▼ The objectives of this study were to develop an in vitro direct adventitious bud induction and an organogenic callus induction and shoot regeneration system via floral organ segments culture for bromeliads, moreover, explore the effect of auxin on plantlet elongation of Guzmania. And further, apply the above micropropagation system to physical and chemical methods to induce somaclonal variances of bromeliad plantlets in vitro for mutation breeding. The explant sources of bromeliads and the components of culture medium were studied to develop a micropropagation system for bromeliads. The results indicated that the 1/3MS basal medium supplemented with a combination of 1.0 mg l-1 BA + 0.5-1.0 mg l-1 NAA, or a combination of 3.0 mg l-1 BA + 0.5 mg l-1 NAA, showed the highest frequency of direct initiation of adventitious buds derived from shoot apex and lateral bud explants of Aechmea fulgens var. fulgens and Guzmania 'Focus'. The best results of adventitious buds induction of the both species were found in the lower lateral bud explants, at 47.5% and 35%, respectively. In addition, the adventitious buds began to form on day 16 after the G. 'Focus' decapitated plantlets had been cultured in medium supplemented with 3.0 mg l-1 BA + 0.5 mg l-1 NAA. However, this phenomenon did not occur in case of undecapitated explants, where only protruding nodules appeared. Petal- and ovary-derived calli of A. fasciata and G. 'Hilda' were induced on 1/2MS basal medium supplemented with 1.0-1.5 mg l-1 2,4-D in combination with 1.0 or 0.5 mg l-1 NAA. Organogenic calli were cultured on medium with 1.0 mg l-1 NAA and 0.5 mg l-1 TDZ could be induced to differentiate and regenerate the adventitious buds. Furthermore, the number of adventitious buds proliferating at the base of the plantlets derived from G. 'Hilda' floral organs, cultured in media with different concentrations of IAA, IBA, NAA, and 8-azaadenine, was only 1-2 adventitious buds individually. This result shows that auxin can indeed suppress cytokinin-effects. The influence on plantlet elongation was greatest in the treatments using 0.5 mg l-1 NAA and 1.0 mg l-1 NAA. After 4 months culture, plantlets grew to 5.73 and 5.62 cm in height, that was 2.22 and 1.95 cm higher than the control, respectively. Plantlets of A. fasciata hardened under the middle (50 Î¼mol m-2s-1) light intensity condition had a higher survival rate, 95%, than that hardened at a low light intensity (1 Î¼mol m-2s-1; 17.5%). The maximum number of newly developing roots, up to 4.15 per shoot, was also observed at the same light intensity treatment. During transplantation, plantlets growing in coir fiber showed the best results in terms of plant growth within 6 months ex vitro culture. The average length of the plantlets was 22.0 cm, and an average of 19.3 leaves per plantlet was achieved. When calli of G. 'Hilda' treated by sodium azide, the survival rate was 0%. The survival rate of decapitated plantlet explants treated with 0.5 mM sodium azide for 60 minutes was 51.3%, about half-lethal dose. ▼ Nasopharyngeal carcinoma (NPC) is a head and neck cancer that rarely occurs in most parts of the world. In contrast, the highest incidence was found in the southern Chinese population of Guangdong, Inuits of Alaska, and native Greenlanders. Pluchea indica (L.) Less is a well-known indigenous medicinal plants that grows in littoral areas of tropical regions of countries such as Malaysia, Indonesia, Australia, Taiwan, India, and Mexico. The plants have been traditionally used as astringent and antipyretic. The present study is to investigate the anti-cancer effect of the ethanolic extract of Pluchea indica (L.) Less root (PIRE) and its possible molecular mechanism in human nasopharyngeal carcinoma (NPC) cells. Cell viability were measured by WST-1 experiments and Colony formation assay. Cell migration were measured by wound healing assay and transwell assay in vitro, respectively. The sub-G1 ratio was examined using flow cytometry and the extent of DNA fragmentation was evaluated by the Tdt-mediated dUTP nick end labelling (TUNEL) assay. The expression of apoptosis-related proteins were assessed by Western blotting. We found that PIRE could strongly inhibit cell viability of NPC-TW01 and NPC-TW04 cells in dose-dependent manner; however, NPC-TW04 cells showed more susceptible to the treatment. Furthermore, cell migrations were also suppressed with exposure to PIRE. This study also found that PIRE could significantly increase the occurrence of the sub-G1 phase and the extent of DNA fragmentation in a dose-dependent manner, which showed that, PIRE significantly increased apoptosis in NPC cells. The apoptotic process triggered by PIRE involved the up-regulation of pro-apoptotic Bax protein and the suppression of anti-apoptotic Bcl-2 protein expression. In the meantime, we also observed that PIRE could increase the ratios of Bax/Bcl-2 protein levels and up-regulation of the p53 protein in a concentration-dependent manner. Therefore, PIRE could induce the apoptosis signaling pathway in NPC cells by activation of p53 and by regulation of apoptosis-related proteins. In summary, this study suggest that Pluchea indica could be further investigated as a new alternative chemotherapeutic agent for NPC. ▼ We studied how the endogenous and exogenous cytokinin and gibberellin functions in floral initiation and development in tuberose. In the aspect of cytokinins. The contents of endogenous cytokinin in tuberose corms (Polianthes tuberosa L. cv. Double) at vegetative, early floral initiation, and flower development stages were investigated. We also determined the influence of exogenous cytokinin treatment on the corm apex at three different growth stages in relation to floral initiation and development in tuberose. The exogenous cytokinin effectively induced floral initiation and development, especially at the early floral initiation and flower development stages. Endogenous cytokinins were higher in early floral initiation and development stages in comparison to the vegetative stage. During floral initiation stage, the zeatin and dihydrozeatin increased significantly, while the cytokinins, zeatin riboside, dihydrozeatin riboside, 6N-(Î´2-isopentenyl) adenine, and 6N-(Î´2-isope -ntenyl) adenine riboside at consistently low levels. The increase of cytokinin levels in tuberose corms during floral induction suggests a role for cytokinins in tuberose apex evocation. Moreover, these results indicate that cytokinins seem to promote the development of flower buds rather than inducing flowering in tuberose. Endogenous gibberellins (GAs) in tuberose corms were isolated using high performance liquid chromatography, bioassay and identified by combined capillary gas chromatography-mass spectrumetry. Gibber -ellins A19, A20 and A53 were quantified at the vegetative, early floral initiation and flower differentiation stages. The identification of these 13-hydroxylated GAs indicates the presence of the early-13-hydroxy -lation in tuberose corms. An increase in GA20 and decrease in GA19 coincided with the transition from the vegetative phase to the stages of early floral initiation and flower differentiation. GA53 maintained at constant levels at three different growth stages. When GA3, GA4, GA20 and GA32 were applied to corms at vegetative stage (plants about 5 cm in height ), floral initiation was induced and/or promoted by several, most notably by GA3, GA20 and GA32. It is suggest that hydroxylated C-19 GAs play an important role in flower induction in tuberose. ▼ Abstract This research is to explore the high school student potential misconceptions on evolution, based on the two-tier diagnostic test and diagnostic interview. Through a proper teaching design, the researcher has developed an untraditional teaching method. The effects of the untraditional teaching method and the traditional teaching method were compared. With the retest of two-tier diagnostic test, a survey was conducted to evaluate the studentsâ achievement resulting from the teaching strategy designed by the researcher. The survey can also be consulted and used in the improvement of future teaching activities. This research adopted a Quasi-experimental research method. Data included the analysis of quantity and the description of quality. The research tool is a two-tier diagnostic questionable. The sample groups consist of two classes in the grade-10 and two classes in the grade-12 with 35 students in each class, taught by the researcher. The results of this research indicate that (1) in general , the students are in lack of the structural knowledge of evolution in their prior learning especiallyâthe relationship between organism and organismâand the concept of human selection; (2) the comparison of learning results brought about between the untraditional teaching method and traditional teaching method, apparently shows that the experimental group students are far better than the control group students in understanding the connection of evolutional concepts and the abundance of conceptive maps; (3) the difference of student learning process of the two teaching methods , compared by the quiz results on each unit, are not significantly different between the two groups of students; and (4) as far as the teaching of the concept of evolution is concerned, the improved teaching method design is appreciated by the experiment group students and is highly expected by the control group students. ▼ The ability of exogenous gibberellin (GA) A3 to substitute for low temperatures in inducing flowering in Phalaenopsis hybrida under high temperatures suggests a similar role for endogenous GAs in normal (low temperature) development of the inflorescence and floral buds. Changes in endogenous GAs in shoot-tips (about 2.0 cm long) were examined when potentially flowering shoot was 2-3 cm and 7-10 cm long, respectively. Treatments included warm control (30/25 â day/night, non-flowering), GA3-treated (30/25 â day/night, flowering) and cool-induced (25/20 â day/night, flowering) plants. GA3 at 0.5 Î¼g•shoot-1 was injected into the cavity of the second visible bud scale below the flowering shoot apex. The contents of the tips of potentially flowering shoot of GA1, GA20, GA19 and GA53 tended to be highest in cool-induced plants, intermediate in GA3-treated plants, and lowest in the warm controls (non-flowering). Tips of potentially flowering shoots in warm controls also contained higher GA8 relative to GA3-treated and cool-induced plants. Tips of potentially flowering shoots (2-3 cm in length with no flower primordium), contained low levels of GAs than older 7-10 cm long flowering shoot (with flower primordium) under cool-inductive conditions. These results suggest that continued inflorescence development and flower bud initiation are closely associated with increases in endogenous GAs, even in GA3-treated plants. They also suggest that a more rapid biosynthetic flow-through from GA19âGA20âGA1âGA8 leaving reduced levels of GA1 may be involved in the inhibition of flowering and shoot elongation seen in warm (30/25 â day/night) control plants. A reduced conversion of âactiveâ GA1 to inactive GA8 may thus be required for successful floral initiation and development whether induced by cool temperature or by applied GA3. These results imply that the high temperature inhibitory effect on flowering is mediated through its effect on lowering the level of endogenous GA1 and its precursors.
