▼ Information on the potential effects of fusariotoxins on reproductive system in ruminants are few. Therefore, the purpose of this study is to evaluate if DON and ZEA major hydroxylated metabolites, α-zearalenol (α-Zol) and β-zearalenone (β-Zol), alone or in combination, can impair bovine reproductive activity via affecting cell proliferation, estradiol (E2) and progesterone (P4) production and gene expression (CYP11A1 and CYP19A1 mRNA abundance) of primary granulosa cells (GC). GC were obtained aspirating aseptically small (1 to 5 mm) (SMGC) and large (8-22 mm) (LGGC) ovarian follicles. GC were cultured for 2 days in medium containing 10% fetal bovine serum followed by 1 or 2 days in serum free medium without (control) or with added treatments. At the end of the experiments, numbers of GC were determined using a Coulter counter, the concentration of steroid hormones was evaluated via radioimmunoassay and RNA analysis was performed. In SMGC cell proliferation was negatively affected after exposure to β-Zol at 31 µM and after exposure to α-Zol (3.1 µM) alone and combined with DON (3.3 µM). Steroidogenesis was differently affected by tested mycotoxins: DON and α-Zol had inhibitory effects while β-Zol at high concentration (31 µM) stimulated steroid production. DON (3.3 µM) and β-Zol (31 µM) increased GC CYP19A1 mRNA abundance. CYP11A1 mRNA abundance was stimulated by DON, alone and combined with α-Zol and β-Zol, whereas was inhibited by β-Zol alone. In LGGC, α-Zol alone and in combination with DON, increased cell proliferation whereas DON strongly inhibited P4 and E2 production mainly at high doses (3.3 µM). In conclusion these results demonstrated that DON and ZEA metabolites may affect in vitro bovine GC function and also provides information on the interaction between DON and ZEA metabolites.

▼ Nowadays meat quality perception is multifactorial and, especially for red meat, consumers are becoming more and more critics about sensory traits and concerned about psychological and ethical aspects, such as sustainability, animal welfare, antibiotics utilization, safety and healthiness. Address to these topics is fundamental for red meat industry, in order to counteract the decreasing consumption in western countries and to improve production efficiency to fulfill the growing demand from developing countries.
Improving all the aspects related to perceived quality will lead to meet consumer satisfaction, with positive impact on demand, and to increase production efficiency and profitability. The following trials aimed to evaluate specific dietary supplements and on farm and post farm strategies to improve animal health, strictly connected with sustainability, animal welfare and production efficiency, and meat appearance and eating quality.
8.1. Trial 1
Bovine respiratory disease (BRD) represents a concern for veal industry, as it causes economic losses, reduces animal welfare and increases antibiotics utilization. A field study to evaluate the effects of multivalent vaccination on veal calves’ health and performance was carried out. 944 healthy calves were randomly allotted into two separate barns. One group was vaccinated 7 days after arrival with quadrivalent vaccine against IBR, PI3, BRSV and BVDV Type 1 viruses plus booster administration 21 days apart (VAC; 675 calves), while the second group was not treated (CON; 269 calves). Vaccination did not affect overall mortality (P=0.75) and mortality due to BRD (P=0.28), while it reduced BRD morbidity (P=0.009). VAC group showed better average daily gain (ADG; P=0.03) and carcass weight (P<0.001). Vaccination delayed the peak of BRD by nearly two weeks (P<0.001) and represented a protective factor against BRD (OR 0.55; P=0.001). BRD increased mortality (P=0.01), lowered ADG (P<0.001) and carcass weight (P<0.001), increased the amount of discounted carcasses (P<0.001). Lung lesions and pleuritis and/or consolidations increased with BRD (P=<0.001) but were unaffected by vaccination (P=0.40 and P=0.43 respectively). In conclusion, BRD impaired veal calves’ health, growth and carcass value and vaccination reduced BRD morbidity, thus improving carcass value.
8.2. Trial 2
Selenium (Se) is involved in several biological functions and its supplementation is necessary for farm animals. Se can be provided in organic or inorganic forms, the former characterized by a higher bioavailability. Despite organic supplementation generally increased the meat Se, inconsistencies in the effects on meat quality have been reported and only one short–term supplementation study has been performed. ▼ Diarrhea is a common problem in recently weaned and young dogs. If the separation from the litter occurs at a time when the immune system is not fully competent or whether as the puppy had a lower colostrum consummation or a bad alimentation, this combination of events increases the susceptibility of the puppy or young dog to a variety of infections and gastrointestinal disturbances. Bacterial pathogens, such as enteropathogenic Escherichia coli and enterotoxigenic E. coli are also associated with enteric disease in young dogs.
Our study about bovine colostrum supplementation aimed to know its effects on recently weaned puppies at 32 days of age. The puppies were divided into 2 groups each composed of 3 and 4 animals, the control one (CT) and the treatment one (TG) randomly. The experimental groups belonged to the same litter. The involved puppies were fed with the same basal diet. A treatment was represented by administering bovine colostrum once a week through a specific oral syringe. Feed intake was registered daily. Individual body weight and average daily gain were measured weekly. We found that its principal benefit was an increased gut flora such as Lactobacillus spp. versus E.coli. in treated puppies.
The purpose of the second study was to evaluate the role of the integration of an alternative source of fiber, i.e. microcrystalline cellulose, in relation to its potential for action in the intestine, in the diet of dogs suffering from acute gastrointestinal symptoms. For the test we took into consideration twenty subjects, ten of which received a simple industrial diet, while the remaining ten were given the same industrial insoluble fiber (microcrystalline cellulose) enriched diet in such quantity that the final ration contained crude fiber of 10% dry matter. All subjects featured a symptomatic gastrointestinal clinical manifestation characterizing variable output of feces, soft feces, formed feces, the feces without definite form until liquid stools.
The third study was about chronic diarrhea as a consequence of food intolerance. The dietary management of these cases varies according to the underlying disease. Generally, the animal should receive a diet with moderate levels of fiber and fat divided in into small portions during the day. The objective of this diet was to facilitate intestinal regulation, beneficially modify the composition and metabolic activity of the intestinal microbiota and exclude antigens from the diet. In our cases the patents showed a good response to these elimination homemade diets.

▼ Effects of different diets in a mouse model of neurodegeneration
Abstract
Tauopathies are neurodegenerative disorders characterized by the accumulation of abnormal Tau protein leading to cognitive and/or motor dysfunction; several studies suggest that dietary manipulations are increasingly viewed as possible approaches to treating neurodegenerative diseases. The mouse model chosen for this study was P301L; mice expressing P301L mutant Tau mimics features of human tauopathies and provides a model for investigating the neuropathogenesis of diseases. In this study we investigate at 7 and 15 months of age the effects of different diets (high fat-protein diet and low fat-protein diet) on P301L TG and CTR mice valuating metabolic, behavioral and cognitive activities; for the metabolic activities we analyzed survival rate, body weight gain and food and water consumption of animals. For behavioral activities we valuated mnemonic, locomotor and exploratory performances of animals; for cognitive activities we investigated the cognitive impairment (identifying agglomerates of hyperphosphorylated tau, neuronal loss, astrogliosis and oxidative damage) using immunohistochemical analysis and neuronal counts. We characterized P301L TG mouse model (trial 1-3) that replicated the impairments found in patients affected by tauophaty in a way age-gender-dependent showing in female TG mice a strong cognitive impairment strictly correlated with an increase in P-Tau, in both cerebral cortex and hippocampus, as well as astrogliosis and oxidative damage. We found an improvement of pathological conditions in TG mice administrating a low fat-protein diet in a way age-gender-dependent (trial 2-3), occurred with an increased lifespan, a reduction of food and water consumption, a reduction of aggregates of P-Tau, neuronal loss, astrogliosis and oxidative damage. We found an interaction between tauophaty and consumption of high fat-protein expressed on peripheral organs as hepatic insulin resistance and fatty accumulation in the liver, which induced nonalcoholic fatty liver disease in TG mice (trial 4). The correlation between P-Tau, insulin/IGF resistance and high fat-protein diet consumption was expressed by a condition of hepatomegaly characterized by the presence of lobular inflammatory infiltrate and deposits of collagen in liver and spleen, an increase of weight and size of liver and spleen, the highest increase of median levels of triglycerides, AST and ALT in P301L TG mice fed with high-fat protein diet in a way age-gender-dependent. In summary, we demonstrated the importance of interaction between nutrition and neurodegeneration and the role that different diets can have on the onset and development of tauophaty, obtaining an improvement of pathological conditions administrating a low fat-protein diet. Calf morbidity and mortality represent major financial losses for veal and dairy producers, especially intestinal bacterial infections are the primary causes of calf mortality. Thus the preventive intervention is a recommended strategy and as a prior measures probiotics and acidifiers is an alternative to the use of antibiotics. Probiotics play a critical role in the completeness and immune response of the intestinal mucosa while the main mode of action of organic acids is through their bacteriostatic and bactericidal action. The purpose of the present trials was the evaluation of the effects both a species-specific probiotic administration in veal (1st trial) and dairy calves (2nd trial) than an acidifier supplementation in veal calves (3rd trial) on health status, gut microbial balance and growth performance. On veal calves the effects of probiotic and acidifier on slaughter performance and histological survey were further evaluated.
The aim of first the study was to evaluate the effects of the administration of a species-specific probiotic supplement to veal calves on performance and microbial parameters in standard rearing conditions. Ninety six male Friesian veal calves (49.31 ± 1.38kg of body weight and 20±5 days of life) were divided at random in two homogeneous groups of 48 animals each from the arrival in the farm and fed either a basal diet (C) or a basal diet plus 1.8x109 CFU/head/day of a probiotic supplement containing Bacillus coagulans, Lactobacillus animalis and Lactobacillus paracasei spp. paracasei in a 35:30:35 ratio (T) for a total of 180 days. Starting from 10th day from arrival and monthly until the end of the trial, individual body weight (BW) was recorded and average daily gain (ADG) was computed. At the same time, on the half of animals per group, faecal samples were collected for faecal score evaluation (FS), Lactobacilli count, Escherichia coli count and Lactobacilli/E.coli ratio, while blood samples were collected for haematological, haematochemical and immunological parameters evaluation. During the whole experimental period daily health status and therapeutic treatments were recorded for General Health Score (GHS) determination. At slaughter 10 animals per group were analysed for macroscopically injuries detection, and gut samples were collected for histological analyses. On each subject carcass weight, dressing percentage, fattening condition, carcass grade (SEUROP classification) and meat pH were recorded. During the trial were considered six times (corresponding to the each month of fattening): from time 0, referred to the 10th day from arrival, to the last considered month (time 6). BW resulted significantly higher (P≤0.05) in C calves than T at the 5th (C=226.76kg vs T=223.27kg) and 6th (C=267.14kg vs T=263.56kg) considered times while ADG was not different between groups. Faecal consistency was, significantly higher in T group at the 4th sampling (P≤0.01) as faecal Lactobacilli content (C=8.37 Log10 vs T=8.49 Log10; P≤0.05). This trend could explain the better GHS…

▼ The objectives of the thesis were: (i) to evaluate the use of automatic systems and the related sensor-based technologies (Precision Dairy Farming – PDF - systems) in three important areas of a dairy farming; (ii) to assess different methods of estimating liner compression (LC) by using a new test device and a novel artificial teat sensor, both specifically designed and built. Four studies were carried out to achieve these goals.
In the first study “Use of a proactive herd management system in a dairy farm of northern Italy: technical and economic results” the reproductive and economical performances of an AMS farm that adopted a proactive herd management system (Herd Navigator™) were analyzed. Reproductive and economic data were recorded before and one year after the installation of Herd Navigator™. Number of days open reduced from 166 to 103 days, number of days between the first and second insemination decreased from 45 to 28 days, and days for identifying an abortion were 80 % less, from 31 to 6 days. The preliminary results highlighted the usefulness of the proactive herd management system implemented for the reproduction management. A basic economic model was proposed to evaluate the potential economic benefits coming from the introduction of this technology. The model considered the benefits deriving from the reduction of reproduction problems and, consequently, of days open. Considering the effects related to the above mentioned aspects in a case study involving 60 dairy cows, a return on investment over 5 years was calculated.
In the second study “Evaluation of an electronic system for automatic calving detection on a dairy farm”, a GSM-based remote alarm system for automatic calving detection was evaluated - in terms of sensitivity and PPV- as useful and reliable tool to detect the exact moment of calving in the field. Up to date, various monitoring technologies and protocols have been proposed to predict the exact moment of the calving but none of them have been adopted widely by producers due to high costs, difficulties of execution or lack of quality staff. Visual observation of the cow’s behavior is still the most frequent. The system object of the study, showed very high sensitivity and PPV, respectively 100% and 95 %, allowing the farm staff to be present at the moment of calving in 100 % of cases when cow were monitored using this system. Cows not monitored by this system, were assisted only in 17% of cases (P<0.001). The farm staff, if present during this crucial and important moment, could assist the animal preventing possible problems for the cow and the calf. This possibility would be of great interest particularly with heifers and with problematic cows.

▼ In these studies, different approaches for the conservation of meat and fish products were deepened. The application of bioprotective cultures and the addition of organic salts were investigated, following the current trends of demanding mild technologies for food, in order to contrast the pathogenic and spoilage microorganisms without significant modifications of the sensorial quality of the products.
In the first study, the anti-listerial activity of L. sakei strains, isolated from salmon scraps, was observed evidencing the importance of the possibility to reduce the risk of food alteration in cold smoked salmon industry and to prevent the replication of L. monocytogenes using autochthonous microorganisms with a potential biopreservative action.
Moreover, in the second study, strains of Lactobacillus animalis and L. paracasei subsp. paracasei, never studied before for bioprotective purpose and characterized by limited knowledge regarding their antagonistic properties, showed promising antimicrobial activity in vitro. This action against a wide number of spoilage and pathogenic bacteria was detected both when the strains were used singularly and was more evident when they were applied as mixture (especially when L. paracasei subsp. paracasei was predominant). The antagonistic effect was due mainly to the competition for nutrients and to the production of organic acids (acetic and lactic) as combined effects.
In the third study, two multistrain mixtures (L. sakei mixture and especially L. curvatus mixture), obtained adding different strains isolated and identified from Argentine vacuum packed meat, showed a promising antimicrobial activity against a wide number of spoilage and pathogenic bacteria, highlighting that the antagonistic effect originates probably from the nutrient competitive exclusion and the possible production of metabolites such organic acids. Moreover, the addition of the two mixtures to sliced vacuum-packed beef determined a better microbial quality of meat as TVC, Enterobacteriaceae and Gram negative bacteria resulted lower if compared to control samples. In any case, if compared with in vitro results, a slighter effect was detected on beef samples; it was probably due to the different competitiveness of the cultures if applied to a complex substrate like meat and to the buffering capacity of meat.
The fourth study was focused on the application of organic acids and salts on non-prepacked hamburgers, as they are known to exert potential benefits in terms of bacterial inhibition and stabilization of sensory characteristics. The addition of mixtures containing sodium ascorbate, sodium citrate and sodium acetate was effective in microbial inhibition of non-prepacked beef hamburgers, especially when the highest acetate dose was used. An evident activity was found against Gram negative bacteria, which represent the most important microorganisms in determining the shelf-life of raw minced meat. A cross-sectional study was conducted with a total of 201 member farmers in Arsi Zone and East Shewa Zone, in Oromia, Ethiopia. The purpose of the study was to a assess hygienic practices during milking, milk collection and bacteriological quality of the milk of raw bovine bulk milk in Arsi and East Shewa Zones milk collection centers; the study targeted smallholder dairy processing activities. In Arsi Zones majority of the household heads were male while in East Shewa Zone female were the majority. Main feed used by the respondents was mineral block, oilseed cake, and barn, followed by hay, crop residue, bran, local brewer’s yeast and stover\straw and some feed were roughage and/or concentrates. Indoor feeding was common while some used only grazing system. Regarding watering, majority of the respondents, in most cases, used tap water but used river water on rare cases. The average age at first calving in Digeluna Tiyo district is 38.33 months relatively longer than 25.95 months in Tiyo district. The longest calving interval 17.17 months was recorded in Lemuna Bilbilo district, while the shortest was in Tiyo district 14.10 months. The average milk production for local dairy cattle in Tiyo district is 1.45 liters/day, which is less compared to 2.55 liters/day in Lemuna Bilbilo district. The average milk production for crossbred dairy cattle ranged from 5.07 liters/day in Digeluna Tiyo district whereas it was 11.73 liters/day in Tiyo district. Predominantly women do milking. Most of the respondents practiced milking their local and crossed cows twice per day (early in the morning and late in the afternoon) respectively. All the respondents stated that they wash their hands but most respondents wash udder of the cow also before milking. Some of the respondents indicated that they use towel for cleaning the teats before milking the cows. Large proportion of respondents uses plastic as milk container followed by metal and traditional pots. A total of 106 samples of milk were collected to assess bacteriological quality of bulk raw milk. Total of 246 bacterial isolates were obtained from milk samples taken. These included Escherichia coli 19.8%, Listeria monocytogenes 1.2%, Staphylococcus aureus 3.2%and Brucella3.3%. Based on the bacterial isolates found a total of 100 samples of milk were collected to analyzeMethicillin Resistant Staphylococcus aureus (MRSA) and Escherichia coli O157 serotype. Among the total milk samples analyzed for each pathogen, 50.5% were Staphylococcus of which Staphylococcus aureus were (5.05%), and (47.5%) Escherichia coli were also analyzed from the (49.5%) Enterobacteriaceae isolated. Out of the Staphylococcus aureus recovered, all the isolates were sensitive to Methicillin Resistant Staphylococcus aureus (MRSA) test and 27.8% of the recovered Escherichia coli were positive for E coli O157 serotype.
