Abstract: One of the major challenges in the field of prostate cancer research is the search for model systems that allow the investigation of pathological, biochemical and genetic factors of this disease. The gerbil Meriones unguiculatus has enabled the evaluation of prostate lesions and evolution from benign to malignant (invasive) stage after a relatively short period of treatment with the carcinogen N-methyl-N-nitrosourea (MNU), a potent causative of DNA methylation by direct action. Thus, this study aimed to: 1) Determine the incidence, multiplicity, and tumor latency of spontaneous and chemically-induced lesions in ventral and dorsolateral gerbils' prostatic lobes; 2) Investigate whether estradiol exerts protective and/or promoter hole on neoplasms induced by MNU; 3) Evaluate the promotional potential of high-fat diet on induced-carcinogenesis in ventral prostate; 4) Analyze the involvement of altered ras gene products and the global DNA methylation status of prostate epithelium on MNU-mediated tumor process. Therefore, we used adult animals subjected to a single intraperitoneal dose (50mg/kg) of MNU, except the control group. The experimental groups were subjected to chronic exposure of androgen, estradiol or high-fat diet for 14 and 28 weeks. The methodologies involved quantitative and statistical analysis of multiplicity and incidence of prostatic lesions, body weight, body fat accumulation, prostate weight, hormonal measurements, proliferative index, karyometry, frequency of AR-positive and basal cells, global methylation status and determination of protein expression. The model of prostatic tumor induction by MNU associated with testosterone in the gerbil was effective because it reduced tumor latency and allows the study of advanced stages of carcinogenesis after short period. Neoplasms manifest initially in dorsolateral lobe and require a longer time to be established in ventral lobe. However, the progression from premalignant to malignant lesions occurs more significantly in the ventral lobe. This indicates that tumor progression occurs differently between prostatic lobes and alternative pathways maybe possibly involved in this process. Long exposure to high doses of estrogen was able to prevent and reduce the rate of tumor growth. Despite therapeutic effects against neoplastic progression, estrogen therapy led to the establishment of an epithelium with distinct characteristics from normal prostate, such as changes in the pattern of DNA methylation and increased amount of basal cell and ARpositive cells. Together, these events contributed to create an unstable epithelial compartment that can cause lesions recurrence in subsequent periods. The association between MNU and high-fat diet promoted an increase in incidence of lesions stimulated by carcinogen alone, which had a higher number of AR-positive cells, disruption of the smooth muscle layer indicating tumor microinvasion and high reactivity for metalloproteinase type 2. Abstract: Ethanol induces reproductive damages, directly in the tissues or indirectly by hormonal imbalance. Prostate, a hormone-dependent gland, is susceptible to effects caused by ethanol. Emerging evidences assign to testosterone an anti-inflammatory role, with growing interest in the action of hormone therapy on the effects of ethanol. Therefore, the current research aimed to assess the effects of hormone therapy with testosterone on the ethanol consumption: 1) in tissue and plasma hormone assays, histopathology and immunolocalization and expression of hormone receptors (manuscript I); plasma levels and expression of pro and anti-inflammatory cytokines (manuscript II) in the ventral prostate of UChB rats (ethanol-preferring rat). UChB rats aged 90 days were divided into two experimental groups (n=20): C: drinking water only and EtOH: drinking 10% (v/v) ethanol at > 2 g/kg body weight/day + water. At 150 days of age, 10 rats from each group received subcutaneous injections of testosterone cypionate (5mg/kg body weight) diluted in corn oil every other day during 4 weeks, constituting T and EtOH+T, while the remaining animals (10/group) received corn oil as vehicle. All animals were euthanized at 180 days old by decapitation. Blood was collected to obtain plasma hormone and cytokines concentrations and ventral prostate was dissected, weighted and processed. Prostate sections were stained with hematoxylin and eosin, Gomori¿s reticulin and toluidine blue. The following techniques were performed: radioimmunoassay to plasma concentrations of testosterone, dihydrotestosterone (DHT), estradiol and intraprostatic testosterone, immunohistochemistry to Ki-67, AR, ER?, ER?, DACH-1, TGF-?1, pSmad 2, e-cadherin e ?-actinin, western blot to AR, ER?, ER?, DACH-1, PAR4, IL-6 IL-10, TNF? and TGF-?1 and elisa to plasma concentrations of IL-6, IL-10, TNF-? and TGF-?1. Testosterone therapy increased the ventral prostate weight. There were reducing of epithelial compartment and increasing of luminal compartment in the EtOH and hormonal therapy was able to reverse this pattern. Stroma compartment of EtOH showed thin and sparse reticular fiber bundles with rupture of smooth muscle cell layer. Inflammatory foci metaplasia, inflammatory reactive atypia, loss of cell-cell adhesion and increasing of degranulated mast cells were frequent in EtOH and absent in EtOH+T. Testosterone and ethanol did not alter the proliferation rate, but high expression of PAR4 was shown in EtOH. There was increasing of plasma and intraprostatic testosterone and plasma DHT in T and EtOH+T and decreasing of estradiol in EtOH+T. The T and EtOH+T exhibited increasing of plasma TNF? and TGF-?1, without differences regarding to IL-6 and IL-10. AR, ER?, DACH-1 and IL-6 expressions were similar among the experimental groups, differently from ER?, TNF? and TGF-?1, which decreased after hormonal therapy. Our results show that ethanol was able to induce the emergence of inflammatory foci, besides suppress the immunorreactivity to e-cadherin and ?-actinin. Abstract: In contrast to the classic boarding, the prostate is not an exclusive gland of the male organism and it can be found in mammal females, including humans and rodents. The precise comprehension of the post-natal development and possible histopathological processes related to this gland are difficult because of its anatomical paraurethral localization and it restricting the study in necropsy material. However, the similarity between the Mongolian gerbil prostate (Meriones unguiculatus) and women one makes it a good animal model to study the morphofunciotional feature of this gland. The analyse of the epithelial and stromal components in the prostate of young, adul and old ages, the histopathological disorders decurrent of aging and their morphological answer related to the hormonal supplementation by deidroepiandrosterona (DHEA) were the objective of this work. The results were based on structural, ultrastructural, morphometric-steriological studies and hormonal analyzes (except the treated animals). The young female prostate showed an early gland functionaly compared to the male in the same age and it was probably associated to the serum stradiol levels. The aging was marked by the reduction of the serum steroids and intensification of the benign, pre-malignant and malignant histopathological alterations. Altought these same changes had already developed in the adult age, they had lesser incidence. Hormonal supplement promoted instability of the gland homeostasis in the young age inducing the development of benign and malign histophatological alterations beyond the high biossinthetic stimulation on the secretory cells. Some modificated cellular phenotypes were identifyed in the gland secretory epithelium and it could be amplifyed during the senesce period and after the hormonal administration. In this way, the real condition of the prostatic histopathology is alarming and few atention is given to this gland wich is too inherent to the female genital system, especially regarding the senescence and therapies of hormonal supplementation. Maybe it is necessary an adaption in the female clinic routin since this female ogan exists and thus the gland could be monitored and some discomforts and damages of the women health could be prevented.
