Abstract: The current study pays attention to changes of the postnatal brainstem angiotensin receptors by maternal protein restriction (LP), and its impact on in uteri programming of hypertension in adult life. The data shows that LP male pup body weight was significantly reduced when compared to that of control (NP) or LP taurine-supplemented (LPT) pups. That fall in in-born LP body weight was rapidly reverted in 12-wk-old LP offspring. The reduced body weight was associated with striking decreases in the neurons number in inborn brainstem LP offspring, when compared to age-matched LP and LPT. Also, the immunohistochemistry analysis demonstrated a significantly decreased expression of type 1 AngII receptors (AT1R) in the entire medial solitary tract nuclei (NTS) of LP rats at 16-week of age compared to age-matched NP and LPT offspring. Conversely, the immunostained type 2 AngII (AT2R) receptors in 16- wk-old LP medial NTS were unchanged, although, they were expressively decreased in rostral and caudal NTS subnuclei. The present investigation shows an early decrease in fractional urinary sodium excretion in maternal proteinrestricted offspring reverted by diet taurine supplementation. The decreased fractional sodium excretion was accompanied by a fall in proximal sodium excretion and occurred despite unchanged creatinine clearance. Abstract: Chronic elevated plasma insulin levels and resistance to the hypoglycemic effect of insulin have been associated with increased blood pressure in human and animal models of hypertension. This observation has led to the speculation that insulin may play a role in the development of increased blood pressure. Although authors have shown that the peripheral action of insulin reduces urinary sodium excretion, suggesting an attractive reciprocal link between insulin¿s renal effect, urinary sodium excretion and the development or maintenance of hypertension, studies from our laboratory have indicated that acute intracerebroventricular injection of insulin significantly increases the output of sodium. Thus, we hypothesized that central resistance to the natriuretic effect of intracerebroventricular administration of insulin could lead to an inability of renal tubules to handle the hydroelectrolyte balance. To test this hypothesis, we investigated the effects of acute intracerebroventricular insulin administration on sodium tubular handling in SHR and WKy, and characterized the direct actions of insulin on the PI 3-kinase and MAP-kinase in the hypothalamus of these rats. Furthermore, we investigated the long-term effect of intracerebroventricular administration of insulin on sodium tubular handling and arterial pressure of normotensive rats. We demonstrated that chronic intracerebroventricular insulin injection attenuated urinary sodium excretion. However, this altered response did not simultaneously change the arterial blood pressure at this time. We functionally characterized the MAPK-ERK-1/2 as the intracellular hypothalamic pathway responsible for insulin-mediated natriuresis in rats. On the other hand, our results show that central insulin administration is less effective in inducing activation of the MAPK pathway and natriuresis in SHR rats, characterizing these rats as centrally resistant to insulin-mediated natriuresis. Abstract: Considering that there is a significant increase in processed foods rich in saturated fat intake, mainly in Western populations, is important to study the effect of this change in the incidence of diseases. Obesity, hyperglycemia and hypertension have been associated with high circulating levels of oxidizing metabolites, such as superoxides. It is known that both maternal and paternal diets, maternal stress and intrauterine ischemia have epigenetic effects that may alter the gene expression of fetuses leading to increased predisposition to diseases in adult life. In this context, the present study aims to analyze maternal long-term consumption of hyperlipidic diet (HFD), associated with the use of the antioxidant Tempol on the mothers and offspring male rats. Sprague-Dawley rats were fed a HFD (60% of calories from lipids), added to Tempol for 6 weeks prior to pregnancy and throughout gestation and lactation. HFD dams showed significantly lower water and food consumption, with an increase in caloric intake and a consequent increase in body mass. HFD added the antioxidant group (DTempol) had a decrease in weight gain during pregnancy and changes in potassium excretion at the end of the lactation period. HFD offspring with or without Tempol (Diet and Dtempol) had a lower birth weight compared to the Control group. After 16 weeks HFD offspring had a reduction in HDL levels and a surprising decrease in the adiposity index, which was normalized by the antioxidant consume. There was an increase in blood pressure in both groups from HFD dams. Glucose and insulin metabolism were altered HFD offspring, with decreased insulin secretion. DTempol also presented a drop in glomerular filtration and modifications in the sodium and potassium renal manipulation at the 16th week. Maternal long term HFD consumption induced a decrease in nephron number in offspring at 12 days, but with normalization in adulthood (16 weeks of life). In addition, offspring of HFD dams presented important renal morphological changes characterized by: inflammatory infiltrate, increased renal collagen deposition, and elevated renal expression of mesenchymal epithelial transition markers such as desmin and vimentin. These changes were accompanied by elevation in urinary protein excretion and appear to be linked to reduced SOD expression and elevated NADPH expression. Also, HFD with Tempol dams treatment promoted a significant attenuation of the described inflammatory and morphological changes. Thus, the study showed that HFD intake can promote fetal programming in male offspring rats with future renal changes characterized by expression of glomerular proteins that suggest that these kidneys are subject to the epithelial-mesenchymal transition.
