▼ All chapters in this thesis revolve around the general theme, stem cells and their electrophysiological characteristics and capacity to induce pro-arrhythmia. The first part of this thesis focusses on key aspects that are relevant to possible pro-arrhythmic effects of stem cell transplantation. An important factor in this context is intercellular coupling through gap junctions. Connexin43, the main cardiac gap junction protein, is poorly expressed in stem cells differentiating into mesodermal cells. We found that the transcription factor Snail1 represses functional connexin43 expression in differentiating stem cells. Furthermore, spontaneous activity as observed in stem cell-derived cardiomyocytes is a clear indication of pro-arrhythmic potential. Studies by our group and others have demonstrated poor expression of the potassium inward rectifier current in stem cell-derived cardiomyocytes. In this thesis, we demonstrate a proof of concept that spontaneous activity can be suppressed through electrotonic interaction by co-culture with Kir2.1 expressing cells. The exposure to increasing levels of catecholamines during development exerts an important influence on maturing cardiomyocytes in vivo. It may be possible that adrenergic stimulation is also beneficial to differentiation of immature cardiomyocytes in vitro. We found that beta-adrenergic stimulation increased conduction velocity, probably via increased expression of cardiac sodium and calcium channels. Evaluation of spontaneous activity and gap junctional communication between grafted cardiomyocytes and host myocardium is not feasible in vivo, therefore we devised an in vitro model that allowed us to integrate the key pro-arrhythmic aspects in an accessible model. In the second part, we describe a new human source for stem cell-derived cardiomyocytes, the human fetal cardiac progenitor cells. Given the fact that these cells originate from the heart, it is likely that they are committed to the mesodermal or cardiac lineage. We found that they can differentiate into cardiomyocytes with efficiencies up to 90% and have an electrical phenotype that is more mature than that observed in other stem cell-derived cardiomyocytes. These findings bring clinical use of autologous stem cells for cardiac regeneration a step closer but also provide us with a valuable in vitro model that may be useful in pharmacological studies.

Les travaux présentés dans ce mémoire ont pour objectif d’approfondir la compréhension de l’altération de la réponse à la stimulation des récepteurs β-adrénergiques dans plusieurs contextes physiopathologiques. La première étude confirme l’existence d’une dysfonction β-adrénergique à l’échelle du cardiomyocyte au cours de la sénescence. Elle met en lumière le rôle de la protéine MRP4 (multidrug resistance associated protein 4) dans cette diminution de réponse inotrope positive à l’isoprotérénol. La deuxième étude évalue la réponse à la stimulation des récepteurs β-adrénergiques dans le syndrome métabolique et montre que la dysfonction est modérée dans ce contexte même en cas de diabète associé à l’obésité. Ces résultats fonctionnels sont expliqués par la diminution d’expression des récepteurs β1- et β2-adrénergiques mais l’absence de surexpression du récepteur β3-adrénergique comme observée dans le diabète de type 1. La troisième étude analyse le rôle de l’atorvastatine sur la réponse β-adrénergique chez les diabétiques et les mécanismes de modulation de cette réponse par une étude du transcriptome cardiaque. Elle montre également que l’inhibition de la production d’oxyde nitrite améliore la réponse β-adrénergique. La quatrième étude a expliqué une part de la dysfonction β-adrénergique chez les diabétiques par la surexpression de MRP4. L’inhibition de MRP4 a permis de restaurer la réponse à l’isoprotérénol au cours de la cardiopathie diabétique. Au total, l’ensemble de nos travaux poursuit la description des mécanismes de la dysfonction β-adrénergique dans la sénescence et le diabète et souligne le rôle de MRP4.

The studies presented in this report looked for a better understanding of the altered response to stimulation of the β-adrenergic receptors in several physiopathological contexts. The first study confirms the alteration of the β-adrenergic response at the cardiomyocyte level in the senescent cardiomyopathy. The role of MRP4 (multidrug resistance associated protein 4) in the reduced inotropic response to isoproterenol is emphasized. The second study evaluates the response to β-adrenoceptors stimulation in the metabolic syndrome and shows mild dysfunction in this context even in obesity associate with diabetes. These functional results are explained by a reduced expression of β1- and β2-adrenergic receptors but no overexpression of β3-adrenoceptor as observed in type 1 diabetes. The third study analyzes the role of atorvastatin on the β-adrenergic response in the diabetic cardiomyopathy and the mechanisms involved by study of the cardiac transcriptome. The inhibition of nitrite oxide production improves the response to β-adrenoceptors stimulation in diabetic heart. The fourth study explained part of the β-adrenergic dysfunction in the diabetic cardiomyopathy by the overexpression of MRP4. The inhibition of this protein restored the response to isoproterenol during diabetic cardiomyopathy. ▼ In this thesis we analyzed genomic integration, transgenic-expression and adaptive transcriptional changes on RNA-basis in a transgenic (tg) rat model expressing the human Endothelin-A-receptor (ETAR). Therefore we used 5 different rat strands (L6341, L6351, L6888, L6353, L6888) generated by pronuclear microinjection which expressed the human ETAR under control of murine sm-22alpha promoter for smooth muscle cell specific expression. Identification of transgenic animals was done by conventional polymerase chain reaction (PCR). Furthermore we established a realtime PCR based assay for quantification of transgenic insertion. With this method we determined 8, 4 and 1 tg-copies in tg-heterozygote animals of L6351, L6888 and L6878 respectively. Furthermore we were able to distinguish homozygote and heterozygote transgenic animals. On RNA level we analyzed the transgenic expression of human ETAR in selected organs and vessels with RT-PCR technique. Whereas in L6341 no significant expression was determined, high expression was found in L6878 and L6351. In the other two lines moderate expression was found. Interestingly there was significant tg-expression in the organs of L6878. For further discrimination we used Realtime-PCR to quantify RNA- expression of tg ETAR, endogenous ETAR und ETBR in mesenteric arteries and aorta of L6878 and L6351 in one month and one year old animals. Whereas in aortic RNA of one month old animals higher transgenic expression was found in L6351, there was much higher transgenic expression in mesenteric RNA of L6878 compared with L6351. The transgenic receptor expression was nearly on the same level as the endogenous receptor expression. In one year old animals of both lines a downregulation of transgenic expression was recognized. Whereas by this time in L6351 almost no RNA-expression was detectable, L6878 still expressed significant levels of transgenic RNA. Interestingly in homozygote animals of L6878 transgenic expression was doubled compared to heterozygote animals of the same line. Furthermore endogenous ETAR expression was significant diminished in aortic RNA of L6878 in one year old animals. No expressional changes of endogenous ETBR-expression have been observed in any of the lines. Besides of described adoptions, one month old animals of both lines showed a higher alpha1B RNA expression in mesenteric arteries. In addition a higher expression of G-protein-coupled receptor kinase 2 (GRK 2) was detected in transgenic animals. Furthermore a strong correlation between alpha1BR and GRK2 RNA expression was recognized for transgenic and non- transgenic animals. These findings establish a basis for further phenotypic analysis of the transgenic rat model and provide possibilities for a deeper understanding of the ETAR function in vessels. ▼ Sudden cardiac arrest (SCA) is a leading killer in the United States. A large number of patients experience SCA outside of hospitals where there is an inherent delay in treatment due to slow first response times. In the out-of-hospital setting, ventricular fibrillation (VF)-induced SCA (VF-SCA) is the most common context in which the event manifests. VF leads to chaotic electrical and hence contractile function of the heart resulting in loss of cardiac output causing ischemia.Left untreated, electrical activity rapidly deteriorates, culminating in complete electrical failure or asystole. Survival rates for patients found in asystole are a mere 1%: asystole is a death sentence.Despite the high event and death rates of VF-SCA, little is known concerning the mechanisms of electrical failure. This dissertation represents the first concerted effort to understand the pathophysiology of electrical failure. The work contained herein can be divided into threeprojects. These projects were conducted in whole animal and isolated whole heart models of VFSCA.The first is a detailed examination of the complex patterns of electrical failure in VF-SCA. I found a highly complex pattern of electrical failure spanning the intra- and interchamber heterogeneities in three-dimensions. Second, I investigated the contributions of two canonical yet untested theories of electrical failure during VF-SCA: hyperkalemia and ATP-sensitive potassium channel (KATP) opening. I reached the surprising conclusion that neither hyperkalemia nor KATP opening—individually or combined—can explain the pattern of electrical failure in VF-SCA. Third, I investigated the effect of increasing metabolic demand on electrical failure during VF-SCAthrough increased p-adrenergic stimulation and excitation rate. The findings point toward a necessity to reexamine current resuscitation protocols. Specifically, the use of epinephrine (an α-and β-adrenergic agonist) may promote electrical failure during treatment of VF-SCA leading to increased mortality.

▼ La Tesis Doctoral versa sobre el tratamiento informativo que reciben las víctimas civiles, las víctimas políticas y las víctimas pertenecientes a las fuerzas de Seguridad del Estado asesinadas por ETA. Acotamos el marco de estudio en tres décadas diferentes: años 70, 80 y 90. Y el vehículo con el que viajar a través de nuestra investigación: la prensa. La muestra representativa se centra en tres periódicos españoles, con líneas editoriales diferentes, de relevancia en el escenario mediático, con una tirada mínima destacable y una larga trayectoria en la historia de la prensa. De ahí que seleccionamos a los siguientes periódicos: ABC, El Diario Vasco y El País. En nuestro objeto de estudio escogimos de entre todos los atentados perpetrados por ETA, aquellos que fueran de mayor transcendencia: informativa, política e histórica. En los años 70 seleccionamos los siguientes atentados: el 13 de septiembre de 1974, el 4 de octubre de 1976, el 5 de diciembre de 1978. En los años 80: el 23 de octubre de 1980, el 19 de junio de 1987, y el 11 de diciembre de 1987. Y en los años 90: el 29 de mayo de 1991, el 26 de julio de 1996, y el 12 de julio de 1997. Las fuentes utilizadas en nuestro trabajo de investigación han sido por una parte, los propios periódicos, que a su vez los utilizamos como muestra de la investigación, y por otra parte, las fuentes bibliográficas, manuales de metodología en ciencias de la información y también, manuales de historia para contextualizar los atentados. La hipótesis principal que nos lleva a emprender este viaje en el camino de la investigación es averiguar cuál es el tratamiento informativo que reciben las víctimas civiles, políticas y miembros de las Fuerzas de Seguridad del Estado desde tres líneas editoriales diferentes y su evolución a medida que avanzamos en el estudio comprendido en tres décadas distintas. De ahí que planteamos la investigación desde una metodología que se divide en dos vertientes, el análisis cualitativo de contenido y el análisis cuantitativo del material textual y gráfico. En la primera vertiente, llevamos a cabo un análisis cualitativo de contenido, para ello nos inspiramos en las denominadas macroproposiciones que apunta Van Dijk en su obra Racismo y análisis crítico. Nuestro punto de partida, saber qué premisas son las más destacadas en las informaciones publicadas. En este mismo apartado nos preguntamos cuáles son los términos utilizados para referirse a las víctimas, los terroristas y el suceso en sí, así como los personajes que se enmarcan en las informaciones de los atentados además de las víctimas y los terroristas. En la segunda vertiente realizamos un análisis cuantitativo inspirado en la obra de Teresa Sábada Framing: el encuadre de las noticias. En este apartado contabilizamos la repercusión informativa en tiempo y espacio, los géneros periodísticos y la atribución de fuentes. ▼ All chapters in this thesis revolve around the general theme, stem cells and their electrophysiological characteristics and capacity to induce pro-arrhythmia. The first part of this thesis focusses on key aspects that are relevant to possible pro-arrhythmic effects of stem cell transplantation. An important factor in this context is intercellular coupling through gap junctions. Connexin43, the main cardiac gap junction protein, is poorly expressed in stem cells differentiating into mesodermal cells. We found that the transcription factor Snail1 represses functional connexin43 expression in differentiating stem cells. Furthermore, spontaneous activity as observed in stem cell-derived cardiomyocytes is a clear indication of pro-arrhythmic potential. Studies by our group and others have demonstrated poor expression of the potassium inward rectifier current in stem cell-derived cardiomyocytes. In this thesis, we demonstrate a proof of concept that spontaneous activity can be suppressed through electrotonic interaction by co-culture with Kir2.1 expressing cells. The exposure to increasing levels of catecholamines during development exerts an important influence on maturing cardiomyocytes in vivo. It may be possible that adrenergic stimulation is also beneficial to differentiation of immature cardiomyocytes in vitro. We found that beta-adrenergic stimulation increased conduction velocity, probably via increased expression of cardiac sodium and calcium channels. Evaluation of spontaneous activity and gap junctional communication between grafted cardiomyocytes and host myocardium is not feasible in vivo, therefore we devised an in vitro model that allowed us to integrate the key pro-arrhythmic aspects in an accessible model. In the second part, we describe a new human source for stem cell-derived cardiomyocytes, the human fetal cardiac progenitor cells. Given the fact that these cells originate from the heart, it is likely that they are committed to the mesodermal or cardiac lineage. We found that they can differentiate into cardiomyocytes with efficiencies up to 90% and have an electrical phenotype that is more mature than that observed in other stem cell-derived cardiomyocytes. These findings bring clinical use of autologous stem cells for cardiac regeneration a step closer but also provide us with a valuable in vitro model that may be useful in pharmacological studies.

Introduction: The Interferential current (IC) is an electrotherapeutic modality widely used for analgesic purposes, however, few studies have sought to evaluate its mechanisms of action to promote the antinociceptive effect. The aim of this study was to investigate the involvement of spinal serotonergic and noradrenergic receptors in the antinociceptive effect of the interferential current in the animal model of articular inflammation in Wistar rats. Material and method: We included in this study 54 male Wistar rats distributed in the groups CI + saline, CI + DMSO 20%, inactive IC + saline, inactive IC + DMSO 20%, CI + methysergide, CI + tropanil dichlorobenzoate, CI + ioimbine, serotonin hydrochloride and hydrochloride of clonidine. At baseline, behavioral tests were performed for sensitivity and motor performance followed by induction of joint inflammation by injection of 3% carragein solution and 3% caolin in the left knee of the animals. After 24 h, the behavior was reevaluated followed by treatment with the current, preceded or not by pharmacological blockade. The pharmacological blockade was performed by means of intrathecal injection, applied before the treatment with the current. One hours after treatment with IC, the behavioral tests were repeated. The values of p < 0.05 were considered significant. Results: There was a significant decrease in the mechanical threshold of left paw withdrawal 24 h after the induction of inflammation in all groups (p < 0.0001). The groups treated with interferential current that received vehicles (saline or DMSO 20%) Significantly increased the mechanical threshold of paw withdrawal in comparison to the pre-treatment moment (p ≤ 0.001). Both with very large effect size according to Cohen's d (d = 1.95 and d = 1.83, respectively). There was blockade of the effect of IC in the groups that received, prior to the treatment with the current, the methysergide (non-selective antagonist of receptors 5-HT1 and 5-HT2) and tropanyl dichlorobenzoate (receptor antagonist 5-HT3). In these two groups, there was no statistically significant difference in the mechanical threshold of left paw withdrawal, when comparing the moments before and after treatment. In addition, after treatment, the pre-treated groups with methysergide or tropanyl dichlorobenzoate did not increase the mechanical threshold of left paw withdrawal, different from the vehicle groups. The animals that received yohimbine (α2-adrenergic antagonist) increased the mechanical threshold for the removal of the inflamed paw when comparing the pre-and post-treatment moments, showing that this pharmacological blockade did not prevent the effect of IC. Intrathecal injection of serotonergic and α2-adrenergic receptor agonists (serotonin hydrochloride and clonidine) significantly increased the mechanical threshold of left paw withdrawal (p ≤ 0.004). Resumo: Embora haja demonstração de que a estimulação simpática tenha efeito facilitatório sobre a indução de atividade espontânea em miocárdio há controvérsia sobre a participação dos tipos de adrenoceptores na mediação deste efeito. No presente trabalho, descrevemos um método desenvolvido para determinação, em átrio esquerdo isolado (AE) de rato, da relação concentração-efeito para agentes que exercem efeito arritmogênico por aumento da mobilização celular de 'Ca POT. 2+¿. O método baseou-se na interposição de pausas estimulatórias, durante as quais registraram-se contrações espontâneas (CE), precedidas ou não por trens estimulatórios de alta freqüência (5 Hz). O protocolo estimulatório foi repetido na presença de diferentes concentrações de agonistas. Para cada concentração de agonista, a resposta arrítmica foi considerada como a soma dos números de CE/min, no total de preparações. Foi analisada também a resposta inotrópica, como sendo o incremento de força ou encurtamento de pico, desenvolvidos em AE e miócitos ventriculares (MV), respectivamente. A relação foi ajustada por uma função sigmóide para cálculo de Min (i.e., valor da variável na ausência do agonista), Rmax (resposta máxima) e pD2 (-log da concentração do agonista que produziu uma resposta igual a 50% de Rmax). Este método foi aplicado no estudo dos tipos de adrenoceptores envolvidos na resposta arrítmica a catecolaminas em AE e MV. A Rmax inotrópica à ativação de adrenoceptores 'alfa IND. 1¿ + 'beta IND. 1¿ foi comparável àquela por ativação de apenas receptores 'beta IND. 1¿, em ambos AE e MV. Já a ativação de adrenoceptores 'alfa IND. 1¿ produziu uma Rmax inotrópica de apenas metade daquela observada pela estimulação 'alfa IND. 1¿ + 'beta IND. 1¿. Da mesma forma, a resposta arrítmica foi semelhante para estimulação de adrenoceptores a1+ß1 e de apenas receptores 'beta IND. 1¿. Entretanto, nenhuma resposta foi obtida pela estimulação de receptores adrenérgico do tipo 'alfa IND. 1¿. Estes resultados indicam que a estimulação de adrenoceptores 'alfa IND. 1¿, apesar de evocar uma resposta inotrópica positiva em ambos AE e MV, não é arritmogênica. A ativação do tipo 'beta¿, por outro lado, parece ser a principal via para estimulação inotrópica simpática e na indução de arritmias. ▼ All chapters in this thesis revolve around the general theme, stem cells and their electrophysiological characteristics and capacity to induce pro-arrhythmia. The first part of this thesis focusses on key aspects that are relevant to possible pro-arrhythmic effects of stem cell transplantation. An important factor in this context is intercellular coupling through gap junctions. Connexin43, the main cardiac gap junction protein, is poorly expressed in stem cells differentiating into mesodermal cells. We found that the transcription factor Snail1 represses functional connexin43 expression in differentiating stem cells. Furthermore, spontaneous activity as observed in stem cell-derived cardiomyocytes is a clear indication of pro-arrhythmic potential. Studies by our group and others have demonstrated poor expression of the potassium inward rectifier current in stem cell-derived cardiomyocytes. In this thesis, we demonstrate a proof of concept that spontaneous activity can be suppressed through electrotonic interaction by co-culture with Kir2.1 expressing cells. The exposure to increasing levels of catecholamines during development exerts an important influence on maturing cardiomyocytes in vivo. It may be possible that adrenergic stimulation is also beneficial to differentiation of immature cardiomyocytes in vitro. We found that beta-adrenergic stimulation increased conduction velocity, probably via increased expression of cardiac sodium and calcium channels. Evaluation of spontaneous activity and gap junctional communication between grafted cardiomyocytes and host myocardium is not feasible in vivo, therefore we devised an in vitro model that allowed us to integrate the key pro-arrhythmic aspects in an accessible model. In the second part, we describe a new human source for stem cell-derived cardiomyocytes, the human fetal cardiac progenitor cells. Given the fact that these cells originate from the heart, it is likely that they are committed to the mesodermal or cardiac lineage. We found that they can differentiate into cardiomyocytes with efficiencies up to 90% and have an electrical phenotype that is more mature than that observed in other stem cell-derived cardiomyocytes. These findings bring clinical use of autologous stem cells for cardiac regeneration a step closer but also provide us with a valuable in vitro model that may be useful in pharmacological studies.

▼ It is common to characterize adrenergic receptors by placing them in one of two categories, alpha or beta. The receptors responsible for adrenergically induced increase in cardiac contractile force and glycogenolysis are usually classified as beta receptors, but such a classification is not definitely established. Therefore, an attempt was made in the present study to characterize the nature of the receptors responsible for increases in cardiac contractile force and glycogenolysis induced by various adrenergic amines, The effects of rapid injections of phenylephrine and norepinephrine (amines which excite only alpha receptors) and epipinephrine (amine which excites alpha and beta receptors) and isoproterenol (amine which excites beta receptors) were measured in dogs, on blood pressure and cardiac contractile force before and after phenoxybenzamine or Hydergine (alpha receptor blocking agents) or dichloroisoproterenol (beta receptor blocking agent) or d-tubocurarine (neuromuscular blocking agent). Cardiac contractile force was measured by a strain gauge sutured on the ventricule wall and average blood pressure was measured from the carotid artery. Phenoxybenzamine did not alter the cardiac contractile effects of any of the amines and d-tubocurarine did not alter the cardiovascular responses of any of the amines. Hydergine inhibited the cardiac contractile force response of phenylephrine and norepinephrine, and dichloroisoproterenol inhibited this response of all four of the amines. Phenoxybenzamine and Hydergine blocked the blood pressure responses of all of the amines except isoproterenol. Dichloroisoproterenol blocked the blood pressure response of only isoproterenol. These results show that both alpha or beta adrenergic amines can increase cardiac contractile force and that dichloroisoproterenol can inhibit this response of all four of the amines, while Hydergine can produce a selective blockade with two of the amines. This led to the conclusion that the cardiac receptor responsible for adrenergically induced increases in cardiac contractile force can be excited by either alpha or beta adrenergic amines and it is therefore suggested that this receptor be reclassified as an undifferentiated receptor. The glycogenolytic effects of adrenergic amines, in dogs, were determined by measuring the elevation of serum levels of of glucose and lactic acid produced by slow infusions of the four adrenergic amines before and after blocking agents. Serum levels of glucose and lactic acid were significantly elevated by epinephrine and isoproterenol, but not by phenylephrine or norepinephrine. Dichloroisoproterenol or Hydergine significantly reduced the hyperglycemic and hyperlactic acidemic responses of epinephrine, but d-tubocurarine or phenoxybenzamine did not. The hyperglycemic and hyperlactic acidemic responses of isoproterenol were inhibited by d-tubocurarine or dichloroisoproterenol, but not by Hydergine. ▼ The hippocampus is a functionally heterogeneous structure with the cognitive and emotional signal processing ascribed to the dorsal (DH) and the ventral hippocampus (VH) respectively. However, the underlying mechanisms are poorly understood. In the present study, which is consisted of three parts, we used field excitatory postsynaptic potentials (fEPSPs) from transverse rat hippocampal slices, which were maintained in a recording chamber, in order to determine which proteins might play a role in dorsoventral diversification. In the first part of this study we examined the short-term synaptic plasticity, which plays fundamental roles in information processing and may be importantly involved in diversifying the properties of local neuronal network along the hippocampus long axis. Therefore, we aimed to examine the properties of the CA1 synapses along the entire dorsoventral axis of the rat hippocampus and a frequency stimulation paradigm. Applying a ten-pulse stimulus train at frequencies from 0.1 to 100 Hz to the Schaffer collaterals we found a gradually diversified pattern of frequency-dependent synaptic effects along the dorsoventral hippocampus axis. The first conditioned response was facilitated along the whole hippocampus for stimulus frequencies 10-40 Hz. However, steady-state responses or averaged responses generally ranged from maximum synaptic facilitation in the most dorsal segment of the hippocampus to maximum synaptic depression in the most ventral hippocampus. In particular, dorsal synapses facilitated for stimulus frequency up to 50 Hz while they depressed at higher frequencies (75-100 Hz). Facilitation at dorsal synapses was maximal at stimulus frequency of 20 Hz. On the contrary, the most ventral synapses showed depression regardless of the stimulus frequency, only displaying a transient facilitation at the beginning of 10-50 Hz stimulation. Importantly, the synapses in the medial hippocampus displayed a transitory behavior. Finally, as a whole the hippocampal synapses maximally facilitated at 20 Hz and increasingly depressed at 50-100 Hz. The aforementioned results indicate that the short-term synaptic dynamics change gradually along the hippocampal long axis conveying distinct properties of information processing to successive segments of the structure crucially supporting functional segregation along the dorsoventral axis of the hippocampus. In the second part of this study we analysed the effects of β adrenergic receptors activation and blockade in long term potentiation, induced after theta burst stimulation and high 5 frequency stimulation in dorsal and ventral pole of the hippocampus. Noradrenaline is released during emotional arousal in the hippocampus and through activation of β adrenergic receptors (β-AR) modulates synaptic plasticity and memory consolidation. ▼ This thesis studies the applications of index theory to positive scalar curvature (PSC), in particular questions of existence and number of path components of the moduli space of PSC metrics. After Atiyah-Singer proved their legendary index theorem [AS63, AS68a, AS68b], many fruitful applications to positive scalar curvature were discovered, for instance Lichnerowicz's obstruction to positive scalar curvature metrics on spin manifolds with nonvanishing A-hat genus [Lic63]. The theorem of Lichnerowicz relies notably on the existence of a spin Dirac operator on any spin manifold|a self-adjoint, elliptic, first order differential operator having marvellous connections to the geometry of the underlying Riemannian manifold. In 1975, Atiyah, Patodi and Singer [APS75a, APS75b, APS76] proved their index theorem for a Dirac operator D on a manifold Z with boundary @Z = Y . This takes a similar form to the Atiyah-Singer index theorem but notably has a correction term n(A) = (nA(0)+h)=2, called the eta invariant, appearing for the boundary. The eta invariant is defined solely in terms of the spectrum of the Dirac operator A on the boundary, so is a spectral invariant. As it stands, this invariant is not at all robust; if one slightly perturbs the metric on Y then most likely one will produce a change in the eta invariant. A notable exception to this is conformal deformations, which leave the Dirac operator unchanged. There is, however, a more robust invariant which can be procured from the eta invariant. If one twists the Dirac operator A on Y by two unitary representations δ1; δ2 : π1(Y ) → U(N) of the fundamental group of Y , one obtains two twisted Dirac operators A1 and A2 on Y which are locally isomorphic. Subtracting the eta invariants of these twisted Dirac operators yields a modified invariant p(δ1; δ2;A), called the rho invariant. The rho invariant still isn't quite robust, but upon taking the mod Z reduction of the rho invariant, many striking invariance properties emerge. For example, writing = 1(Y ), the rho invariant descends to a well-defined map on geometric K-homology [HR10]: p(δ1; δ2) : K1(Bπ) → R=/Z. The rho invariant can be used to further study the properties of PSC metrics on manifolds. Whereas the Atiyah-Singer theorem is mostly useful for even-dimensional manifolds, the APS index theorem allows one to obtain results for odd-dimensional manifolds by considering them as boundaries of even-dimensional manifolds. In particular, one can obtain obstructions to PSC [HR10], and study the number of path components of the moduli space of PSC metrics on a manifold [BG95]. More recently, Mrowka, Ruberman and Saveliev [MRS16] discovered and proved a new index theorem for manifolds with periodic ends. Roughly speaking, these are manifolds Z∞ which have a compact piece Z, attached to which are one or more ends which repeat themselves periodically off to infinity. ▼ Eta Carinae (! Car) is the most massive, most luminous star in our region of the Galaxy. It is an evolved massive star system and therefore provides many clues to the fate of the most massive stars. In the 1840s its unstable nature culminated in the Great Eruption when it briefly became the second brightest star in the sky and ejected more than ten solar masses, which today enshroud the surviving star as a bipolar nebula. The “supernova impostor” phenomenon and its aftermath constitute a major gap in the theory of massive stars, and ! Car is the only example that can be studied in detail. Its recovery has been unsteady with unexplained photometric and spectral changes in the 1890s and 1940s. Combining data from HST STIS and Gemini-S GMOS between 1998 and 2010, I analyzed several spectroscopic cycles that occur every 5.54 years. In addition, I used some data from the VLT UVES, Magellan II MIKE, and Ir´en´ee du Pont B&C instruments. Observations with a variety of different slit position angles made it possible to map the emission across the nebula and the complex outer ejecta of ! Car permit to observe the star at different stellar latitudes via reflected light. In order to study the distribution of gas and ionizing radiation around ! Car and their implications for its likely companion star, I examined several high-excitation emission lines. The principal results are: (1) The high-excitation fluxes varied systematically and non-trivially throughout ! Car’s 5.5-year spectroscopic cycle. (2) A brief, strong secondary maximum occurred just before the 2003.5 spectroscopic event. (3) These emission lines are strongly concentrated at the Weigelt knots several hundred AU northwest of the star. With less certainty, [Ne III] appears to be somewhat more concentrated than [Fe III]. (4) A faster, blueshifted component appears concentrated near the star and elongated perpendicular to the system’s bipolar axis. (5) Using the photoionization program Cloudy, I estimated the range of parameters for the hot secondary star that would give satisfactory high-excitation line ratios in the close ejecta; Teff ! 40, 000 K, L " 4×105 L!, and Minit " 40–50 M!, for example, would be satisfactory. The allowed region in parameter space is wider and mostly less luminous than some previous authors suggested. Spectra obtained with Gemini GMOS throughout 2007–2010 were used to observe the 2009 spectroscopic event from different stellar latitudes. The He II "4687 emission, only observed during the “events,” was analyzed in spectra in direct view and in reflected polar-on spectra at FOS4. The time-delay of He II at FOS4 is about 18 days and therefore consistent with the predicted additional time-travel time at FOS4. The equivalent width and radial velocity behavior of He II at FOS4 mirrors the behavior observed in direct view. ▼ El terrorismo vasco ha marcado la actualidad de este país tanto en el plano nacional como internacional durante más de 40 años, ha ido construyendo la agenda mediática española que y creando un lenguaje propio y una visión del denominado “conflicto vasco” así como una forma de entender el mismo. La sociedad conoce la historia de ETA más que por los libros por los informativos, por la prensa, por la radio, por la red... Todo ello ha ido conformando nuestro imaginario colectivo en torno a esta cuestión y nos ha hecho participes en muchas ocasiones de esa forma de entender el problema tan mediática. El cine como constructor de realidades es capaz de crear nuevos universos, teletransportarnos a otros lugares, crear sensaciones y emociones y en definitiva hacernos sentir y sobre todo provocarnos pensar.Existe una creencia bastante generalizada de que el cine de alguna manera ha dado la espalda al tema de ETA. Numerosos artículos publicados fundamentalmente en prensa han criticado ferozmente la falta de títulos cinematográficos realizados en España que tratasen de una manera directa y contundente el tema del terrorismo en nuestro país.El documental es un género que supuestamente trabaja sobre realidades. Al género de no ficción se le presupone una serie de requisitos que lo convierten en un género informativo. El documental en aras de mostrar una realidad concreta, con personajes reales y con la utilización de recursos como las imágenes de archivo, trata de abordar una cuestión desde perspectivas que en un principio no deberían distorsionar la realidad en cuestión sino más bien aclararla y provocar una reacción en el espectador que puede pasar desde el reforzamiento de una idea previa sobre el tema o desmontar una teoría sobre el mismo o aportar un punto de vista novedoso sobre una cuestión en concreto.El grueso de las películas seleccionadas lo conforman el trabajo de directores como Iñaki Arteta y el tandem formado por Eterio Ortega y Elías Querejeta ya que son dos realizadores que han centrado su carrera en el cine documental sobre el ETA y el “conflicto vasco” Advisors/Committee Members: Veres Cortés, Luis (advisor).
