Abstract: Regression of atherosclerosis is still a very broad field to be searched, with mechanisms to be explored, the rabbit is an animal used internationally for research in this area. This work was carried out aiming at checking the progress and regression of atherosclerosis in hypercholesterolemic rabbits and its relation to endothelial function, lipid peroxidation and plasma and tissue. We used 30 New Zealand white rabbits initially weighing 3.0 to 3.5 kg and divided into six groups (n=5): G1: not hypercholesterolemic animals, G2: hypercholesterolemic animals, G3: hypercholesterolemic animals, followed by four months of not hypercholesterolemic diet; G4: hypercholesterolemic rats, followed by a month of not hypercholesterolemic diet and treatment with rosuvastatin (5.0mg/animal/day), G5: hypercholesterolemic rabbits, followed by two months of diet does not hypercholesterolemic and treatment with rosuvastatin (5.0mg/animal/day) G6: hypercholesterolemic animals, followed by four months of diet does not hypercholesterolemic and treatment with rosuvastatin. (5.0mg/animal/day). The animals in groups G2 to G6 were fed a hypercholesterolemic diet composed by 0.5% cholesterol and 10% coconut oil for a period of four months. At the end of the experiment were measured in plasma: total cholesterol, triglycerides and HDL-C. The LDL-C was estimated by the Friedwald formula. In aortic segments dosed 0cholesterol and MDA, as well as measured endothelial function through curves of concentration-effect with acetylcholine. Quantification of atherosclerosis in the aorta was observed macroscopically stained sudam IV, and microscopic fragments stained with hematoxylin-eosin and studied by light microscopy. Quantification was performed by specific software. The values of total cholesterol, LDL-C, HDL-C, triglycerides, cholesterol and tissue peroxidation in different groups of animals were examined by nonparametric statistical test Kruskal-Walls, followed by multiple comparison test of Dunn. The curves expressing endothelial function were compared by analysis of variance (ANOVA) for repeated measures. Considering the significant values "p"<0.05. The parameters, both biochemical and tissue were increased for G2 compared to G1. The suspension of the hypercholesterolemic diet was not enough parameters to return the tissue to the G1. The groups treated with rosuvastatin G4, G5 and G6, plasma lipids were significantly decreased, but in relation to cholesterol peroxidation and tissue in animals only significant decrease in the G6. Regarding the endothelial function, only the animals of the G6 group showed significant improvement. Only the suspension of the diet was not effective for improving the tissue values of cholesterol, lipid peroxidation, endothelial function and regress atherosclerosis. Abstract: Background: Heart failure (HF) is associated with a skeletal muscle myopathy with increased expression of fast myosin heavy chains (MHC) and extracellular matrix (ECM) alterations. The skeletal muscle-specific molecular regulatory mechanisms controlling MHC expression during HF have not been described. Myogenic regulatory factors (MRF), a family of transcriptional factors that control the expression of several skeletal muscle-specific genes, may be related to these alterations. The ECM alterations may be associated with enhanced mRNA expression and activity of matrix metalloproteinases (MMP), a family of zinc-dependent endopeptidases that degrade most ECM components and appear indispensable for the breakdown of the connective tissue surrounding muscle fibers. Objectives: This investigation was undertaken in order to examine in Wistar rat skeletal muscle with monocrotaline-induced HF: 1) potential relationships between MRF mRNA expression and MHC protein isoforms and atrophy in Soleus (SOL) and extensor digitorum longus (EDL) muscles; 2) MMP mRNA expression and their potential relationships with changes in MMP activity in SOL, EDL, and diaphragm (DIA) muscles. Methods: MyoD, myogenin, MRF4, MMP2, and MMP9 were determined by using RTPCR; MHC isoforms were separated by using polyacrylamide gel electrophoresis, and MMP activity by electrophoresis in gelatin-containing polyacrylamide gel in the presence of SDS under nonreducing conditions. Results: 1) Despite no change in MHC composition of Wistar rat skeletal muscles with HF, the mRNA relative expression of MyoD in SOL and EDL muscles and that of MRF4 in SOL muscle were significantly reduced, whereas myogenin was not changed in both muscles. This down-regulation in the mRNA relative expression of MRF4 in SOL was associated with atrophy in response to HF. 2) HF increased MMP9 mRNA expression and activity in SOL, EDL, and DIA and MMP2 mRNA expression in DIA.
