A competitive inhibition enzyme linked immunosorbent assay was validated and used to estimate the prevalence of MCF-group virus infections in a sample of Alberta farmed bison. Prevalence among 995 slaughterhouse specimens was 21.8%. Among 953 samples from a serum bank the prevalence was 23.9%, and among 646 samples from five bison herds the prevalence was 23.4%. Test results from samples from one isolated bison herd collected over a period of six years provided evidence that an MCF-group virus was being transmitted across generations in the absence of exposure to any other ruminant sources of MCF-group viruses. Study of an outbreak of MCF in bison following a brief exposure to sheep provided very strong evidence that transmission of MCF among bison does not occur. Culturing lymphocytes from 12 healthy adult bison in the presence of the phorbol ester, 12-0-tetradecanoyl phorbol-13-acetate and testing of DNA extracted from these cultures with consensus herpesvirus PCR allowed the identification of viruses very similar to ovine herpesvirus two in five of the bison samples. 2. Machin, Karen L. Effects of surgery, anesthesia and pain on reproduction and behaviour of captive and free-ranging ducks.

Intra-abdominal radio transmitters are used to provide valuable information on reproduction, movement patterns, habitat use, and survival in a variety of wildlife species, including waterfowl. Pain and stress associated during capture, handling, anesthesia, and with instrumentation during surgery may have sublethal consequences, which may interfere with normal behaviour. As pain is produced by any procedure or injury that causes tissue damage, it is likely that waterfowl implanted with radio transmitters would experience pain. Also, placement of transmitters during incubation necessitates an incision into the brood patch which may have significant implications, such as altered incubation patterns resulting in delayed hatch. Therefore this study attempted to quantify the effects of pain and its treatment on captive and free-ranging waterfowl. Plasma thromboxane B2 levels indicated that two nonsteroidal anti-inflammatory drugs (flunixin and ketoprofen) may exert pharmacological effects for at least 12 hours in mallard ducks. However, use of flunixin in waterfowl cannot be recommended because of extensive muscular necrosis. Treatment isoflurane-anesthetized mallards with ketoprofen demonstrated a significant analgesic through reduction heart and respiratory rate changes in response to painful stimuli. In free-ranging mallards, longer surgeries were correlated with increased time to first nesting attempt after intra-­abdominal transmitter placement. Females that received ketoprofen took 3.5 days less to the first nesting attempt than females that received saline, indicating that analgesia was beneficial. There was no evidence to suggest that ketoprofen was harmful. Bupivacaine (local anesthetic) may be shorter acting in ducks than in mammals. Sequestration and redistribution of bupivacaine may result in delayed toxicity but mechanisms are unknown. In ruddy ducks, bupivacaine did not appear to achieve long-term analgesia or prevention of post-operative pain-related behaviours. In nesting female mallards, surgery altered incubation patterns in the 24 hour post-operative period, regardless of analgesic (ketoprofen or bupivacaine). Incubation period duration was extended in bupivacaine-treated females compared to ketoprofen-treated females, indicating that analgesia may interfere with brood patch sensation. Increases in corticosterone and progesterone were detected following surgery which may indicate stress and/or pain. The benefits of administering analgesia cannot be overlooked in minimizing effects of placement of radio transmitters on free-ranging waterfowl. 3. Helbig, Larissa. Onset of puberty and seasonal fertility in bison bulls.

Onset of puberty was observed in 12 bison bulls by the collection of semen at monthly intervals for 12 months beginning at 13 months of age. Onset of puberty was defined as the time in development when an ejaculate contained a minimum of 50x106 sperm showing at least 10% progressive motility. At each collection, data were recorded on body weight, semen quality, fecal testosterone concentration, and physical development. Semen was evaluated for gross motility, individual progressive motility, sperm morphology, sperm concentration and volume. From these data bison bulls attained onset of puberty at an average age of 16.5 months and an average body weight of 353 ± 52.8 kg. Age was the greatest determining factor for onset of puberty in this group of bulls. Monthly abattoir collections of epididymal sperm (n=288) and testicular tissue (n=120) were evaluated to determine if bison bulls undergo seasonal changes in sperm production. Although epididymal sperm morphology did not give any indication of seasonal variation, the histological study of testicular tissue showed greater seminiferous tubule diameter (27.0 ± 4.3 ìm) during the breeding months (July, August and September) than during any other seasons. Semen collected at 4 different occasions during the year (June, November, January, and April) from live mature breeding bulls (n=21) was used to verify data collected from abattoir samples. Semen from mature bulls showed a significantly greater proportion of normal sperm in June than in November (73.8 ± 9.1%; 44.1 ± 24.3%), respectively. There was little improvement in sperm morphology at the January sampling but in April morphology improved to a level close to that observed in June. Fecal testosterone concentrations were highest in June (128.6 ± 67.4 ng/g) and lowest in April (48.5 ± 33.3 ng/g). Although there was no clear seasonal trend in sperm morphology from bulls sampled at the abattoir, mature bulls showed slight seasonal variations in semen quality.
