▼ Abstract: We conducted cytogenetic, phylogenetic and phylogeographical studies to investigate aspects of chromosomal and molecular evolution in the Neotropical tree frogs of the genus Phyllomedusa genus (Hylidae, Phyllomedusinae), which have been the subject of extensive taxonomical debate. In the first chapter, the study focuses on the morphological variation found in populations attributed to P. hypochondrialis, and examines whether this variation is interspecific or inter-population. The results of this analysis support the need for a thorough revision of the phenotypic features used to discriminate P. azurea and P. hypochondrialis, given the considerable overlap found in these characters when more populations are samples. We also report inter- and intra-population variability in the NOR of P. hypochondrialis. In the second chapter, we use phylogeographic approaches to evaluate the source of the karyotype variation found within P. hypochondrialis, provide a better delimitation of the Brazilian populations of P. azurea, and estimate the genetic diversity within P. nordestina. These three species are closely related, but are associated with distinct South American morphoclimatic domains. Our results reveal a deep genetic structuring in the P. hypochondrialis populations, based four well-supported clades with a low migration signal among haplogroups. The evolutionary history of this species has been influenced by the interaction between the geomorphological changes occurring in the Miocene-Pliocene that have persisted through the episodes of climatic fluctuation that occurred during the Pleistocene. The populations of the Cerrado domain appear to have colonized this region during the Pleistocene period, when successive cycles of forest expansion and retraction may have facilitated expansion into this biome via gallery forests. We were able to recognize two cryptic lineages within the P. nordestina populations, differentiated by the São Francisco River in northeastern of Brazil. Estimates of coalescence time indicated a process of divergence between two geographic clades of P. nordestina during the Pliocene-Pleistocene transition, which is temporally congruent with historical modifications to the course of the São Francisco River. This geographic barrier has had a primary role in the divergence of lineages, supported by the high differentiation between populations from opposite margins of the river and the absence of gene flow between haploclades. A strong signature of genetic decline marks the evolutionary history of the Brazilians populations of P. azurea. While more extensive surveys are needed, Brazilian populations of this species appear to be restricted to the Pantanal biome. ▼ Abstract: The anurans of the family Leptodactylidae are distributed in three subfamilies ¿Leiuperinae, Leptodactylinae and Paratelmatobiinae. The spermatozoa of 23 leptodactylid species were analyzed under an electron microscope in order to provide evidence for the better understanding of a number of phylogenetic uncertainties. In the case of the leiuperine genus Pseudopaludicola, the sperm of the representative of the P. pusilla group and those of four clades identified in the molecular phylogeny, which correspond to well-defined chromosomal groups with 2n=22, 20, 18, and 16, were compared. The analysis of the different Pseudopaludicola species found spermatozoa with heads of similar shape, which contain an acrosomal vesicle covering the subacrosomal cone. However, the tails present marked differences. In the species with 2n=22, that is, Pseudopaludicola sp. 1 (P. pusilla group), P. saltica, P. falcipes and P. mineira the tails present accessory fibers (juxtaxonemal fiber, undulating membrane and axial fiber). Fibers of this type are also observed in P. ternetzi and P. ameghini (representatives of the 2n=20 clade), although the axial fiber is more developed. In the specimens of the 2n = 18 clade (P. canga, P. facureae, P. giarettai, P. atragula and Pseudopaludicola sp. 2) and the 2n=16 clade (P. mystacalis), the tail has a thick undulating membrane, characterized by a paraxonemal rod, which is shorter than the undulating membrane of the tail of Pseudopaludicola (2n=22 and 20). The ultrastructural morphological differences observed in the tails of the spermatozoa indicates that these traits are phylogenetically informative, given their partial correspondence to the four clades identified in the molecular studies, and provide evidence of a progressive simplification of the spermatozoa during the evolution of the genus Pseudopaludicola. The comparative analysis of the leiuperine genera indicates that in Physalaemus (P. albifrons, P. centralis, P. cicada and P. cuvieri), the tail of the spermatozoon also shows accessory fibers, such as the juxtaxonemal and the undulating membrane, although in this genus, the membrane is longer than that observed in Pseudopaludicola. In addition, there is no axial fiber on the tail of the spermatozoon in Physalaemus. In the case of Engystomops, sister group of Physalaemus, the species E. petersi, E. freibergi, and E. puyango the sperm tail presents all three types of fiber (juxtaxonemal, undulating membrane and axial fiber). The tail of these two genera also differ each other for the shorter undulating membrane in Engystomops, corroborating the revalidation of the genus Engystomops. Additionaly, the tail of E.puyango differs from E. petersi and E. freibergi, Duovox clade, and it is similar to E. pustulosus, Edentulus clade, indicating the need of more studies in this genus. ▼ Abstract: The genomes of eukaryotic organisms are rich in repetitive DNA sequences, which can be dispersed or arrayed in tandem. The tandem repeat sequences include the satellite DNA, the ribosomal DNA, and paralogous copies. Satellite DNA is the main component of constitutive heterochromatin, while the ribosomal genes encode the rRNAs that make up the ribosomes; they are divided into two families, the 45S and 5S rRNA. Different types of 5S rDNA have been identified in a single organism, proving that there is diversity in this type of DNA sequence. In the anuran Physalaemus cuvieri, a satellite DNA family called PcP190 has been identified, which is thought to have derived from the 5S rDNA of an ancestor species. In different populations of P. cuvieri that were previously studied with conventional cytogenetic tools, an accentuated interpopulational variation among chromosomes harboring the NOR was observed, while in other species of the P. cuvieri group, interspecific traits were found. However, the karyotypes in these species are very similar. The aim of this thesis was to expand the cytogenetic studies on P. cuvieri species that needed further chromosomal description, such as P. kroyeri and P. cicada. Another objective was to analyze the PcP190 satellite DNA at the chromosomal and molecular level, as well as to study the molecular organization and the diversity of 5S rDNA sequences in the genomes of the Physalaemus species and other species of related genera. We present three chapters as a result of this research: (i) "Long-time evolution and highly dynamic satellite DNA in frogs," which demonstrates that the PcP190 satellite DNA is widely conserved and was recognized in representatives of two anurans families, Leptodactylidae and Hylodidae. Moreover, it also demonstrates that these sequences are highly dynamic within the chromosomes of the P. cuvieri species group. (ii) "5S rDNA in Leiuperinae (Anura): new insights on its evolution." The results show that in the genomes of these anurans there is wider diversity among this multigenic family than previously assumed and that these 5S rDNA sequences have remained conserved since the evolutionary divergence of Actinopterygii and Sarcopterygii. (iii) "Karyotype and repetitive DNA mapping of the Physalaemus kroyeri and P. cicada (Anura Leptodactylidae)," which demonstrates that the 5p chromosomal band of P. kroyeri is a good chromosomal marker for species from the P. cuvieri group, indicating that it is a chromosomal synapomorphy. Conversely, the absence of this 5p band in P. cicada does not provide evidence for the inclusion of this species in the P. cuvieri group or any other species group

▼ Abstract: The taxonomic position, phylogenetic relationships, synonymization of species considered to be distinct, existence of subspecies, and the proposals of intrageneric groups of the genera Pseudis and Lysapsus have always been a matter of discussion among herpetologists. The genera Pseudis and Lysapsus have already been included in the family Pseudidae and also in the subfamily Hylidae. Recently, these two genera have been allocated to the subfamily Hylinae as distinct genera. Pseudis paradoxa, P. minuta and Lysapsus limellus, cytogenetically analyzed by other investigators, show a chromosome number of 2n=24, and Pseudis sp. (aff. minuta), now cytogenetically confirmed as the recently described P. cardosoi, has 2n=28 chromosomes, including four additional pairs of telocentric chromosomes. In the present study, we analyzed the pattem of heterochromatin distribution and the number and location of the nucleolar organizer region (NOR) by conventional chromosome staining in species of the genera Pseudis and Lysapsus, except for L. laevis, in order to add cytogenetic traits to the systematics and to the study of the phylogeny of the two genera, in addition to providing evidence for the understanding of the processes involved in the chromosome evolution of these groups. Cytogenetic analysis revealed that the complement of 2n=24 chromosomes is a plesiomorphic condition both in the genus Pseudis and in the genus Lysapsus, and confirmed the hypothesis that the origin of the 2n=28 karyotype is the same as that of the 2n=24 karyotype of P. minuta since the heterochromatic marker bands in the two karyotypes were not detected in any of the species analyzed in the present study. Analysis of the chromosome morphology and the pattem of heterochromatin distribution permitted the inter- and intrageneric separation of the two genera, except for the subspecies Pseudis paradoxa paradoxa and P. p. platensis which presented common cytogenetic data. In addition to permitting the separation of Lysapsus and Pseudis species, the differences detected in the pattem of heterochromatin distribution also suggested the reassessment of the taxonomic status of the subspecies L. limellus limellus and L. I. bolivianus, especially of the L. I. bolivianus population from Guajará-Mirim, which differed in the morphology and banding pattem of chromosomes 7 and 8 in relation to the other populations of the same subspecies. The presence of the NOR on the long arms of the chromosomes of pair 7 was a plesiomorphic trait both in the genus Pseudis and in the genus Lysapsus, and the position the NOR occupies on the long arm is an important cytogenetic characteristic for the separation of Pseudis species. Chromosome morphometry, banding pattern and NOR position on chromosomes 7 also permitted the detection of heteromorphic sex chromosomes in the ZZlZW system of P. tocantins, with evidence that mechanisms of inversion and heterochromatinization caused the differentiation of the Z and W chromosomes

▼ Abstract: The proximity between Paratelmatobius and Scythrophrys has been suggested by several authors, even though these genera are currently classified in two subfamilies of Leptodactylidae. In the present work, the living species of Paratelmatobius, P. cardoso i and P. poecilogaster, and of the monotypic genus Scythrophrys, S. sawayae, were studied cytogenetically. Some restriction enzyme sites of ribosomal gene 28S were also analyzed. The karyotypic study indicated the presence of a new species of Parate/matobiu.'i;, Parate/matobius sp. (aff cardoso i), and two groups of Scythrophrys, a finding supported by some morphological and bioacoustical analyses carried out by others. The diploid complement of all these species and that already described for P. lutzii consists of 24 chromosomes, several of which were similar in morphology, especially those of P. poeci/ogaster, P. /utzii and group II of Scythrophrys. A diploid number of 2n=24 is rare among leptodactylids and the karyotypes of species with this chromosome number share fewer similarities with Paratelmatobius e Scythrophrys. Analysis of the nucleolus organizer regions (NORs) suggested that the presence of an NOR in a short metacentric chromosome, as seen in P. poecilogaster, Parate/matobius sp. (aff cardosoi) and group II of Scythrophrys, is a plesiomorphic state relative to the NOR location in these genera. The C-banding suggested possible sinapomorphies between P. cardosoi and Parate/matobius sp. (aff. cardoso i), and between the groups of Scythrophrys. This technique also revealed non-centromeric heterochromatin in the short arm of chromosome 1 in all the karyotypes, a1though this region showed interspecific variation in size. Molecular analysis of ribossomal DNA revealed a Pvu II site exclusive to populations of group I of Scythrophrys, which agrees with the proposal for two taxonomic groups in this genus. The species of Paratelmatobius and Scythrophrys, as well as Cycloramphus izeckshoni and Hylodes asper, failed to show a Bst ElI site considered by others to be indicative of possible synapomorphy between the subfamilies Leptodactylinae and Telmatobiinae. The restriction maps for Paratelmatobius and Scythrophrys showed interspecific variation in the size of some rDNA fragments, indicating that such regions may be useful in future molecular studies

▼ Abstract: The Terrarana is the largest taxon of the Anura order, comprising 900 species. Recently this anuran group has undergone a series of taxonomical rearrangements. At first, these species were removed from the Leptodactylidae family and transferred to the synonymy of Brachycephalidae, and lately they were placed into a new taxon named Terrarana. Currently, the Terrarana species are distributed in the families, Eleutherodactylidae, Craugastoridae, Strabomantidae and Brachycephalidae. Despite being intensively studied, many aspects of their inter- and intrageneric relationships remain unclear. The South American Strabomantidae and Brachycephalidae are the least known clades regarding molecular phylogeny. In the present work, Brazilian Terrarana species of Pristimantis, Ischnocnema, Barycholos, Haddadus and Brachycephalus were studied by means of cytogenetical and sperm ultrastructural characteristics. Metaphases obtained from suspensions of intestinal epithelium were stained with Giemsa or submitted to silver staining (Ag- NOR) and fluorescence in situ hybridization (FISH) techniques, in order to detect the nucleolus organizing region, and to C-banding for heterochromatin localization. Sperm ultrastructure was analyzed by transmission electron microscopy. The studied species were P. fenestratus (Borba and Manaus at Reserva Florestal Adolfo Ducke, Amazonas, and Rio Branco, Acre), P. dundeei (Chapada dos Guimarães, Rondonópolis, and Aripuanã, Mato Grosso), P. crepitans (Chapada dos Guimarães and São Vicente, Cuiabá, Mato Grosso), Ischnocnema paulodutrai (Ilhéus, Bahia), I. juipoca (Atibaia and Campos do Jordão, São Paulo), I. guentheri and I. parva (Mogi das Cruzes, São Paulo), B. ephippium (Mogi das Cruzes and Atibaia, São Paulo), Barycholos ternetzi (Uberlândia, Minas Gerais, and Porto Nacional, Tocantins), and Haddadus binotatus (Mogi das Cruzes and Ilhabela, São Paulo). The cytogenetical analyses allowed chromosomal characterization of the species P. fenestratus 2n=34 chromosomes, P. crepitans 2n=22, P. dundeei 2n=28 and I. paulodutrai 2n=30, which have not been previously analyzed in phylogenetical studies, in addition to B. ternetzi and I. juipoca 2n=22. The data revealed differences in C-banding and Ag-NOR, which indicated the existence of not yet described Pristimantis species in the Manaus, Rio Branco, and Aripuanã localities, and of Barycholos (from Tocantins) uncorrectly identified as P. fenestratus, P. dundeei and B. ternetzi. The cytogenetical analysis also indicates that the low chromosome number in P. crepitans is unique within this genus and suggests that this species is not closely related to its congeneric species. The karyotypical similarities between I. paulodutrai and P. dundeei indicate that these species could be close relatives. ▼ Abstract: Even though many amphibian species are among the most endangered animals, the knowledge about their genetic variability and population genetic structure is still limited. In the present work, natural populations of the barker frog Physalaemus cuvieri
were analyzed for genetic variability and differentiation using microsatellites, considering its wide geographic distribution, interpopulational morphological variation and cytogenetic intra- and interpopulational variation. Analyzes comprised specimens from ten P. cuvieri populations sampled in the following regions of Brazil: São Pedro da Água Branca (MA), Urbano Santo (MA), Porto Nacional (TO), Uberlândia (MG), Vitória Brasil (SP), Palestina (SP), Nova Itapirema (SP), Chapada dos Guimarães (MT), Vitória da Conquista (BA) and Passo Fundo (RS). Ten microsatellite loci were isolated for P. cuvieri from a library enriched in (CA)8 and (GT)8 repeats. In 160 P. cuvieri individuals, the average number of alleles per locus was 5.7 and the expected heterozygosity ranged from 0.30 to 0.85. Although most of the genetic variation was found within populations, the overall FST value indicated high genetic differentiation among the populations. The low gene flow among populations suggested low dispersion capability within the geographic scale pattern used in this work, which is compatible with the anuran philopatric behavior. In addition, there was no correlation between geographic and genetic distances of these populations, suggesting fidelity of P. cuvieri populations to their spawning place. These results indicated that the geographic distance alone does not explain the genetic variability observed among these populations. The data also indicated that the populations from São Pedro da Água Branca (MA) and Nova Itapirema (SP), respectively in the Northeast and Southeast regions of Brazil, have likely gone through a recent population bottleneck effect. Destruction of natural habitats leading to population fragmentation could be a major cause of this suggested bottleneck effect. Of the ten microsatellite loci developed for P. cuvieri, nine were used to investigate cross-amplification and suitability for population genetic structure studies in two other species of the cuvieri group closely related to P. cuvieri. These species are: P. ephippifer (15 specimens) and P. albonotatus (11 specimens). These two populations
remained isolated from all populations of P. cuvieri, demonstrating its high genetic differentiation, pointing that these two species are distinct from P. cuvieri. The Crateús (CE) population was grouped with Urbano Santos (MA) population (both P. cuvieri). This result corroborated previous cytogenetic results. The Porto Nacional (TO) and Passo Fundo (RS) populations were divided in two sets of individuals each one. ▼ Abstract: The ultrastructure of bivalve spermatozoa can be species-specific and often provide valuable taxonomic traits for systematic reviews and phylogenetic reconstructions. Transmission and scanning electron microscopy was an efficient tool to differentiate the spermatozoan structure of species sharing many conchological characters, which could lead to equivocal identification. We analyzed sperm ultrastructural features that were useful for taxonomic issues with regard to Isognomon bicolor and Isognomon alatus (Isognomonidae); Macoma constricta, Macoma biota and Tellina lineata (Tellinidae); Donax hanleyanus and Donax gemmula (Donacidae). The data revealed that the bivalve species studied here, except T. lineata, produce primitive-type spermatozoa, typical of marine invertebrates in which eggs are fertilized in the surrounding water. We, also, compared the ultrastructure of spermatozoa from the bivalves Anomalocardia brasiliana and Tivela mactroides (Veneridae). The spermatozoa of both species contain a curved nucleus with a short cone-shaped acrosome. There were six mitochondria and glycogen clusters in the middle piece of the T. mactroides sperm cell. Possibly, the glycogen clusters and the higher mitochondria number correspond to an adaptive advantage in turbulent waters. Increasing the sperm life expectance also increases the odds of finding the eggs and accomplishing fertilization. Oysters of the genus Crassostrea and mussels of the genus Mytella, commercially important in Brazil, were also investigated in this thesis. The sperm ultrastructure of the specimens described herein and molecular studies, confirm the presence of Crassostrea brasiliana (Ostreidae) in Cananéia. Spermatozoa of mussel specimens of the genus Mytella, from two populations living in distinct habitats (muddy-sand sediment and rocky shores), were examined by transmission electron microscopy. The spermatozoa of all specimens shared the same ultrastructural pattern suggesting that the analyzed specimens, which live in distinct habitats, indeed belong to a same species, which was identified as M. charruana (Mytilidae) based on conchological analysis. The presence of an axial rod in sperm cells upholds the inclusion of M. charruana in the subfamily Mytilinae. In conclusion the electron microscopy was an efficient method to solve some particular taxonomic issues. The present spermatozoan ultrastructural studies have provided new and welcome data about the families Isognomonidae, Veneridae, Tellinidae, Donacidae, Ostreidae and Mytilidae. The results showed in this thesis could be valuable in future phylogenetic analyses.

▼ Abstract: In the present study we characterized and compared cytogenetically populations of seven closely related Dendropsophus (Hylinae), a group of 30 chromosomes species, that show similar external morphology: D. nanus (Botucatu, SP; Bodoquena, MS; Telêmaco Borba, PR; Bacabal, MA), D. walfordi (between Rio Madeira and Pacaás, RO), D. sanborni (Torres, RS), D. jimi (Uberlândia, MG), D. rubicundulus (Uberlândia, MG), D. elianeae (Botucatu, SP; Nova Itapirema, SP; Bodoquena, MS) e D. minutus (Vitória Brasil, SP; Bodoquena, MG). The metaphases were obtained from intestinal epithelium and testis cell suspensions. The slides were stained with 10% Giemsa solution or submitted to the C-band (King, 1980), silver staining (Ag-NOR) and in situ fluorescence hybridization techniques. All species had 2n=30 chromosomes, except some individuals of D. nanus, which showed up to three little B-chromosomes, all telocentrics. These chromosomes were univalent in meiosis. All the species presented a little amount of heterochromatin, located only in the centromeric region. However, some individuals of D. nanus from Telêmaco Borba (PR) and of D. rubicundulus from Uberlândia (MG) had a little block of interstitial heterochromatin in the pair 3 (D. nanus) and in the pair 12 (D. rubicundulus), characterizing an intra-populational variation. The nucleolar organizing region (NOR) was detected in only one chromosomal pair in all species. An individual of D. sanborni had one heteromorphism due to a duplication of the NOR in one of the homologues of the pair 12. Dendropsophus nanus from Telêmaco Borba (PR), Bacabal (MA) and Serra da Bodoquena (MS) and of D. walfordi collected between the Madeira and Pacaás rivers (RO) showed identical karyotypes, with 2n=30 and NF=52. A specimen of D. nanus from Serra da Bodoquena presented NF=51 due to an heteromorphism in the pair 6, in which one of the homologues is metacentric and the other is telocentric, probably because of a pericentric inversion. In all the specimens, the NOR was located in the metacentric pair 13. Dendropsophus sanborni from Botucatu (SP) and Torres (RS), and D. jimi from Uberlândia (MG) also had identical karyotypes, with 2n=30 e NF=50, differing from the other two species by the presence of five telocentrics instead of four, with the NOR located in the telocentric pair 12. We did not observe citogenetic differences between the populations of D. nanus and of D. sanborni, neither differences among these populations and others already described in the literature. The B-chromosomes found in two populations of D. nanus did not showed heterochromatin neither ribosomal genes. Dendropsophus elianeae had three pairs of telocentric and the NOR in the pair 11. In contrast, D. rubicundulus presented only two telocentrics and the NOR in the pair 5. ▼ Abstract: The genus Physalaemus, belonging to the family Leptodactylidae, shows extensive polymorphism and an abundance of cryptic species. At least four species are currently recognized in this genus. In this work, we investigated some aspects of the cytogenetics and spermiogenesis of several species in this genus, with particular emphasis on P. fuscomaculatus and P. biligonigerus. These two species are difficult to distinguish, mainly because of their considerable morphological similarities. A further problem is that the specific name fuscomaculatus has been applied to several populations of biligonigerus. AlI the specimens of P. fuscomaculatus, P. biligonigerus and P. aff. biligonigerus analyzed had 2n=22 chromosomes. All of the chromosomes had the same morphology, with the exception of pair 3 of P. fuscomaculatus, which was submetacentric, but metacentric in the other two species,. The short arm of chromosome 3 contained a block of heterochromatin which was Cband positive and was slightly longer in P. biligonigerus and P. aff. biligonigerus than in P. fuscomaculatus. The divergenge in morphology of chromosome 3 can be explained by the addition or delection of heterochromatin. The three species also showed a large block of pericentromeric heterochromatin in the pair 8, a small interstitial C-band, on the long arm of pairs 5 and 7, as well as a uni que ribossomal cistron, on the long arm of chromosome 9, coincident with the secondary constriction. P. aff. biligonigerus was the only species to have a large block of pericentromeric heterochromatin in the short arm of chromosome 9 and another one in the telomeric band of the long arme The nuclear basophilia during spermiogenesis in P. fuscomaculatus and P. biligonigerus (biligonigerus group), P. riograndensis, P. centralis, P. lisei, P. gracilis and P. cuvieri (cuvieri group) and P. petersi (pustulosus group) showed different responses with toluidine blue (TB) pH 4.0 and alkaline fast-green (AFG) after deamination. The spermatozoa of P.fuscomaculatus, P. biligonigerus, P. cuvieri and P. gracilis stained violet, whereas those of the other species showed different responses to TB. Except in P. gracilis, the spermatozoa of all species were positive to AFG, even after deamination. These results show that the DNA in the spermatozoa ofthese species is differently associated with proteins. This is the first description of such intrageneric variability in the Leptodactylidae. An ultrastructural analysis of the spermatozoa of P. fuscomaculatus, P. biligonigerus, P. gracilis and Pseudopaludicola falcipes, a genus considered to be a sister-group of Physalaemus, showed that the three species of

▼ Abstract: In the present study, evaluations of the susceptibility to organophosphorus, carbamate and pirethroid insecticides of Aedes aegypti and Culex quinquefasciatus were made by means of bioassays. This allowed monitoring the resistance in populations of these species in
Campinas, SP, Campo Grande, MS and Cuiabá, MT. Moreover, a cytological evaluation of the polytene chromosomes from Malpighi tubules was undertaken in larvae, pupae and adults from A. aegypti populations:
Campinas, SP (Aea-campus, Aea-Lab, Aea-Unicamp) and Campo Grande, MS (AeaUFMS, Aea-cemetery) - Brazil, for the populations Aea-INS-Bogota and Aea-Palmira, VA - Colombia, and for the Rockefeller susceptible standard strain. In the same way the polytene chromosomes from C. quinquefasciatus populations were appraised:
Campinas, SP (UNI-R1, UNI-R2), Campo Grande, MS (UFMS-R) and Cuiabá, MT (Cuiabá-R) - Brazil, Cxq-Palmira, VA and Cxq-INS-Bogota - Colombia, and in the IAL susceptible standard strain. Starting with larvae reared in low temperatures (18-24°C), well fed and with little stress due to overcrowding, cytological studies were made using the conventional technique of orcein staining. The best results for obtaining and reproducing the polytene chromosomes of A. aegypti and C. quinquefasciatus were reached with Malpighi tubules of recently emerged big female pupae (O to 4 h / 18-24°C). Adequate slides were also obtained with female adults and 4th advanced instar larvae. It can be considered that the difficulties registered in the literature for these species were avoided in 80% for the strains evaluated in this work. Polytene chromosome maps from A. aegypti and C. quinquefasciatus were also elaborated. This allowed identifying chromosomallandmarks for use in the differentiation of the chromosomes and cytological characterization of species. In C. quinquefasciatus the centromere regions and the nucleolar organizing region (NOR) are the main landmarks to identify the chromosomes. In this species, other groups of bands and puffs are also characteristic landmarks. The degree of politenization varied among the strains of C. quinquefasciatus, being smaller in the standard lineage. Although the pattern of bands in C. quinquefasciatus provides greater resolution than that of A. aegypti, in the two species identification of characteristic and specific chromosome landmarks was possible. In the maps of the resistant strains of C. quinquefasciatus, chromosomal marker of the amplified gene for esterase was associated with resistance to the organophosphorus insecticides and observed in section 31 of chromosome arm 2L. The monitoring of medically important mosquito populations appears to be viable by means of bioassay as well as cytological evaluations, showing changes in the susceptibility to the insecticides and demonstrating the need of surveillance of resistance in the appraised populations. It is suggested that in this way, population studies and physical mapping of genes in species of Culicinae, can be undertaken based on polytene chromosomes

▼ Abstract: Polymorphic populations of Hyla semiguttata and related species in the Hyla pulchella group were analyzed cytogenetically and by comparing the sequences of the cytochrome b gene and the control region of mitochondrial DNA. All H semiguttata populations showed polymorphism in color pattern, body size and acoustics. The cytogenetic analysis showed a karyotype with 2n = 24 chromosomes and a very conserved chromosomal morphology in all of the species. H semiguttata from São Francisco
de Paula and Cambará do Sul, RS and Piraquara, PR, H marginata, and Hyla sp. (aff. semiguttata) from Misiones, Argentina had the same C-banding pattern. The nucleolar organizer region (NOR) located in the long arm of pair 10 in H marginata and in the short arm of pair 1 in the other specie the on1y character that differentiated these species cytogenetically. These results do not support the suggestion of some authors for the synonymization of H semiguttata and H joaquini with H marginata. The phylogenetic analysis based on 361 bp of the cytochromo b gene and 744 bp of the control region of mitochonsdrial DNA showed that the H semiguttata population at São Francisco
de Paula was very close to H joaquini, in contrast to the phylogenetic conclusions based on morphological and biological characters, which indicated a greater proximity with the population at Cambará do Sul. The populations of H semiguttata at Palmeira and Piraquara were very close to each other and distant form the other populations. Although H marginata differs morphologically and biologically from Hyla aff. polytaenia, H bishoffi and H guentheri, our analysis showed them to be very closely related. The presence of Hyla sp. (aff. polytaenia) (H polytaenia group) in this clado indicates that the H pulchella group may be paraphyletic. Natural hybridization between H bischoffi and Hyla sp. n. demonstrates that cladogenic events may occurr between sympatric taxa and that the similar vocalization patterns of the two species facilitates this process. As already suggested by other many cladogenic events have been involved in the speciation of neotropical hylids, which makes it difficult a study polymorphic populations, especially when the polymorphism is related to the pattern of coloration

▼ Abstract: The Dendrobatidae is a large neotropical anuran family for which the evolutionary relationships are still unclear. Affinities with frogs belonging to the Ranoidea neobatrachian lineage, specifically with ranid frogs related to the genus Petropedetes, have been proposed. Alternatively, relationships with hylodine leptodactylids (Bufonoidea) have also been considered. The aim of this work was to examine the relationships between the Hylodinae and Dendrobatidae based on cytogenetic and ultrastructural studies. The karyotypes of foUf dendrobatid species of the genus Epipedobates (E. jlavopictus, E. trivittatus, E. femoralis and E. hahneli), as well as seven Hylodinae species (Hylodes phyllodes, H asper, Crossodactylus sp. n., C. cf. caramaschi, Megaelosia massarti, M boticariana and M lutzae), were analyzed using conventional Giemsa staining, C-banding and Ag-NOR techniques. An ultrastructural analysis of spermatozoa was done for the four species of Epipedobates (listed above) and for Hylodes phyllodes, Crossodactylus sp. n. and Megaelosia massarti. The cytogenetic results showed a conserved chromosome number (2n = 24) in Epipedobates, although there was considerable variation in chromosome morphology, in the amount and distribution of heterochromatin and in the location of the NORs. Within the Hylodinae, the diploid number was the same (2n = 26) in Hylodes and Crossodactylus but varied among the three species of Megaelosia (2n = 28, 30 and 32). Crossodactylus had the most conserved karyotype in terms of chromosome morphology and C-banding pattern, whereas there were differences between the two species of Hylodes. The NOR location did not vary within each genus. On the other hand, all of these parameters varied among the three species of Megaelosia. Ultrastructural analysis showed that Epipedobates femoralis was peculiar in that it possessed biflagellate spermatozoa. The presence of mitochondria within the undulating membrane of the other species also differentiated them from E. femoralis. Despite the great similarity among the spermatozoa of the three genera of Hylodinae, Crossodactylus sp. n. was most similar to other leptodactylids and to most of the remaining bufonoids. In contrast, M massarti and H phyllodes shared a similar shape in their axial and juxtaxonemal fibers. These results elucidated some inter- and intrageneric aspects within each group (Hylodinae and Dendrobatidae) but did not provide much information on the relationships between the two groups. Even with the use of banding techniques, the karyotypical analysis provided no unambiguous homeologies indicative of the possible relationships between the Hylodinae and Dendrobatidae. On the other hand, the karyotypic peculiarities of E. femoralis suggested a unique position within the genus. ▼ Abstract: The family Dendrobatidae consists of approximately 230 species that are grouped in nine genera. Colostethus is the largest genus with 128 nominal species. Some Colostethus species groups have been proposed and, although used, have been criticized because of the lack of apomorphic characters to support the species groupings. In this work, we examined the cytogenetics and sperm ultrastructure of Colostethus as part of an investigation into the taxonomic and intrageneric relationships of this genus. The karyotypes of Colostethus brunneus, C. marchesianus, C. caeruleodactylus, Colostethus sp. 1 (aff. marchesianus), Colostethus sp. 2 (aff. marchesianus), Colostethus sp. (aff. trilineatus) and Colostethus sp.
were analyzed using conventional Giemsa staining, C-banding and Ag-NOR techniques. Fluorescence in situ hybridization (FISH) with an rDNA probe was also used when necessary. The ultrastructure of spermatozoa from all species, except for Colostethus caeruleodactylus and Colostethus sp. 2 (aff. marchesianus) and including C. nidicola and C. stepheni, were also examined. Cytogenetic analysis showed that C. brunneus had 24 chromosomes whereas the other species had 2n = 22 chromosomes. Variations in the Cbanding patterns of the karyotypes were seen in all of the species. However, the Colostethus species with 2n = 22 showed a faintly stained band on the long arm of pair 7, which probably represent a homeology among these species. The nucleolus organizer region (NOR) was located on the long arm of pair 4 of C. marchesianus, Colostethus sp. 1
(aff. marchesianus) and C. caeruleodactylus and on the short arm of the same pair of the Colostethus sp. (aff. trilineatus). In C. brunneus, the NOR was located on the long arm of pair 3, which was morphologically similar to pair 4 in the other species. In Colostethus sp. 2 (aff. marchesianus) and Colostethus sp. the NOR was located on different chromosomal pairs. Additional NOR sites and size heteromorphism were also seen in some species, and were confirmed by FISH with an rDNA probe. However, in one specimen of Colostethus sp. 2 (aff. marchesianus), FISH revealed an additional site that was never seen by silverstaining. The difference in chromosomal number, heterochromatin distribution and NOR localization, made it possible to distinguish among the species. Ultrastructural analysis of the spermatozoa showed the presence of biflagellated spermatozoa in all species, except for C. stepheni. The acrosomal complex structure and flagellar apparatus of the spermatozoa of the species examined resembled those of bufonoids. In contrast to the flagella of the other species, the single flagellum of C. stepheni spermatozoa contained mitochondria in the undulating membrane and a very thickened axial sheath. The results of the cytogenetic analysis were useful for distinguishing among these morphologically similar species.
