1. Introíni, Gisele Orlandi. Analise ultra-estrutural de espermatozoides de especies de bivalves marinhos: Spermatozoan ultrastructural analyses of marine bivalve species.

Degree: 2009, Universidade Estadual de Campinas

Abstract: The ultrastructure of bivalve spermatozoa can be species-specific and often provide valuable taxonomic traits for systematic reviews and phylogenetic reconstructions. Transmission and scanning electron microscopy was an efficient tool to differentiate the spermatozoan structure of species sharing many conchological characters, which could lead to equivocal identification. We analyzed sperm ultrastructural features that were useful for taxonomic issues with regard to Isognomon bicolor and Isognomon alatus (Isognomonidae); Macoma constricta, Macoma biota and Tellina lineata (Tellinidae); Donax hanleyanus and Donax gemmula (Donacidae). The data revealed that the bivalve species studied here, except T. lineata, produce primitive-type spermatozoa, typical of marine invertebrates in which eggs are fertilized in the surrounding water. We, also, compared the ultrastructure of spermatozoa from the bivalves Anomalocardia brasiliana and Tivela mactroides (Veneridae). The spermatozoa of both species contain a curved nucleus with a short cone-shaped acrosome. There were six mitochondria and glycogen clusters in the middle piece of the T. mactroides sperm cell. Possibly, the glycogen clusters and the higher mitochondria number correspond to an adaptive advantage in turbulent waters. Increasing the sperm life expectance also increases the odds of finding the eggs and accomplishing fertilization. Oysters of the genus Crassostrea and mussels of the genus Mytella, commercially important in Brazil, were also investigated in this thesis. The sperm ultrastructure of the specimens described herein and molecular studies, confirm the presence of Crassostrea brasiliana (Ostreidae) in Cananéia. Spermatozoa of mussel specimens of the genus Mytella, from two populations living in distinct habitats (muddy-sand sediment and rocky shores), were examined by transmission electron microscopy. The spermatozoa of all specimens shared the same ultrastructural pattern suggesting that the analyzed specimens, which live in distinct habitats, indeed belong to a same species, which was identified as M. charruana (Mytilidae) based on conchological analysis. The presence of an axial rod in sperm cells upholds the inclusion of M. charruana in the subfamily Mytilinae. In conclusion the electron microscopy was an efficient method to solve some particular taxonomic issues. The present spermatozoan ultrastructural studies have provided new and welcome data about the families Isognomonidae, Veneridae, Tellinidae, Donacidae, Ostreidae and Mytilidae. The results showed in this thesis could be valuable in future phylogenetic analyses. 2. Serrano, Deborah Regina. Comportamento, em camundongos, do Schistosoma mansoni oriundo de moluscos tratados com hidrocortisona.

Degree: 2002, Universidade Estadual de Campinas

Abstract: The pathogenicity of schistosomes is suggested to be influenced by the susceptibility of the intennediary host. Since hydrocortisone increases the susceptibility of mollusks to infection, we examined the behavior of Schistosoma mansoni in the final host, as well as the pathogenicity of parasites derived ftom mollusks. The treatment of mollusks with hydrocortisone did not alter the ability of cercariae to penetrate mice (Mus musculus) or the total number of wonns. The number of eggs in the feces was greater in mice infected with cercariae ftom hydrocortisone treated mollusks. Mice infected with cercariae ftom hydrocortisone treated mollusks showed more granulomas in the tiver, spleen and intestine than did mice infected with cercariae ftom non treated mollusks. In the pancreas, this difference was not marked while in the lungs the ftequency of granulomas was similar in both groups. The size of the granulomas in the spleen, lung and intestine was similar in both groups, whereas those in the tiver and pancreas were smaller in mice infected with cercariae ftom hydrocortisone treated mollusks. The spleen weight was smallest in mice infected with cercariae ftom hydrocortisone treated mollusks but there was no difference in the survival or body weights of the two groups of mice Advisors/Committee Members: UNIVERSIDADE ESTADUAL DE CAMPINAS (CRUESP), Zanotti-Magalhães, Eliana Maria, 1953- (advisor), Universidade Estadual de Campinas. 3. Franjola Tepper, Jorge Blas Ladislao. Resposta tissular em infecções por Fasciola hepatica em Lymnaea viatrix. Epidemiologia da fasciolose no gado bovino da provincia de valdivia, Chile.

Degree: 2000, Universidade Estadual de Campinas

Abstract: The adult trematode Fasciola hepatica lives in the bile ducts of herbivorous and omnivorous animais realizing an indirect life cycle for its complete development. The intermediate host is a pulmonate snail of the genus Lymnaea. In Chile, Lymnaea viatrix is the main intermediate host. The tissue response and histological modifications of L. viatrix against the infection of Fasciola hepatica miracidia during the first 72 hours of infection were observed in the present investigation. The tissue reaction started 48 hours after infection. A dead sporocyst was observed 72 hours after infection due to its encapsulation. Epidemiological study of fascioliasis was also performed in the Province of Valdivia, Chile, in 1997. The prevalence of 30.9 percent of infected animais was detected among those bought and processed in the slaughter-house of the city of Valdivia. The highest rate of fasciolosis were observed in March, April, September and October. Puerto Nuevo, Pichoy, Calle-Calle and Antilhue showed highest percentages of bovine fascioliasis although Rio Bueno presented the greatest number of infected cattle (2297). Among the cattle, male calves had the highest degree of infection. LField survey of L. viatrix was performed in 53 sites of the Province of Valdivia. From ali the sites visited only in 28 of them, samples of earth were collected for snail search. L. viatrix were not found in any of these sites probably due to the dry survey period. Snails naturally infected with F. hepatica larvae were found in the field of the Austral University of Chile (3.3%). L. viatrix used in the laboratory experiments were obtained from Teja Island, where belongs to the Austral University of Chile. It was determined that this species grows in the mud and probably feeds with micro-algae, especially of the genus Lyngbya. It was also observed that L. viatrix is the intermediate host of F. hepatica in the studied region. The samples of L. viatrix collected in field and obtained in the laboratory were smaller than those described by Paraense. In a personal communication, this author reported that the prostate of these snails were proportionally larger than those described by him Advisors/Committee Members: UNIVERSIDADE ESTADUAL DE CAMPINAS (CRUESP), Magalhães, Luiz Augusto, 1932-2017 (advisor), Universidade Estadual de Campinas.
