The STR markers linked to the X chromosome can be used for complement the analysis of autosomal markers, especially in complex cases of kinship testing, in cases of post-morten identification and in paternity testing, when the disputed child is a girl. The aim of this work was investigate five STR X-chromosome markers (DXS10079, DXS10074, DXS10075, DXS7424 and DXS101) in the population of Alagoas, Brazil and analyze their frequencies for forensic purposes. The sample was composed of 404 unrelated individuals, 203 males and 201 females. The DNA was extracted using Chelex procedure and amplification was performed by PCR in a pentaplex system and the fragments were separated by capillary electrophoresis. For the studied STR markers, it was calculated the allele and haplotype frequencies, the observed and expected Heterozygosity values, the Hardy Weinberg equilibrium (HWE), the genetic diversity, the Mean Exclusion Chance of trios involving daughters (MECT) as well as in father/daughter duos (MECD). Also, it was calculated Power of Discrimination in males (PDM) and in females (PDF) and the Polymorphism Information Content (PIC). The forensic efficiency values demonstrate that DXS101 is a highly informative marker, followed by DXS10074, DXS10079, DXS7424 and DXS10075. The polymorphism information content ranged from 0.7470 to 0.8858. For the pentaplex evaluated, the combined values of PDM and PDF were 0, 9998947 and 0, 9999998, respectively and the combined MEC in trios involving daughters and in father/daughter duos were 0,999817 and 0,998042, respectively. No deviations from the Hardy Weinberg equilibrium were observed. We concluded that the five ChrX STRs analyzed are highly informative markers for kinship testing and constitute a powerful tool for forensic practice in our population.

Os marcadores STRs ligados ao cromossomo X podem ser utilizados para complementar as análises de marcadores autossômicos, especialmente em casos complexos de vínculo genético, em casos de identificação post-morten e em testes de paternidade, quando a criança analisada é uma menina. O objetivo desse trabalho foi investigar cinco marcadores STRs do cromossomo X (DXS10079, DXS10074, DXS10075, DXS7424 e DXS101) na população de Alagoas, Brasil, e analisar suas freqüências para propósitos forenses. A amostra foi composta de 404 indivíduos não aparentados, sendo 203 do sexo masculino e 201 do sexo feminino. O DNA foi extraído através do método Chelex-100 e a amplificação foi realizada por PCR em um sistema pentaplex, sendo os fragmentos separados por eletroferese de capilar. Several approaches using molecular biology techniques and the social sciences has been employed in order to clarify the involvement of Africans and their descendants in the history of Brazil. The African presence in America dates back to the sixteenth century when men and women were sold as slaves to serve as manpower in the colonies of Europe. It is estimated that since that time more than 15 million people have been brought forcibly to the Americas, 40% had Brazil as a destination. These Africans and their descendants started swapping genes with Europeans and Indians who inhabited the region. However, is not yet known in detail the participation of Africans in shaping history and genetics of population. Difficult access to detailed history of Africans and their descendants in Brazil is due largely to lack of documents and other historical data and geography, and only recently modern techniques of molecular biology have allowed a detailed analysis of the origin and migration of peoples. Quilombo communities are remnants of Quilombo, which were groups of runaway slaves and freedmen, located in remote and inaccessible. Quilombo is a Bantu word which means warrior camp in the forest. This study aimed to determine the polymorphism of 11 microsatellite markers on the Y chromosome in the maroon communities of the State of Alagoas. We analyzed 11 microsatellite loci of 211 men in nine maroon communities of the State of Alagoas, identified 108 haplotypes. The haplotype diversity varied from 0.7464 ± 0.0907 (Muquém) to 0.9794 ± 0.0594 (Carrasco), suggesting that there is significant founder effect in these communities. Analysis of molecular variance (AMOVA) revealed that populations have maroon marked heterogeneity, with 25.4% of the variation due to differences among the 74.6% and intrapopulation differences. The ancient African lineages, Amerindian and European is quite varied, noting that some populations are genetically closer to European and other populations of African populations.

Diversas abordagens utilizando técnicas de biologia molecular e de ciências sociais tem sido empregadas com a finalidade de esclarecer a participação dos africanos e seus descendentes na historia do Brasil. A presença do africano na América remonta ao século XVI, quando homens e mulheres foram comercializados como escravos para servirem de mão-de-obra nas colônias européias. Estima se que desde essa época mais de 15 milhões de pessoas tenham sido trazidas forçadamente para o continente americano, sendo que 40% delas tiveram o Brasil como destino. Esses africanos e seus descendentes iniciaram troca de genes com europeus e índios que já habitavam a região. The slave trade across the Atlantic brought an estimated four million Africans to Brazil since the beginning of the XVI century until mid-XIX century, thus contributing to the significant miscegenation of Brazil s population. The current genetic condition is a result of the interbreeding process between the Native American (Amerindian), European populations and the African population. Presently, despite this population s elevated heterogeneity, small isolated groups can still be found, as is the case of the Quilombolas, who are the product of the resistant movement against slavery imposed by Portuguese colonization. With the objective of conducting research of the genetic composition and the origin of paternal lineages in nine Afro-descendent communities of Alagoas, Brazil, 15 Y-SNP markers were analyzed by applying the SNaPshot (Applied Biosystems) method. Utilizing the updated nomenclature of the Y-Chromosome Phylogenetic Tree, it was possible to identify nine haplogroups of the Y-Chromosome in a total of 209 individuals. The calculated genetic diversity ranged from 0.2000 to 0.7190, thus evidencing against a standardized composition of this population. As a result of patriarchal domination of the Portuguese economic model of colonization in Brazil, haplogroup R1b1b2*-M269, of European origin, was observed in all populations at a frequency of 5.26 - 79.17%. Haplogroup F*(xK)-M213 was found at a rate of 4.17% and 36.84%, suggesting an origin of European male ancestry in these individuals. Seven populations corresponded to haplogroup E1b1a1*-M2, of Sub-Saharan African descent, which presented a frequency above Alagoas population diversity, varying from 13.3% to 90.0%. However, Q1a3a*-M3, of the Amerindian group was observed in only 2 chromosomes. Sub-clades of Haplogroup E, of African descent, including E1b1b1b1*M81, E1b1b1a1*-M78 and E1b1b1c*-M123 were considerably frequent among the Portuguese. Applying AMOVA test, occurrences of heterogeneity among Alagoas Quilombola population (FST=0.23964, P=0,00000±0,00000) were verified, had a genetic intrapopulation variation of 23.96%. The study revealed no significant genetic variances between populations from Alagoas, Rio de Janeiro, Portugal and the Quilombola populations of Jacu, Palmeira dos Negros, Paus Pretos, Poços do Lunga and Carrasco. However, Cajá dos Negros, Muquém, Filus and Povoado Cruz presented distinct genetic constituents, thereby highlighting the hypothesis that the aforementioned communities were originally places of refuge for slaves. An analysis of hierarchies pertaining to Y-SNP markers improved the basis of knowledge of Afro-descendant populations in Alagoas, and in union with historical and anthropological facts and demographics, establishes itself as an important instrument in the field of population genetics.
