Human gastrointestinal tract is colonized by intestinal microbiota. The intestinal epithelium acts as a physical barrier and represents the area where host-bacteria interaction occurs. The selective pressure in this interface determines that both parties have developed many ways to communicate. In this work we have addressed two aspects of host-bacteria interaction. As a model for metabolic adaptation of Escherichia coli adherence to enterocyte, we have studied the regulation of lldPRD operon. As a mechanism that bacteria use to interact with the host, we have analysed the secretion mechanisms of the multifunctional protein glyceraldehyde-3-phosphate dehydrogenase (GAPDH) of pathogenic and probiotic strains. lldR encodes the regulator protein of the lldPRD operon, which is responsible for the metabolism of L-lactate in E. coli. We have demonstrated that LldR plays a dual role as an activator of transcription of the operon in the presence of L-lactate, and as a repressor in absence of the inducer through the binding of LldR to two operator sequences and the formation of a DNA loop. The fact that the lldPRD operon is overexpressed in attached bacteria might be a mechanism used by bacteria to adapt to increased local concentration of L-lactate on the intestinal surface. We have determined the participation of two alternative mechanisms in bacterial GAPDH secretion. The T3SS injectisome is active in pathogenic strains growing in eukaryotic culture medium. Another mechanism is active in pathogenic and probiotic strains (E. coli Nissle 1917) growing in rich bacteriological medium. No secretion was detected in natural non-pathogenic isolates, indicating that secretion or exposure of the protein on the surface confers advantages to pathogenic and probiotic bacteria in terms of adhesion and colonization. We have also determined that GAPDH binds fibrinogen and plasminogen. Moreover, we have demonstrated that GAPDH possesses ADPrybosiltranferase activity, which might determine its participation in the modification of host proteins when translocated to the cytoplasm of eukaryotic cells. In the context of host-bacteria communication, these results provide insight into the adaptation mechanisms that allow some pathogenic and/or probiotic E. coli strains to adapt more effectively to the intestinal environment than bacteria lacking these mechanisms.
