Small to Medium Enterprise (SME) performance is a function of the composite of the decision-making processes within the enterprise. Owner managers are conscious of the gap in their decision-making efficacy and acknowledge that the process is often unstructured and over reliant on ‘gut feel’. While there is a drive to introduce better decision aids, such as ERP implementations, their propensity to provide data rather than information does not speak coherently to the decision context within the SME. Owner managers typically resort to ex-post accounting measures (often on spreadsheets) as the basis for enterprise decision-making. This would suggest that the understanding of the SME operational decision needs has not been well articulated either in the literature or in current practice. A better understanding of the decision needs structure would facilitate the selection and deployment of more effective decision aids within SMEs. The purpose of this thesis is to provide such a structure via a Method for Structured Operational Decision Making (MSODM), which allows the user to ascertain relative decision difficulty and prioritise the operational decisions requiring management intervention. Relative decision difficulty is determined using the Decision Difficulty Taxonomic Technique (DDTT), which consists of a detailed analysis of the three main elements of a decision, namely the decision itself, the decision maker and the decision environment. Decision prioritisation is based on the assessment of the impact of decision scenarios (clusters of related operational decisions) on enterprise performance measures. The method also incorporates a decision improvement cycle which identifies, develops and deploys decision specific support solutions and then measures the ‘before and after’ impact on the decision difficulty. The resulting MSODM not only ensures resources are focussed on the areas of most concern to the SME owner manager providing coherent guidance as to the most opportune areas to apply decision aids, but also provides a measure of the relative impact of any solutions deployed.

The focus of this thesis is to determine if there exists a set of dynamically available dominant, operational, ex-post performance measures for owner managed SMEs (small and medium enterprises). There is a predominance of performance measurement literature that focuses solely on corporate strategy with little or no literature speaking to advance operational performance. This thesis focuses on SMEs and the identification and application of performance measures for the daily operational decision making of an SME owner manager. The majority of the research developments in relation to manufacturing have examined idealised corporate strategy models that bear little relation to the resource constraints and constrained problems that are experienced in SMEs. Few of these developments are inter-changeable among large enterprises and fewer still are suitable for SMEs. The concepts contained in this document were developed through a review of literature and a methodology that involves significant practitioner input through case studies, a survey, a workshop and direct action research within 2 SMEs. SMEs spend the majority of their time dealing with operational concerns revolving around availability of time, money and people. SME owner managers require a method to adequately monitor and control operational concerns before they focus time and money on dealing with strategy. This research focuses on 3 objectives that will contribute to SME management capability; the identification of a set of dominant performance measures, the application of dominant performance measures and the validation of those measures in a more global context. These objectives must be fulfilled in order for the owner manager of an SME to introduce performance measurement in a manner that will promote the opportunity for better management and decisions without the introduction of additional time constraints. The research outlined in this thesis facilitates the identification, application and validation of 6 top-level ex-post dominant performance measures of common concern for SME owner managers. The approach that this thesis takes is to engage with owner managers directly to make the research as relevant as possible to real world environments. These owner managers were engaged using various methods including workshops, case study interviews, surveys and action research. This research was then validated with statistical significance using a combination of action research and a simulation model. This overall approach created a closed loop around experienced SME owner managers, whose primary concern is ‘Fire fighting’ with limited resources and multiple time conflicts. These dominant operational performance measures are designed to help reduce ‘Fire fighting’ through immediate information availability enhancing the opportunity for improved decision making. It will provide an opportunity for owner managers to spend less time on operational issues and to focus more on the strategic direction of the SME.

Recently, photoautotrophic cyanobacteria have gained much attention due to their ability to convert carbon dioxide using light energy into carbon metabolites, some with potential biofuel applications. This is significant due to the sustainable nature of production and the global limitation of fossil fuels in the future. Model organisms such as Synechocystis sp. PCC 6803 have been described as “cell factories” for this reason and can be genetically modified and manipulated to produce compounds such as ethanol. It was decided to construct a recombinant strain of Synechocystis 6803 expressing a Zymomonas mobilis pyruvate decarboxylase (pdc) and an alcohol dehydrogenase (adh) native to Synechocystis 6803 which is capable of autotrophic conversion of pyruvate to ethanol. In an attempt to improve ethanol yield it was hypothesised that utilising a PDC with a reported lower Km might improve flux to ethanol and have identified and utilised a novel pdc gene from Zymobacter palmae in this respect. A pET22b (+) expression vector with a strong T7 promoter was primarily used to overexpress the ZpPDC for enzymatic analysis and purification purposes for potential structural studies. The pdc gene from Zymobacter palmae was amplified and a codon optimised version was also synthesised. Two constructs were created which contained the native/codon optimised Zppdc and the adh native to Synechocystis 6803 under the control of the strong light driven pPSBAII promoter. These cassette constructs were transformed into wildtype Synechocystis 6803 and integration of the cassettes into the polyploidy chromosome at the PSBAII neutral site was confirmed via DNA sequencing and PCR screening. Strains confirmed as verified recombinants were then characterised for growth, biomass and ethanol productivity relative to UL004 (expressing the Zymomonas mobilis pdc). Unfortunately and to our surprise, no increase in ethanol production was observed in the fully segregated strains in comparison to UL004, the original production strain expressing the ZmPDC despite testing a number of different segregants and utilising repeated assays. Although there may in fact be several reasons for this it was hypothesised that the ZpPDC may be less compatible than the ZmPDC in terms of its pH optimum, functional linkage to the native ADH or just sensitivity to ethanol. Going forward, it may be worthwhile focusing on other PDC enzymes from organisms such as Gluconacetobacter diazotrophicus or Gluconobacter oxydans which have recently been reported to display even lower Km values than the PDC from Zymobacter palmae.
