▼ Austerity is among contemporary public management’s defining features. Austerity management research traditionally focuses on the contents of budgetary changes and organisations’ broad strategies. While valuable, this high-level strategic lens comes at the expense of accounting for the austere situation’s turbulence and the grounded practices through which austerity is managed. Consequently, this thesis reconceptualises austerity management through Thévenot’s 'regimes of engagement' (RE). This pragmatist lens better conceptualises actors’ situated dilemmas and consequent practices. Thévenot’s approach particularly suits austere environments, whose scarcity surfaces public organisations’ latent inter-goal conflicts: RE differentiates modes of action by the different goods people seek. To capture the national and organisational debate in whose context public managers operate, RE was combined with Critical Discourse Analysis. The resultant study focuses on a sector in which austerity management research is overdue: UK healthcare. It combines a case study of national policy with three organisational cases. Observations, interviews and documents are analysed discursively, processually and pragmatically. The resultant findings enable three theoretical contributions. The first emphasises the linguistic situating work we do to establish the nature of situations we encounter. It particularly emphasises its importance in shaping the national austerity situation. The second highlights an annual cycle of instability as organisations’ emphasis shifts between reporting acceptable financial plans and making those often highly optimistic numbers real. The third demonstrates that austerity destabilises the very coordination devices which people deploy to manage it. Together, these contributions conceptualise austerity management as a set of responses to a situation. It notes how austerity’s evaluative and situational instability undermine traditional ways of coordinating work despite uncertainty. This recasts austerity management as not only a set of financial choices but also a search for coordination. Such an approach is needed to do justice to the messy realities of austerity which public managers know so well.

▼ The outsourcing of military responsibilities to private contractors is most comprehensively encapsulated not in armed security contracting, which dominates the literature, but in the supply of the armed forces. Military logistics, broadly conceived, stretches back the furthest in history and involves among the largest manpower and sums of money expended by defence enterprises. Drawing on the United States of America (USA) and the United Kingdom (UK) since the end of the Second World War, this dissertation develops a holistic understanding of why states outsource military capability, the politics and processes which produce the decisions (not) to acquire military services from the market, and the longer-term impact and trajectory of defence services acquisition as a result of the way states use private contractors in their defence enterprises. Most fundamental to understanding why states outsource military capability is an appreciation of the dominant ideas and norms that guide policy-makers and constrain decision-making. In defence these are, in particular, a state's defence strategy and posture, strategic culture, and political economy. These factors strongly determine the type, size, and shape of the armed forces, the weapon systems and services required for their supply, and the sources of these products and services. Together with the formal and informal political structures of the state, these factors also heavily determine who participates in the defence policy process. In the case of the USA and the UK, the general tendencies to espouse global defence postures, draw on private enterprise for the supply of goods and services, and rely on highly sophisticated, hi-tech weaponry in the conduct of war clashed with a lack of resources which were - under these influences - sought to be overcome by relying on private providers. The policy processes are similarly biased towards business ideas, solutions, and providers while exhibiting a remarkable lack of veto-points and veto-players. "More of the same", i.e. the increasingly routineised use of private contractors in the generating of military capability, is therefore the unsurprising outcome of the past decades which witnessed the growing reliance on private service providers in the defence enterprise. Outsourcing is not only "here to stay", as authors often conclude; the USA and the UK are formally and doctrinally integrating their military and contractor workforces into joint logistics forces that will support and supply their armies for decades to come, and concomitantly transferring knowledge and assets out of the military and into the private sector for the long term. Being improbable from within, change in the USA and the UK would only come about as a result of strategic, economic, or ideational "shocks" to this otherwise very stable ecosystem. ▼ This study is concerned with the experience of secondary trainee teachers following full time postgraduate training routes in England. The research illuminates the differing approaches to learning and teaching practices in three settings offering postgraduate teaching qualifications: a university; a school-centred initial teacher training site and a blended programme that combines school-led and online learning. The study’s primary focus is on how trainee secondary English teachers learn and develop in the contrasting settings. Alongside this, consideration is given to how trainees’ identities as teachers develop during the training year. Analysis of the literature and the ramifications of government legislation on teacher training in England contributes to an understanding of the positioning of trainee teachers in the shifting educational landscape. Issues of identity are explored, addressing how trainee teachers define and redefine themselves through the everyday life events encountered in their professional and training contexts. The lived experience of the trainees is captured using ethnographic methods. Observational data of the three contrasting cohorts is combined with classroom observations of identified participants. This is supported by semi-structured interviews staged throughout the year and consideration of the academic assignments completed as part of the training programme. The reporting of the ethnographic data is organised around a series of structuring concepts. This non-linear approach serves to highlight the recursive and sometimes regressive nature of the process of learning to teach. The yearlong immersion in three different research sites results in a richness of data whilst the subsequent analysis provides a significant contribution to our knowledge of what it looks and feels like to learn to teach in the increasingly marketized environment of teacher training in England. The selected locations straddle the binary line between university-led and school-led training, although a stark contrast in outcomes between trainees following different routes was not apparent. The findings highlight qualitative differences in the provision between sites but also identify the shared significance of the school experience in shaping the identity and progression of trainees during the year. The analysis of the data culminates in the formation of a conceptual framework addressing issues of conflict, transition and agency. The framework offers a mechanism for the analysis of the trainee experience across sites, subjects and key stages and has the potential to inform the practice of teacher educators and policy makers in England and beyond.

▼ Drug delivery via the skin is attractive, but it is challenging and thus it is an area of continual research. Challenges arise in this field because only a narrow range of drugs naturally diffuse into the cutaneous tissue. This natural passive skin diffusion process is largely dictated by the drugs physicochemical characteristics. Only moderately lipophilic, low molecular weight agents accumulate in sufficient concentrations in the systemic circulation after topical delivery to elicit a clinical effect. The inability of drugs to pass into the skin has been addressed in previous work through chemical and physical methods of penetration enhancement, but using the currently available technologies it remains problematic to force compounds through the skin without damaging the tissue. The aim of this project was to investigate if the application of topical hypobaric pressure, a new physical technique to enhance topical drug delivery, could be used to facilitate drug delivery into the skin without skin damage. To study the effect of local hypobaric pressure changes upon drug penetration into the skin three model systems were selected based on their different physicochemical properties; minoxidil (a small drug that passes into the skin via the hair follicles), dextran (a model macromolecule), and nanosized carriers (to simulate a drug delivery system). Application of the device (built-in-house) gave a stable pressure reading for 60 min. Histology, showed hypobaric pressure up to 400 ± 50 mBar did not damage the skin. The application of hypobaric pressure (400 ± 50 mbar) was shown to be significantly (P < 0.05) enhance drug deposition in the skin for the majority of test systems. For example, an enhancement ratio of 16.5, 5.3 and 5.2 was observed for minoxidil in 'stratum corneum', epidermis and dermis respectively. For solid lipid nanoparticles an enhancement ratio of 1.2, 8.8 and 7.8 was recorded for 'stratum corneum', epidermis and dermis respectively. For the dextran (FD-4) the enhancement ratios were 2.9, 2.0 and 1.2 for 'stratum corneum', epidermis and dermis respectively. The hypobaric enhanced skin permeation was dependent on the test systems size, surface charge, and hydrophobicity. Confocal microscope images showed the solid lipid nanoparticle enhancement was through the intercellular and follicular pathways, whereas the dextran enhancement was through the intercellular pathway when hypobaric pressure was applied. In conclusion the results obtained from this project suggested that the application of hypobaric pressure was an effective, safe and promising method to enhance the delivery of drugs into the skin and it warrants further assessment in vivo to enable its clinical use.

▼ The coherent collective oscillation of free electrons in metal nanoparticles under the external illumination is referred to as localised surface plasmon resonance. They may decay either radiatively in light or non-radiatively, producing high energy electron-hole pairs, termed hot carriers. These hot carriers may relax via electron-phonon coupling locally heating the particle or reach the particle surface and transition into unoccupied levels of acceptor adsorbates and trigger chemical reactions. Photochemistry represents one of the most promising fields of study in hot carrier technologies, where hot carriers can catalyse chemical reactions by interacting with external molecules at the particle surface. This includes degradation of organic pollutants from wastewaters, hydrogen generation by solar water splitting and reduction of CO₂, amongst others. The photocatalytic efficiency depends on various factors, such as hot carrier generation rate, hot carrier energy distribution, rate of adsorption of molecules and chemical stability of the photocatalyst. In some cases, plasmonic metals are paired with semiconductors in order to improve the efficiency by extending hot carrier lifetimes, improve optical absorption and promote molecular adsorption. In this work, silica nanoparticles decorated with plasmonic Au nanoparticles and Pt as co-catalyst were designed, characterised and optimised for the enhancement of hot electron generation and extraction. The resulting heterostructure was systematically investigated by varying Au and Pt loading, allowing assessment of the hot carrier extraction via observation of photodegradation of methylene blue, an organic molecule extensively used by the textile industry and that is usually found in wastewaters. An optimal heterostructure design was found while studying the response of the photocatalyst to diffeerent metal loadings, maximising hot carrier generation from Au, and also maximising the catalytic effects. In the case of optimised heterostructures, Pt layers formed on the Au nanoparticles. Since the Pt layers have a rough surface with small Pt clusters which locally support electric field enhancement, the catalytic efficiency was improved due to the efficient hot-carrier generation and extraction. It was found that hot electrons are responsible for triggering the degradation of methylene blue by interacting with oxygen molecules and exciting them to generate superoxide ion species, which in turn chemically react and degrade methylene blue molecules. The combination of plasmonic/catalytic metals in nano-heterostructured devices provide an important impact on photocatalytic processes, and may be of significant interest for future clean technologies.

▼ Background: Pain is a commonly reported and disabling symptom of inflammatory bowel disease (IBD) and has a significant impact on a patient’s quality of life, mood and functioning. While over two thirds will experience pain during active disease, a significant proportion (20-50%) of patients report pain in remission. A lack of clinical and endoscopic markers of active disease in the presence of pain is distressing for patients and challenging for clinicians. Preliminary research demonstrates that IBD-pain is a multifactorial construct, however further research is needed to better understand and effectively manage pain in IBD. Aim: The aim of this thesis was to develop and feasibility test a self-management intervention for chronic pain in IBD. Methods: The Medical Research Council (MRC) framework and Behavioural Inhibition System/Behavioural Activation System (BIS/BAS) theory of chronic pain were used to guide intervention development. A systematic review examined psychosocial factors associated with pain in IBD. A cross-sectional study evaluated the association between psychosocial factors identified in the review and other psychosocial factors. A qualitative study explored experiences, current management strategies and needs for an intervention for pain. Finally, a feasibility study assessed the feasibility and acceptability of a self-management intervention and gained preliminary estimates of efficacy on pain and psychosocial processes. Results: • The systematic review including 15 studies (paper 1) found cognitive, emotional and behavioural processes associated with pain, yet highlighted that further research was required using validated pain measures and to investigate psychosocial factors identified but not yet explored in IBD. • The cross-sectional study (paper 2) (n=297) confirmed psychosocial factors identified in the review such as depression, anxiety and pain catastrophising. It also demonstrated cognitive and behavioural processes such as symptom focusing, avoidance resting behaviour and pain self-efficacy were associated with pain. • The qualitative study (paper 3) (n = 14) identified three themes; i) vicious cycles ii) finding solutions and iii) attitudes. It found that pain is rarely experienced in isolation but with other symptoms of fatigue and urgency in IBD. • Cognitive behavioural therapy techniques were used to target psychosocial processes in a BIS/BAS model of IBD-pain. Patient public involvement ensured that the intervention was tailored to people with IBD-pain. The intervention was delivered online with facilitator support over 9 weeks. • A feasibility study (n=20) (paper 4 – submitted) demonstrated that the online intervention was acceptable, as participants understood the workings of the intervention and felt confident in their ability to complete sessions and tasks. Despite low recruitment rates, feasibility was demonstrated through usage and completion of the intervention and adequate provision of facilitator support. ▼ The mouse is commonly used in cardiovascular disease research. Therefore, accurate and reliable measurement of blood pressure in conscious mice is essential. Blood pressure can be measured directly from a major systemic blood vessel using implantable devices (telemetry probes) or noninvasively from a peripheral site such as the tail. Telemetry allows continuous collection of data in freely moving mice, whereas the mice are restrained during the tail-cuff procedure. However major surgery and higher cost associated with telemetry result in the fact that the tail-cuff technique remains widely used. The tail-cuff technique is known to cause stress in the animals and therefore its reliability and validity are questioned. The hypothesis is that the handling technique may reduce stress associated with the tail cuff technique. The aim of this thesis was to perform a detailed analysis of different handling techniques and the stages of the tail-cuff technique in their impact on central blood pressure, heart rate and core body temperature using telemetry. I have revealed that all interventions caused significant increases in these parameters, with the restraint step inducing the largest change, which is maintained throughout the restraint period. These were not further exacerbated by increasing temperature or tail-cuff inflations. I saw no differences due to different handling techniques or the operator’s gender. Having compared the simultaneous recordings obtained by telemetry and the tail-cuff in the same mouse, I found that the tail-cuff system consistently produced lower readings than telemetry. However, the readings obtained by the tail-cuff were similar to those obtained by telemetry in non-disturbed mice on the same day in normal and hypertensive mice. The results show that the tail-cuff technique induces a stress response in mice, which is not alleviated by repeated exposure. Although these results support the use of the tail-cuff for monitoring blood pressure in the hypertension model, they also highlight the fact that measuring the blood pressure from the tail may not be representative of the systemic blood pressure. This is important new knowledge for researchers who use murine models of hypertension.

▼ The term software-based art has emerged from conservation practice over the past decade to describe artworks for which software forms the primary artistic medium. Such works present new challenges for those engaged in the long-term care of collections of modern and contemporary art. They are often technically complex and may employ many inter-related (and sometimes bespoke) components, embedded in a specific technical environment. As a result, software-based artworks are particularly at risk from processes of loss and obsolescence. While progress has been made toward the development of practical strategies for their preservation, how to effectively document them in a conservation context remains poorly understood. In this thesis, I describe practice-led research which has sought to address this gap using a constructive research approach. I first develop a conceptual framework through which to better understand the problem space, consisting of two parts: an in-depth examination of the characteristics of software as a medium; and an exploration of the document concept and its meaning in relation to the role of the conservator. Using this conceptual framework to further refine my research aims, I examine three topics in detail, seeking to develop practical solutions for each: the analysis and representation of software structures; the extent to which notions of significance and artwork identity might be formalised as documentation; and how the patterns of change which occur in the life of a software-based artwork might be understood and recorded. In addressing each of these aims, I draw on insights gained in the in-depth study of a set of software-based artwork case studies from the Tate collection and the synthesis of existing theory from a number of related domains. The outcomes of the research have direct relevance to conservation practice, not as formal templates, but rather as a set of flexible and reusable principles and methods that might be applied individually or in conjunction to effectively document a diversity of software-based artwork types.

▼ Producing single-sex litters can have a positive impact for many applications, including reducing disease-spreading vectors, increasing effective breeding populations for conservation, in agricultural biology, and in research; reducing unnecessary animal culling, in accordance with the Home Office 3Rs. In the laboratory mouse model, Mus musculus, males carry the sex chromosome complement XY. The daughters inherit the X chromosome, and the sons inherit the Y chromosome. In this thesis I exploit the unique inheritance of the X and Y chromosome from the male heterogametic sex to investigate multiple methods of generating all-male or all-female litters. Co-inheritance of two alleles, one from the father and one from the mother, which only upon the presence of both alleles causes embryonic lethality, was called a bi-component system. I first performed a proof-of-principle strategy utilising two pre-existing mouse models, X-Cre and an inducible Rosa26-DTA. Diphtheria toxin A (DTA) expression was only active upon excision of a floxed-STOP cassette by Cre recombinase. Co-inheritance of the XCre allele from the father, and Rosa26-DTA allele from the mother resulted in activation of the DTA and female-specific embryonic non-viability. In an alternative approach, I used CRISPR-Cas9 to generate mutations in a target housekeeping gene by co-inheritance of a Cas9 and sgRNA transgene. I first investigated the mutation efficiency of multiple sgRNAs in vitro, targeting an essential gene Topoisomerase 1. Highly-mutagenic sgRNA constructs were then carried forward to generate sgRNA-expressing mouse lines via recombination-mediated transgenesis. The sgRNA-expressing mice were mated with a pre-existing Rosa26-Cas9 mouse line. I showed that co-inheritance of the Cas9 and sgRNA transgene induced mutations in the target Topoisomerase 1 locus. Co-inheritance of the two transgenes also resulted in embryonic non-viability due to loss-of-function of Topoisomerase 1, whilst inheritance of a single transgene did not result in loss-offunction mutations. Lastly, in order to generate sex-specific mutations by CRISPR-Cas9, I generated sex chromosome-linked Cas9 transgenes. I showed that the X-Cas9 transgene expressed Cas9 in vitro and in vivo, and in the future this line may be a useful tool for generating mutations in a target gene in a sex-specific manner.

▼ Recent years have seen a resurgence of interest in fundamental questions regarding the out-of-equilibrium behaviour of isolated quantum systems. Due to both new experimental techniques, such as those involving cold atomic gases, and progress in the theoretical understanding of closed quantum systems, it is now possible to explore fundamental questions about thermalisation and the applicability of statistical mechanics. The interplay between disorder, interactions and quantum interference gives rise to interesting phenomena, such as many-body localisation (MBL), which has a key role in the non-equilibrium behaviour of such systems. Realistic models of disordered, interacting quantum systems are largely intractable analytically. Their numerical study is limited by the difficulty of simulating quantum systems with classical computers, making it challenging to separate genuine "thermodynamic" results from finite size effects. In this thesis we consider some more tractable mean- field models, as a starting point to investigate specific aspects of ergodicity and localisation transitions in many-body systems. The first part is devoted to the outstanding question of whether MBL systems undergo two separate ergodicity and localisation transitions when disorder strength is increased. The two transitions are separated by a putative "bad metal", nonergodic extended (NEE) phase, in which ergodicity is broken but the eigenstates are not exponentially localised. We show explicitly the existence of the NEE phase in a random matrix model, and characterise it using the local resolvent in an unusual scaling limit. In the second part we consider a spin glass model, in which quantum effects are introduced by a transverse magnetic field. A refined equilibrium phase diagram, going beyond the quasi-static approximation, is obtained with a numerically exact diagrammatic Monte Carlo approach. We discuss the difference between the ergodic, eigenstate and clustering transitions. Finally, we consider a quantum model that shares some of the peculiar low-temperature properties which have brought the Sachdev-Ye-Kitaev (SYK) model under the spotlight of the string theory community. In our model such properties are understood in terms of the glassy dynamics of a corresponding classical stochastic system.

▼ The majority of online products and services, such as search engines, social networks, digital maps and electronic communications apps, are offered free of charge. But there is a catch. In exchange for the use of their 'free' services, online firms collect their users' personal data and process it for myriad of commercial purposes. The ability of firms to collect vast amounts of data and process them through sophisticated algorithms to reveal patterns, trends and associations (Big Data) has opened new routes for dominant firms to abuse their market power, and at the same time, has given rise to pressing online privacy and consumer protection issues which, whilst falling outside the scope of competition law, have nevertheless an impact on the competitive process in online markets. The aim of this thesis is to uphold the importance of Article 102 TFEU enforcement for the healthy operation of competition in online markets. To this effect, it explores recent proposals for the passing of a new sector-specific regulation applicable to online platforms and for lax antitrust enforcement in the digital economy, exposes their flaws, and demonstrates that the enforcement of Article 102 TFEU can be flexible enough to account for the challenges posed by online markets. The key is to identify the features that differentiate online competition from competition as traditionally conceived in competition analysis, and to determine how such differences translate into new emphases, tests and approaches. In addition, acknowledging the interrelated nature of competition, data protection and consumer protection issues that arise from data-driven competition, this thesis proposes the creation of a new category of abuse of dominance based on infringements of data protection and consumer protection law, when such infringements are linked to the infringer's dominant position, adversely affect innovation and choice, and/or derive in the exploitation of consumers.

▼ Over the course of the last few decades, Internet has evolved to form a major technological component in our society. Accompanying this exponential growth are many challenges embracing both users expectations and ambitions of network providers. Internet Protocol (IP) is continuously being extended from the native Internet towards ubiquitous packet delivery models for the next generation of cellular networks. The two families of networks with their means of access, technologies and architectures are converging. The mutable access network structures, deployment practicalities and global traffic increase, necessitate advancements of packet delivery models. Evolution of the cellular network architectures introduces the flat-IP structure and novel topological arrangements in the backhaul. A flexible routing within an efficient Traffic Engineering (TE) is needed for novel random all-IP access network topologies with heterogeneous wireless accesses. These requirements converge with the conventional IP intra-domain routing concepts. Multi-Plane Routing (MPR) is a TE approach comprised of offline and online features tailored for all-IP access networks. Diversity is leveraged upon via multiple paths between traffic sources and destinations rendered by slicing the physical topology into multiple logical routing planes. Performance is enhanced utilising the entire topology while minimising the MPR-incurred protocol overhead. In this thesis, MPR extensions reflecting the evolution of access networks and emerging traffic types are studied. Minimum Set Cover (MSC) mathematical approach for random graphs representations of IP access network is applied accommodating novel features in offline TE: capacity planning/projections in conjunction with path correlations. Initially, a MPR utilization of the whole topology is validated via extensive simulations for diverse models of IP packet delivery improving upon key performance criteria and supporting meshing. The study proceeds to broader cases of random topologies of different sizes and sparseness where MSC-approach proves as superior by managing such complex routing resources. Additionally, Quality of Service (QoS) is tested for many traffic types including the Tactile traffic with its stringent requirements.

▼ The transcription factor Cdx2 is required for the formation of several developmental lineages in the mouse embryo, and is thought to play different roles within the same lineage at different stages. The role of Cdx2 in establishing trophoblast fate has been extensively studied, but the reason for its continued expression in established trophoblast cells is less well understood. To explore this question, I generated ATAC-seq and RNA-seq libraries from wild-type and Cdx2 knockdown trophoblast stem cells (TSCs) in vitro. ATAC-seq experiments show, as expected, that loss of Cdx2 causes decreased accessibility at CDX2 consensus binding sites. However, of the regions showing differential chromatin accessibility in Cdx2 knockdown cells, two thirds have increased accessibility. These sites are enriched for the TFAP2C consensus motif and footprint; Tfap2c is a TSC marker that plays a part in trophoblast differentiation by driving genome-wide increases in accessibility. Gene ontology analysis of sites with increased accessibility in Cdx2 knockdown cells suggests that these regulatory regions are associated with trophoblast giant cell (TGC) differentiation. Consistent with this observation, continued Cdx2 knockdown drives homogenous differentiation of trophoblast stem cells into trophoblast giant cells within days. Similarly, homozygous Cdx2 knockout TSCs are unstable and spontaneously differentiate into TGCs, although in this context cells appear to retain some ability to compensate. TSC differentiation can also be initiated in vitro by growth factor withdrawal, albeit in a heterogeneous manner. However, the sites of increased chromatin accessibility observed under these circumstances differ from those observed in Cdx2 knockdown cells. Moreover, although RNA-seq analysis shows that 87% of genes whose expression changes in Cdx2 knockdown cells are also mis-regulated during conventional differentiation, 31% of these transcripts change in opposite directions. This work suggests that Cdx2 normally maintains 'stemness' in TSCs by preventing them from differentiating directly and homogeneously into TGCs. This gatekeeping role of Cdx2 differs from its earlier function in repressing pluripotency genes and suggests that Cdx2 is repurposed to play different roles within the trophoblast lineage. Furthermore, our work may finally provide a plausible mechanism by which half of non-progenitor derived parietal TGCs are generated.

▼ This thesis investigates active and/or passive utilization of robot morphology to achieve effective and safe interaction with its environment. The proposed approach is based on replicating certain morphological features observed in nature to enhance robotic performance during contact. The term morphology herein refers not only to the shape, but also physical properties such as stiffness, as well as actuator and sensor distribution. Within the context of “Soft Robotics”, two paradigms entailing contact are considered: a soft continuum manipulator for soft interaction and haptic perception; and a rigid-link arm with internal actuator compliance for potential industrial collaborative tasks. Firstly, A fluid-actuated intrinsically-soft continuum robot is proposed as a bioinspired finger-like appendage. A novel analytical model is presented to solve the continuum kinematics based on the Cosserat Beam Theory and the Principle of Virtual Work, yielding complete pose and force data across the backbone hence attaining embodied sensory information. In addition, elasticity versus hyperelasticity considerations are discussed, showing the validity of elastic assumptions within its operational range. Secondly, for the first time, the soft robotic appendage is used for haptic perception in soft tissue palpation for anomaly detection. Using Silicone as a tissue phantom, different actuation pressures are applied to the robot for mapping the tissue surface and detecting a hard nodule embedded in the phantom while estimating the tip force of the robotic appendage. Thirdly, an innovative design is introduced to generate variable impedance for the soft robotic appendage. Inspired by muscular movement in nature, a hybrid actuation scheme is proposed by introducing embedded tendons inside the manipulator’s wall to enable stiffness adjustment, which also improves manoeuvrability. Experiments are carried out by applying an external force in different configurations while changing the stiffness by the two actuation mechanisms, demonstrating that dual antagonistic actuation increases load bearing capacities in this class of manipulators. Subsequently, the tendon-augmented robotic appendage is enhanced by multiplying the tendons to produce a more uniform longitudinal tension distribution. Sweeping tests of a Silicone phantom with an embedded nodule are again carried out with different actuation pressures and tendon tensions. Utilising force sensors placed at the tip and at the base (representing tactile and proprioceptive cues, respectively), the behaviour of selected sensing modalities are investigated and compared. This novel setting delivers insights into the role of internal impedance regulation in perceptive exploration and how the tactile and proprioceptive sensory cues evolve by alteration in the morphology of the appendage. ▼ Many epidemiological and clinical studies suggest a link between skin function and glucose homeostasis. For example, the inflammatory skin disease psoriasis is an independent risk-factor for Type 2 diabetes (T2D) development, and presence of psoriasis is a strong predictor of T2D progression and severity. Moreover, some skin-specific transgenic mouse models display altered whole-body glucose homeostasis. This suggests an important role for skin in regulating whole-body glucose metabolism. However, the underlying mechanisms are unclear. We hypothesised that the skin plays an important regulatory role in whole body glucose homeostasis, via the actions of skin-derived proteins and lipids, termed the skin secretome. Secondly, we hypothesised that the skin secretome becomes dysregulated in psoriasis, leading to impaired metabolic homeostasis. To investigate this hypothesis we adopted two main approaches: (1) The imiquimod (IMQ)-mouse model of psoriasis was used to investigate the effects of skin inflammation on whole-body metabolic phenotype in vivo. Glucose and insulin tolerance tests were conducted to assess whole-body glucose metabolism and key metabolic tissues - particularly pancreatic islets and subcutaneous adipose tissue (sAT) - were collected to assess tissue specific effects on skin inflammation. (2) Human explant skin was cultured and treated with imiquimod to induce a psoriasis-like phenotype. Skin conditioned media (CM) was collected and used to treat sAT and pancreatic islets. Islets and adipose tissue were then analysed to assess the regulatory effects of skin-derived factors present in the CM. IMQ-mice displayed signs of systemic and sAT inflammation and insulin resistance. Furthermore, pancreatic β-cells showed signs of compensation to insulin resistance in the form of improved glucose tolerance and increased β-cell function and proliferation. Consistent with a role for the skin secretome in these effects, incubation with skin-CM (IMQ) induced inflammation in healthy sAT. In contrast to the in vivo findings, incubation with skin-CM (IMQ) induced signs of β-cell dysfunction in healthy islets. In conclusion, this data demonstrates that skin inflammation alone can regulate whole-body glucose metabolism, via actions on key metabolic tissues including pancreatic islets and sAT. The effects on sAT appear to occur via direct actions of the skin secretome, whilst the effects on islets are potentially dependent on more complex inter-organ crosstalk. Fully characterising the skin secretome could lead to the identification of novel psoriasis co-morbidity biomarkers and therapeutic targets.

▼ This thesis focuses on engineering a repair strategy for use in peripheral nerve tissue after transection injuries. With an estimated 300,000 new cases of peripheral nerve injury reported in Europe each year, the socioeconomic cost of these injuries is multifactorial (Chiardelli and Chiono, 2006). Loss of nerve function often leads to long-term disability and chronic neuropathic pain, which dramatically reduces the number of quality adjusted life years (QALYs) post-injury (Siemionow and Brzezicki., 2009; Ciaramitaro et al., 2010). Some research suggests that nerve repair surgery results in a cost effective increase in number of QALYs and suggests that this field warrants further research (Wali et al., 2017). Further, nerve injury has been shown to correlate with post-traumatic stress disorder, especially when loss of function persists longer than 12 months (Miller et al., 2017). 360,000 people in the US suffer paralysis resulting from upper limb trauma alone, which suggests that the total number of patients with peripheral nerve injury (PNI) is much larger and means a large proportion of the population suffer with a nerve defects and their complications. Often, PNI is concomitant with multiple other acute traumas resulting from incidents such as motor vehicle accidents. Therefore peripheral nerve repair (PNR) is withheld as repair of other traumas is prioritised (Gin-Shaw and Jorden, 2002). Evidence suggests withholding PNR reduces the efficacy of regeneration and if subfunctional repair occurs this has a long-term negative effect on patient well-being and ability to self-care (Faroni et al., 2015). Nerve tissue injury leads to degeneration of functional neural cells and the loss of effective communication between the peripheral nerve and nervous system at large, meaning many vital neurological reflexes are lost. Effective nerve repair strategies that expedite recovery are vital to preventing long term disability and improving patient quality of life. The current gold-standard of PNR involves use of autologous nerve tissue, which results in comorbidities, and lack of reliable results (autograft repair methods) (di Summa et al., 2014). However, the development of tissue engineering constructs to encourage peripheral nerve regeneration represents an alternative therapy to autograft. Provision of a synthetic implant, a Nerve Guidance Conduit (NGC), is designed to mimic nerve tissue and encourage infiltration by endogenous glial cells (Schwann cells) and neural cell processes (axons) across the graft (Barton et al., 2017). In contrast to studies that solely focus on the biological events that take part following peripheral nerve injury, this thesis presents an engineering approach to create a more functional design for a synthetic NGC. ▼ Cysteinyl leukotriene (LTC4, LTD4 and LTE4) are proinflammatory lipid mediators involved in the pathogenesis of asthma. LTE4 the most stable of the cysteinyl leukotrienes elicits bronchoconstriction, and unlike LTC4 and LTD4 it enhances bronchial hyperresponsiveness. LTE4 can also mediate other features of asthma, such as eosinophilia, increased mucus secretion and vascular permeability. None of these LTE4 activities can be explained by current understanding of classical cysteinyl leukotriene receptors, CysLT1 and CysLT2, suggesting the presence of a novel receptor that preferentially responds to LTE4. LTE4 mediated pulmonary inflammation in a murine model was shown to remain intact in the absence of CysLT1 and CysLT2, and was abrogated by the knockout of the purinergic receptor, P2Y12. This study aimed to elucidate whether LTE4 directly signalled through the P2Y12 receptor. Models of human CysLT1, CysLT2 and P2Y12 overexpressed in HEK293T, CHO cells and human platelets were used to characterise responsiveness to cysteinyl leukotrienes. LTE4 failed to induce any specific signalling responses in cells expressing P2Y12 or in human platelets, showing that LTE4 does not directly signal through P2Y12. Previous reports have shown that LTE4 is a potent agonist in human mast cells and so the aim was to identify the receptor responsible for LTE4-induced responses using two human mast cell lines, LAD2 and LUVA. When intracellular signalling and gene expression were compared in response to stimulation, LUVA cells responded to LTC4 and LTD4 but lacked the potent responses to LTE4 observed in LAD2 cells. Gene screening by microarrays was used to identify 4 target GPCRs which were cloned and their responsiveness to cysteinyl leukotrienes analysed in HEK293T overexpression model. CysLT1 was identified as the only GPCR responding to cysteinyl leukotrienes and differently expressed in LAD2 cells. Stable knockdowns were created and analysed to reveal CysLT1 was responsible for the augmented responses to LTE4 observed in LAD2 cells.

▼ This thesis investigates how symbolic and economic value is generated in the artworld, through a complex system that I have conceptualised as the art machine. This mechanism is made up of artists, dealers and galleries, art schools, critics, auction houses, museum and independent curators, private and corporate collectors as well as foundations and government entities, who instinctively collaborate in creating a name and a market for visual artists. Within this art machine, there exists not only an interdependency between the various functional parts, but also an innate fragility when the local mechanism is subjected to significant changes in its macroenvironment. My research uses this conceptual framework of the art machine to study in detail the workings of the Venezuelan art market, which, with the arrival of the Regime of the late Hugo Chávez, has witnessed significant structural changes, most notably to the existing institutional framework designed to legitimise the visual arts. Despite these recent socio-political changes as the State purposefully sheds a capitalist model of value-creation for the visual arts, the local art machine continues to operate using an alternative institutionalism that allows its continued operation under these adverse conditions. The thesis uses qualitative research methods (semistructured interviews, participant observation, and illustrative photography) in examining the various complexities of the art market in Venezuela. This inductive approach is underpinned by Constructivism, where realities (value, reputation and demand for art) are constructed in the minds of individuals and groups, enabling the research to reconstruct Venezuela's artworld via a selective sample of key participants, including artists (upcoming and established), commercial galleries, private and public curators, critics, corporate and private collections, and auction houses. As well as providing insights into the field of (arts) marketing, my research highlights the significance of institutionalism to understanding the impact of the macroenvironment on marketing and consumption.

▼ This thesis uses individual level data to analyse the potential impacts of temporary migration in Eastern Europe. The empirical analysis looks at: (i) the patterns of selection into migration and return, (ii) the labour market returns to return migration and (iii) the impact of a change in natives' attitudes on the intended duration of stay abroad. The analysis commences by presenting cross-country evidence on the labour market outcomes of recent return migrants in Eastern Europe (Chapter 2). It addresses both the income premia received by returnees and their occupational choices upon return. The rest of the thesis goes into more details by analysing the wage earning ability and the selection of Romanian migrants and returnees (Chapters 3 and 4). The results suggest that the selection and sorting of migrants by skills is driven by different returns in countries of destination. These findings are consistent with a model of rational choice in migration decisions. They imply that for a source country like Romania relatively high rates of temporary migration might have positive long- run effects on average skills (schooling) and wages. These effects crucially depend on the magnitude and selection of return migration flows. The last part of the thesis (Chapter 5) addresses therefore one widespread consequence of recent East-West migration in the aftermath of economic recession: the anti-immigrant backlash sweeping across major destination countries in Europe. A widely documented crime in Italy provides a quasi-experimental setting to identify the impact of natives' attitudes on the return plans of Romanian migrants. Our results suggest a significant effect of anti-immigrant attitudes on the intended duration of stay in the host country. The impact is more pronounced for low-skilled migrants. This in turn has important consequences for how migration affects the long run convergence between sending and destination countries.

▼ The films of Lars von Trier are consistently described as excessive, categorized as extreme cinema. Dense, dark, but rarely titillating, they enhance conflict and impact by simultaneously folding together multiple layers of excess both stylistic and visceral within complex narrative frameworks, all of which lack clear narrative resolution but culminate in death. Von Trier’s films induce trauma, and emotional rupture rarely paralleled in cinema yet challenge audiences to respond to cinema in new ways. Although often noted, the film’s spiritual, religious nuances rarely have extended analysis beyond Breaking the Waves (1996) and Dogville (2003). I will argue that the spiritual, biblical content of the films holds a key to making sense of von Trier’s oeuvre of excess. In this perspective, the excess is subsumed under spiritual conflict—beyond a human to human struggle—which acts as a centrifugal force, exploding outward. Thus, spiritual conflict is the mechanism that unpacks the films’ notorious excess. Von Trier's films, in my view, truthfully and prophetically expose the complicity of the spectator in personal and structural evil, forcing self-examination related to the films' theological themes. As such, von Trier’s films and concerns may be considered analogous to the extreme, transgressive prophetic voice of the Hebrew prophet Ezekiel, understood in light of insights drawn from his prophecy and its form of delivery. I will explore the prophetic voice in von Trier's films by juxtaposing them with Ezekiel's prophecy through Paul Ricoeur’s hermeneutics and Ben Quash’s theological aesthetics. Ezekiel and von Trier both have a reputation as destabilizing critics of their cultures, von Trier as the provocateur of extreme cinema, and Ezekiel as the outrageous prophet who acts and verbalizes the unspeakable in God's name. The focus of this thesis however, is not von Trier’s status as an artist-prophet, but a prophetic voice that issues forth in the films, juxtaposed not with Ezekiel the prophet, but Ezekiel’s prophecy and prophetic acts. There is not sufficient evidence to validate von Trier as artist-prophet, and the layers of history and redaction limit our knowledge of the person Ezekiel.

▼ This thesis argues for an identifiable literature of the bedchamber in late medieval England. Arising from the socio-economic context in which the private bedchamber became more accessible to wealthy merchants, as well as the nobility, than it had previously been, the literature of the bedchamber comprises romance, fabliaux, dream poems and visionary literature. The cross-genre nature of such a body of literature has previously obscured the centrality of the bedchamber to the imaginative worlds of the texts that constitute it. Using a material cultural and socio-historical approach, I aim to examine the cultural signifiers attached to the bedchamber, to do with privacy, gender, the life-cycle, and its status as a literary phenomenon. The bedchamber is a creative space, from within which texts are structured, put into contact with each other, and created. This thesis is divided into four chapters, each one focusing on a text or set of texts that illustrates a specific feature of the literature of the bedchamber. In the first chapter of this thesis, I look at privacy in Troilus and Criseyde, both in terms of how it is performed and how it is experienced. Troilus and Criseyde are each operating within different generic concepts of privacy that are ultimately irreconcilable with each other. The second chapter turns to Sir Gawain and the Green Knight, offering an analysis of the bedchamber textiles. I argue that these textiles generate an aesthetic of enclosure that structures the poem as a whole but that also destabilises Gawain’s knightly identity, providing the means through which his ‘trawþe’ is tested. In the third chapter, I examine The Book of Margery Kempe and the centrality of the bedchamber to Margery’s devotional behaviour. By drawing on different models of devotion, Margery uses the bedchamber to establish her spiritual identity and authority. Chapter four takes Chaucer’s dream poems as its focus and looks at the portraits of the private reader that he creates in them. Using the bedchamber as a space for a secular version of meditative reading, Chaucer positions reading as a creative activity that decentres the authority of the author. The thread running through each chapter, and defining the literature of the bedchamber, is the use of the bedchamber as a space in which to generate narratives, whether through the tension between the private space of the bedchamber and the public world outside, or through bringing different genres and the expectations arising from them into contact with each other. The literature of the bedchamber is about bedchambers and created within them.

▼ Skin is the primary interface to the external environment, protecting us from harmful compounds and infection. Over many millennia, coevolution has culminated in a mutualistic relationship between ourselves and a specialised, yet diverse resident community of microorganisms. This microbiota is thought to exist in a delicate balance to maintain homeostatic equilibrium, and plays a role in the shaping of our immune system. Skin diseases are some of the most common human disorders and present a considerable economic burden. There is now growing appreciation of the role the cutaneous microbiome plays in disease, and how host-microbe interplay is associated with disorders of the immune system. The cutaneous microbiota as well as the host transcriptome and the interactions between them is the focus of this thesis. Computational methods were used to examine the microbiota and transcriptomes of a large cohort of matched samples from healthy volunteers and patients with Atopic Dermatitis (AD) and Psoriasis (PSO). The community composition of inflamed and healthy skin was assessed and the specific species which are over-represented on diseased skin were identified. In parallel, the host gene expression was pro led to identify common and disease specific transcriptional signatures revealing clinically relevant pathways. Next, using schemes of dimensionality reduction and computational methods, the associations between microbes, disease severity and host transcription was interrogated. <i>Staphylococcus aureus</i> demonstrated an impressive relationship with AD associated gene signatures, whereas host-microbe associations in PSO were inconclusive. Using Weighted Gene Co-expression Networks Analysis (WGCNA), gene-gene interaction networks were reconstructed and differentially connected modules between healthy and inflammatory states were identified. Modules encoding processes for the epidermal barrier, extracellular matrix, non-coding RNA metabolism and immune system processes were all associated with the relative abundance of S. aureus. Overall, whilst associations in psoriasis were inconclusive, a range of host-microbe interactions were uncovered in AD. The results presented in this thesis contribute towards a greater understanding of the differences in the cutaneous microbiome and the transcriptional mechanisms which underlie allergic and autoimmune inflammation.

▼ The compositions in this portfolio explore the expressive potential of ‘vocality’ and its opposite, the avoidance of vocal qualities. Most prominently, in 'Four Rhapsodies, Poems without Words, Two Poems' and 'Poemas de amor' the reciting voice provides a model for instrumental composition. Conversely, in 'Sto periyáli to krifó' and 'Two Impressions', the undermining of the vocality of the singing voice renders a further spectrum of expressive possibilities. Furthermore, in 'Canto' and 'Sonatina' ‘vocality’ is combined with quotation, re-contextualisation and paraphrase. In the instrumental works recitation often involves lines arising through processes of melodic fusion and fission, as well as frameworks derived from the sonic and semantic features of particular poems. These types of melodic materials are present not only in compositions that represent the reciting voice, but also in Aedh’s 'Kaleidoscope, Tre per due' and 'Sto periyáli to krifó'. Poetic frameworks underlie the creation of all compositions except 'Canto, Sonatina' and 'Tre per due'. Generally, harmonic colour and tension, varied return and metric regularity complement considerations about vocality. The compositions in this portfolio engage to various extents with both the classical repertory (contemporary and historical) and non-classical music. More specifically, 'Tre per due' and 'Sto periyáli to krifó' draw on elements of Turkish modal music, while 'Sonatina' borrows from Colombian and Brazilian song and dance forms.

▼ In this work, we provide a theoretical study of physical layer security (PHYLS) performance in full-duplex (FD) small-cell networks. Here, the base stations (BSs) and user equipments (UEs) follow from the homogeneous Poisson point process (PPP)-based abstraction model. First we consider the network where BSs serve a single user in one resource block where PHYLS in both scenarios of HD and FD UEs are analysed and compared. We derive the ergodic secrecy rate in both downlink (DL) and uplink (UL) in presence of a field of passive eavesdroppers (EDs). To facilitate FD communications, we take into account (i) successive interference cancellation (SIC) capability at the UE side via guard regions of arbitrary radii, and (ii) residual self-interference (SI) at the BS side using Rician fading distribution with arbitrary statistics. We study the affect of density of BSs and EDs in the FD mode and compare it to its HD counterpart. Next, we consider the case where BSs serve several HD single-antenna users at the same time and frequency with assistance of multiple antennas. We investigate the small-cell network PHYLS performance in the presence of a Poisson field of EDs, under the different scenarios of passive and colluding eavesdropping. Considering linear zero-forcing (ZF) beam-forming in the multiple-input multiple-output (MIMO) scenario, we characterize the UL and DL ergodic secrecy rates and derive closed-form expressions for the different useful and interference signals statistics. Moreover, we look into the impact of SIC and SI capabilities in the UE and BS side, respectively. Thereafter, we derive explicit expressions of the key performance indicators using tools from machine learning. In particular, for certain special cases of interest, we apply supervised learning-based non-linear curve-fitting techniques to large sets of (exact) theoretical data in order to obtain closed-form approximations for the different ergodic rates and ergodic secrecy rates under consideration. Our findings, obtained from theoretical analysis and system-level simulations, indicate that the FD functionality, in addition to enhancing the spectral efficiency (SE), can significantly improve the PHYLS performance, especially with the aid of multi-antenna communications and interference cancellation schemes. Finally, we highlight several promising future research directions in relation to the outcomes of this work, including the application of physical layer security and full-duplex operation in the context of 5G networks and beyond.

▼ This thesis is an examination of the cultural and political climate of the medieval Welsh Marches. The investigation is structured around three case studies: Hereford, c. 1180-c. 1210; Ludlow, c. 1310-c. 1350; Cwm Tawe, c. 1380-c. 1410. Using these three case studies, the thesis develops a critique of the core-periphery model that has dominated modern conceptualisations of medieval political and cultural geographies. In its place, I formulate an alternative model based on an engagement with social theories of the network, including Manuel Castells's 'network society' and, in particular, Bruno Latour's work on 'actor-network theory'. Reading with networks might, I contend, provide a new, more ethical interpretative model, one capable of restoring cultural and political agency to erstwhile 'peripheral' regions. The first chapter traces the multifarious textual networks in which the three case-study locales were active: it thereby identifies the corpus of the following three chapters (i.e. the texts circulated, composed, copied, or translated in each locale), and situates them in their manuscript contexts. The second chapter develops 'networked' readings of the texts themselves: I analyse the ways in which the texts position their local environments in relation to the global networks described in their narratives, such that they not only disprove the 'peripheral' status ascribed to the Marches, but more searchingly question the validity of the model that produces such ascriptions. Chapters 3 and 4 take a thematic approach, using networks to analyse the political modalities of the texts' representations of nonhuman agency (Chapter 3) and of issues surrounding language, translation, and multilingualism (Chapter 4). Chapter 5 opens up the corpus to investigate representations of the Welsh Marches in Arthurian literature: I suggest that not only do the Marches emerge as highly connected regions, but that Arthurian literature more fundamentally imagines a networked model of political and cultural geography. The thesis concludes by turning to the Hereford 'mappa mundi' as a succinct image of the investment of the so-called 'periphery' in a truly global worldview, and by reasserting the importance of reading with networks in the current political climate.

▼ Mesenchymal stem cells (MSCs) are adult stem cells which are characterised by self-renewal and differentiation capacity into three mesenchymal lineages: osteoblast, chondrocyte and adipocytes. MicroRNAs (miRNAs) are a class of non-coding short RNAs (20-22 nucleotides) which target messenger RNAs to supress protein synthesis and regulate cell function. There is increasing evidence that miRNAs can play key roles in the regulation of multiple biological processes include stem cell regulation. Therefore, the aim of this study was to investigate the potential role of miRNAs in the regulation of MSC fate decisions. Firstly, 4 different MSC cell lines were tested for their differentiation capacity following induction with osteogenic, adipogenic, or chondrogenic media. From these results one cell line was selected for use in further studies. The expression of 9 candidate miRNAs which had previously been shown to be associated with regulation of other stem cells were tested during osteogenic or adipogenic induction. To identify novel miRNAs associated with MSC regulation, clonally derived MSCs with osteogenic, adipogenic or bipotent differentiation patterns were tested using miRNA array. From the results of the array 2 family members of the miRNA-302 (a and b) were selected for further investigation. MiR-302a and b expression was validated by qRT-PCR in the clonally derived cell lines and in primary MSC cultures. To determine the role of the miR-302 family during MSC differentiation, their functional activity was tested by knock-down and over expression, via transfection of a miRNA-302 family inhibitor or individual miR-302a and miR-302b mimics into MSCs and the cells were treated with osteogenic, adipogenic or growth medium as control for 14 days. Then lineage specific gene expression was measured by qRT-PCR. Expression patterns of the 9 candidate genes showed no reproducible significant effects during osteoblastic or adipocytic differentiation. MiRNA-302a and b were both down regulated during differentiation of MSCs into both osteogenic and adipogenic lineages. Knock-down of miRNA-302 family in MSCs caused a significant upregulation of between 2-5 times of RUNX2 expression and between 3-10 times of ALP expression during osteogenic differentiation of MSCs. Overexpression of miR-302b did not have any significant reproducible effects on osteogenic differentiation of MSCs. In contrast, over expression of miR-302a resulted in increased RUNX2 expression by 1.5-2 fold on day 2 and ALP expression by almost 50 fold after day 4. Inhibition or over expression of miR-302 family had no early effects on adipogenesis of MSCs, but at day 14 FAB4 was upregulated nearly 40% in inhibited cells and between 20-30% in miR-302a overexpressed cells. In contrast overexpression of miR-302b downregulated FAB4 by a range of between 10-80% at day 14. ▼ Prevalence of liver disease is rising in the UK. Public health strategies suggested to reduce the burden of liver-related morbidity and mortality include management of obesity and type 2 diabetes in addition to screening for liver fibrosis in at-risk groups. Increased prevalence of liver-related mortality has been reported in people with severe psoriasis since the late 1980s. Historically, this increased risk was attributed to methotrexate use. In the last decade, studies have shown that people with severe psoriasis are also at increased risk for obesity, metabolic syndrome, non-alcoholic fatty liver disease and alcohol excess which are all risks for the development of liver fibrosis in the general population. Despite this, there have been no studies to investigate the natural history of liver fibrosis in people with severe psoriasis. Therefore, who within the severe psoriasis population is most at risk for liver fibrosis is not understood. Liver fibrosis is usually asymptomatic until cirrhosis develops. Liver biopsy is the gold standard test for diagnosis of fibrosis; however, the biopsy procedure is associated with a significant morbidity and mortality, which has limited its use as a research tool. The advent of transient elastography (TE) has enabled liver fibrosis estimation without any associated risks to the patient. Consequently, the risks associated with fibrosis have been studied in other populations such as non-alcoholic fatty liver disease (NAFLD) and viral hepatitis, although not yet in people with severe psoriasis. European dermatology guidelines recommend serial procollagen-3 levels (P3NP) to screen for liver fibrosis in people with psoriasis taking methotrexate, based on studies performed in the 1990s. This is in contrast to guidelines for NAFLD which recommend screening for liver fibrosis using TE and/or panels of serological tests developed as surrogate markers for fibrosis. These composite tests have not been prospectively evaluated in a severe psoriasis population. Earlier detection of liver fibrosis would provide an opportunity to reduce liver-related morbidity and mortality in people with severe psoriasis. Therefore, the work in this thesis aimed to address three research questions. Firstly, to understand how common liver fibrosis is in people with severe psoriasis. Secondly to determine who within the severe psoriasis cohort is at increased risk of liver fibrosis by identification of risk factors associated with liver fibrosis in this group. Thirdly to evaluate the diagnostic accuracy of TE and serological tests to detect liver fibrosis in people with severe psoriasis. ▼ Reconcilability between family and work is an issue thematised in various literatures. What is often missing from accounts of Feminist Political Economy and Economics, as well as Welfare State Research, is an engagement with the everyday and how 'time' is experienced as a multifaceted issue for carers. Focussing in on time, this dissertation addresses parenthood in sequences, from the wish for children, through the first year after birth, and in the transition from familial to public childcare. Tracing the 'asynchronous' temporal structures of labour markets and public childcare in everyday life in Cologne, Germany, it reveals how 'temporal disempowerment' is affecting carers. In terms of a conflict between neo-familialist and third-way ideologies, findings suggest that the current state of affairs has not stabilised towards any hegemony. The results are broken institutional interfaces in the shape of scheduling and synchronisation problems, as well as psychological issues arising from a mismatch between experiences and expectations around work and family reconcilability.

▼ IL-17A is a well-characterised pro-inflammatory cytokine, which has been implicated in the immunopathology of inflammatory arthritis. IL-17F is less well-studied. This thesis describes investigations into the phenotype of IL-17F+ CD4+ T cells, what drives IL-17F expression in CD4+ T cells and how IL-17F may contribute to inflammation. Healthy donor CD4+ T cells were cultured with IL-1β, IL-23, anti-CD3 mAb and anti-CD28 mAb, and cytokine expression was analysed by flow cytometry. In comparison to IL-17A+ IL-17F- CD4+ T cells, IL-17F+IL-17A- and IL-17A+IL-17F+ CD4+ T cells contained lower proportions of cells co-expressing IL-10 and higher proportions of cells co-expressing IFNγ. Titration of anti-CD28 mAb into healthy donor CD4+ T cell cultures revealed that strong co-stimulation increased the frequency of IL-17F+IL-17A- and IL-17A+IL-17F+ CD4+ T cells, whereas IL-17A+IL-17F- CD4+ T cell frequencies decreased. This was shown to be via an IL-2 dependent mechanism. To allow for the further characterisation of different IL-17A+ and IL-17F+ CD4+ T cell populations an IL-17A and IL-17F capture assay was developed in this thesis. Addition of human recombinant IL-17A, IL-17F and TNFα to synovial fibroblasts from patients with psoriatic arthritis (PsA) and rheumatoid arthritis (RA) resulted in significant production of IL-6. Combined blockade of IL-17A and IL-17F reduced IL-6 production to a larger extent than blockade of IL-17A alone, indicating IL-17F can contribute to inflammation. While IL-17A protein and IL-17A+ T cells were detected at increased levels in the inflamed joint of RA and PsA patients, little IL-17F protein and few IL-17F+ T cells were detected. Collectively, data presented in this thesis indicate that IL-17A and IL-17F are differentially regulated and that strong T cell stimulation induces IL-17F. Additionally, data suggests IL-17A+IL-17F-, IL-17F+IL-17A- and IL-17A+ IL-17F+ CD4+ T cells are functionally distinct. Finally, it is shown that in an environment where IL-17A and IL-17F are present dual blockade of IL-17A and IL-17F is more effective at reducing inflammation.

▼ This thesis focuses on the extent and determinants of intergenerational income mobility in Indonesia between 1993 and 2014. Most developing countries and even some developed countries exhibit high levels of intergenerational persistency (rather than mobility) which implies that a child’s future income is to a great extent determined by their parental income. Such a scenario violates the notion of equal opportunity for all children regardless of their parental background. Estimates of intergenerational mobility are extensive in terms of levels, measures, causes, and implications of relative and absolute economic mobility across generations within and between countries. However, such studies, to date have not focused in-depth on the case of Indonesia. The thesis addresses this gap in knowledge and makes use of five waves of the Indonesia Family Life Survey (IFLS), a longitudinal study of socio-economic status in Indonesia covering 7,224 households in the 1993 wave distributed across 13 provinces. The thesis does the following: (i) makes estimates of relative and absolute income mobility; (ii) compares estimates for Indonesia with international estimates; (iii) assesses how income mobility differs across cohorts, genders, and provinces; and (iv) identifies the determinant of income mobility in Indonesia. To generate income mobility estimates for Indonesia a pooling sample connecting economic outcomes of parents and their respective adult-children is constructed from five waves of the IFLS. Relative mobility is measured by regressing children’s outcomes on the parent’s outcomes of those children. Absolute mobility is measured by estimating the conditional probability of children being 'trapped' (children being in a low-income position if their parent was also in a low-income position); the conditional probability of children being 'upwardly mobile' (children being in a middle-income position even if their parent was also in a low-income position); the conditional probability of children being 'privileged' (children being in a high-income position if their parent was also in a high-income position); and the unconditional probability of children having a higher income than their parents. After providing robust estimates of income mobility, further analyses are conducted as follows: (i) mobility indices are compared to estimates of other countries; (ii) the pooling sample is disaggregated into different cohorts, genders, and provinces, to investigate how mobility rates differ across those groupings; and (iii) the determinants of relative and absolute mobility are analysed with a focus on the effects of children’s circumstances on income mobility.
