Abstract: MAGP-1 is a glycoprotein present in the elastic fibers, functioning as part of the microfibrils components. Studies have shown that MAGP-1 expression is critical for vascular development and is associated with decreased interaction between integrins and the extracellular matrix in zebra fish. Also MAGP-1 interacts with von Willebrand factor and the active form of TGF-? and BMP. In mice lacking MAGP-1, thrombus formation is delayed, increasing the occlusion time of carotid artery despite normal blood coagulation in vitro. MAGP-containing microfibrils may play a role in hemostasis and thrombosis. In this work, we evaluated the function of MAGP-1 and its relation to TGF-? in the arterial thrombosis process. For this purpose, we obtained and analyzed a mutated MAGP1 (VG1) protein, its participation in the arterial thrombosis formation and its interaction with important proteins in the coagulation process. We analyzed thrombus formation time in wild type and MAGP-1-deficency mice. The potential participation of TGF-? in this process was accessed when we treated both wild type and MAGP-1-/- mice with losartan and captopril. Besides, we evaluated the gelatinolytic activity in the arterial walls. The MAGP-1 mutation avoided its thrombus formation time recovery, although the mutated MGP-1 still interacting with vWF and fibronectin. Losartan and captopril were able to recover the thrombus formation time without changing blood pressure, aPTT and PT, platelet aggregation and adhesion, but increased gelatinases activity. Abstract: Recent works show that an increased activation of the TGF-? is associated with most of the symptoms of the Marfan syndrome. Studies using mouse models of Marfan treated with losartan have been shown to prevent the degradation of the elastic matrix. Other investigators suggest that the metalloproteinases and the noncanonical ERK signaling are involved in the breakdown of the elastic fiber, which results in the aneurysm. In this study we investigated the role of fibrillin-1 in the arterial thrombosis model using mouse models of Marfan. We analyzed thrombus formation, platelet levels, APTT, PT and TT time, hemodynamic parameter, TGF-? levels, MMP activity as well as platelets, elastic fibers morphology and thrombus. This study demonstrated that Fbn1mg?/+ mice take about 120±21,07 minutes for the thrombus to be formed when compared with wild type, 58±7,16. When these Fbn1mg?/+ mice were treated with antihypertensive losartan and captopril the time taken for the thrombus formation was reduced in 57,5% and 67,5%, respectively. The activity of metalloproteinases and activity TGF-? was increased in the Fbn1mg?/+, however no significant difference between the hemodynamic parameters, levels of totals TGF-?, morphological platelets, elastic fiber and thrombus were observed. Finally, the results suggested that fibrillin-1 interferes with this process and antihypertensive affect the physiology of Fbn1mg?/+ mice. Abstract: The aims of this work were to purify and to characterize biochemically a lectin and to determine the primary structure of a Kunitz-type serineproteinase inhibitor from seeds of Delonix regia specie (Leguminosae-Caesalpinioideae) and also to verify the in vitro effect of the inhibitor towards to digestive enzymes activities of the lepidoptera insects A. kuehniella and C. cephalonica. For lectin and serine proteinase inhibitor purification, the seeds were tritured and the flour was submitted to saline extraction 10%. After centrifugation , the supernatant (total extract) was dialised, Iyophylized and applied in molecular exclusion, ion exchange and reverse phase C18 HPLC columns. The eluted fractions were monitored by absorbance at 280 nm. The lectin and the inhibitor purified were designated DRL (Delonix regia lectin) and DrTI (Delonix regia trypsin inhibitor), respectively. After identification and purification, the lectin was analyzed in SDS-PAGE 12.5%, showing that this protein has an apparent molecular mass of 12 kDa under reduced (DTT 0.1 M) and non reduced conditions. Analysis of native gel revealed that DRL is a protein constituted by only one polypeptide chain. The inhibition tests of hemagglutinating activity were realized with rat erytrocytes, using the following sugars: D-glucose, D-glucosamine, D-fructose, Dmannose, D-sucrose, D-galactose, D-galactosamine, D-raffinose, D-Iactose and Dmaltose and we verified that hemagglutinating activity of DRL was more specifically inhibited by glucose at 0.048 mM concentration, indicating this lectin belongs to the glucose-mannose group. The lectin aminoacid composition showed high content of aspartic acid, glutamic acid and glycine, but cysteine residue was not detected. The determination of the N-terminal region of the lectin revealed high homology with other leguminous lectins such as, con A, lentil and pea. The pH range for the lectin activity was 8-9 and DRL lost activity totally in range of acid pH (2-4). Furthermore, the lectin maintained activity after thermal treatment to 70°C, during 30 minutes, with lost of 50% in activity at 100°C. DRL does not require calcium for the activity but is manganese dependent, suggesting that DRL is a metalloprotein. The treatment of DRL with urea resulted in total lost of hemagglutinating activity, while in the presence of DTT, the lectin maintained normal activity probably due absence disulfide bridges in the molecule. The primary structure determination of DrTI, proved that this inhibitor belongs to the Kunitz family of proteinase inhibitors. The region of reactive site presented a glutamic acid residue instead of an arginine or Iysine, but the molecule presented two "loops", composed by four cysteine residues, with high similarity with other Kunitz type inhibitors. The in vitro inhibition studies with insect proteases from A. kuehniella and C. cephalonica showed at concentration of only 5mg, DrTI inhibited about of 92% of the proteases these insects, indicating high affinity of the inhibitor by these enzymes.
