Abstract: Candida infections have high prevalence; therefore, better understanding of the development mechanisms of biofilms is required for the reduction of virulence and for more appropriated clinical management. The aim of this study was to analyse the virulence of Candida albicans biofilms developed under different conditions. The role of biomaterial surfaces, salivary pellicle, and stage of biofilm development has been reported in Chapter 1. The influence of the presence of other microorganisms in virulence of C. albicans and on the interaction with the epithelium was evaluated in Chapter 2. The role of the atmosphere and of the biofilm¿s bacterial population was investigated in Chapter 3. In Chapter 1, C. albicans biofilms were developed on acrylic resin and titanium discs coated with saliva pellicle, or saliva with blood plasma pellicle. The materials¿ surfaces were analysed with regards to surface roughness and surface free energy (SFE). We evaluated the number of viable organisms, the DNA concentration, the metabolic activity, the expression of virulence factors and the architecture of the biofilms. There were no differences in the surface roughness of materials. The pellicles eliminated the differences between SFE of materials. Higher SFE was observed for discs coated with pellicles of saliva with plasma. The number of viable organisms, the DNA concentration and the metabolic activity increased throughout the biofilm maturation. Higher metabolic activity, increased expression of ALS1, ALS3 and HWP1, and more hyphae were observed in biofilms grown in the presence of saliva with plasma pellicle. There were no differences between the materials. In conclusion, the presence of saliva with plasma pellicle increases the C. albicans virulence. In chapter 2, mature biofilms (72 h) of C. albicans, or C. albicans with bacteria, were developed on the surface of acrylic resin discs. These biofilms were then analysed or used to infect a reconstituted human oral epithelium model (RHOE). Biofilms were evaluated according to the number of hyphae and the expression of virulence factors. The epithelial response was evaluated by the expression of IL-18 and Dectin1, by the activity of lactate dehydrogenase enzyme (LDH), and with regards the epithelial invasion. Increased number of hyphae and increased expression of HWP1, SAP4 and SAP6 were observed in multi-species biofilms developed on acrylic. Multi-species biofilms that have infected RHOE also had a greater number of hyphae and increased expression of ALS3, EPA1, SAP6 and HWP1. Consequently, these epithelia showed greater tissue invasion, higher LDH activity and higher IL-18 expression. We concluded, therefore, that the presence of bacteria promotes the virulence and pathogenicity of C. albicans. Abstract: The development of Candida albicans biofilms may be mediated by a differential expression of proteic ligand sites in the acquired pellicle formed onto the oral prosthesis surfaces. Therefore, the main objective of this study was to evaluate the influence of salivary and plasmatic-derived proteins on the development of C. albicans biofilms. In the first chapter the state-of-the-art was reviewed for methodologies applied to the analysis of proteins. From the knowledge of the methodologies, the research was realized aiming to characterize the protein profile of acquired pellicles (ADP) on poly(methylmetacrilate) surfaces' specimens in the presence of saliva or saliva supplemented with blood plasma. ADP composition was analyzed by mass spectrometry techniques, which results demonstrated a significant difference in the obtained proteome between the groups. Moreover, the influence of these pellicles was evaluated for the surface free energy (SFE) and expression of virulence factors by C. albicans biofilms. The findings demonstrated that the binding of plasmatic proteins in the ADP determined increases in both experimental variables. From these data, the individual effect of some identified ADP proteins was evaluated on C. albicans biofilm development. With this objective, C. albicans biofilms were developed (90 min, 24, 48 and 72 h) over mono-protein pellicles of albumin; mucin I and II; lactoferrin; fibrinogen; C3b; IgA and histatin 5. For each time point, biofilms were analyzed for metabolic activity (XTT assay), structural biofilm conformation (Confocal scanning laser microscopy) and, expression of virulence factors by means of enzymatic activity assays. Abstract: Over the past two decades, there has been substantial increase in concern about the impact of oral health-related quality of life (OHRQoL) of children and adolescents, which has been due to the fact that oral diseases significantly compromise the physical, emotional and social needs of these individuals. Thus, this research aimed to evaluate the OHRQoL and associated factors in 8- to 14-yr-old children and preadolescents, of both genders, students of public schools of Piracicaba, SP. To evaluate the oral health, the following variables were considered: presence and severity of dental caries, malocclusion, fluorosis, gingivitis according to World Health Organization, and signs and symptoms of temporomandibular disorder (TMD), using the Research Diagnostic Criteria/Temporomandibular Disorders - axis I. Self-perceived of OHRQoL was assessed using age-specific questionnaires, the Brazilian versions of the Child Perceptions Questionnaire (CPQ) for 8- to 10 yr-old children (CPQ8-10) and 11- to 14-yr-old preadolescents (CPQ11-14). Two questions of these questionnaires were also used to assess the global ratings of oral health (OH) and overall well-being (OWB). Sociodemographic characteristics (the child?s age and gender, the number of adults in the household and the mother's educational level), dental service utilization (past and current actual experience) and the child?s oral hygiene habits (tooth brushing frequency) were evaluated as part of the factors associated with OHRQoL. Symptoms of anxiety and depression were assessed using the Portuguese versions of the Revised Children's Manifest Anxiety Scale (RCMAS) and the Children's Depression Inventory (CDI), respectively. To evaluate the salivary cortisol concentrations, saliva sampling was collected 30 minutes after awakening (fasting) and at night (bedtime) to determine the diurnal decline of salivary cortisol (in ?g/dl), then, the difference between the values of the first and second samples were obtained. Salivary cortisol concentrations were determined by enzyme immunoassay analysis. The data were discussed in four studies, denominated as chapters in this work. The specific objectives of the first (Evaluating oral health-related quality of life measure for children and preadolescents with temporomandibular disorder) and second (Oral health-related quality of life in children and preadolescents with dental caries, malocclusions or temporomandibular disorders) chapters were to evaluate the psychometric properties of the questionnaires for using in children and preadolescents with signs and symptoms of TMD, to compare the OHRQoL of groups with different oral conditions (caries, malocclusion and TMD) and controls, respectively. The second study also aimed to identify the concepts associated with the responses of each clinical group to OH and OWB.
