▼ YDL100cp is the ArsA homologous protein found in Saccharomyces cerevisiae. Previous studies show that deletion of YDL100c was not lethal but unable to grow at 40°C. To study the role of YDL100c in response to lethal heat shock, the wild type strain (WT) and YDL100c disrupted strain (KO) were exposed to 50°C for 30 min. The growth and survival rate of KO cells at 30°C after heat-shock was lower than that of WT cells, and the difference was complementated by introducing the plasmid carrying YDL100c. The oxidative stress has been shown to be involved in the heat-induced cell death in S. cerevisiae. Therefore, the intracellular molecular oxidation level, expression of antioxidant genes, trehalose accumulation, and glutathione (GSH) content were further examined. The intracellular molecular oxidation was increased in KO compared to WT when exposed to 50°C, suggesting heat-shock-induced cell death is related to oxidation of intracellular components. The results also demonstrated that both WT and KO had a decreased GSH content and trehalose accumulation after heat-shock, indicating that GSH and trehalose are not directly involved in the slow growth of KO after heat-shock. However, CTT1 expression is decreased in KO compared to WT when exposed to 50°C, suggesting that decreased CTT1 expression resulted in the increased intracellular oxidation and YDL100c is likely involved in the activation of CTT1 expression.

Cisplatin is one of the most effective chemotherapeutic agents used in treatment of a variety of human solid tumors. The most common adverse side effect limiting the use of Cisplatin is nephrotoxicity. Recent studies indicate that inflammatory and oxidative signaling play critical role in pathogenesis of Cisplatin related nephrotoxicity. Cisplatin-induced nephrotoxicity is associated with lipid peroxidation and the superoxide anion and hydroxyl radical scavenger could prevent acute renal failure through both attenuation of tubular damage and enhanced regenerative response of the damaged tubular cells. It has been shown that green tea polyphenols with antioxidant properties inhibit inflammatory and oxidative responses in mice. However, the evidence indicating the protective effect of epigallocatechin gallate (EGCG), the major polyphenol found in green tea, on Cisplatin-induced nephrotoxicity is lacking. The present study is to evaluate the effect of EGCG injection on Cisplatin-induced nephrotoxicity in rats. The male rats were divided into four groups (n = 6 each); control group, Cisplatin group, EGCG group and Cisplatin + EGCG group. The control group received only intraperitoneal normal saline injection. Cisplatin (6 mg/kg) was given single dose intraperitoneally at day 0, EGCG (10 mg/time) was given subcutaneously at day 4, day 2 and day 0 before Cisplatin challenge and day 2 and day 4 after Cisplatin injection. Nephrotoxicity was evaluated by biochemical analysis of blood and histopathological observations of rat kidney. Nuclear factor-kappa B (NF-κB) activation, inducible nitric oxide synthase (iNOS) expression and malonyldialdehyde (MDA) content were also determined in rat kidney. Cisplatin injection induced an increase in serum blood urea nitrogen, creatinine and tubular necrosis, and upregulation of NF-κB and iNOS expression and MDA content in kidney. All the increases were significantly inhibited by EGCG treatment. The results suggest that EGCG may attenuate Cisplatin induced nephrotoxicity through the anti-inflammatory/oxidative effects.

▼ Many studies show that silver is an effective antimicrobial agent. Thin film metallic glasses (TFMGs) exhibit excellent corrosion resistance and much better hydrophobic property. It follows that TFMGs might be promising for improving the surface properties and antimicrobial ability of substrates in biomedical applications. However, the development and investigation of antimicrobial ability and biocompatibility for metallic glasses are still rare. Besides, the mechanism of antimicrobial effects of TFMGs is still unknown. To understand the interaction between bacteria and TFMGs, it is investigated by comparing the antimicrobial activity between the Al48Ag37Ti15 TFMG, Zr54Ti35Si11 TFMG, and Zr59Ti22Ag19 TFMG. In this study, all the TFMGs were deposited on glass by co-sputtering processes. Afterward, the superior TFMGs were also deposited onto the modified 316L stainless steel (SS). The antimicrobial performances of the TFMGs are tested with Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa which are the most common nosocomial infection pathogens. The experimental results show that the antimicrobial effect of Al48Ag37Ti15 and Zr59Ti22Ag19 TFMG is almost similar to thin film of pure Ag. Moreover, the result of Zr54Ti35Si11 TFMG is shown to be non-cytotoxic to mouse bone marrow stem cells using an MTS assay for proliferation. These results are expected to impose significant contributions to the development of biomedical devices.

▼ Since Louise Joy Brown, the first baby of In Vitro Fertilization-Embryo Transfer (IVF-ET), was born in the United Kingdom in 1978, there had been over five million successful cases over the course of 35 years. There are two commercially available culture media in the market: Single Step Medium (Global Medium) and Sequential Medium (Sage Medium). The development of these two media had enhanced the quality of embryo and increased the pregnancy rate in IVF-ET. In this collection of 210 cases of IVF-ET, oocytes were fertilized by intracytoplasmic sperm injectionï¼ICSIï¼and randomly assigned to the two commercially-available culture media. The quantity and quality of embryos were observed and recorded for three consecutive days in order to compare the development of embryos in the two commercial culture medium. A fertilized rate of 75.5% in Global Medium and 77.5% in Sage Medium were observed on the first day. There was a higher number of cleaved cells in Global Medium and embryos of Grade 1 cells than those in Sage Medium both on the 2nd and the 3rd day, though no significant differences were noted in the pregnancy rates between the two samples. In the group of those over 38 years old, there was a higher number of cells in Global Medium, but exhibited no difference in either embryonic pattern or in the pregnancy rate. Though the development of commercial culture medium had enabled a longer cultivation, some cases still failed be cultured to the blastocytic stage due to other limitations. It had been attempted with morphologic selection in embryos by the 3rd day of culture, with a criteria of over six cells per embryo and minimal cell fragmentation, to reach compatible pregnancy rate of embryo implantation of the 5th day by conventional method. Our study indicated that the there was higher number of Grade 1 embryos with more than 6 cells and lower embryo fragmentation in Single Step Medium, thus yielded a better overall quality of embryo than Sequential Medium. It can be speculated, therefore, that the Single Step Medium would offer a valuable choice in patients with difficulty in reaching blastocytic stage by conventional method due to various factors.

▼ Use of nitrifying and denitrifying bacteria to remove ammonia from waste water had been studied for a long time due to their high efficiency and low cost. Nitrifying bacteria not only grow slowly but also require high concentration of oxygen to facilitate the nitrifying process. Moreover, the followed denitrifying process needs the supply of adequate carbon sources for denitrifying bacteria to avoid greenhouse gas emission from the system. It shows the operational control to remove ammonia from waste water would be very difficult. Therefore, it is important to study the physiological and biochemical characteristics of those nitrifying and denitrifying bacteria closely. In 1995, Mulder discovered the disappearance of ammonium at the expense of nitrate and nitrogen production from their denitrifying pilot plant in the Netherlands, then van de Graaf verified an ANAMMOX reaction in the laboratory. Further studies that have revealed the combination of aerobic nitrification and anaerobic ammonium oxidation is more efficient to remove ammonia than most conventional methods. The ANAMMOX process is performed by a group of Planctomycete which involves the oxidation of ammonia anaerobically with nitrite as the final electron acceptor to yield gaseous nitrogen. Since this process is no need of supply external carbon source and oxygen, the ANAMMOX system can offer the advantages of less cost, less microbial contamination and less N2O and NO emission to the environment. This study is to summarize the bacterial species diversity, distribution in nature, their physiological characteristics, and potential biochemical pathways of those nitrogen converting microorganisms. In addition, several novel nitrogen removal technologies are also discussed for further understanding of the process optimization under both aerobic and anaerobic conditions.

▼ Microbacteria are everywhere even they canât been seen by eyes directly. There are seventy-one percent of the earthâs surface covered by ocean. Within different location or ocean current etc. causing intense different environment. Marine bacteria have been through evolution all the time begin. We can discover the existence of bacterial no matter at the deep-sea with high pressure, or volcano on the sea-bed. There will be development different bacterial community structure according to the environmental factors. So we can analysis the relationship between the area and bacterial diversity by collecting the sample in different location. In this study, we sampled the surface seawater and the sediment from Dongsha island to research the bacterial community structure. But due to 99% of marine bacteria can not be incubated in laboratory, we use high throughput sequencing technique to analysis uncultured marine bacteria. Dongsha island doesnât open to the public, so we infer the microbiota may be more close to the origin environment. Pass through the sequencing analysis we found that the Gammaproteobacteria is the dominant class not only in the sediment but also in seawater. But the most common dominant bacterium in the normal sea water is Alphaproteobacteria not Gammaproteobacteria. We infer that Dongsha is a island in rich of coral reef and the Gammaproteobacteria is one of the dominant bacterium associate with coral or they were been washed away from land in to aqua environment. So we can find abundance Gammaproteobacteria in our sample no matter in sediment or sea water. The subdominant bacterium is Alphaproteobacteria in seawater and Bacilli in sediment. Maybe it is come from the organic product of plant on the land by wade.

▼ Diabetes mellitus is characterized by elevated plasma glucose and increased rate of skin infection. Altered immune responses have been suggested to contribute to this prevalent complication. High glucose treatment reduced human β-defensin-3 (hBD3) expression of cultured keratinocytes. This pathogenic process involved inhibition of p38MAPK signaling, an event that resulted from increased formation of advanced glycation end product and showed worse anti-Staphylococcus aureus activity. In addition, high-glucose cultivated keratinocytes expressed reduced levels of human β-defensin-2 (hBD2) and pSTAT-1. Besides the impact on keratinocyte cultured under high glucose, the suboptimal interaction between keratinocytes and inflammatory cells also contributed to impaired diabetic wound healing. High-glucose environment enhanced interleukin (IL)-8 production via EGFR-ERK pathway in a ROS-dependent manner in keratinocytes. Treating diabetic rats with neutrophil inhibitor improved the healing. On the other hands, high glucose cultivated monocytes have significantly reduced production of IL-22, a molecule that is responsible for promoting keratinocyte migration. In summary, high glucose environment impared keratinocyte function directly or indirectly that contributed to impaired diabetic wound healing, and focusing on these defects will present a therapeutic approach to promote diabetic healing.

▼ The purpose of this study is to investigate the treatment efficiencies of PCDD/Fs and Hg contaminated soil by phytoremediation, and to learn the translocation and distribution of PCDD/Fs and Hg in the plant tissues. In addition, the object of tests of adding amendments (citric acid, P. mendocina
NSYSU and EDTA) into the testing systems are to assess the effects of plants on removal rates of PCDD/Fs and Hg from the contaminated soil. The results of this research were expected to provide important reference for remediating similar contaminated sites in the future. In this study, the pot tests in greenhouse were conducted by using the contaminated soils sampled from China Petrochemical Development Corporation (CPDC) An-shun site located in Tainan City, Taiwan. The selected plant species included Bidens pilosa, Leucaena glauca, Vetiver, Paspalum vaginatum, Commelina diffusa, Maize, Physic Nut and Squash. These plant species were all characterized as rapid growth, so it would be helpful and convenient for using them in different stages of test runs.
The experimental results showed that the toxicity of PCDD/Fs and Hg might inhibit the growth of plants. The higher the concentrations of PCDD/Fs were controlled in soils, the less the height and biomass of the plant were observed. In addition, the plant roots were found able to promote the activities and diversity of microbial in the areas of rhizosphere. Besides, the congener profiles and concentration levels of PCDD/Fs between the contaminated soil and the shoot and root of plant species are highly positive correlation. In this study, it is also confirmed that the PCDD/Fs and Hg were first phytoextracted into the plant roots, and then were translocated into the shoot. However, the concentrations of PCDD/Fs and Hg of root were measured much higher than the shoot. The results of tests of adding amendments showed that citric acid, P. mendocina
NSYSU and EDTA would promote the uptake ability of plants for PCDD/Fs and Hg, especially the plant species of vetiver and maize. In additions, adding EDTA into test systems presented effective improvement for the plant species of vetiver and maize uptaking Hg. The experimental results for PCDD/Fs and Hg accumulating in plant tissues showed that the species of vetiver performed better than the species of maize, which meant that the species of vetiver exhibited better resistance and higher biomass than the species of maize. Moreover, the removal rates of PCDD/Fs from soils were found higher for the species of maize than the species of vetiver. However, for Hg removal rates, the experimental results indicated that the species of vetiver was found greater than the species of maize. Thus, the removal effects of PCDD/Fs and Hg from contaminated soils would vary with the different plant species. It was concluded that the higher capability of extracting PCDD/Fs and Hg into plant tissues and the ability of promoting the rhizosphere microbial activity make both species of vetiver and maize become suitable candidates for phytoremediation in soils…

▼ OSTC, a Homo sapiens OSTC/DC2 protein, belongs to a new subunit of mammalian oligosaccharyltransferase (OST). OST transfers the high-mannose sugars to nascent polypeptides during N-linked glycosylation in the lumen of the rough endoplasmic reticulum. In this study, we used synthetic peptides (RMKLPGYLMG) as an antigen to develop a polyclonal antibody against OSTC/DC2. After heat treatment of Balb/c mice, OSTC/DC2 showed up-regulated in some tissues of brain, and peaked at the 12th hour of recover stage. Furthermore, OSTC/DC2 was also up-regulated in cell lines during heat-shock treatment. The strikingly immediate up-regulation of OSTC/DC2 in response to heat shock is only seen in the cortex, suggesting that OSTC/DC2 may have unique functions in neurons. In addition, OSTC/DC2 induction is synchronous to increased expression of HSP70 and activation of AKT after heat shock, supporting the notion that OSTC/DC2 may play a significant role in heat shock response.

▼ The offspring of maternal immune activation (MIA) mice induced by environmental allergen lipopolysaccharides (LPS) is an animal model to reflect the observation of anxiety- and autism-like behaviors in human. In this project, we hypothesized that, during pregnancy, mouse exposure to low doses of LPS directly lead to the offspring occuring the neuronal damages, the changes of intenstinal microflora, aberrance of immune response and neurotransmistters, and ultimately result in developing anxiety- and autism-like bahaviors. The abilities in activation of splenocytes, proliferation of bone marrow cells as well as raised serum cytokines and chemokines were firstly evaluated for endotoxemic mice that respectively induced by Burkholderia spp. and Escherichia coli LPS at low doses. Although B. multivorans LPS potentially had the strongest immune stimulation, E. coli LPS can induce an increase of CD16/32+CD45lo brain infiltrating leukocytes. Thus, MIA were induced by E. coli LPS at 100 ïg/Kg, totally. The appearance of anxiety-like behaviors in MIA offspring were evaluated using open-field test, dark/light boxes test and elevated plus maze test as well as of autism-like behaviors by home cage test and 3-chamber behavior test. Those mice with anxiety- and autism-like behaviors exhibited (1) a down-regulation of cerebral serotonin (5-HT), 5-HT synthetic enzyme gene (tph2) and 5-HT transporter gene (slc6a) in combination with decrease of the number and distributed density of 5-HT neurons, (2) alteration of intestinal microbiota, pariticularly is predominately Clostridium cocleatum, (3) the changes of interferon-induced genes (gbp3, irgm1 and ifi44), IL-2-induced gene (jak3), apoptotic genes (tnfrsf1, nlrp3 and nr4a1) and cell adhesion molecule genes (sell and selp) in the brains, and (4) up-regulation of cerebral IL-6 at adolescent and adulthood but decrease of MCP-1 at adulthood as well as down-regulation of L-selectin at adolescent and adulthood. Moreover, TNF-ï¡, IL-6, IFN-ï§, MCP-1, L-selectin and P-selectin in the brain were increased for MIA offspring re-stimulated by LPS as compared to PBS offspring re-stimulated. Our results indicated that the MIA offspring appeared an imbalance of neurotransmitters and brain development on shor-term effect as well as the changes of intestinal microbiota that directly and indirectly effected on aberrant expression of IL-6, MCP-1 and L-selectin and development of 5-HT neuron and neurotransmitters at long-term effect. Ultimately, anxiety- and autism-like behaviors were generated in MIA offspring.

▼ Chlorinated aliphatic hydrocarbons are frequently found as contaminants of soil and groundwater as a result of their widespread use in various industrial processes and improper disposal methods. When they are released into the subsurface, they tend to adsorb onto the soils and cause the appearance of DNAPL (dense-non-aqueous phase liquid) pool. The use of enhanced bioremediation technology for soils and groundwater sites contaminated with chlorinated organic compounds is a feasible technology for site remediation. During the bioremediation process, bioavailable substrates can be injected into soil and groundwater to stimulate local microbial growth and create an ideal environment for the growth of dechlorinating bacteria for pollution remediation. This study used a chlorinated organic compound contaminated site as a studied site. Site investigation and remedial system design were performed. In this study, the causes of groundwater pollution was also assessed. Site investigation results show that some of the groundwater samples were contaminated with chlorinated organic compounds. Enhanced bioremediation has been applied for groundwater remediation. Biodegradable substrates have been injected into the subsurface to remediate the contaminated groundwater. After the remediation process, concentrations of chlorinated organic compounds dropped to below the groundwater standards.

▼ This research is focus on the analysis of microbial diversity and existance of methanotrophic microbial strains in Wu-Shan-Ding mud volcano areas. The microbial distribution and diversity in soil surrounding the mud volcano were analyzed by polymerase chain reaction (PCR) and denaturing gradient gel electrophoresis (DGGE). The 16S rDNA sequences from PCR-DGGE bands were used to identify the bacterial strains with NCBI gene bank data. The results showed that the microbial diversity in the fresh erupted mud sample varied with other soil samples. The degrees of diversity were propotion to the distance away from the mud volcano. The main bacterial species found in the mud sample are Brevundimonas terrae, Ralstonia solanacearum, Ralstonia taiwanensis, Pseudonocardiaceae bacterium, Ochrobactrum anthropi, Burkholderia phytofirmans, Stenotrophmonas sp., Methylobacterium sp., Cryseobacterium sp., Sphingobacterium sp., Stenotrophomonas sp. MG-3, and Stenotrophomonas sp. EP01. According to documents, all of the above strains are tolerant to high salt and alkaline. The Ochrobactrum anthropi and Burkholderia phytofirmans were found only in the fresh erupted mud sample and the 5-meter soil sample, while Ralstonia mannitolilytica, Ralstonia pickettii, Amycolatopsis rugosa, and Maricaulis sp. were found only in 10-meter soil sample. In other word, all of the soil samples we examed in this study have their own specified bacterial strains. Two methanotrophic microganisms, Methylobacterium organophilum and Ochrobactrum anthropi were also successfully isolated in pure cultures. The biochemical characteristics of these two strains had been studied. These two strains show a potential might be able to use in the bioremediation of petroleum pollution.

▼ This study investigated the microbiota in ten different drinking water treatment pools, particles in the Biological Activated Carbon Filtration (BACF) bed, and two mimic columns in the Cheng-Ching Lake Water Treatment Plant. Assimilable organic carbon (AOC) is one of the main nutrition sources for microbes to survive in tap water. Over growing microbes not only decrease the water quality, but also contaminate the water treatment system and distribution system. In this study, we used two molecular biology techniques, the polymerase chain reaction (PCR) and denaturing gradient gel electrophoresis (DGGE), to analyze the dynamic microbial communities and biodiversities in the drinking water cleaning system and the micorbiota that exist in the BAC and anthracite filtration pellets. The bacterial 16S rDNA sequences resulted from PCR-DGGE were compared with the data in the Ribosomal Database Project Bank to construct a phylogenetic tree which allowed us to understand the microbial communities and biodiversities in the drinking water treatment pools and the filtration pellets. The total bacterial count and PCR-DGGE profiles showed that the drinking water quality had been improved during the treating processes and most of the microbes in raw water were removed. The scanning electron microscopy clearly indicated the biofilms were developed on the pellet surface. From the mimic column studies, the PCR-DGGE profiles suggested that various microbial communities were present on different depth of the columns samples. In comparing the 16S rDNA sequences with Gene Bank, many are new category bacteria were found and most of them are unculturable. Most of these microbes belong to the beta-proteobacterium. Although many bacteria were located on the surface of the filtration pellet, the BAC and anthracite could still absorb AOC efficiently to enhance the bacteria growth. The over growing bacteria might release out and contaminate the drinking water. Therefore, we suggest that it is important to backwash the filter bed frequently in order to diminish microbes of the filtration pellet and avoid re-contaminate the drinking water.

Deinococcus radiodurans is highly resistant to radiation when it grown in tryptone-glucose-yeast extraxt (TGY) medium. It oxidized glucose slowly mainly by the pentose phosphate pathway (PPP) and showed little glycolytic Embden-Meyerhof pathway (EMP) activity. The addition of 10 µM Mn(II) into the stationary phase cultures, could induced new round of cell division (Mn-CD effect) and the EMP activity. Glucose metabolized by Mn-CD cells at a EMP:PPP=6:1 ratio. In analyzing the metabolites for DNA repair, we found that after the addition of Mn(II) , the concentrations of PPP metabolites such as insione monophosphate (IMP)ãuridine monophosphate (UMP) and NAD (nicotine adenine dinucleotide) were greatly reduced. This event is also occurred when replacing the glucose by fructose, sodium acetate, or removing glucose from the TGY culture medium. Besides, we also found that the TGY and TFY grown cells contained more PPP metabolites than those of TAY and TY cells. This finding suggested that glucose and fructose were metabolized by the PPP pathway in D. radiodurans. Finally, the concentrations of IMPãUMP and NAD in the cells were greatly decreased after UV irradiation. This indicated that these metabolites were probably employed to repair the DNA damage causing by UV irradiation.

Ras proto-oncogenes are members of the superfamily of GTP-binding proteins. Many tyrosine kinase receptors, including those for epidermal growth factor, insulin, and nerve growth factor, signal through RAS proteins. The product of members of this oncogene family (H-, K-, N-ras) is a 21 kD protein with nucleotide binding activity, involved in the transduction of information from the cell surface to the nucleus. The three RAS proteins exit in two states: a resting state in which they are bound to GDP and an active state in which they bind GTP. The most common form of mutational activation of Ras oncogenes in human tumors is through single base substations affecting either the GTP-binding of main (codons 12 and 13) or the GTPase domain (codon 61) of the protein. Thus, mutant RAS proteins result in constitutive activation of the downstream signaling cascade because their affinity for GTP is increased or their GTPase activity is decreased, so that the protein cannot return to the resting state. To investigate we have screened 89 thyroid tumor specimens, which include 8 follicular carcinomas (FC), 42 papillary carcinoma (PTC), 2 anaplastic carcinoma (AC),5 Hurthle cell adenoma (HA), 12 follicular adenoma (FA) and 20 nodular goiter (NG), for mutation in three Ras genes using PCR and automatic sequencing. Four tumors contained Ras gene mutation. Of these, three were identified among FC (37.5%), which mutation were in the codon 61 of each Ras genes. One mutation were at codon 61 of N-ras in FA specimens (8.3%). In addition, 33.7% (30/89) of specimens contain H-ras codon 27 polymorphism. In conclusion, our data indicated that the prevalence rates of Ras gene mutation were 5.8% and 2.7% in thyroid carcinoma and thyroid benign adenoma, respectively. Other environmental or genetic factors might also involved in the thyroid tumorigenesis and worth further investigation. The data were further confirmed using the combination of the PCR and denaturing gradient gel electrophoresis (DGGE). Four more cases of possible Ras mutation were detected which did not revealed by automatic sequencing , indicating that DGGE is a more sensitive method in detecting single nucleotide mutation. DGGE analysis should increase the detection rate of Ras gene mutation in our analysis.

▼ Taura syndrome virus (TSV), the epidemic agent of Taura syndrome (TS), was tentatively classified in the family Dicistroviridae, caused disastrous losses with high mortality rates from 60 to >90% of affected pond-cultured Litopenaeus vannamei. The amino acid sequence alignments of ORF1 revealed sequence motifs characteristic of a helicase, a protease and an RNA-dependent RNA polymerase, similar to the non-structural proteins of picornavirus, suggesting perhaps a similar to picornavirus polyprotein processing. Proteolytic processing of the TSV polyprotein and protease is still unclear. In order to further understand the role of protease interaction with polyprotein, several clones were constructed. In this study, we constructed several clones that expressed the TSV virus coat protein(VP-polyprotein)ï¼pTSVVPï¼, putative 3C proteaseï¼pTSV3Cï¼, proteaseï¼RdRp-polyprotein(pET3CD) and both VP+3C proteins(pTSVVP3C) in E. coli. The VP-polyprotein band was excised from SDS-polyacrylamide gels and electroeluted for antibody production in mice. Processing of the VP-polyprotein by 3C protease was carried out in the in vitro co-transcription/translation system and detected by Western blot. The results showed there were protein bands corresponding to the sizes of VP2+VP1 and VP1+VP3 indicating that the processing might have been partial or incomplete.

▼ Incidence of dengue fever (DF) has been estimated a 30 fold increase in the past 50 years. Clinical manifestations of DF range from a simple febrile illness with physical soreness to life-threatening dengue hemorrhagic fever (DHF). The need for a better classification of the severity in DEN infections has been proposed to clarify the immunopathogenesis for the prevention and management of serious DEN infections. We attempted to investigate whether different mechanisms involved in the varied manifestations of bleeding tendency and vascular leakage in DF. In a hospital-based study, we first compared clinical features as well as laboratory data including virus load, T helper (Th1/Th2) cytokines, and vascular leakage-related mediators between patients with DHF and DF. Moreover, we defined another class of patients associated with bleeding tendency but not fulfilled with DHF criteria, called DF w/B, for a further comparison. The virus load in blood was not significantly different among DF, DHF and DF w/B. DF patients had a higher Th1 cytokine, IFNr, expression (70.0 ± 10.7 vs. 33.1 ± 8.0 vs. 33.0 ± 7.1 pg/ml; DF vs. DF w/B, p = 0.009; DF vs. DHF, p = 0.002), and both DHF and DF w/B patients had a significantly higher IL-10 levels (14.3 ± 4.1 vs. 26.2 ± 3.3 vs. 26.0 ± 3.5 pg/ml; DF vs. DF w/B, p = 0.023; DF vs. DHF, p = 0.016) than DF patients. Both DHF and DF w/B patients also had a higher rate of secondary dengue infection (DF w/B vs. DHF vs. DF: 50.0%, 74.4% and 14.3%ï¼ p < 0.001). By contrast, DHF but not DF w/B patients had significantly higher vascular leakage-related mediators: sVCAM-1, PGE2 and TNFα levels than DF patients. Patients with DF w/B had a higher platelet counts (DF w/B vs. DHF: 66.0 ± 8.3 vs. 20.7 ± 2.1 x109/L, p < 0.001) but lower ALT levels than those with DHF (DF w/B vs. DHF: 56.3 ± 7.7 and 144.7 ± 20.5 IU/L). This study provides new insight to different immune mechanisms involved in patients with DF, DF w/B, and DHF. DF involves augmented Th1 reaction, and DF w/B involves altered Th2 reaction, but DHF involves both altered Th2 reaction and augmented vascular insult. Clarification of the immune mechanisms among DF, DFw/B and DHF will facilitate certain specific treatment and prevention of DF patients from varied bleeding tendency and vascular leakage manifestations.
