Abstract: O ambiente marinho representa uma importante fonte de diversidade biológica com grande potencial para descoberta de novos metabólitos secundários biologicamente ativos. Dentro desse ambiente, pode-se destacar os actinomicetos marinhos, cujos estudos podem contribuir para a melhor compreensão de suas funções ecológicas e para seu uso como uma fonte importante de novos metabólitos. A taxonomia dos actinomicetos é bastante complexa e a metodologia de MLSA, ferramenta alternativa em estudos de sistemática microbiana, pode ser uma técnica importante na caracterização de actinomicetos. Novos metabólitos secundários tem sido isolados de actinomicetos marinhos, sendo as atividades antibacteriana e anticâncer as principais descritas,porém pouco se conhece sobre a atividade antiviral deste grupo de micro-organimos. Neste contexto, o presente trabalho teve por objetivo avaliar a diversidade de actinomicetos associados ao ambiente marinho, utilizando uma abordagem polifásica, associada à avaliação das atividades antimicrobiana e antiviral destas bactérias. No total, foram obtidos 579 isolados de bactérias oriundos de macro-organismos marinhos coletados no litoral norte do Estado de São Paulo, Brasil. Desses, 72 foram identificados por análise de sequência do gene RNA ribossomal 16S, como actinomicetos, e nove isolados foram descritos como novas espécies de actinobactérias pertencentes aos gêneros Gordonia (B204), Marmoricola (B374), Williamsia (B138, B375 e B452), Serinicoccus (B736), Kineococcus (B366), Knoellia (B175) e Janibacter (B742). A caracterização polifásica dessas novas espécies de actinobactérias foi realizada pela técnica de hibridação DNA-DNA, Multilocus Sequence Analysis (MLSA) utilizando genes conservados (rpoB, rpoA, gyrB, recA e trpB), análise de perfil de ácidos graxos e microscopia eletrônica. A caracterização dos isolados foi complementada por testes fisiológicos e quimiotaxonômicos, tais como a determinação do conteúdo de GC, lipídios polares, menaquinonas, testes de utilização de fontes de carbono, atividade enzimática, degradação de compostos e tolerância a antibióticos, pH e temperatura. Quanto ao potencial biotecnológico dos 72 isolados de actinobactérias avaliados, 16 isolados apresentaram potencial antimicrobiano e 13 potencial antiviral. Os extratos dos isolados B175 (CIM = 1 mg/mL), B375 (CIM = 2 mg/mL), B138 (CIM = 1 mg/mL) e B366 (CIM = 2 mg/mL) apresentaram atividade antimicrobiana frente a Staphylococcus aureus ATCC 6538. O extrato do isolado B374 apresentou atividade antiviral frente ao vírus Metapneumovírus aviário (aMPV), os isolados B366 e B742, frente ao herpes vírus simplex do tipo 1 (HSV-1), os isolados B138 e B452 frente ao vírus Calicivírus Felino (FCV) e o isolado B204 frente ao vírus da diarréia viral bovina (BVDV). Abstract: Endophytic microorganisms are those that inhabit the interior of the host without causing damage or apparent symptoms, in mutualistic or commensal relations. The objective of this work was to analyze the diversity of the endophytic bacteria present in the foliar tissues of the medicinal plant "Passiflora incarnata". A total of 141 endophytic bacteria affiliated with four phyla, 17 families and 23 genera were identified, including the genera Sphingomonas, Methylobacterium and Curtobacterium, responsible for a total of 24 different species. After an initial phenotypic screening, ten isolates showed antagonistic activity against the main phytopathogens of the passion fruit crop: Bacillus sp. (1), Brevundimonas albigilva (1), Curtobacterium pusillum (2), Methylobacterium rhodesianum (1), Methylobacterium radiotolerans (3), Sphingomonas leidyi (1) and Sphingomonas yabuuchiae (1). The isolate EP58 (Bacillus sp.) was able to inhibit in up to 66.6% the fungal mycelial growth and to form inhibition halos of 30 mm against the pathogenic bacterium Xanthomonas axonopodis pv. passiflorae, demonstrating the potential to be used as a candidate for biocontrol agent. The organic extracts obtained from the most promising isolates were analyzed by HPLC-MS / MS and compounds from different classes of natural products, such as alkaloids, antibiotics and biosurfactants were detected, confirming the potential of these endophytes as producers of biomolecules of interest. A comparison of the endophytic bacterial communities of P. incarnata by culture-dependent and -independent methods as a function of the developmental stage of the plant (vegetative and reproductive) was also carried out. The results showed a great diversity of these microorganisms and the existence of specific endophytic communities according to the plant developmental stage, suggesting a modulation of the taxonomic and functional diversity by the host according to its physiological needs. Finally, the potential of P. incarnata endophytes for flavonoid biosynthesis was investigated through genotypic and phenotypic tests, since this plant is known to be a significant producer of this compound. Abstract: The Antarctic continent is considered a region with extreme climatic conditions, with low temperatures, high incidence of UV radiation and water deficit. It has a very unique biome, which means that much of its biological heritage can not be found elsewhere on the planet. Under these conditions it is expected to find adapted microorganisms (extremophiles) that can be exploited for the identification of unique and versatile metabolic arsenals with potential biotechnological applications in several areas. The development of recent research in our laboratory with bacteria and fungi in Antarctica allowed the isolation of a large number (around 1200) of microorganisms, many of them possibly indigenous, not yet cataloged or studied for the production of potential bioactive compounds. Based on this repertoire, this study aimed to identify bacterial isolates and discover natural products of biotechnological interest, focusing on antibiotics and pigments for application in the pharmaceutical or cosmetic industry. Thus, a total of 326 bacterial isolates, distributed in 39 different genera, were recovered and identified based on 16S rRNA gene sequencing. Antimicrobial screening revealed fifteen isolates capable of inhibiting the growth of at least one of the indicator strains Escherichia coli, Micrococcus luteus, Staphylococcus aureus, Bacillus subtilis and Candida albicans. A psychrotolerant bacterium, Pseudomonas sp. isolate 99, showed broad antimicrobial spectrum, besides antiproliferative and antiparasitic activity. Regarding pigments, four bacteria were shown to better withstand UV radiation and their pigments were extracted and chemically identified. All selected pigments were identified as carotenoids and exhibited high antioxidant activity and good stability under ultraviolet light exposure.
