Abstract: Cisplatin is one of the most widely used and effective chemotherapeutic agents to treat several human malignancies. Nevertheless, its use is often hampered by the onset of serious side effects, especially on reproduction. Despite the widespread use of cisplatin for treatment of testicular cancer, which affects mainly young men, no reports were found about late reproductive effects caused by treatment during peri-puberty. This chemotherapeutic cause cross-links with sperm DNA, which may affect progeny of cancer survivors. Thus, the goals of the present study were to evaluate effects of the cisplatin administration during peri-puberty on several reproductive endpoints and the reversibility of these effects in adulthood. Moreover, implications of paternal treatment with cisplatin on progeny outcome, including reproduction of the adult male offspring, were evaluated. Peri-pubertal Wistar male rats (45 days old) were distributed into 2 groups. Control and Cisplatin (CP: 1mg/kg/day, 5 consecutive days/week, for 3 weeks, ip.). The study was conducted in two steps and evaluations were performed at ages of 66 (post-pubertal age, short-term evaluation) and 140 (adult age, long-term evaluation) days on: 1) organ weights, serum gonadotropins and testosterone levels, sperm counts, motility and morphology, testicular histo-morphometry, spermatogenesis kinetics, Sertoli cell number and apoptosis of germ cells. 2) sexual behavior, fertility and intratesticular testosterone were evaluated in the treated male and fetal development, postnatal growth and sexual development in its male and female progeny. In addition, fertility and other reproductive endpoints were examined in adult male offspring. At the end of CP-therapy, rats showed reductions in sperm production and reserves, sperm with progressive movement, tubular diameter, intratesticular testosterone and fertility potential, but increased numbers of TUNEL-positive seminiferous tubules, immotile sperm and pre-implantation losses compared to control. Furthermore, CP-treated post-pubertal rats displayed impaired testicular histology and sexual behavior. Serum gonadotropins and testosterone levels, sperm morphology, spermatogenesis kinetics and Sertoli cell number were comparable between experimental groups at both ages. No adverse effects in fetal development and puberty onset were seen in the offspring from CP-treated group. However, testicular descent was delayed and postnatal growth was impaired in these animals at short-term evaluation. Moreover, seminal vesicle weight, epididymal sperm count and testicular histology from adult progeny were affected by paternal exposure to cisplatin at short-term evaluation. In progeny, spermatogenesis was the unique parameter changed at long-term evaluation. Alterations found in post-pubertal CP-treated male were recovered at adulthood, except for sperm motility and damage to testicular histology. Abstract: The deleterious effects of endocrine-disrupting chemicals (EDC) and high-fat diets on male reproductive parameters are serious problems in modern life. Di-n-butyl phthalate (DBP) is an EDC known to cause severe reproductive anomalies when administered during sexual differentiation, but the damage caused by such exposure to the sperm parameters of the gerbil remains little known. High lipid intake in the diet has also been associated with impairments in male fertility, since it compromises sperm quantity and quality. However, lipid activity on reproductive functions varies according to the amount and nature of the lipid. The present study evaluated the consequences of gestational exposure to DBP for testicular function and sperm parameters of the Mongolian gerbil and the interference in the DBP effects of the diluent vehicle corn oil, widely used in the administration of liposoluble substances. The study also investigated the effects of a high-saturated-fat diet (lard), from weaning up to adulthood, on these reproductive aspects. Adult gerbils (16 weeks of age), born from mothers fed a standard diet, and intact (C), exposed only to corn oil (O), or to corn oil containing DBP (Ph) were used. Half of the offspring of intact mothers received a diet containing high-saturated fat, from weaning up to adulthood (D). DBP (100 mg/kg /day) and corn oil (0.1 ml) were administered to mothers, via gavage, from gestational day 8 to 23. Twelve animals were used per experimental group. Gestational exposure to DBP caused dyslipidemia, increased serum and intratesticular estradiol levels, enhanced the levels of oxidized biomolecules in the blood, and reduced sperm reserve and motility in gerbil. Maternal corn oil consumption increased, even if only marginally, the non-HDL cholesterol levels, enhanced serum estradiol levels and testicular ERalpha content, and decreased both sperm reserve and the rate of sperm without progressive movement in the adult offspring. Postnatal consumption of a lard diet increased serum levels of total cholesterol and non-HDL cholesterol, decreased intratesticular testosterone levels and increased intratesticular estradiol levels (p=0.052). These changes were accompanied by an increase in the testicular content of the nuclear receptors ERalpha and LXRalpha, as well as a reduction in sperm motility. It was concluded that gestational exposure to DBP disrupts the lipid metabolism and steroidogenesis and compromises the sperm reserve and motility of the adult gerbil. In addition, corn oil per se altered the lipid and hormonal profile and impaired sperm quantity and quality, suggesting that this vehicle interferes with the reproductive DBP response. Abstract: Ethanol induces reproductive damages, directly in the tissues or indirectly by hormonal imbalance. Prostate, a hormone-dependent gland, is susceptible to effects caused by ethanol. Emerging evidences assign to testosterone an anti-inflammatory role, with growing interest in the action of hormone therapy on the effects of ethanol. Therefore, the current research aimed to assess the effects of hormone therapy with testosterone on the ethanol consumption: 1) in tissue and plasma hormone assays, histopathology and immunolocalization and expression of hormone receptors (manuscript I); plasma levels and expression of pro and anti-inflammatory cytokines (manuscript II) in the ventral prostate of UChB rats (ethanol-preferring rat). UChB rats aged 90 days were divided into two experimental groups (n=20): C: drinking water only and EtOH: drinking 10% (v/v) ethanol at > 2 g/kg body weight/day + water. At 150 days of age, 10 rats from each group received subcutaneous injections of testosterone cypionate (5mg/kg body weight) diluted in corn oil every other day during 4 weeks, constituting T and EtOH+T, while the remaining animals (10/group) received corn oil as vehicle. All animals were euthanized at 180 days old by decapitation. Blood was collected to obtain plasma hormone and cytokines concentrations and ventral prostate was dissected, weighted and processed. Prostate sections were stained with hematoxylin and eosin, Gomori¿s reticulin and toluidine blue. The following techniques were performed: radioimmunoassay to plasma concentrations of testosterone, dihydrotestosterone (DHT), estradiol and intraprostatic testosterone, immunohistochemistry to Ki-67, AR, ER?, ER?, DACH-1, TGF-?1, pSmad 2, e-cadherin e ?-actinin, western blot to AR, ER?, ER?, DACH-1, PAR4, IL-6 IL-10, TNF? and TGF-?1 and elisa to plasma concentrations of IL-6, IL-10, TNF-? and TGF-?1. Testosterone therapy increased the ventral prostate weight. There were reducing of epithelial compartment and increasing of luminal compartment in the EtOH and hormonal therapy was able to reverse this pattern. Stroma compartment of EtOH showed thin and sparse reticular fiber bundles with rupture of smooth muscle cell layer. Inflammatory foci metaplasia, inflammatory reactive atypia, loss of cell-cell adhesion and increasing of degranulated mast cells were frequent in EtOH and absent in EtOH+T. Testosterone and ethanol did not alter the proliferation rate, but high expression of PAR4 was shown in EtOH. There was increasing of plasma and intraprostatic testosterone and plasma DHT in T and EtOH+T and decreasing of estradiol in EtOH+T. The T and EtOH+T exhibited increasing of plasma TNF? and TGF-?1, without differences regarding to IL-6 and IL-10. AR, ER?, DACH-1 and IL-6 expressions were similar among the experimental groups, differently from ER?, TNF? and TGF-?1, which decreased after hormonal therapy. Our results show that ethanol was able to induce the emergence of inflammatory foci, besides suppress the immunorreactivity to e-cadherin and ?-actinin.
