▼ Diabetes mellitus is correlated with modifications in bone microarchitectural and mechanical strength, leading to increased bone fragility. The incretin hormones, with a classical effect to increase insulin secretion following food ingestion, are now postulated to have important direct effects on bone. As such, glucose-dependent insulinotropic polypeptide (GIP) has dual actions on bone cells; enhancing bone-forming activity of osteoblasts and suppressing bone resorption by osteoclasts. The sister incretin of GIP, glucagon-like peptide-1 (GLP-1), is also suspected to directly influence bone health in a beneficial manner, although mechanism are less clear at present. The physiological actions of incretins are attenuated by dipeptidyl peptidase (DPP-4) activity and it is speculated that introduction of DPP-4 inhibitor may also positively affect quality of the skeleton. As such, this thesis evaluates the potential beneficial effects of a DPP-4 resistant GIP analogue, namely [D-Ala2]GIP, on osteoblastic-derived, SaOS-2 cells, and also preliminary in vivo studies on the impact of genetic deficiencies of GIPRs and GLP-1Rs on bone mineral density and content. Further studies characterised the beneficial effects of incretin-based therapies on metabolic control, bone microstructure and bone mechanical integrity in animal models of pharmacologically-, genetically- and environmentally-induced diabetes. GIP and related stable analogue increased bone-forming biomarkers in SaOS-2 cells and importantly, [D-Ala2]GIP was shown to be more potent than native GIP. Knockout mouse studies revealed that both GIPR and GLP-1R signaling are important for optimum bone mass. All diabetic mouse models displayed reduced bone mass, altered bone micromorphology and impairment of bone mechanical strength, similar to the human situation, confirming their appropriateness. The incretin-based therapeutics, [D-Ala2]GIP and Liraglutide, in streptozotocin-diabetic significantly increased bone matrix properties, indicating recovery of bone strength at the tissue level. The beneficial effects of administration of [D-Ala2]GIP-oxyntomodulin on bone health in db/db mice were more prominent as the Oxm analogue did not only improve bone strength at tissue level, but also at whole-bone level. These modifications were independent of metabolic status. Twice-daily Exendin-4 therapy improved glycaemic control and increased work required to resist bone fracture in high-fat fed mice. It was also established that Sitagliptin had neutral effects on bone microstructure and mechanical strength in high-fat mice. In

summary, these data demonstrate the negative impact of diabetes mellitus on normal skeleton development and bone quality. Moreover, this thesis highlights the growing potential of incretin-based therapies for ameliorating bone defects and improving the increased fragility fracture risk associated with diabetes.

▼ Northern Ireland’s recent Property Market Bubble of 2007 has been particularly severe in comparison to other historical and international experiences (Reinhart and Rogoff, 2009). Despite this there is currently, a dearth of literature and understanding relative to the Northern Ireland case, which poses a number of challenges for key stakeholders attempting to mitigate the adverse outcomes from the event and, prevent a future occurrence. Therefore, this research is positioned to address this gap. More specifically, informed by neoclassical and Keynesian theories on asset market bubbles and organisational theory, the study explores the bubble phenomenon in Northern Ireland, the key contributory factors that led to its emergence and, seeks to provide some insight into its consequences. To examine this topic area in more detail, underpinned by a pragmatic stance, a sequential mixed methods approach was employed. First, a quantitative recursive regression technique pioneered by Phillips and Yu (2011) was utilised to confirm and model the bubble and to date the phenomenon. Building on this, next, in-depth, semi-structured interviews were used in an exploratory triangulated manner to gain a comprehensive understanding of the key contributory factors and consequences from a stakeholder perspective. The findings obtained confirm bubble behaviour in the context of Northern Ireland from 2005 Q3 to 2009 Q1 and illustrate that the reasons for the bubble are complex and wide-ranging. A number of contributory factors were identified including, spill-over effects from the Republic of Ireland, inadequate risk management and lax mortgage and commercial lending practices, issues associated with the internal organisational structures of the banks and, the nature of the planning system in operation over the period. However, behavioural biases in particular, herding at the individual and institutional levels were identified as playing a significant underlying role by influencing the behaviours of key participants including, banks and property developers. As such, the main conclusion drawn from the analysis is that Keynesian theory may be more applicable in understanding Northern Ireland’s recent Property Market Bubble. Based on the findings a number of important theoretical implications and practical recommendations were developed. Theoretically, the study contributes and extends the literature in the broad area of asset market bubbles and in particular, relative to Keynesian theory. It also contributes to the knowledge base in the field of organisational theory. On a more practical level, several recommendations relative to banks, regulatory and supervisory authorities and government are forwarded.

▼ The diagnostic criteria for posttraumatic stress disorder (PTSD) have been a topic of heated debates for several decades. In 2013, the fifth edition of the Diagnostic and Statistical Manual of Mental Disorders (DSM-5) introduced significant changes to the way in which this disorder is diagnosed. For the first time in the history of PTSD in the DSM, the disorder was characterized by four latent factors (i.e., symptom clusters) and a dissociative subtype of PTSD was added as a diagnostic sub-category. Consequently, studies examining the latent structure of PTSD and the existence of the dissociative PTSD subtype started emerging all around the world, but primarily in the United States. The aim of the current thesis was to contribute to and extend this emerging literature by examining these two major changes in the diagnostic criteria for PTSD in Slovakia; a country in Eastern Europe, where DSM-related research has been largely neglected. A systematic literature review of PTSD’s latent structure (Chapter 3) and the reviews in Chapters 4 and 6 of this thesis showed that there have been no studies on this topic conducted in Slovakia. The empirical chapters of this thesis utilized data collected from students at six universities in Slovakia. Contrary to the existing literature and the DSM- 5, the best-fitting model of PTSD was the six-factor Anhedonia model (Chapter 4), the construct validity of which was supported in relation to the external variables of anxiety and depression (Chapter 5). Chapters 6, 7 and 8 o f this thesis examined the dissociative PTSD subtype and supported the existence of this diagnostic construct in Slovakia. However, the analyses suggested that the DSM-5 definition of the dissociative PTSD subtype may be inadequate. The results have important implications for future revisions of the PTSD diagnostic criteria in the DSM.

▼ The ethno-nationalist conflict in Israel/Palestine has been the subject of significant academic interrogation. However, the political ecological dimensions of that conflict, despite their importance, have gone largely unnoticed. The natural environment, for example, is central to national identity constructs that have long been a source of contestation and friction in the region. The scarcity of vital natural resources (both land and water) needed to sustain nation-building efforts continue to occasion conflict. Access to and control of such resources is divisive and the environment has become a weapon through which to contest the ‘other’. Whilst there is a deep-rooted attachment and importance given to the environment, it has also been a victim to the conflict itself often being seconded to the ‘high political’ aspects prioritised at the governmental level At the opposite end of the spectrum, there are also numerous efforts which attempt to use the environment to promote cooperation/peacebuilding between actors in the conflict. This thesis attempts to unpack the often-overlooked political ecological dimensions of the Israeli-Palestinian conflict. To do so, it focuses on a number of specific, localized and illustrating case studies in a series of three papers. The first analyses the impact of the Israeli West Bank separation barrier on Palestinian agricultural systems and processes. The second paper, analyses local attempts at Israeli-Palestinian ‘environmental cooperation’ and asserts the need for delving deeper into ‘cooperative’ interactions in order to determine their true nature and effectiveness. The third paper takes the same greater Bethlehem case studies and explores their possible peacebuilding contributions, before suggesting ways to improve their potential in that regard. As well as filling a significant vacuum in Israel/Palestine scholarship, the thesis has broader theoretical and practical relevance. It adds greatly to vibrant contemporary academic discussions, debates, and lines of enquiry pertaining to the relationships between the environment, conflict and peace. It also speaks to recent calls for geographers to research and contribute to ‘a geography of peace’. In a desire to employ a holistic lens (in this case a political ecological one), an inductive approach was pursued to facilitate the emergence of new ideas. That is, instead of having rigid, predefined theories to test, the approach, whilst including key ideas related to the broad conceptual framework, was to remain open-minded to what emerged in discussions with those involved in the scenarios, and through the analysis of a multitude of information sources. Key methods of primary data collection included semi-structured interviews and ethnographic observation. Primary data collection occurred remotely via ‘e-interviews’ between 2012 and 2013, and directly during a period of fieldwork to Israel/Palestine in 2014. A total of 40 interviews were completed. ▼ Proposing an ontological division based on the terms photobook and photobookwork, the main goal of this thesis is to examine and ascertain the historical, material and conceptual characteristics of the latter book practices. Conducted from a critical perspective, the analysis that structures this project aims in the first place to demonstrate how a series of interconnected material, cultural and aesthetic developments enabled the emergence and maturation of photobook practice between the late 1830s and the early 1890s. The project also explores how the materialization of photographically illustrated cultural magazines in the late nineteenth-century catalysed a transformation of visual discourse and literacy that later enabled the full development of photobookwork practice. Importantly, this thesis suggests that photographically illustrated book making fully branched into two distinct forms in the mid 1920s. Whereas until that period photographic illustration in book form was predominantly rooted in the single photographic image, modernist photobook makers, greatly influenced by the visual discourse of photographically illustrated periodicals, developed a photographically illustrated book practice centred on cumulative and relational photographic narratives that traversed the entirety of the book. Ultimately, this project attempts to recalibrate the somewhat monolithic and nebulous perception of photobook history proposed in recent general ‘photobook’ scholarship, while simultaneously contributing towards a better understanding of the historical, material and ontological characteristics that define the two forms of photographic illustration in book form discussed throughout this thesis.

▼ Following the expansion of the European Union (EU) in 2004 with the accession of the A8 countries namely, Poland, Lithuania, Latvia, Hungary, Slovakia, Slovenia, Czech Republic and Estonia, the United Kingdom (UK) experienced an influx of nationals from these countries. This was down to two main factors, firstly the UK was one of only three EU nations, alongside Sweden and Republic of Ireland, to place no restrictions on the right of these individuals to travel and secondly, they were given the right to join the labour market and seek work. This study used both quantitative and qualitative methods to investigate the impact these arriving A8 migrant workers had on those organisations with the manufacturing industry in Northern Ireland who employed them. An initial phase of quantitative data collection was utilised to create a research framework, that was then used to select those organisations and individuals who participated in the qualitative phase(s). It considers management and organisational issues by considering the literature on the use of human resources giving competitive advantage (SHRM) and the key aspects of diversity and equality including sameness/difference, the business, social justice and legal cases for diversity management. Results showed that issues such as language are key in aiding inclusion and reducing friction in the workplace, indigenous workers can feel threatened when migrants speak in the native language and conversely on many occasions, migrant workers do not have the required language skills to communicate confidently without an interpreter. This lead to the evolvement of ‘informal leaders’, those migrant workers who, found themselves acting as spokesperson for their colleagues and used as interpreters and or a negotiator. In conclusion, an employer typology is developed that visualises the behaviour and outlook of organisations in relation to the key aspects of diversity as previously mentioned.

▼ An investigation was carried out to quantify and prioritise the importance of factors that influence energy related decisions in Northern Ireland households. This was achieved by using the largest energy use population survey to date in Northern Ireland, analysis of actual electricity consumption data and analysis using analytical hierarchical modelling. The use of Analytical Hierarchical modelling has been developed for this investigation and has provided a novel way of quantifying the importance of various factors that influence household energy use. 85% of households are interested in reducing their energy use motivated mainly by a desire to save money (86%); by comparison, only 9% are interested in reducing their carbon footprint and 5% due to a sense of obligation. The per person electricity consumption by apartment dwellers was the smallest compared to households in other types of homes. Those in the rental group found measures where there is a sacrifice in comfort or potential well-being were more acceptable as a strategy to reduce energy use compared to owner occupiers, and the latter would prefer to invest in 'fit and forget' measures. Sacrificial measures were the last acceptable strategy for the older age bracket, identifying behavioural change measures as the most acceptable strategy. The group as a whole prioritised the importance of energy related considerations (with the quantified weightings shown in brackets) and are as follows: implementing measures that will reduce household running costs (22.4%); measures with minimal maintenance requirements (11.4%); measures that will improve comfort (10.4%); lower cost measures (10.4%); measures that will bring added convenience (10.4%); measures that will modernise the home (10.4%); measures with a short payback period (10%); measures that will cause minimum disruption during installation (9.3%); and measures that will contribute to a low carbon footprint (5.4%). Low carbon footprint is the least important consideration. 9% were motivated by a need to reduce their carbon footprint. When considered in isolation, this group found the need to lower their carbon footprint a high priority, with a weighting of 15.5% the second most important consideration after the need to reduce household running costs, which had a weighting of 16%. The promotion of multi-generational households and encouraging downsizing from large under-occupied homes to more size appropriate housing units combined with specific legislation requiring a minimum standard of energy efficiency for homes in the private rented sector is needed, in light of the high levels of fuel poverty in Northern Ireland.

▼ There is an urgent need for site-specific drug delivery systems or new treatment options with improved targeting capability. This thesis investigated new methods for targeted delivery of sensitisers and drugs to solid tumours. Chapter 3 details the development of a pH responsive sensitiser to exploit the lower pH present in cancer tissue when compared to non-cancer tissue as a targeting method. Modifying pH enabled control over ROS generation and cellular toxicity which was confirmed both in vitro and in vivo. In Chapter 4, an amphiphilic polymer was used to encapsulate the photosensitiser hypericin and enable its dispersion in aqueous media. Targeting was increased by attaching folic acid to the polymeric micelle and its effectiveness compared to a non-labelled analogue was determined through studying the PDT mediated toxicity in folate receptor positive (FR+ve) and folate receptor negative (FR-ve) cell lines. A significantly improved efficacy was observed in the FR+ve cell line for the labelled complex while no significant difference was observed in the FR-ve cell line. Chapter 5 describes the synthesis of a near infrared (NIR) absorbing sensitiser containing multiple iodine atoms, to overcome the issue of light penetration while also limiting the problem associated with activation by ambient light. The efficacy was determined in two pancreatic cancer cell lines and an in vivo murine model of the disease. A significant NIR activated PDT mediated reduction in cell viability was found compared to the non-iodinated analogue. The di-iodinated derivative also produced significant PDT mediated reductions in vivo. In Chapter 6 an azide functionalised doxorubicin prodrug was prepared and encapsulated in a polymeric micelle. When delivered to HeLa cells along with a similar micelle containing PPh3 in the core, a significant reduction in cell viability was observed, as the prodrug is effectively converted to the active doxorubicin via an intracellular Staudinger reaction.

▼ Sleep is a vital physiological process, which is essential for health and wellbeing. Reported sleep problems are, however, commonplace within the general population. Currently, the literature indicates clinically validated, long term sleep monitoring is best achieved using Wrist Based Sleep Actigraphy (WBSA). WBSA, however, is prohibitively expensive to the general population and requires contact with the body to operate, leading to accessibility and user compliance issues. Correspondingly, a literature review found that neither research nor commercial domains have suitably explored the potential for non-contact, actigraphy driven, sleep monitoring. Accordingly, this Thesis presents a series of investigative studies into the development of Passive Sleep Actigraphy (PSA), a bed-based, non-contact method of recording actigraphic data from a bed occupant for the purpose of sleep profiling. The first investigation considered suitable hardware for recording in-bed movement without body contact, where it was discovered that a range of in-bed postural changes could be recorded using a tri-axial accelerometer. A follow up study considered whether PSA data could be used to identify periods of sleep and wake using a WBSA like approach. To evaluate this, 118 hours of validated data was recorded, then used to develop and test a PSA sleep scoring framework. This resulted in metrics of 0.96 sensitivity (sleep detection) and 0.79 specificity (wake detection). A third study focused on optimising the sleep scoring framework, to improve overall performance. This included an evaluation of vector-based features, addressing sleep/wake class imbalance and identifying optimal accelerometer locations. Accordingly, a further 291 hours of validated data was recorded and used to evaluate the revised framework. This presented improvements of up to 16% in specificity, with nominal changes to sensitivity. The final study investigated sleep statistics generated from the PSA data, against those from a number of existing sleep monitoring tools. Observations here indicated that the PSA platform appears to provide comparable measures of sleep to existing approaches. In summary, the work presented throughout this Thesis culminated in a prototype platform for actigraphy driven, non-contact sleep monitoring. Accordingly, PSA addresses several limitations associated with WBSA and offers a novel contribution to objective sleep monitoring through its findings and associated datasets.

▼ I am a photographer. In photographing the city, I remap it intuitively and libidinally. I distort it. Places in the city are turned inward, into a mental space, into a projection of my own desires and deceptions. I do not speak of this city, of which I know very little, but speak from within the city. I am a tourist. For me the location is full of fantasies, dreams. It reminds me of art, of other photographs and films. I am with myself in this unknown and yet so familiar landscape. A place already close to me, already seen. It feels uncanny. It feels as if I were at the confines of memory, in what is left under forms of impressions, traces of stories, fantasy, and dreams, but also deceptions, frustrations, fears and symptoms. I look at the city through moments of deja vu. This thesis examines the relationship between photography and the city through the phenomenon of deja vu, hence engaging the artistic practice with contemporary theories of space, time and consciousness. After a conceptualisation of key concepts, deja vu, the cinematic and the excess, the thesis progresses through a speculative reading of the images exploring the boundaries between photographic practice, contemporary philosophy, and fiction. In interweaving those elements, at times dissonantly, the thesis investigates how deja vu can implicate one's subjectivity into the sense of duration, in a Bergsonian sense, and how this creates heterogeneous worlds rather than a homogeneous space. By fragmenting thought and dislocating the persona of the researcher, the thesis attempts to respond to such vision. Deja vu permits a displacement of the relationship between photography and the city into questions of time. The thesis examines this displacement creatively and moves into a Deleuzian philosophy of becoming.

▼ Henan University of Urban Construction (HUUC) is a provincial university funded by Henan Province, China. It features Civil Engineering, Construction, and Building Environment majors. The researcher is a lecturer, who has taught College English (CE) more than ten years in the Foreign Languages School of HUUC. CE, understood as general English, is a compulsory subject for the undergraduates of non-English majors during the first and second academic years in Chinese institutions of Higher Education (HE) including HUUC. The author explored to discover whether Task-based Learning (TBL) mitigated the current problems for CE, which had been identified on the initiative of a few administrative staff in the HUUC context. Action Research (AR) was used to implement TBL over a 12-week period during the 2014—2015 academic year. The sample consisted of four CE teachers (including the researcher), 326 first- year undergraduates of non-English majors in three experimental classes, and 267 students in three control classes. Quantitative and qualitative data were collected, and then analysed using statistical and interpretive methods. Findings indicated that TBL, was not well suited to the HUUC context, because of numerous personal and contextual factors such as students’ limitations in motivation, overall abilities and language skills as elements of personal factors, and examination-based syllabus, grammar-based examination, large class sizes, etc. as elements of contextual factors. TBL was, however, effective from the personal point of view in that it led to improvement in teachers’ motivation and total abilities, and students’ motivation, overall abilities and language skills when using TBL. The study provides some findings to teachers, students, researchers, and policy-makers in the development of plans for CE in future. Some recommendations are also provided by the researcher for future research on TBL in China and elsewhere, which have similar contexts for English language teaching and learning.

▼ This study employed event study methodology to examine the role of institutions, resources and dynamic capabilities in the relationship between ISO 14001 adoption and firm performance. Utilising financial data from 140 ISO 14001 certified firms and 320 noncertified firms, the results of the study suggested that UK and Irish manufacturers were not implementing ISO 14001 solely to gain legitimacy. In contrast, the results demonstrated that firms were fully integrating the ISO 14001 standard within their operations as certified firms were able to improve both financial and operating performance when compared to noncertified firms. However, while there were significant and long lasting improvements for employee productivity, manufacturing cost efficiency, return on assets and sales turnover, the sample firms operating cycle and fixed asset efficiency displayed evidence of diminishing returns in the long-run, underling the observation that no operating advantage based on incremental improvements can be everlasting. Hence, there is an argument for investing in dynamic capabilities which help renew and refresh the resource base and help the firm adapt to changing environments. Indeed, the results of the regression analysis suggest that dynamic capabilities for innovation acted as a moderator in the relationship between ISO 14001 certification and firm performance. However, while it was shown that firm innovation aided both short term and long term ROA performance, adaptive market capabilities only aided firms in the short-term at the marketing strategy deployment stage. Finally, the results have important implications for firms operating in an economic recession as the results suggest that firms should scale back investment in R&D while operating in an economic downturn. Conversely, under normal trading conditions, consistent and long term investments in R&D was found to moderate the relationship between ISO 14001 certification and firm performance. Hence, the results of the study have important implications for academics and management alike.

▼ In addition to the classical islet hormones (insulin and glucagon) several other regulatory peptides from the gut and neurons, are known to affect the functioning of islets directly or indirectly. These peptides include glucagon-like-peptides (GLP-1 and GLP-2), glucose dependent insulinotropic polypeptide (GIP), PYY, neurotensin, xenin, CCK and gastrin. These bioactive peptides are best known for the secretion from extra-pancreatic sites but their expression in islet lineage during embryogenesis may prove important for understanding their local involvement in beta-cell function, islet growth and beta-cell proliferation. Mouse model of diabetes and cellular stress generated with multiple low dose strcptozotocin presented with severe loss of beta-cell mass accompanied by notable increases in alpha and PP cell numbers. In contrast, hydrocortisone-induced insulin resistance increased islet number and beta-cell mass. PYY expression was consistently observed in alpha-, PP- and delta-, but not beta-cells. Streptozotocin decreased islet PYY co-localisation with PP and somatostatin whilst hydrocortisone increased PYY co-localisation with glucagon in mice. We also demonstrated the colocalization of neuronal peptides (NPY, neurotensin and xenin), proglucagon derived peptide (GLP-2) and gastrin family of peptides (cholecystokinin and gastrin) in various islet cell types. Expression of PYY, NPY, neurotensin, COPA (encodes xenin), CCK and gastrin genes were demonstrated in mouse islets. Consistent with a functional role their receptor expression was also established in mouse islets and cultured beta-cell lines. Xenin, CCK and gastrin stimulated basal and glucose-stimulated insulin secretion from immortalised rodent and human beta-cells whereas PYY and NPY inhibited glucose-stimulated insulin release from BRIN BD11 and 1.1 B4 cells. Xenin and CCK improved glucose disposal and insulin secretion following an i.p. glucose challenge in vivo. Intraperitoneal administration of PYY(3- 36), neurotensin, GLP-2 and CCK significantly reduced food intake whereas NPY demonstrated orexigenic effects in overnight fasted mice. These peptides also stimulated proliferation frequency and protected cultured beta-cells against cytotoxic insult. Taken together, these studies reveal that the peptides investigated in this thesis can be considered as non-classical islet peptides and receptor modulation of the pathways triggered by their actions may play a significant role in beta-cell regulation and afford a novel means for the treatment, or prevention, of diabetes.

▼ Quorum sensing (QS) acts as an important communication wiring in Pseudomonas aeruginosa. The bacteria use the system to regulate production of autoinducers for several adaptation and survival advantages. The system controls the production of key factors like proteases and rhamnolipid. Due to the substantial genome size and plasticity of P. aeruginosa, other factors are increasingly being linked with QS calling for the need of a better understanding of this complex system. The initial part of the thesis acknowledges the QS adaptation in P. aeruginosa clinical isolates from Cystic Fibrosis (CF) patients. Much of the factors that are deemed important for early infection establishment become redundant, as observed through loss of QS behaviour in most of the late isolates investigated in this study. This was particularly validated through complete loss of QS gene expressions and virulence factors like elastase, pyocyanin and rhamnolipid in a late clinical isolate, PA80. However, this cannot be regarded as an "universal adage" as isolates with and without QS activity were detected from the same patient in late or chronic stage of infection. The observations recorded in the study helps us to sketch a natural system where isolates having both QS activity and inactivity can co-exist in harmony in a chronic CF lung. In a stressful lung environment, it would be beneficial to have only certain isolates producing the expensive QS metabolites, in concentrations sufficient for the entire community, allowing the cell energy to be reserved for other functions. P. aeruginosa is a notorious nosocomial pathogen which is intractable with most of the common antibiotics. The resistance towards most antibiotics emphasises the need for alternative strategies to reduce virulence without eliciting a selection pressure on the bacterial population. Targeting QS with natural compounds is a promising strategy. This work, using a reliable and accurate RT-qPCR method developed inhouse, demonstrated that trans-cinnamaldehyde and salicylic acid could attenuate expressions of QS genes at sub-inhibitory concentrations. Trans-cinnamaldehyde was effective in downregulating the Iasi and lasR genes by 13- and 7-fold respectively while salicylic acid caused smaller reductions of 3-fold in Iasi and 2-fold in lasR compared to untreated samples. The ability to interfere with the master QS genes had a consequent inhibitory effect on the production levels of QS controlled virulence factors (elastase, protease, pyocyanin). Rhamnolipid, an important QS metabolite, was extensively studied and showed to be significantly downregulated following treatment with the inhibitors. The use of QS inhibitors alone may not completely abrade the infection but can reduce the pathogenic load by decreasing the levels of QS regulated virulence factors. This could allow for effective use of antibiotics at much lower concentrations than used now. ▼ Diabetes is considered as a prominent threat to the worldwide population, and if left untreated may result in a number of severe complications, cognitive deterioration, and death. For the most prevalent form of the disease, type 2 diabetes, the main contributing factors to the development of the disease is being overweight and obese. There are a number of mediating factors regarding the relationship between excessive weight gain and subsequent development of diabetes, the most widely supported of which is the typical eating behaviours of the individual. While the physiological characteristics of diabetes are well-known, much less is understood in relation to the behavioural consequences. Animal models may aid our understanding of a disease, but the animal model used should not only replicate the physiological symptoms but also the behavioural characteristics. The studies discussed within this thesis encompass a thorough behavioural evaluation of two of the most commonly used diet-induced and chemically-induced animal models of diabetes; the high-fat diet model of obesity induced diabetes and Streptozotocin induced diabetes. The main focus of investigation was on the impact of the disease regarding the food preferences and motivations of mice to obtain food, using a classical method for assessing preference (T-maze) and motivation to respond for different appetitive stimuli on an operant conditioning schedule of food reinforcement (progressive ratio schedule, PR). During the latter phase of experiments, a range of behavioural processes (e.g. food deprivation alterations, prefeeding with or without satiation, and extinction) were also assessed in relation to how these may be affected by the presence of diabetes. In the final study, a new model of PR performance was applied to the results, with the intention of addressing issues relating to the accurate measurement of motivation under a PR schedule. Orderly behavioural data were obtained but neither model of diabetes showed clear effects on food motivation. Results are presented and discussed in relation to the limitations and implications of the research.

▼ Background: The Republic of Ireland has become a more ethnically diverse society with inward migration of men from geographically disparate regions such as Eastern Europe and Africa. These men now constitute ten percent of the Irish male population. Migrant men report poorer health than their indigenous counterparts for many reasons including socio-economic disadvantage, cultural and linguistic barriers. Migrants also report reduced engagement with health services in their host country Aim: To explore the health of Polish men resident in the Republic of Ireland. Methodology: This study used a survey design, with a convenience sample (n =900) of Polish migrant men drawn from across seven Irish urban centres. A self-administered questionnaire with three distinctive scales was used to measure respondents’: health status, health beliefs, health behaviours and demographic profile. Ethical approval was granted by the Ethical Governance Committee of the University. Data were analysed using the Statistical Package for the Social Sciences (SPSS version 18.0). Findings: A response rate of 47% (n=421) was achieved. Respondents reported significantly lower educational levels than those of indigenous men or Polish men resident in Poland. The health behaviours of this cohort were similar to those of their Irish counterparts. There were a number of notable exceptions: 63% (n=265) of respondents smoked, 23% (n=97%) reported feeling nervous or depressed but over 50% (n=213) reported they were unlikely to access mental health services. Conclusions: Socio-economic disadvantages are apparent and these disparities need to be addressed in order to meet the evolving health needs of Polish migrant men. Smoking cessation needs to be considered including, how these activities will be initiated, methods used to impart information and follow up care for those who cease. Greater information about mental health care facilities, accessibility and the support services available need to be made more explicit to populations such as these.

▼ Over the course of Northern Ireland's recent political conflict, categorised as 'The Troubles', a number of research studies into the involvement of former paramilitaries have been conducted. For example, various studies have focused on the reintegration of former paramilitaries back into their communities, while others focused on the involvement of former paramilitaries in conflict resolution and DOW former paramilitaries are dealing with the past. However, to this date, none have explored in any detail the situational forces and systemic influences that contributed to the transformation of ordinary young men into paramilitary killers. This six year, Ph. D. study seeks to fill this void by investigating what led ordinary young men to join paramilitary groups and perpetrate violence. Throughout this thesis, I will persistently repeat that seeking to understand such situational and systemic influences, on the participants in this investigation, is not an attempt to excuse them from, or absolve them of, their actions. This sociological study examines the biographical narratives of former Loyalist paramilitaries from inner city Belfast. The individuals under investigation are former political prisoners who were members of the paramilitary group known as the Ulster Volunteer Force, (UVF) or its associated groups namely the Young Citizen Volunteers, (YCV) and Red Hand Commando (RHC). Fourteen narratives have been captured using the method known as the Biographical Narrative Interpretive Method (BNIM) containing the personal accounts of the respondents. Two of the fourteen narratives are presented as case studies and are the main focus of the thesis. These oral histories offer a perspective on the past now which the narrators did not have at the time but which has emerged in the reflection on and telling of their stories. The additional twelve narratives are considered satellite cases from which material is used in support or otherwise of the two case studies. The retrospective period being investigated is from their earliest memories until their time of arrest. The specific relevance of this study is that it focuses on the situational factors which are influential in seeking to understand why ordinary young men, some of whom were still at school, engaged in the conflict at its most violent period, (1972-1975), and became killers. In addition, it aims to contribute an additional perspective to the body of literature on paramilitarism in Northern Ireland. The study is borne out of the personal experience of being a former political prisoner myself and has emerged out of consistent contact with former political prisoners over the past twenty years, during the course of my community development practice.

▼ The need to achieve a greater understanding of the potential for shared learning between scientists, policy-makers and practitioners for Disaster Risk Reduction (DRR) is internationally recognised through the Sendai Framework for DRR. This is complemented by academic calls for interdisciplinarity in addressing disasters along with a growing interest in the concept of social learning as a lens through which to explore processes of collaboration in environmental management. Adopting a social learning approach this research uses a Community of Practice Framework to explore the potential for shared learning between earthquake scientists and humanitarian Non-Governmental Organisations (NGOs) based in the UK and Ireland. The thesis is based on qualitative interviews carried out with earthquake scientists and humanitarian practitioners, as well as individuals working at the interface of the two groups. Additional empirical evidence includes workshop recordings from a particular example of earthquake science-humanitarian collaboration. The analysis defines earthquake science and humanitarian NGOs as separate Communities of Practice (CoPs) and seeks to understand the elements uniting community members around a shared sense of enterprise, as well as gaining insight into the learning practices unique to each group. It highlights the crucial role of legitimacy in driving one's motivation to participate within a specific CoP or engage in cross-community learning. It finds high degrees of difference in community knowledge and culture to be presenting barriers to knowledge exchange. Mapping the multiple ways in which boundary work takes place between the CoPs the thesis finds those engaged in cross-community activities to be experiencing marginalisation, and current attempts to overcoming marginality to be ineffective. Recognising that the inherent challenge of evaluating boundary activities lies at the root of why they tend to be unsupported, the study seeks to adopt a postpositivist approach to evaluation, exploring alternative methods of critiquing boundary work in the short term. Wider political and economic factors are seen to underpin the potential for earthquake scientists and humanitarian practitioners to undertake fruitful collaboration in the long-term. To conclude, the thesis discusses structural implications and recommendations for supporting shared learning and legitimising boundary work between the earthquake science and humanitarian NGO communities.

▼ Type 2 diabetes mellitus (T2DM) is characterised by an impaired incretin effect, related to reduced glucagon-like-peptide-1 (GLP-1) secretion and impaired glucose- dependent insulinotropic (GIP) action. The inherent reduction in GLP-1 secretion can be overcome by clinically approved GLP-1 mimetics, whereas there is still need to develop novel therapeutic approaches to restore the blunted insulinotropic effect of GIP. Xenin has been recently reported to potentiate the biological actions of GIP. To date, no peptide mimetic has been able to surpass the beneficial antidiabetic effects of bariatric surgeries. In relation to this, altered secretion of various gut hormones are linked to superior metabolic control associated with bariatric surgeries. This thesis characterised the biological activity and therapeutic applicability of xenin based hybrid peptides in obesity-diabetes. These hybrid peptides included (DAla2)GIP/xenin-8-Gln, exendin-4/xenin-8-Gln, exendin-4/gastrin/xenin-8-Gln and cxendin-4/gastrin/xenin-8-Gln-Lys27PAL. Importantly, all hybrid peptides were resistant to enzymatic degradation and enhanced insulin secretion in vitro. All selected hybrid peptides displayed glucose lowering, insulin secretory and satiety effects. Importantly, there was a clear augmentation of the biological action of native GIP in all hybrid peptide treated mice, suggestive of restored GIP effectiveness. Twice daily administration of (DAla2)GIP/xenin-8-Gln for 21 days improved glucose tolerance, insulin resistance and pancreatic islet morphology in high-fat fed mice. Similarly, twice daily treatment with exendin-4/gastrin/xenin-8-Gln and exendin-4/xenin-8-Gln significantly improved metabolic control in dietary-induced diabetes. The superior metabolic profile of exendin-4/gastrin/xenin-8-Gln prompted further development of its related fatty acid derivative analogues. The protracted response of exendin- 4/gastrin/xenin-8-Gln-Lys27PAL was evident for up to 12 hours. Furthermore, subchronic administration of both peptides offered enhanced biological potency in terms of glycaemic control, insulin secretory responsiveness, insulin sensitivity and potentiation of GIP action in ob/ob mice. Similarly, combination therapy of exendin- 4/gastrin and xenin-8-Gln enhanced biological response to native GIP, improved insulin sensitivity, glucose homeostasis, reduced food intake and body weight. Additionally, these beneficial metabolic effects persisted for 21 days after cessation of treatment. Overall, these findings provide strong encouragement for the development of xenin based hybrid peptides as a new class of multiple targeting peptides for alleviation of insulin resistance and beta cell dysfunction in T2DM.

▼ It is believed that damage occurring to the earth's environment and the quality of life for future generations will be irreparable (Lozano et al., 2013). Sustainable development as a concept has been used as a means of integrating strategies to reduce this degradation. Education has been deemed to be key to promoting this sustainable future, and thus the term education for sustainable development (ESD) was coined. Research proposes that higher education institutions in particular could be significant contributors to ESD (Karatzoglou, 2013). Literature surrounding ESD has increased within recent years due to the UNESCO Decade of Education for Sustainable Development (2005-2014). Particular emphasis has been placed on reorientation of education towards sustainability (Sterling, 2001). It is believed that the curriculum should be holistic, participatory, promote critical thinking and values that promote sustainability (Firth and Smith, 2013). However, changes in the curriculum to date have been deemed minimal and the effectiveness of the Decade of Education for Sustainable Development questioned (McKeown and Day, 2015). This research argues that ESD does not have a distinct pedagogy, but instead is made up of various component parts. These component parts take the form of learning pedagogies presented as (andragogy, social learning, experiential learning and transformative learning). A qualitative approach was used to collect data for this research with emphasis being placed on using the four mentioned learning theories to evaluate ESD within different learning settings (classroom, fieldwork, volunteer setting and work based learning environment). This thesis identifies that each learning theory researched promotes good pedagogical practice whilst incorporating ESD indicators. The research concludes that a careful balance of different elements of these learning theories is needed to effectively deliver ESD within higher education.

▼ The aim of this doctoral programme of research, consisting of four separate studies, was to determine the perceived benefits of sport based interventions on the psychological well-being of prisoners. A methodical review was undertaken in Study 1 to assess the current evidence base and identify subsequent research questions to be addressed. Study 2 then focused on increasing knowledge of how and why sport based interventions can positively impact on psychological well-being within prison. Studies 3 and 4 were conducted in response to the need for robust prison based intervention studies, adopting specific well-being measures and follow-up, to test for immediate and long-term impacts on psychological well-being. Study 1 established positive impacts on psychological well-being within prison in 12 from 14 sport based interventions. However inconsistent definitions of psychological well-being, measurement inconsistencies and limited follow-up led to the conclusion that sport can have positive effects, but raised questions regarding how and under what conditions? There was also a consistent absence of psychological theory to explain and help replicate any positive impacts of sport observed. In response to the limitations highlighted in Study 1, Study 2 engaged with 16 stakeholders responsible for the design, delivery and oversight of sport based interventions in prison. A thematic framework was presented, linked to three psychological theories, to increase knowledge of how sport based interventions within prison can effectively impact upon psychological well-being. In Study 3 the effect of a 6-week sport based intervention in prison was considered. Positive effects on short-term psychological well-being during participation was shown, but failed to demonstrate any substantial longer term vi impacts. Study 3 also identified environmental barriers to effective implementation of sport based interventions, resulting in prisoner frustration at times. The inclusion of psychological theory highlighted in Study 2, and incorporated into Study 3, was inconclusive and requires further investigation. In response to the feasibility issues identified, Study 4 focused on testing the perceived benefits of an alternative short form sport-based intervention, aimed at directly improving mental health and psychological well-being within the male prison population. Statistical analysis revealed a short-term positive impact on mental health awareness. Results from the thematic analysis of focus group data also revealed participants perceived the intervention as a novel, appropriate and engaging format, and reported increased intentions to seek help and sense of hope for the future. ▼ Language repeats. And repeats. And repeats. Language exhales a repetitive reality that we, as its speakers and writers, are bound to and captured by. The repetition of language is a metronome that chimes a spatial and temporal conditioning, the territorialising performance of an aesthetic experience that is unlimited yet limiting, which invades our very being. A contagious praxis that arranges a material structuring, not concerned with meaning but only its own propagation, language organises an institutionalising ideology of repetition and reproduction which denies ontological potential in deference to the finitude of possibility. A homeostatic regime that we occupy and are occupied by, the material topology of language’s repetition appears inescapable. Caught in a loop, we are captivated by language, a conditioning that pulses and reverberates, structured by reference and deference. But if speaking is mere repeating, the stammer and the echo are a repeated repeating; procedures not of stale reproduction but of vibrant production through repetition. Using iteration against itself, the stammer and the echo present modes of transformative mediality that motivate alternative material and ontological practices in abeyance of language and its regime of reproduction. Stammers & Echoes: A manifesto is an arts research thesis that unfolds the philosophy of repetition in language. A synthesis of Gilles Deleuze’s transcendental materialism and Jacques Derrida’s deconstruction, this text will establish ontological and material procedures for alternative patterns of repetitive speech. In developing what will be termed linguistic materialism and the determination of language as a hyperobject (Timothy Morton), the stammer and echo are proposed as itinerant (anti)methodologies that not only expose the materialism of language but are agencies that provide access to an ontological potential in excess of the possible. This research thesis is also a manifesto; a performative document and an art object. The speculative condition of this research is embedded in the very reading and writing of this text. Repetition is deployed in the making of a rhythmic reading of research, in which circularity and reiteration manifest the material performance of a distinct spatial and temporal experience that reconfigures the difference of repetition itself. This text proposes not only an exemplar for doctoral artistic research, but a manifesto for practices and procedures to be deployed against language and other regimes or ideologies of repetition and reproduction.

▼ This research investigated the potential to deliver substituted CaP thin films onto medically relevant substrates, in a one step process, via RFMS, so as to enhance the substrates surface topography and chemistry to stimulate an enhanced osteoblastic response. RFMS was utilised initially to sputter deposit (substituted-Sr and/ Zn) CaP onto Ti. Chemical and topographical surface properties in both the as-deposited and annealed state at 500 °C were investigated. Studies were undertaken which placed special emphasis on how the chemistry and morphology of (substituted) CaP coatings, sputter deposited onto PEEK, altered in relation to the deposition parameters. It was realised that a gradient interlayer was created between the PEEK substrate and the (substituted) CaP coating due to the calcium, phosphate and substituted ions being implanted within the PEEK to different depths in an erosion/ deposition process and that all of the coatings produced were amorphous, this created chemical and morphological changes to the PEEK substrate, but no damage to the bulk material. With the addition of impurity ion(s) the thin film formation took longer indicating a slower development rate. The chemical and morphological stability of the various (substituted) CaP coatings deposited onto PEEK were studied under physiological conditions, each coating had the ability to withstand this environment for 14 days. The in vitro response to the neat PEEK and (substituted) CaP surfaces was investigated up to 21 days, with all sample types proving their capability in supporting early cell attachment and proliferation. ALP measurements indicated high levels of initial differential potential on the Sr substituted CaP modified PEEK and the crystalline (substituted) CaP modified Si. By day 14 the (substituted) CaP modified PEEK samples maintained the highest levels of osteogenic differentiation. The Sr substituted CaP modified PEEK substrates promoted earlier and prolonged differentiation potential when compared to the CaP coated PEEK.

▼ Human-managed grasslands deliver a wide range of ecosystem services including food production and the regulation of nitrogen (N) and carbon (C) cycles between aboveground and belowground compartments. Common management practices, however, such as animal grazing, agricultural liming and nutrient fertilization can greatly influence the functioning of grassland ecosystems and reduce their long-term sustainability. This PhD study has specifically addressed how grazing, liming and nutrient additions might affect soil biogeochemistry and aboveground-belowground interactions in a grassland experiment established in 1991 at Silwood Park, Berkshire, UK. Key findings from this study show how repeated liming applications have significantly (1) increased archaeal and bacterial (B) abundance while reducing fungal (F) abundance, (2) increased the C pool of soil smaller soil aggregate fractions (e.g. micro and silt clay aggregates), and (3) enhanced plant nitrogen use efficiency (NUE). Chronic nutrient fertilization has also significantly increased the abundance of ammonia oxidizing bacteria and decreased plant NUE. A novel result of this study is no change in Fungal and Bacterial gene copies in response to multiple nutrient additions. This suggests that fungal taxa are benefitting from labile resources and not all functional bacterial groups are benefitting from an increase in labile resources returned to the soil. These findings are state of the art and in line with papers that are questioning the classic model of the soil food web where bacteria predominantly metabolise labile detritus and fungi predominantly metabolise recalcitrant detritus. These resources were thought to form part of two distinct basal energy channels, however, the findings in this study support an emerging conceptual framework that understands the food web to have one continuous C pool that can at different times benefit both bacteria and fungi.

▼ Analyses of deformations associated with earthquakes are calculated in terms of plate motions and co-seismic stress changes, which assume the crust responds as an elastic medium. However, the lower crust and region of ductile mantle behaves viscoealastically over much longer time scales. In other words, lithospheric stress change following earthquakes is time dependent process. In order to draw a complete picture of total deformation over a seismic cycle, we cannot ignore the considerable influence of the viscoelastic relaxation processes. The objective of this thesis is to assess the role of post-seismic viscoelastic relaxation processes following earthquakes. First, I investigate the post-seismic viscoelastic flow in the lower crust and upper mantle of large earthquakes in eastern Turkey, during the 19th and 20th century. Using three possible rheological models, the viscoelastic, post-seismic deformation is analysed to assess the extent to which these events influence the velocity fields at GPS sites in the region. Models show that the post-seismic signal currently contributes to the observed deformation in the eastern part of the North Anatolian fault and northern and middle parts of the East Anatolian Fault Zone, primarily due to the long-lasting effect of the Ms 7.9 1939 earthquake. Study results identify a post-seismic signal in the region and could contribute to between 3% and 25% of the observed GPS measurements. Next, in order to assess the importance of the post-seismic viscoelastic relaxations over the last seismic cycle, I investigate stress accumulation over the East Anatolian Fault Zone by modelling the stress changes associated with 16 large (M>6.8) earthquakes that occurred in the region since 1822. Two areas of increased stress in the region are identified; the first in a fault segment that runs from the southern part of the city of Kahramanmaras to the southern part of the city of Malatya (KM Segment) and the second is the Yedisu segment at the eastern terminus of the North Anatolian Fault Zone. It is also observed that the stress accumulation associated with the post-seismic viscoelastic stress equals or larger than the co-seismic stress values in these segments. Finally, I assess the extent of time dependent viscoelastic post-seismic stress influences on the static shear stress change on the rate of triggered events that located in ± 2° after a main shock. Receiver faults that have calculated shear stress changes (on their nodal planes) between ±0.01 and ±1 MPa are considered as potentially triggered by the source earthquakes. The stress changes associated with 244 main shocks (M>7.0) from Global Centroid Moment Tensor Catalogue are modelled. A 10-year time window after the main shocks is used for the subsequent analysis to allow the numbers of aftershocks to decaying to the background seismicity rate. Resident participation in tourism planning and decision-making is thought to be beneficial for destination communities. This study presents an analysis of tourism development and the dimensions of community participation in decision-making and empowerment for local communities in the Sporades Islands, in Greece. The research location, containing four islands, was selected in order to provide an opportunity for comparison between communities with different levels of tourism development. The main focus of the study was to investigate community empowerment through the process of decision-making with respect to tourism planning and development. Seasonality as a factor was taken into consideration as an important determinant of tourism development for the islands. More specifically, the study aimed to examine the residents' attitudes towards tourism development, to determine the extent of resident participation regarding tourism decisions, to evaluate the degree of community empowerment that exists, as well as identifying those factors that promote or restrict it. This led to the development of a conceptual model of the process toward community empowerment and this was applied to the Sporades Islands. A mixed-methods methodology was chosen, achieving 887 completed questionnaires across Sporades during both the high and low season, and 26 in-depth interviews were undertaken with key stakeholders and residents from within the research location. The findings demonstrated that tourism is well-supported amongst the residents but also identified a series of barriers that restrict the involvement of community members in tourism-related decisions. Some of the barriers were associated with limited resident access to information concerning tourism development and the apathy that local authorities identified in residents. Based on the findings a conceptual model of the process toward empowerment emerges that provides a blueprint toward enhancing residents with greater involvement in tourism decision-making.

▼ This thesis presents the findings of a study undertaken with Polish male prisoners in Maghaberry and Magilligan prisons in Northern Ireland. In a jurisdiction emerging from years of armed conflict, the prison system is currently undergoing changes which attempt to deal with the structural problems stemming from that conflict's legacy. Alongside that reform, another transition is evident: that of a rapidly increasing national and linguistic diversity of the prison population. It was therefore important to study how this change impacts on the prisons in Northern Ireland where as recently as 2011 their practice in relation to the treatment of national and ethnic minorities was assessed as being "culturally blind” (PRT, 2011b: 39]. Using data obtained in a series of semi-structured interviews with eighteen prisoners, a number of semi-structured interviews with staff and representatives of independent prison monitoring bodies, this thesis examines how Polish prisoners negotiate daily life in prison custody in Northern Ireland. Analysing the ways in which they experience deprivations of prison life, the thesis is also concerned with the prisoners’ methods of adaptation to prison regimes in an environment which they often struggle to understand. Looking at the ways in which they forge relationships with other prisoners and staff, the thesis concludes that many of the male Polish prisoners in Northern Ireland live in a prison within a prison, with a high wall of communication barriers around them, suspended between their entry into custody and the ever-looming moment of deportation. The prison system, largely unprepared to deal with more nationally and linguistically diverse populations, facilitates their existence in 'mono- cultural boxes’ in the meantime.

▼ Pressure ulceration remains a significant problem within healthcare and over recent years has been elevated to a position of priority within national quality improvement agendas. This thesis will contextualise key issues, determine barriers and facilitators in relation to pressure ulcer prevention and make recommendations for service improvement. Several methodologies were used to explore different aspects of the structure, process and outcome measures involved with pressure ulcer prevention. Initially a retrospective data analysis reviewed incidence, reporting and validation of hospital acquired pressure damage. This showed the importance of a robust reporting and validation system. Qualitative methods were then used with staff throughout the organisational hierarchy to explore values and beliefs about pressure ulcer prevention. This showed that the context and key drivers behind the pressure ulcer prevention quality initiative were highly valued by those in positions of seniority. The ‘feedback loop’ was also viewed as of paramount importance by those in key positions of responsibility, but did not appear to be valued equally by clinically based ward staff. The importance of cohesive multidisciplinary ‘teamwork’ was valued by all levels of staff and equally ‘education and documentation’ were described as key facets of preventative care. ‘Accountability’ with regards to responsibility and ownership was also highlighted however a variation in views was evident among different staff groups. Whilst a high priority was attributed to pressure ulcer prevention by everyone, competing interests meant that this fundamental aspect of care was frequently delegated to healthcare assistants with no formal training in this area of practice. An experimental study design was therefore used to evaluate the efficacy of an educational intervention for healthcare assistants. Results show that training has improved knowledge levels, attitude and self-report of clinical practice. It is recommended that further research is conducted to substantiate the findings.

▼ Alzheimer's disease is a neurodegenerative condition, characterised by cognitive decline and aggregations of misfolded proteins in the brain. With improving healthcare and extension of life expectancy, prevalence of Alzheimer’s disease continues to rise as a consequence of advanced age. Some drugs are available that offer symptomatic relief in Alzheimer's disease, however, none exist that alter the disease course. Diabetes and obesity increase the risk of developing Alzheimer’s disease; inflammation and insulin resistance are thought to mediate this association. Data presented in the following chapters demonstrate a time course of disturbed cognition, inflammation and insulin signalling in the brain that progresses in the early stages of the development of high-fat diet-induced obesity. Notable decline in working and spatial memory was induced in chronically high-fat diet-fed mice and these deficits were not corrected in mice treated with Xenin- 25 [Lys (13) PAL], a long-acting analogue of gut hormone xenin-25. Genetic and protein markers of inflammation and insulin signalling were also dysregulated in the brains of 21-week high-fat diet-fed mice, many of which were corrected following Xenin-25 [Lys(13)PAL] treatment, representing previously unreported findings. Using an aged APP/PS1 mouse model of Alzheimer’s disease, striking similarities with age-matched wild-types were apparent. However, spatial learning was impaired and increased insulin resistance was apparent in the brains of APP/PS1 mice. Three gut hormone analogues, liraglutide, NAcGIP[Lys(37)PAL] and Xenin-25rLys(13]PAL] were shown to have positive effects on cognitive function and reduced amyloid-(3 burden in the brains of APP/PS1 mice. Finally, a novel immunomodulatory polysaccharide that enhances the adaptive immune system showed promising effects in the APP/PS1 model, underscoring the therapeutic potential of this compound, suggesting that augmentation of certain immune responses may be a viable treatment approach for Alzheimer's disease. This thesis demonstrates similarities between the pathogenesis of diabetes, obesity and Alzheimer's disease. Results here show that brain insulin resistance in APP/PS1 mice occurs in the absence of peripheral metabolic dysregulation, suggesting that brain insulin resistance in Alzheimer's disease is a distinct pathological phenomenon, divergent from typical metabolic disease. Modulation of inflammatory and insulin signalling pathways, using pharmacological peptides and natural compounds has potential to alter the disease course of Alzheimer's disease.

▼ Rhamnolipids are amphiphilic surface active molecules that are currently receiving major interest from industry as a sustainable alternative to chemical surfactants produced using petrochemical feedstock's. Whilst the majority of research has been carried out on P. aeruginosa in terms of industrial scale RL production the organisms' pathogenic nature makes its use on an industrial scale uneconomic. The main focus of this thesis was therefore to explore whether B, thailandensis, a non-pathogenic alternative to P. aeruginosa, could be characterized and manipulated in order to produced RLs at a level equal to or greater than that of P. aeruginosa. The first part of this thesis was used to characterize RL production in B. thailandensis and to gauge its potential as a possible industrial scale RL producer. As very little work has been carried out on B. thailandensis in terms of RL production this chapter was used to generate a detailed, quantitative outlineof RL production in B. thailandensis under specific growth conditions as well as giving an insight into some of the phenotypic traits RLs are involved in. In addition, quantitative expression analysis of RL synthesis genes also provides a transcriptional profile of RL production providing a complete and in depth profile of RL biosynthesis in B. thailandensis E264.
