All cells have specific procedures to degrade proteins to free amino acids, and thus regulate the proteome. ATP-dependent protease complexes are generally compartmentalized formed by several subunits regulated by post-translation modification and requires energy from ATP hydrolysis. Among these complexes, two are threonine protease: the complex HslV/U and 20S proteasome. Trypanosoma cruzi is a rare example of organisms that have genes for these two proteolytic complexes. In this study, we used two approaches to infer the coexpression of these complexes in epimastigotes from Berenice 62, Berenice 78, Cl, Colombian and Y T. cruzi populations. Initially, we determined the transcription profile of the complex HslV/U and 20S proteasome beta-1, -2 and -5 subunits using qRT-PCR. The results showed that mRNA levels of hslv are two times smaller in Be-78 population when compared to others. Moreover, the transcription level of hslu not varies between populations. The expression profile for beta -1 and -5 genes did not show significant variations, however, the beta -2 gene was 4.5 times less expressed in Be-62 when compared to other populations. Subsequently, we evaluated the protein levels of the complex HslV/U and 20S proteasome using antibodies for the alpha-7 and HslV specific to T. cruzi. The results showed that the relationship HslV/proteasome follows two distinct profiles: one for the Y population, which has equivalent levels for these two proteases and the other for Be-62, Be-78, Cl and Colombiana populations, with predominance of the 20S proteasome. Also we have shown a translational regulation of the 20S proteasome when epimastigotes were grown in the presence of PSI, a specific proteasome inhibitor. Together, the results suggest the coexistence of the protease HslV/U and 20S proteasome in T. cruzi reinforcing the hypothesis that the non-lysosomal proteolysis mediated by dependent and independent of ubiquitin mechanisms plays an important role in cell biology of this parasite.

Todas as células possuem processos específicos para degradar proteínas à aminoácidos livres, e assim, regular o proteoma. Proteases ATP- dependentes são geralmente complexos compartimentalizados, altamente regulados e formados por várias subunidades, cuja função requer energia proveniente da hidrólise de ATP. Dentre esses complexos compartimentalizados, podemos citar duas treoninas proteases, o complexo HslV/U e o proteassoma 20S. O Trypanosoma cruzi é um dos raros exemplos de organismos que apresentam genes para estes dois complexos proteolíticos. Neste estudo, nós utilizamos duas abordagens para inferir a coexpressão destes complexos em epimastigotas obtidos das populações Berenice 62, Berenice 78, Cl, Colombiana e Y do T. cruzi.
