The annually recurring regeneration of deer antlers, accompanied by rapid growth of around 2 cm per day, is of great interest as a research model due to the structural and histological similarities between antlers and the bones of other mammals. The aim of the study was to investigate the effect of cell culture supernatant obtained from fallow deer antler cells on the proliferation and differentiation of human mesenchymale cells (HMCs) taken from the iliac crest. Here, in particular, the question was whether the supernatant was able to accelerate the differentiation of these cells into mature osteoblasts. The results show that the proliferation of the mesenchymale stem cells could be positively influenced by directly applying a supernatant from an antler cell culture. The mesenchymale cells reached up to 25 times the initial cell number, cultivated in a culture medium with a ratio of either 1:1 or 1:2 of antler cell supernatant. Furthermore, an effect on the differentiation behaviour of mesenchymale cells into mature osteoblasts was observed by detecting the expression of the bone markers like Cbfa1 (synonym Runx2), osteocalcin, and type I collagen. Osteocalcin was shown to be consumed by the HMCs within the medium, even though its expression was detected. This suggests that osteocalcin plays a role in the energy metabolism of these cells as well as in the mineralization process of the extracellular matrix.
