Abstract: The Sun has a highly active atmosphere, featuring several events that directly affect all solar system bodies. The space weather refers to any conditions and events in the Sun, including the solar wind, impacting on the space near Earth, and the Earth's atmosphere. Disturbances in space weather can damage several fields, including aviation and aerospace, satellites, oil and gas industries, and electrical systems, leading to economic and commercial losses. Solar flares - one of the most significant events - comprehend sudden releases of radiation and particles affecting the Earth's atmosphere in a few minutes after occurrence. As such, it is imperative to create systems to forecast them. Although many ad hoc forecast approaches have been proposed in the literature in recent years, few authors have focused their research on establishing an automated design process to develop such predictors with flexibility and optimized performance. Accordingly, in this thesis, we propose an automated novel methodology for designing solar flare classifiers under Python's Scikit-learn framework. Our methodology intends to offer a comprehensive pipeline of machine learning processes relying on good practices for space weather forecasting. Overall, such methodology comprehends: (i) proper data splitting; (ii) model selection; (iii) feature selection; (iv) hyperparameter optimization; (v) analysis of classifiers' cost function; (vi) data resampling; (vii) adjustment of cut-off point of classifiers; and (viii) evaluation of validation and test data. We investigated some distinct case studies to design classifiers for forecasting >= C- and >= M-class flare events (higher than or equal to C- and M-class, respectively) up to three days ahead to validate our methodology. The designed classifiers' results agreed with the best performing forecast approaches in the literature. Accordingly, for >= C events, our models scored true skill statistics (TSS) scores of 0.69 (next 24 h), 0.70 (next 48 h), and 0.73 (next 72 h), agreeing with their high corresponding true positive rates (TPR) of 0.86 (next 24 h) and 0.89 (next 48 and 72 h). On the other hand, for >= M events, our models scored TSS = 0.53 (next 24 and 72 h) and 0.55 (next 48 h), also agreeing with their high corresponding TPRs of 0.83 (next 24 h), 0.85 (next 48 h), and 0.80 (next 72 h). Besides, results from our models' area under the curve (AUC) scores have confirmed the usefulness of the positive forecasts: AUC = 0.93 (next 24 h) and 0.94 (next 48 and 72 h), for >= C flares, and AUC = 0.84 (next 24 h) and 0.85 (next 48 and 72 h). Automated machine learning efforts can be useful to provide state-of-the-art learning methods and design processes to researchers interested in applying concepts rather than knowing the technologies in their in-depth details. Abstract: In this investigation the evaluation of some mechanical properties for aluminum matrix composite is focused. For this purpose, commercially pure Al powders with certain particle sizes are used. Particularly, commercially pure (c.p.) silicon and copper contents are used. Composites designated as Al/2Si/2Cu and Al/4Si/4Cu are constituted. Additionally, other powders with similar nature to Si are also used, i.e., SiO2, SiO2+TiO2 (commercially designated as Tritan® glass, quartz, and TiO2) powders are utilized. Different compaction pressures, sintering, and heat treatments are developed. In accordance with the results, the increase in Si and Cu contents induces to an increase in the tensile strength. Both Cu content and intermetallic Al2Cu are responsible for the improvement in the mechanical behavior of the composites examined. Although two distinct particle size ratios between Al and Si are used, i.e., 8: 1 and 30: 1, no substantial modifications or discrepancies in the tensile strength results are indicated. In the sintering process, the samples are treated at 580 °C. This is chosen due to a possible eutectic transformation above this temperature. Three different sintering times are carried out, i.e., 0.5h, 1h, and 3h. The best sintering time is that of 1h in terms of the obtained compressive strength (CS) results. Abstract: The interaction of graphene oxide materials (GO) with fetal bovine serum (FBS) in cell culture medium was evaluated in order to understand the protein corona formation and its impacts on cytotoxicity. First, it was performed a comparative study on single-layer GO (SL-GO) and multi-layer GO (ML-GO) interaction with FBS, and it was observed that: (i) the protein corona composition depends not only on GO physicochemical properties, but also on GOs behavior in the cell culture medium; (ii) the proteins adsorbed by GO materials have varied in quantity (394 on SL-GO and 290 on ML-GO), type (115 exclusive proteins on SL-GO and only 11 on ML-GO) and biological functions (SL-GO enriched proteins involved in metabolic processes and signal transduction, while ML-GO enriched proteins involved in cellular development/structure, and lipid transport/metabolic processes). Then, the cytotoxicity of SL-GO was evaluated for human kerationocytes (HaCat), in the absence or presence of a protein coating from FBS proteins or bovine serum albumin (BSA). Our results have shown that: (i) the protein corona formation has reduced, but not completely avoided the GO cytotoxicity, once it was observed a decrease in cell viability after a 72h exposure and a variation on proteins secreted by cells; (ii) the GO internalization, the proteins secreted by cells exposed to GO, and the signaling pathways/toxic mechanisms activation or inibhition have varied as function of the presence or absence, as well as, the composition of the protein corona; (iii) besides the already recognized mechanisms such as oxidative stress, cell cycle arrest and mitochondrial disfuntion, the secretome analysis has identified new mechanisms/signaling pathways such as the epithelial-mesenchimal transition and the aryl hydrocarbon receptor signaling. Thus, our data highlight the importance to perform an advanced characterization of GO materials and their interaction with cell culture medium and FBS during in vitro nanotoxicological studies, once these interactions can impact on dosimetry (the effective dose of GO delivered to cells) and cellular response as function of protein corona composition.
