Abstract: The pathogenicity of schistosomes is suggested to be influenced by the susceptibility of the intennediary host. Since hydrocortisone increases the susceptibility of mollusks to infection, we examined the behavior of Schistosoma mansoni in the final host, as well as the pathogenicity of parasites derived ftom mollusks. The treatment of mollusks with hydrocortisone did not alter the ability of cercariae to penetrate mice (Mus musculus) or the total number of wonns. The number of eggs in the feces was greater in mice infected with cercariae ftom hydrocortisone treated mollusks. Mice infected with cercariae ftom hydrocortisone treated mollusks showed more granulomas in the tiver, spleen and intestine than did mice infected with cercariae ftom non treated mollusks. In the pancreas, this difference was not marked while in the lungs the ftequency of granulomas was similar in both groups. The size of the granulomas in the spleen, lung and intestine was similar in both groups, whereas those in the tiver and pancreas were smaller in mice infected with cercariae ftom hydrocortisone treated mollusks. Abstract: Phenotypical variation intra and inter-populational of Schistosoma mansoni ocurred in several geographic areas. The genetic heterogenicity of the parasite contributes for phenotypic variation in the parasite-host interaction, representing an important role in the disease epidemiology. The more studied phenotypic difference, however, is the response to chemotherapy, so that is very relevant in the schistosomiasis contro!. The DNA was isolated fiom a pool of worms of the strains: BH, SJ1, SJ2, 000, LE and MAP, with the finality to observe and evaluate the genetic variability among brazilian strains, with different characteristics, and infer phylogenetic relations among them. The RAPD was processed with 5 random primers. The RAPD-PCR was realized with the fmality to observe the variability in codificators regions of genome. The two methods showed to be able to distinguish, generating molecular markers of strains. One of this markers was sequenced and showed high homology of nucleotides (99%) comparing to a human sequence and 89% with human glyceraldehyde-3-phosphate dehydrogenase, protein already relationed to the induction of resistance to the reinfection with S. mansoni and S. haematobium. The marker G400-9 showed high homology of nucleotides confirmaded for high homology of amino acid with transposase of TNI0. Other marker (CBH300-l0) was sequenced and apresented high homology with ribosomal 28 S gene Other interesting markers were observed in tolerant strain 000: 000250-10 and 000200-11, with the primers RdlO (GGGTAACGCC) and Rdll (GTGACTGCAG). Specific markers were not detected by us for MAP strain, resistant to oxamniquine, and a distinct PCR product profile with the primer Rd7 (TGCCGAGCTG). In the construction of phylogenetic trees Neighbor-joining, afier the geographic areas of strains were infered, a dendogram was obtained formed by two differents groups, one of paulistas strains and another of mineiras. When the type of intermediate host was infered the obtained dendogram revealed the formation of three groups. The LE strain kept in laboratory without retro-feed, belong to a external group very different fiom the others. The BH, SJ1 and SJ2 strains were grouped among them, possibly, in fimction of the genic flux existent among them, and the strain 000 was established in other group. This last two groups, with recent laboratory maintenance strains or with retro-feed, are more related among them, than to LE strain. We believed that the major genetic distance from this strain in relation to the others should be because the long maintenance in laboratory with the establishment of the allels. Abstract: Protease is an important biological process in different parasites, especially in enzymatic complex like 26S proteasome. The objective of the present work was to initiate the study of the 26S proteasome in the nematode Strongyloides venezuelensis using biochemical experiments and identifying some components of this multienzymatic complex. This nematode has been used as a model and tool for the studies on strongyloidiasis, a helminthiasis of high prevalence in tropical and subtropical regions. In this study, two development stages of S. venezuelensis, namely infective larvae (third-stage larvae) and parasitic stages, were used for experimental infections in Rattus norvegicus. In biochemical experiments the crude extracts of the both stages were submitted at proteolytic activities (chymotrypsin-like and trypsin-like) using fluorogenic peptides in presence and absence of proteasome inhibitors (MG132 and PSI). Moreover, the crude extracts were submitted at SDS-PAGE and western-blot methods using specifics antibodies. In molecular experiments, RNA of both stages were amplifed with specific oligonucleotides to alfa 6 and beta 6 subunits of the 20S proteasome (proteolytic core) and ATPase 3 and S5a of the regulatory particle (19S). The genomic DNA was amplifed to all subunits, excepting of S5a subunit. Results show the presence of the 26S proteasome in proteolytic activities from S. venezulensis of the chymotrypsin-like and trypsin-like and the presence of the labeled bands by the antibodies, as well as molecular analysis of the subunits.
