▼ A utilização de antígenos específicos do M. tuberculosis é de extrema importância para o diagnóstico precoce da tuberculose e para o controle da propagação da micobactéria na população. O CFP-10 é um antígeno que está presente no M. tuberculosis, porém está ausente no M. bovis da BCG e nas micobactérias ambientais. Portanto, o objetivo deste trabalho foi verificar se a resposta imune contra o antígeno recombinante CFP-10 do M. tuberculosis é capaz de diferenciar a imunidade desenvolvida pela vacina BCG daquela devido à infecção pela micobactéria. Para isto, foi analisada a resposta imune celular de 18 pacientes com TB ativa, 16 pacientes tratados e 19 indivíduos saudáveis PPD-positivo. A resposta imune celular foi analisada através de cultura de PBMC para ensaios de proliferação celular e produção de citocinas Th1 (IFN-γ e TNF-α), Th2 (IL-4, IL-13) e a citocina reguladora IL-10 frente aos estímulos de PHA, BCG e CFP-10. Foi possível observar um aumento significativo na resposta proliferativa das células após cultivo com PHA entre os indivíduos tratados e os saudáveis quando comparados aos pacientes ativos. A comparação dos índices de proliferação entre os três grupos frente ao BCG e ao antígeno CFP-10 não foi estatisticamente significativa. Em relação aos níveis plasmáticos das citocinas, os níveis de IFN-γ mostraram-se significativamente mais elevados nos pacientes com TB ativa em relação aos pacientes tratados e aos indivíduos saudáveis. Os níveis plasmáticos de IL-13 foram significativamente maiores entre os pacientes com TB ativa em relação aos indivíduos saudáveis. Os níveis plasmáticos de IL-10 foram significativamente maiores entre os pacientes com doença ativa em relação aos pacientes tratados. Já em relação ao TNF-α e à IL-4, não foram detectadas diferenças nos níveis plasmáticos entre os três grupos. Em relação à produção das citocinas nos sobrenadantes de cultura, houve produção significativamente maior de IFN-γ frente ao BCG entre os indivíduos saudáveis quando comparados aos pacientes ativos e tratados. A produção de IL-4 foi significativamente maior entre os pacientes com TB ativa em relação aos indivíduos saudáveis nos sobrenadantes de culturas sem estímulo e após os estímulos com BCG e CFP-10. Já a produção de IL-4 após o estímulo de PHA foi significativamente maior entre os pacientes ativos e tratados quando comparados aos indivíduos saudáveis. A produção de IL-13 foi significativamente maior nos grupos dos pacientes ativos e tratados em relação aos indivíduos saudáveis após estímulo com PHA. Em relação à produção de IL-10, foi significativamente maior nos grupos dos indivíduos saudáveis e dos pacientes tratados quando comparados aos pacientes com TB ativa nas culturas sem estímulo e após estímulo com PHA. One-third of the world s population is currently infected with Mycobacterium tuberculosis. The development of new methods of diagnosis and vaccines against tuberculosis (TB) are necessary for disease control. The immune response against M. tuberculosis involves the production of many cytokines and chemokines, but their interaction during the infection is complex. The objective of this work was to study the production of cytokines and chemokines in pulmonary TB under stimulation of M. tuberculosis antigens. The method of Cytometric Bead Array (CBA) was used to evaluate plasma levels of chemokines MIG, IP-10, IL-8, MCP-1 and RANTES in untreated patients with active TB (ATB), in patients during treatment (DTB) and in treated patients (TTB) and healthy controls individuals (HC). We also evaluated, by CBA, the levels of cytokines IFN-g, TNF-a, IL-6, IL-2, IL-10 and IL-4, and chemokines MIG, IP-10, IL-8, MCP-1 and RANTES, in culture of peripheral blood mononuclear cells (PBMC) after stimulation with the antigens ESAT-6/CFP-10 and 16kDa, in groups of untreated patients with active TB (ATB), in treated patients (TTB), healthy controls individuals (HC) and in contact individuals of patients with TB (CT). The production of plasma chemokines, the levels of MIG, IP-10 and IL-8 were higher in the group ATB, compared to the other (HC, DTB and TTB), and the levels of chemokines analyzed in plasma decreased after the beginning of the treatment. In assays with human PBMC, the cytokine IFN-g, TNF-a, IL-6, IL-2 and chemokines MIG, IP-10, IL-8 and RANTES were stimulated at higher levels, by the fusion protein ESAT-6 / CFP-10, in groups ATB and CT in relation to the controls and treated patients. The stimulation with antigen 16kDa did not allow for differentiation of these cytokines and chemokines between the groups. IL-10, IL-4 and MCP-1 showed high levels in the cultures of PBMC from healthy control individuals and contacts when stimulated with the studied M. tuberculosis antigens compared to patients with TB. The antigen ESAT-6/CFP-10 differentiated active TB with regard to the production of cytokines IFN-g, TNF-a, IL-6 and IL-2 and chemokines MIG, IP-10, IL-8 and RANTES, which can contribute to a protective response in TB. The antigen 16kDa was a strong stimulator of IL-4, IL-10 and MCP-1 molecules that can regulate or inhibit the immune response against M. tuberculosis infection, demonstrating the important role of this antigen in promoting the persistence of the bacillus in the host. The plasma levels of chemokines can be effective in differentiating patients with active TB from the healthy control individuals, and can also be useful in monitoring the effect of anti-TB chemotherapy. The comprehension of the dynamics of the immune response against M. tuberculosis antigens can aid the development of new strategies to combat tuberculosis

▼ South Africa has faced many changes in the last 20 years. There are some glaringly obvious ones such as democracy while others, such as the considerable increase of the legislative and regulatory requirements in the financial-services industry, are more subtle. Both however, have far-reaching consequences for both individuals and businesses. In Schaeffer's keynote address at the 2011 Financial Planning Convention, the global trends currently influencing the financial-planning profession were pointed out. These included the move towards a more relationship-based approach between financial planners and their clients, with holistic service offerings and regulatory change. Currently close to 100 000 Financial Services Board (FSB)-registered brokers/ advisors/ planners/ consultants claim to offer financial advice. Of these representatives only 4 600 (approximately) are Certified Financial Planner® professionals. The Financial Planning Institute of Southern Africa (FPI) maintains that the regulatory environment governing the financial industry is dynamic. Coupled with a tumultuous economic environment, this enforces the importance of an expert to assist individuals and businesses in planning for the future. The primary purpose of the Financial Advisory and Intermediaries Services Act (FAIS Act) is to: 1. protect the client against indecorous conduct by the financial service provider (FSP) and its representatives when providing financial advice and/or rendering intermediary services to clients; 2. ensure economic efficiency of the industry; 3. ensure that clients of FSPs are provided with sufficient information regarding their financial products, the representatives selling the products, the represented FSP and the product provider offering the products. The effect of the FAIS Act is that key individuals and representatives are now being held responsible for their actions, which may include providing incorrect information or not having sufficient qualifications or experience. According to the FSB the FAIS Fit and Proper requirements determine a set of requirements that all FSPs, key individuals and representatives need to comply with. The Determination of Fit and Proper Requirements of 2008 outlines the categories of fit and proper requirements for FSPs, their key individuals and representatives. The categories of the FAIS Fit and Proper requirements are: - honesty and integrity requirements for FSPs, key individuals and representatives; - competency requirements of FSPs, key individuals and representatives; - experience requirements of FSPs, key individuals and representatives; - qualifications requirements of key individuals and representatives; - regulatory examinations requirements of key individuals and representatives; - continuous professional development requirements of FSPs, key individuals and representatives; - operational ability of FSPs, key individuals and representatives; and - solvency of FSPs. ▼ Alzheimer’s disease (AD) is a neurodegenerative disorder frequently characterized by the presence of senile amyloid plaques. These plaques are formed through the aggregation of the amyloid β (Aβ) peptide. Aβ is formed through the sequential proteolysis of amyloid precursor protein (APP) by the β-site APP cleaving enzyme-1 (BACE1) and γ-secretase. As the rate-limiting enzyme of Aβ formation, the amyloidogenic properties of BACE1 have been widely examined. Recently, there has been evidence that suggests BACE1 exists as a dimer that is significantly more catalytic than the monomeric form. However, as of yet, there have been no complimentary studies that have explored the dimerization properties of the BACE1 homolog, BACE2. The BACE1 and BACE2 enzymes make up a unique class of membrane-bound aspartyl proteases that share similar extracellular, transmembrane, and cytosolic domains. Interestingly, it is only the BACE1 enzyme that exhibits amyloidogenic properties. Contrarily, BACE2 demonstrates distinct anti-AD characteristics such as the non-amyloidogenic cleavage of APP at α- and θ-sites while also functioning as an efficient Aβ-degrading enzyme. Elucidating the dimerization of BACE2 can help us understand whether dimerization is an inherent characteristic of this unique class of proteases or, alternatively, if dimerization is an attribute required for the amyloidogenic properties of BACE1. In this study, cultured human embryonic kidney 293 (HEK293) cells were transfected with BACE2-CFP fusion protein and imaged using differential interference contrast (DIC) and confocal microscopy to examine sub-cellular localization of the fluorescent construct. Subsequently, two-photon fluorescence lifetime imaging microscopy (2P-FLIM) was used to examine changes in the fluorescence lifetime distribution of transfected cells. The results of this study suggest that the BACE2 enzyme does not undergo dimerization in contrast to BACE1, as demonstrated in previous studies.

A detecção de anticorpos específicos pode representar um instrumento valioso no diagnóstico da tuberculose (TB). Neste trabalho níveis de anticorpos IgG contra os antígenos ESAT-6, CFP10, 85B, 16kDa e HBHA do M. tuberculosis foram avaliados em 102 amostras de soro de pacientes com TB pulmonar agrupados como: 1) pacientes com TB ativa (n=33); 2) pacientes com 1 a 3 meses de tratamento (n=47); 3) pacientes que completaram 6 meses de quimioterapia (n=22). Foi incluído um grupo controle formado por 38 indivíduos saudáveis vacinados com BCG. Níveis de IgG1, IgG2 e IgG4 foram quantificados no soro de pacientes com TB ativa contra os antígenos ESAT-6 e CFP-10. Foram detectados níveis elevados de anticorpos IgG contra ESAT-6, CFP-10, 85B, 16kDa e HBHA no soro de pacientes com TB ativa em comparação com o grupo de pacientes que completaram o tratamento (p<0,05), e em relação ao grupo controle sadio (p<0,05). Níveis de IgG contra 16kDa mostrou um pico após 1-3 meses de quimioterapia. A análise da curva ROC indicou sensibilidade de 85% (ESAT-6), 94% (CFP-10), 91% (85B), 61% (16K) e 70% (HBHA), e especificidade de 89% (ESAT-6), 87% (CFP-10), 82% (85B), 89% (16K) e 84% (HBHA). A área sob a curva ROC foi maior para os antígenos ESAT-6 (área = 0,946, IC 95%: 0,865 a 0,985) e CFP-10 (área = 0,942, IC 95%: 0,859 a 0,983), quando comparada com os antígenos 16kDa (área = 0,809, IC 95%: 0,699 a 0,893) e HBHA (área = 0,805, IC 95%: 0,694 a 0,890). O antígeno 85B (área = 0,909, IC 95%: 0,817 a 0,964) não apresentou diferença estatística em relação ao antígeno 16kDa na análise da curva ROC. Uma predominância de anticorpos IgG1 contra os antígenos ESAT-6 e CFP-10 foi observada no soro de pacientes com TB ativa. Detection of specific antibodies may represent a valuable tool in the diagnosis of tuberculosis (TB). In this work levels of IgG antibodies against ESAT-6, CFP-10, 85B, 16kDa and HBHA M. tuberculosis antigens were measured in 102 serum samples from pulmonary TB patients grouped as: 1) active TB patients (n=33); 2) patients who had received 1 to 3 months of anti-TB chemotherapy (n=47); and, 3) patients who had completed 6 months of chemotherapy (n=22). A control group formed by 38 BCG vaccinated healthy controls was included. levels of IgG1, IgG2 and IgG4 were also quantified in the serum of patients with active TB against the ESAT-6 and CFP-10 antigens. Elevated levels of IgG against ESAT-6, CFP-10, 85B, 16kDa and HBHA were detected in active TB patients in comparison with the group of patients that completed the treatment (p<0,05) and in comparison with the healthy control group (p<0,05). Syftet var också att undersöka om tillväxtfasen påverkar styrkan på sambandet. Metod: Studien antar en positivistisk forskningsfilosofi med en hypotetisk-deduktiv ansats. Sekundärdata har samlats in från databasen Thomson Reuters. Studien är av longitudinell design där data har inhämtats från elva år och sedan analyserats i SPSS. Resultat & slutsats: Resultatet ger bevis för att de immateriella tillgångarna agerar som en partiell medlande funktion i det positivt dubbelriktade sambandet mellan CSP och CFP. De immateriella tillgångarnas påverkan på CSP eller CFP beroende på tillväxtfas visar dock inte något entydigt resultat och kan således inte styrkas. Examensarbetets bidrag: Studien ger bevis för att det finns en partiell medlande effekt från de immateriella tillgångarna i det dubbelriktade sambandet mellan CSP och CFP. Även om vårt resultat med tillväxt som variabel inte styrks kan detta ligga till grund för vidare forskning och har trots allt bidragit till mer kunskap. Vårt resultat bidrar till teoretisk kunskap som framtida forskning kan bygga vidare på samtidigt som företagsledare kan satsa på immateriella tillgångar och nå finansiella mål. Förslag till fortsatt forskning: Ett förslag till fortsatt forskning är att göra en studie som också inkluderar de företag som inte självmant lämnat in sin rapportering. Detta kan skapa ett mer jämförbart resultat. Eftersom vår studie inte får något resultat på företagets tillväxttakt hade det varit intressant om vidare forskning ser detta ur en annan aspekt. Eftersom det tar tid att bygga upp immateriella tillgångar och detta kan förklara varför företag i tillväxt inte har stärkt effekt från de immateriella tillgångarna. Således kan det vara relevant att göra ett urval av de företag som har haft tid att bygga upp sina immateriella tillgångar för att se om dessa företag visar starkare effekt under några år med hög tillväxt för att sedan visa sämre effekt vid en stagnerad tillväxt.

▼ In recent decades, thermochemical conversion of biomass, such as pyrolysis, has gained popularity as a source for renewable materials. Pyrolysis generates three products: biochar that has shown promise as a soil amendment and carbon sequestration agent, bio-oil that could supplement petroleum-based products and transportation fuel, and syngas that is useful for ammonia, methanol, or hydrocarbon/aromatic production. At the molecular level, these samples are complex and difficult to analyze, which creates a bottleneck for thorough understanding. This dissertation utilizes high-resolution mass spectrometry (HRMS) to overcome the sample complexity and improve understanding at the molecular level. HRMS was applied to understanding organic molecules entrapped in biochar during pyrolysis and gasification of switchgrass. Extraction of organic molecules used toluene and a mixture of water/methanol for hydrophobic aromatic compounds and hydrophilic polar compounds, respectively. Orbitrap mass spectrometric data acquisition revealed that molecular compounds previously known in bio-oils were observed for fast pyrolysis biochar, whereas polycyclic aromatic hydrocarbons (PAHs) with various ring sizes were observed for gasification and slow pyrolysis biochars. Bio-oils from fast pyrolysis of switchgrass harvested at various times throughout the year were studied using high-resolution mass spectrometry. Nearly three hundred total nitrogen-containing species were detected through efficient ionization and accurate mass information. Nitrogen-containing species, particularly N2 compounds, were highly abundant for early summer bio-oils, but decrease significantly in later harvest times. Contour plots of double bond equivalent (DBE) versus carbon number and tandem mass spectrometric analysis were utilized to determine the major structural motif for N1 and NO class compounds as pyridine and N2 class compounds as imidazole. The dramatic decrease in nitrogen compounds correlates to the decomposition of proteins as the perennial plant senesces. Catalytic deoxygenation of cellulose pyrolysis was evaluated using micropyrolyzer-gas chromatography (µPy-GC) coupled to dopant-assisted atmospheric pressure chemical ionization (dAPCI) time-of-flight mass spectrometry (TOF MS). A vast majority of compounds produced via catalysis and/or pyrolysis cannot be found in the database. However, dAPCI-TOF MS produces soft ionization and accurate mass measurement for direct chemical composition analysis of GC-separated molecules. This analytical technique demonstrated the ability to evaluate catalytic efficiency and monitor the change in reaction products. A total of 142 compounds could be analyzed with this approach compared to 38 compounds in traditional Py-GC-EI-MS analysis. Finally, HRMS is utilized for the real-time monitoring of fast pyrolysis products of glucose-based carbohydrates. ▼ This study investigates how Certified Financial Planner (CFP®) professionals experience professionalism. The study was carried out to provide insight for the international financial planning community, including the Financial Planning Standards Board and the CFP Board of Standards in establishing internationally relevant, accurate and meaningful professional standards for financial planners. This is a qualitative study following the phenomenographic research tradition which aims to describe the qualitatively different ways in which people understand or experience a phenomenon. This study is unique for two reasons; firstly as a study undertaken independently of the financial planning standard setting bodies seeking to understand more about the professionalism of financial planners; and secondly as a study that takes a second order perspective where the researcher attempted to see the world through the eyes of CFP® professionals who experience the phenomenon of professionalism. The utterances of CFP® professionals from Australia, Hong Kong and America have been analysed to produce categories of description comprising ‘what’ aspects and ‘how’ aspects of the phenomenon of professionalism. The ‘what’ aspect is comprised of participant understandings of what professionalism means to them, and the ‘how’ aspect comprises conceptualised acts that explain how participants think about acting professionally. The relationship of the ‘what’ and ‘how’ categories form a conceptual map that for the purpose of this study have been presented as an outcome space. The research findings have produced a high degree of similarity and correlation between how CFP® professionals in each of the three countries experience professionalism. The different labels and descriptions given to the conceptions reflect differences which can be explained largely by country-specific cultural, regulatory and practice differences. A common conception among CFP® professionals from the three countries relates to putting client’s interests first. In Australia, CFP® professionals’ awareness of professionalism is affected as much by Government regulation as it is by CFP® certification requirements. In Hong Kong, CFP® professionals have to reconcile and manage employers’ expectations of selling financial products with the professional standards of being a CFP® professional. In the United States, CFP® professionals have a more holistic and altruistic experience of professionalism. A full and open account of the research study, member checking and presentation of the research findings in peer-reviewed conference proceedings were used as methods to validate and test the reliability of the results. The significance of this study is that it gives voice to financial planners who hold the professional designation – Certified Financial Planner. This will enable relevant standard setting bodies to understand professionalism through the eyes of CFP® professionals. Les modifications post-traductionelles des histones contribuent à l’expression génique et à la stabilité du génome. La méthylation de la lysine 4 de l’histone H3 (H3K4me), une marque associée aux promoteurs de gènes transcrits, est déposé par les methyltransferases hautement conservées de la famille SET1, dans le contexte du complexe COMPASS. SET-2, l’homologue de SET1 chez Caenorhabditis elegans, est responsable de la déposition de H3K4me dans la lignée germinale, et son inactivation provoque une perte progressive de la fertilité. Le but de mon travail de thèse a été d’étudier comment SET-2 et la méthylation de H3K4 contribuent au maintien de la lignée germinale. J’ai montré que l’absence de SET-2 provoque une sensibilité accrue aux dommages à l’ADN. Cependant, les voies de signalisation et de réparation de ces dommages sont fonctionnelles dans le mutant set-2. Par séquençage de l’ADN, j’ai par ailleurs montré que la stérilité progressive observée en l’absence de set-2 n’est pas due à une capacité de réparation réduite. L’ensemble de mes résultats suggère que H3K4me pourrait agir en aval de la signalisation de dommages à l’ADN, en influençant l’organisation de la chromatine aux sites des cassures double brin. J’ai d’autre part mis en évidence une nouvelle fonction pour la méthylation de H3K4 dans l’organisation de la chromatine en montrant que set-2 interagit génétiquement avec le complexe Condensine II et la Topoisomérase II, facteurs clefs de l’organisation mitotique des chromosomes. Des expériences de microscopie par FLIM-FRET ont d’ailleurs validé une fonction de H3K4 méthylée dans l’organisation de la chromatine dans la lignée germinale. Enfin, j’ai montré par analyses transcriptomiques que la protéine CFP-1 du complexe COMPASS est impliquée dans la régulation du programme transcriptionnel de la lignée germinale et que cette fonction est indépendante de SET-2. L’ensemble de mes résultats montre comment la régulation chromatinienne impacte le maintien d’une lignée germinale fonctionnelle à plusieurs niveaux.

Methylation of lysine 4 of histone H3 (H3K4me), a mark associated with actively transcribed genes, is deposited by the highly conserved SET1 family methyltransferases acting in COMPASS related complexes. The purpose of my PhD work was to study how SET-2 and the methylation of H3K4 contribute to the maintenance of the germ line. Syfte: Ett flertal studier har undersökt om socialt ansvarstagande (CSR) är lönsamt, men inga entydiga bevis finns. Tidigare undersökningar har dock implicit antagit att CSR-aktiviteter är lönsamma oavsett hur de implementeras. Vi ifrågasätter detta antagande och undersöker hur implementeringen av CSR påverkar dess lönsamhet. Vi undersöker om variablerna takt, konsekvens och väg förklarar relationen mellan CSR och bolagens lönsamhet. Metod: Studien är utförd enligt ett positivistiskt perspektiv med en hypotetisk-deduktiv ansats och en longitudinell design med fem års observationer. Data har insamlats från Thomson Reuters Eikon och Thomson Reuters Datastream. Data har analyserats med statistiska metoder. Resultat & slutsats: Studien visaratt implementeringen av CSR påverkar bolags CFP och studien diskuterar de teoretiska förutsättningarna för sådana effekter. Bolag kan påverka lönsamheten av CSRgenom att implementera den på rätt sätt. Förslag till fortsatt forskning: Ett förslag till fortsatta studier av CSR-implementering är att inte bara mätabolagens redovisningsmässiga mått utan även se på de marknadsmässiga. Att mäta bolag CSR-aktiviteter med start i tidigare årtal än vi gjort kan dessutom öka möjligheten att mäta effekter av implementering. Uppsatsens bidrag: Föreliggande studie utvidgar CSR-litteraturen och redovisningslitteraturen genom att (1) diskutera hur implementering av CSR kan påverka dess lönsamhet och (2) empiriskt undersöka effekten av implementering av CSR på lönsamhet. Det praktiska bidraget är att kunskap om hur implementering av CSR kan påverka lönsamheten är till nytta för all implementering av CSR och för behovet av lagstiftning om socialt ansvarstagande.

Aim: A number of studies have investigated whether social responsibility (CSR) is profitable, but no unambiguous evidence exists. However, previous surveys have implicitly assumed that CSR activities are profitable regardless of how they are implemented. We question this assumption and investigate how the implementation of CSR affects its profitability. We investigate whether the variables pace, consistency and path explain the relationship between CSR and the company's profitability. ▼ Under de senaste årtiondena har ansvarfullt företagande (CSR) varit ett ämne som erhållit allt mer fokus inom forskning och således även för företag och samhället i stort. CSR är ett ämne som är högst aktuellt då globalisering är ett faktum och medför att de företag som är verksamma idag bär ett globalt ansvar, då dessa är stora aktörer i en värld som kantas av ekonomiska kriser, miljöproblem och humanitär försummelse. Det ökade intresset från samhället för CSR har medfört att företagen idag ser detta som en strategisk investering och företags satsning i hållbarhetsaktiviteter har kommit att erhålla en allt större del av företags verksamhet. Frågan kvarstår om detta endast är en investering som ökar företagets etik och moral eller om det faktiskt är lönsamt, och även om lönsamma företag ökar sin investering i hållbarhetsarbete. En stor mängd forskning har utförts för att undersöka förhållandena mellan hållbarhet och lönsamhet, dock utan att konsensus i forskningsgrenen har uppkommit. Grundat på den nuvarande forskningssituationen syftade denna studie till att undersöka vilken effekt ansvarsfullt företagande har på olika lönsamhetsmått, samt vilken effekt de olika lönsamhetsmåtten har på ansvarsfullt företagande, i företag registrerade på large cap och mid cap på Nasdaq OMX Stockholm. För att besvara studiens problemformulering och uppfylla studiens syfte har en kvantitativ forskningsansats brukats och ett urval av företag listade på large och mid cap Nasdaq OMX Stockholm har undersökts. För att sätta ett mått på ansvarsfullt företagande (CSP) har en innehållsanalys av företags årsrapporter och hållbarhetsrapporter genomförts, lönsamhetsmåtten (CFP) har utgått från redovisningsbaserade mått (ROE och ROA) samt ett marknadsbaserat mått (total avkastning). Vidare undersöks även relationen mellan variablerna, CSP och CFP, med en tidsaspekt och kontrollvariablerna storlek, risk, FoU och industritillhörighet har brukats. De teorier som har använts för att förklara CFP:s effekt på CSP är slappa resurser och direktörsopportunism, och effekten CSP har på CFP har teorierna intressentteorin, resursbaserad synvinkel och kompromissteorin använts. Det resultat som framkommit genom de multipla regressionerna som testat studiens hypoteser visar att ROE:s effekt på CSP är positiv, medan både ROA:s och avkastningens effekt på CSP är neutral. Slutsatsen blir således delad då lönsamhetsmåttet ROE visar att företag som har en högre lönsamhet kommer investera mer i hållbarhet, denna effekt knyts samman med teorin om slappa resurser. Å andra sidan visar både ROA och avkastning på en neutral effekt på CSP vilket ej kan knytas till varken slappa resurser eller direktöropportunism. Vidare när istället CSP:s effekt på ROE, ROA och avkastning undersökts visar samtliga på en neutral påverkan. ▼ ENGLISH SUMMARY: Mycobacterium bovis (M. bovis) forms part of the Mycobacterium tuberculosis complex (MTC), a group of genetically related bacteria that causes tuberculosis in humans and animals. African buffaloes (Syncerus caffer) are maintenance hosts of M. bovis, the causative organism of bovine tuberculosis (bTB). Since this species acts as a bTB reservoir for a wide range of domestic and wildlife species, the detection of M. bovis-infected animals is essential to control spread of the disease. However, diagnostic assays used for bTB management and control programmes, such as the single intradermal comparative tuberculin test (SICTT) and commercially available interferon-gamma release assays (IGRAs), are still believed to be sub-optimal for the diagnosis of bTB in bovids. A potential approach to improving detection of M. bovis infection could be the use of novel diagnostic antigens or the identification of alternative or ancillary biomarkers to interferon-gamma (IFN-γ). The studies presented in this dissertation aim to identify, develop and evaluate novel approaches for improving the detection of M. bovis infection in African buffaloes. The first objective was to evaluate the performance of two new commercially available IGRAs, the Bovigam® PC-EC assay and the Bovigam® PC-HP assay, for the first time in buffaloes and compare their performance to that of two versions of an adapted human IGRA, the modified QuantiFERON® TB-Gold (mQFT) assay. In addition, the effect of increased blood incubation time on sensitivity of the mQFT assay was assessed along with whether centrifugation was a necessary step prior to harvesting the plasma fraction. Furthermore, the relative sensitivities of Bovigam® assays, a modified Bovigam assay that contains an additional stimulation criteria with Mycobacterium fortuitum tuberculin and the SICTT were compared in identified M. bovis-infected buffaloes. Combinations of these assays were evaluated to identify the optimal test algorithm (i.e., highest sensitivity) for detection of bTB in African buffaloes. Commercially available bovine ELISAs as well as a human IP-10 ELISA were used to identify selected candidate biomarkers of M. bovis-specific immune activation and evaluate their diagnostic utility. Lastly, this study investigated what effect storing stimulated whole blood plasma under various conditions would have on the diagnostic performance of IP-10 assays in African buffaloes. Agreement between the Bovigam® PC-EC and the Bovigam® PC-HP assays was high (κ = 0.86, 95% CI 0.75–0.97) and these detected the greatest number of test-positive animals suggesting that they were the most sensitive assays. Agreement between two versions of the mQFT assay was also high (κ=0.88, 95% CI 0.77-0.98); however, all buffaloes with discordant mQFT results (n=6), including 3 confirmed M. bovis-infected animals, were positive at 30 hours incubation and negative at 20 hours. ▼ After partial denervation of mature muscle, the remaining motoneurons grow terminal sprouts, which reinnervate denervated muscle fibers. In neonatal rat soleus muscle however, not only do the remaining motoneurons fail to reinnervate nearby muscle fibers, but they also display clear morphological and physiological evidence of terminal disruption. My ultimate aim is to attain a better understanding of this disruption by performing repeated in vivo observations of individually identified terminals in transgenic mice expressing fluorescent proteins in their motoneurons. For technical reasons, these in vivo imaging studies would ideally be conducted on CFP mouse sternomastoid (SM) muscle. I therefore tested whether neonatal partial denervation of CFP mouse SM muscle also causes terminal disruption. Two to five days after neonatal partial denervation, I fluorescently labeled nerve terminals and acetylcholine receptors (AChRs) to assess the effect on terminal morphology. Terminal disruption after partial denervation did not occur as extensively in CFP mouse SM muscle (a fast-twitch muscle) as it did in rat soleus muscle (a slow-twitch muscle). This relative absence of terminal disruption in CFP mouse SM suggested that the extent/occurrence of terminal disruption after neonatal partial denervation could depend on either the species being studied or muscle fiber type. To distinguish between these possibilities, I investigated terminal disruption after partial denervation of rat SM muscle. In both soleus and sternomastoid muscles of rats, neonatal partial denervation resulted in disruption of about half of the terminals, whereas in SM muscles from CFP mice only 8% of terminals showed evidence of disruption. Therefore, it appears that the nature of terminal disruption that takes place after partial denervation of neonatal muscles is not related to muscle fiber type but may be species-specific. ▼ High-resolution three-dimensional (3D) shape reconstruction of objects has huge potential for applications in the field of design, security, entertainment, biomedicine, industrial quality control etc. Of the available techniques, optical methods have the distinctive advantage of facilitating non-contact and non-intrusive measurements. Of late, integration of optical measurement system with the computer based data processing has improved the quality of the results. Of the available techniques, structured-light illumination (Fringe Projection) is the most effective, owing its simplistic experimental architecture and analysis. Traditional Fringe Projection techniques function with the use of fringes generated using interferometric methods. With the advent of digital light projectors, digitally generated fringes have taken the place of interferometry based fringes. Despite the technological advances that this field has witnessed over last couple of decades, digital fringe projection technique still suffers from various shortcomings. This thesis presents a strategic solution to the challenges faced by the technique in its application to out-of-plane deformation measurement of objects under impact loading. First part of the thesis reports the developmental work on building an LED-Grating based optical projection system for implementation of linear-fringe projection profilometry. Successful use of the developed system in measuring out-of-plane deformation experienced by multiple targets under impact loading with a time sapling of 20,000 frames per second is re-ported. However, for performing ballistic impact measurements using the liner-fringe projection method, an order of magnitude higher time-sampling is needed. This is due to the disadvantages associated with linear fringe projection technique: (1) results in wrapped phase map (2p ambiguity) estimation, and (2) the deformation/shift of the recorded fringe pattern in the modulation direction sets a limit on unambiguously measurable whole-plane displacement. Typically, fringe pitch dictates the limit of maximum detectable displacement, and thus to be able to capture larger deformation from the earlier state, coarser fringe pitch is required to be projected; while this adversely affects the resolution of measurement system. Hence, there is a need to develop a fringe projection system which has capability for whole-plane displacement without affecting the resolution and/or necessitating higher temporal-sampling. Circular Fringe Projection (CFP) technique is proposed in the second part of the thesis as a novel solution to address the above issues. CFP technique offers additional advantage of relaxing the temporal resolution requirements of the imaging system by decoupling the maximum measurable deformation rate and the frame rate of camera. ▼ Syfte: I ett flertal tidigare studier har positiva samband mellan hållbarhetsprestationer (CRP) och finansiella resultat (CFP) hittats och försökt förklaras. Dock har endast ett fåtal antagit synsättet att immateriella resurser fungerar som medlare för dessa samband. Vi menar att de immateriella resurserna har en betydande roll, för sambandet. Med hjälp av de immateriella resurserna formas en positiv spiral som ökar hållbarhetsprestationer och finansiellt resultat i båda kausala riktningarna. Vi vill belysa huruvida detta positiva, medlande samband existerar samt om tillväxt stärker sambandet mellan CRP och CFP, genom de immateriella resurserna. Metod: Studien har antagit en positivistisk forskningsfilosofi med en hypotetisk-deduktiv ansats. En kvantitativ metod har använts genom att sekundärdata samlats in från databasen Thomson Reuters Eikon. I en longitudinell design har fem års observationer använts för ett urval av 113 publika industriföretag där inhämtat data har analyserats statistiskt. Resultat & slutsats: Denna studie fann övervägande negativa samband och kan inte visa att de immateriella resurserna medlar kopplingen mellan CRP och CFP i en positiv spiral. Studiens resultat visar dock att immateriella resurser har betydelse för uppbyggnaden av CRP och CFP men de fungerar inte som verktyg som påverkar prestationsmåtten i båda kausala riktningarna. De har en egen effekt på CRP och CFP direkt vars verkan skiljer sig mellan de olika immateriella resurserna. Endast i tillväxt har studiens resultat visat på en positiv påverkan av CRP på CFP och tvärt om. Examensarbetets bidrag: Ur ett teoretiskt perspektiv bidrar studien till insikter om att immateriella resurser och satsningar på CRP är vägar att öka finansiella prestationer hos industriföretag i tillväxt. Praktiskt kan detta vara användbar kunskap för företagsledare i beslut om satsningar på immateriella resurser. Förslag till fortsatt forskning: Denna studie bygger på mätdata från en femårsperiod som sträcker sig över en tidsperiod då det råder lågkonjunktur. Studien bygger också på ett relativt litet urval som försvårar möjligheten att dra slutsatser med precision. Studien undersöker också endast fyra immateriella resurser. En studie med ett större urval under en längre tidsperiod där fler immateriella resurser tas med i beräkningsmodellen kan kasta ljus över hur de immateriella resurserna bidrar till förändringar i CRP och CFP.
