimentos relativos à determinação dos desreguladores endócrinos
através da espectrometria de massas por detector tipo tandem quadrupolo, onde
também foram realizados os procedimentos de validação e determinação dos
compostos bisfenol A e estrona que não foram suscetíveis a efeito de matriz durante
a ionização, que acabou impedindo a determinação de outros desreguladores
endócrinos. Os resultados indicam que não há redução da presença dos
desreguladores endócrinos detectados, e de outros compostos orgânicos também
detectados, mas não identificados, entre os pontos a montante e a jusante da
Unidade de Tratamento do Rio Arroio Fundo. Todavia em função do grande efeito de
matriz oriundo do fato da amostra ser de extrema complexidade, não foi possível
realizar uma identificação e quantificação de todos os compostos alvos.
Endocrine disrupters are compounds micropollutants of deleterious effects that
cause effects to animals and humans has brought these problems to several
decades related to poor training or infertility of various species. The presence of
these compounds in the ng L-1 order is already capable of generating negative effects
on the endocrine system. This work aimed at determining the presence and amount
of some of these compounds as 17α-ethinylestradiol, bisphenol A and estrone
among others, in the waters of the river Arroio Fundo, located in Jacarepaguá in Rio
de Janeiro that has a bad water quality index. The first stage of this work was the
implementation and validation of a method for determination of 17α-ethinylestradiol
by liquid chromatography-fluorescence detection which were established limit of
detection, quantification and linearity as directed by Inmetro (2010). Besides the

▼ Although wastewater treatment plants are a line of defense in minimizing indiscriminate output of microconstituents to natural waters, we do not possess a fundamental understanding of the mechanisms involved in microconstituent removal during wastewater treatment. With this in mind, experiments were designed to investigate the factors that can influence the fate of four microconstituents, carbamazepine (CBZ), 17alpha-
ethinylestradiol (EE2), iopromide (IOP), and trimethoprim (TMP), during biological suspended culture treatment. Specifically, the role that various ecological members of biological treatment systems play in biotransforming these compounds was evaluated.
Sorption assays were performed with inactivated biomass samples (ammonia oxidizing bacteria (AOB), laboratory enriched heterotrophic cultures free of active nitrifiers with low (Ox⁻) or high (Ox⁺) oxygenase activity, and a nitrifying activated sludge (NAS) from a full-scale wastewater treatment plant) to determine whether partitioning dictates removal of individual microconstituents. No microconstituents sorbed to the AOB culture. Neither CBZ nor IOP sorbed to Ox⁻, Ox⁺ and NAS cultures; however, EE2 and TMP sorbed to the Ox⁻, Ox⁺ and NAS biomass. Sorption was positively influenced by the presence of exopolymeric substances (EPS) associated with the cultures. The protein content of EPS affected EE2 and TMP sorption more appreciably than the polysaccharide content of EPS.
Further experiments were performed to investigate microconstituent biodegradation by AOBs, Ox⁻ and Ox⁺ cultures. The influence of growth state and oxygenase activity on biotransformation by each culture was also evaluated. Results indicate that EE2 was the only microconstituent that was amenable to biotransformation by batch cultured AOB and heterotrophic cultures. EE2 was biotransformed but not mineralized by AOB chemostat and batch cultures. TMP was not transformed by AOB batch or chemostat cultures; however both EE2 and TMP were transformed by Ox⁻ and Ox⁺ chemostat cultures. Radiolabeled studies showed that EE2 was mineralized by this culture. Kinetically, AOBs dominated EE2 transformation to monohydroxylated metabolites; however, both Ox⁻ and Ox⁺ cultures further degraded and mineralized EE2 and metabolites generated by AOBs. These results indicate that biotransformation of EE2 by NAS may be limited by heterotrophic activity whereas TMP fate may be a function of heterotrophic activity only. Oxygenase activity did not limit EE2 or TMP biotransformation in chemostat cultures.
Subsequent experiments that were performed to identify the factors that influence heterotrophic degradation of EE2 and TMP indicated that the presence of readily biodegradable substrates slows EE2 and TMP biotransformation. The impact of slowly biodegradable substrates like EPS on EE2 and TMP degradation was unclear. These results suggest that EE2 and TMP are most amenable to biodegradation in bioreactors where endogenous conditions dominate.

▼ Anthropogenic sources of endocrine disrupting compounds (EDCs) are prevalent in aquatic ecosystems. As a result, these compounds target the most vulnerable physiological mechanisms of reproductive development in fish. The main focus of this study was to assess the impact of two sources of EDCs; a single potent EDC, 17α-
ethinylestradiol (EE2), as well as an agricultural source of an EDC mixture, poultry litter leachate, on the reproductive processes of the fathead minnow,
Pimephales promelas. The experimental paradigms included a 21-day laboratory reproductive assay, a larval exposure including a depuration step until 8 months of age to determine the response in the adult animal, and a whole-organism, partial life-cycle assessment of reproductively active adult fish and development of their offspring.
In the first experiment, breeding groups of fathead minnow were exposed to 0, 10, and 40 ng/L of EE2 for 21 days. A significant decrease in testosterone (T), the 17β-estradiol (E2)/T ratio and 11-ketotestosterone in males was observed in the 10 ng/L and 40 ng/L EE2 treatments compared to the control. Male secondary sex characteristics declined with increasing concentrations of EE2. Moreover, 43% of the males in the highest EE2 group (40 ng/L EE2) developed ovipositors. In experiment two, E2 was detected at approximately 200-400 pg/mL and T at approximately 100-400 pg/mL in the poultry litter leachate. Gender ratio was skewed toward female in the two highest concentrations of the poultry litter leachate treatment groups. In experiment three, with a transgenerational design, adult fish had reduced fecundity and males had a significant reduction in male traits when exposed to poultry litter leachate. Vitellogenin production was observed in male fish. In the offspring, both the Low and E2 fish from the same treated adults had significantly lower GSI values and aromatase activity levels. Exploration of different developmental stages and exposures across generations improves our understanding of the impact anthropogenic EDCs can have on wild fish populations.

A presença no meio hídrico de contaminantes orgânicos com atividade
estrogênica é um tema que tem gerado crescente preocupação na comunidade científica, uma vez que tais contaminantes são capazes de desencadear efeitos adversos no sistema endócrino de várias espécies. O estrogênio sintético 17α-etinilestradiol (principal componente utilizado em formulações de contraceptivos orais) tem sido apontado como um dos principais compostos responsáveis por provocar tais alterações. O principal objetivo deste estudo foi investigar, através de um questionário, o padrão de consumo do 17α-etinilestradiol presente em contraceptivos orais e medicamentos utilizados na TRH feminina, e estimar o aporte deste estrogênio nos corpos hídricos da área urbana do 1° distrito do município de Santa Maria Madalena. Para o desenvolvimento deste trabalho utilizou-se como metodologia um estudo do tipo inquérito realizado com 325 mulheres de 15 a 59 anos de idade, residentes na área de estudo, no período de setembro a dezembro do ano de 2013. Das 325 mulheres entrevistadas,
constatou-se que 95 fazem o uso de contraceptivo oral, sendo que destas, 91 fazem uso de formulações que contêm o 17α-etinilestradiol. Tomando a amostra (...) como estatisticamente representativa da área de estudo, pôde-se extrapolar o valor de consumo do 17 α-etinilestradiol para a população total de mulheres (...) na faixa etária estabelecida. Com base nas informações obtidas a partir da elaboração do presente estudo foi possível estimar que há um considerável aporte desta substância nos corpos hídricos da área urbana do 1° distrito do município de Santa Maria Madalena (16 mg/dia), o que sugere a possibilidade de efeitos adversos à biota aquática local. Uma vez que o presente estudo não encerra a discussão sobre o assunto, ao fim desta dissertação há uma proposição para um trabalho futuro no sentido de se avançar neste tema.

This work presents a study for preparation of thin films obtained with a natural and synthetic polyelectrolyte, such as humic acid (HA) and by the hybrid prepared by encapsulation of platinum nanoparticles in the matrix of chloride 3-n- propylpyridinium silsesquioxane (Pt-SiPy+Cl-). Even when dealing with molecules of complex nature, the film deposition occurred regardless of the type of substrate. It was found that there was growth of films by alternate deposition of polyelectrolytes HA and Pt-SiPy+Cl-, governed largely by electrostatic interactions between functional groups COOH, OH-phenolic of HA with pyridinium ring of SiPy+Cl-. The presence of polyelectrolytes in LbL films built on
molecular architectures (HA/Pt-SiPy+Cl-)n and (Pt-SiPy+Cl-/HA)n, was obtained by spectroscopic measurements. From the shifts of the bands in the absorption spectra in the UV-Vis region of LbL films in relation to the precursor solutions, it was inferred on the interactions between these molecules. Measurements of absorption spectroscopy FTIR and Raman scattering of the LbL film were observed by displacement or disappearance of absorption bands due to the presence of precursors of Pt-NPs cavities of SiPy+Cl-. It was found that bands were more intense and defined in the spectra of the film (HA/Pt-SiPy+Cl-)30 in relation to the (Pt-SiPy+Cl-/HA)30 film. This observation corroborates the results of UV-Vis showing that from 12 bilayers no linear growth of absorbance as a function of number of bilayers (Pt-SiPy+Cl-/HA)12. The AFM images showed a linear increase when the film (HA/Pt-SiPy+Cl-)30 was obtained with a thickness of 2.6 nm per bilayer, confirming the nanostructured nature provided
by the molecular organization of polyelectrolytes. Morphological characterization showed that the homogeneity of the surface in function of the amount of deposited materials influence of surface phenomena, such as the number of active sites, as evidenced by electrochemical action of the hormone 17α - ethinylestradiol (EE2). From the diferential pulse voltammetric response of the investigated in the presence of EE2 6.25 x 10-5 mol L-1, it was found that the film (HA/Pt-SiPy+Cl-)3 showed high peak current (Ipa = 17.26 μA) and minor oxidation potential (Epa = 0,74 V vs Ag/AgCl) in relation to no modified electrode (FTO) (Ipa = 8,97 μA e Epa = 1,1 V). Under these conditions , the electrode (HA/Pt-SiPy+Cl-)3 obtained a linear response in the concentration range of 1.37 x 10-6 to 2.14 x 10-5 mol L- 1, yielding a sensitivity 0.68 mmol L-1 and limits of detection ( LOD) and quantification (LOQ ) equal to 1.06 x 10-6 and 3.52 x 10-6 mol L-1 . ▼ Oral contraceptive use is associated with venous thrombosis. However, the mechanism behind this remains unclear. The aim of this thesis was to evaluate genetic variation in the first-pass metabolism of contraceptives, to identify the clinical implications of hormonal contraceptive use after a thrombotic event and to provide an overview of the risk of venous thrombosis per combined oral contraceptive. We found that the UGT2B7 gene in the first-pass metabolism may at least in part explain the risk of venous thrombosis in oral contraceptive users. Furthermore, 20% of women continued or switched to a different contraceptive after a thrombotic event. Contraceptive use after an event was associated with a threefold increased risk of recurrent venous thrombosis, which depended on the ethinylestradiol dose, type of progestagen and whether the contraceptive was triphasic or not. All ten combined oral contraceptives commonly used and included in the analysis increased the risk of a first venous thrombosis. The highest risk of venous thrombosis was found among users of drospirenone with 30 µg ethinylestradiol, cyproterone acetate with 35 µg ethinylestradiol and levonorgestrel with 50 µg ethinylestradiol. Users of levonorgestrel with 20 µg ethinylestradiol had the lowest risk of venous thrombosis among combined oral contraceptive users.

▼ Organic contaminants from industrial and/or domestic effluents may be harmful to humans directly or indirectly by degrading the quality of the aquatic environment. Consequently these contaminants must be reduced to levels that are not harmful to humans and the environment before disposal. Chemical, physical and biological methods exist for the removal of these pollutants from effluents. Among the available chemical methods, heterogeneous photocatalytic oxidation has been found particularly effective in removing a large number of persistent organics in water. In this study, photocatalytic degradation was explored for the removal of reactive azo-dye (textile dye), triclocarban (disinfectant), clopyralid (herbicide) and three endocrine disrupting compounds (EDCs) (estrone, 17ß-estradiol and 17α-ethinylestradiol) from synthetic effluents. The major factors affecting the photocatalytic processes including the initial concentration of the target compounds, the amount of catalyst, the light intensity, the type of catalyst, the electron acceptor, the irradiation time and the pH were studied. Other oxidation techniques including (O3, H2O2, UV) were also studied. Generally UV light is used in combination with titanium dioxide, as photocatalyst, to generate photoinduced charge separation leading to the creation of electron-hole pairs. The holes act as electron acceptors hence the oxidation of organics occur at these sites. These holes can also lead to the formation of hydroxyl radicals which are also effective oxidants capable of degrading the organics. The results obtained in this study indicated that photolysis (i.e. UV only) was found to have no effect on the degradation of reactive azo-dye (RO16). However, complete photocatalytic degradation of 20 mg/L (3.24×10-2 mM) RO16 was achieved in 20 minutes in the presence of 1g/L TiO2 Degussa P25 at pH 5.5. Comparison between various types of catalysts (i.e. Degussa P25, VP Aeroperl, Hombifine N) gave varied results but Degussa P25 was the most effective photocatalyst hence it was selected for this study. For RO16 the optimum catalyst concentration was 0.5 g/L TiO2 with initial concentration of 20 mg/L RO16. It was found that the disappearance of RO16 satisfactorily followed the pseudo first-order kinetics according to Langmuir-Hinshelwood (L-H) model. The rate constant was k= 0.0928 mol/min. Photodegradation of TCC was studied in 70%v acetonitrile: 30%v water solutions. UV light degraded TCC effectively and the reaction rates increased with decreasing initial concentration of TCC. UV/TiO2 gave unsatisfactory degradation of triclocarban (TCC) since only 36% were removed in 60 minutes with initial concentration of TCC 20 mg/L. The degradation of clopyralid and the EDCs was studied using three oxidation systems UV/TiO2, UV/H2O2 and O3. Complete degradation of clopyralid (3,6-DCP) was achieved with UV/TiO2 in about 90 minutes at an optimum catalyst concentration of 1g/L. Diversos estudos vêm
apontando sobre a ocorrência de contaminantes emergentes nas matrizes aquáticas. Esses poluentes ainda não foram regulamentados, mas têm potencial de afetar a saúde humana e ambiental. Neste grupo de contaminantes, destaca-se o 17αetinilestradiol (EE2), hormônio sintético presente em pílulas anticoncepcionais, com alto potencial estrogênico e de bioacumulação. Dentre seus efeitos como desregulador endócrino, o mais notório é a feminização de peixes. A ocorrência de poluentes emergentes no Lago Paranoá (Distrito Federal – Brasil) é presumida devido ao lançamento de efluentes provenientes de estações de tratamento nesse corpo hídrico e pelo fato dessas estações não terem sido projetadas para a remoção desses tipos de contaminantes. Nesse contexto, considerando que o Lago Paranoá é um manancial, os objetivos desse trabalho foram: quantificar as concentrações de EE2 nesse reservatório, avaliar o desempenho das estações de tratamento de esgoto e de água quanto à remoção de EE2, e
caracterizar o risco de exposição involuntária ambiental. Utilizou-se o método ELISA (ensaio de imunoabsorção enzimática) para a realização das análises. Concentrações de EE2 foram quantificadas em todas as amostras de esgoto bruto (1,31 – 2,08 ng. L -1 ) e de efluente tratado (0,23 – 1,50 ng. L -1 ), e em uma das amostras de água superficial do Lago Paranoá (0,07 ng. L -1 ). Nas amostras de água tratada a concentração ficou abaixo do limite mínimo de quantificação do método. O desempenho máximo de remoção de EE2 pelas ETEs foi de 88%, alcançado pela ETE Riacho Fundo, que opera em sistema RBS que remove fósforo. Por outro lado, as demais ETEs que operam com sistema Bardenpho, seguido de coagulação/floculação/ flotação, a remoção foi em torno de 30%. A concentração de 0,07 ng. L -1 quantificada no Lago Paranoá, ficou abaixo das concentrações médias de efeito observado descritas na literatura para animais aquáticos representantes do segundo e do terceiro nível trófico, mas superior às
