▼ High mobility group box 1 (HMGB-1) protein is a non-histone chromosomal protein. As a DNA binding protein, HMGB-1 is involved in the maintenance of nucleosome structure, regulation of gene transcription and it is active in DNA recombination and repair. It has been known that HMGB-1 is a late mediator of endotoxemia and sepsis. HMGB-1 is released from activated macrophages, induces the release of other proinflammatory mediators, and mediates cell death when overexpressed.
We speculated that the course of sepsis maybe different without the involvement of HMGB-1. The aims of this study are to investigate the role of HMGB-1 in mediating sepsis and to observe the effects by using RNAi to affect the production of HMGB-1. Lipopolysaccharide (LPS) was used to simulate sepsis in culture as well as stimulate the release of HMGB-1 from RAW 264.7 cells. Levels of HMGB-1 in the culture medium were subsequently measured by Western blot. Other proinflammatory cytokines (TNF-α, IL-6 and TGF-β) were measured by ELISA. HMGB-1 could not be detected in the culture medium in the absence of LPS stimuli, but after 0.5 μg/ml LPS treatment HMGB-1 release could be detected. HMGB-1 the amount of released from LPS activated RAW 264.7 cells was in a time- and dose-dependent manner. The present study demonstrated that RNAi in the treatment of LPS-stimulated RAW264.7 cells resulted in the blockade of HMGB-1 and decreased LPS-induced inflammatory response. The results demonstrated that HMGB-1 plays a pivotal role in macrophage inflammatory responses by modulating the production of inflammatory mediators.

▼ Head and neck cancer (HNC) makes up 6 ï¼ of the cancer patients
in the world every year. This disease usually occurs in males and the
incidence is increasing year by year. According to statistical analysis,
HNC has less than 50 ï¼ five-year survival rate. Therefore, the research
of HNC seems imminent so that may lead to the development of new
approaches of diagnosis and therapy.
Recent research had shown that expression of podoplanin caused
cellular proliferation, and may be associated with tumor invasion,
metastasis and malignant prognosis. Podoplanin, a mucin-type
transmembrane sialoglycoprotein, is highly expressed in lymphatic
endothelial cells but not expressed in vascular endothelial cells. The
purpose of this study was to determine the clinical and pathological
significance of podoplanin in oral squamous cell carcinoma (OSCC).
Therefore, we collected clinical specimen and associated patient history
of OSCC. Further, we used the human cell lines of HNSCC (Fadu, Hep2)
to investigate the molecular regulation of podoplanin.
Podoplanin expression was analyzed by RT-PCR and Western blot assay
firstly. As shown, podoplanin was found to be overexpressed in tumors compared with normal adjacent tissues. Further, immunohistochemical
analysis revealed the location of podoplanin expression in OSCC tissues.
The results showed that podoplanin had higher expression in T4 stage
tumor section than in normal adjacent tissues of OSCC samples, or in T1
stage. Here, podoplanin was highly expressed in the OSCC tumor cell
and lymphatics of stage T4 OSCC tissue. Furthermore, we found that
overexpression of podoplanin in OSCC patients was associated with
decreased five-years survival rate. In the univariate analysis, several
factors were statistically significantly associated with disease-specific
survival rate, including Tumor stage, Nodal stage, and podoplanin
expression. In the subsequent multivariate analysis, only Tumor stage and
Nodal stage showed a trend toward worse disease-specific survival. To
further investigate the regulatory mechanism of podoplanin and its
position of expression within the cell, immunofluorescence and
transfection were utilized to assay. The results showed that podoplanin
was expressed in the nuclear membrane of the Fadu and Hep2 cell lines,
and the PI3K/AKT signaling pathway was involved. We suggest that the
role of podoplanin in OSCC should be further investigated for potential
future treatment.

▼ OSTC, a Homo sapiens OSTC/DC2 protein, belongs to a new subunit of mammalian oligosaccharyltransferase (OST). OST transfers the high-mannose sugars to nascent polypeptides during N-linked glycosylation in the lumen of the rough endoplasmic reticulum. In this study, we used synthetic peptides (RMKLPGYLMG) as an antigen to develop a polyclonal antibody against OSTC/DC2. After heat treatment of Balb/c mice, OSTC/DC2 showed up-regulated in some tissues of brain, and peaked at the 12th hour of recover stage. Furthermore, OSTC/DC2 was also up-regulated in cell lines during heat-shock treatment. The strikingly immediate up-regulation of OSTC/DC2 in response to heat shock is only seen in the cortex, suggesting that OSTC/DC2 may have unique functions in neurons. In addition, OSTC/DC2 induction is synchronous to increased expression of HSP70 and activation of AKT after heat shock, supporting the notion that OSTC/DC2 may play a significant role in heat shock response.

Emerging diseases such as middle east respiratory syndrome, severe acute respiratory syndrome or other infections induced by Ebola virus, Bacillus anthracis, and Legionnaireâs disease which are all related to lung inflammation and lung edema with high mortality. The mechanisms involved in these emerging diseases could be related to the microbes infection and the inflammation reactions involved in the lung. The inflammation of the lung could be induced by sequestered of the inflammatory cells in the lung, so that oxidative stress increased, pulmonary arterial pressure increased, and the permeability changes in pulmonary capillaries. Besides, blood cells sequestered in the lung-induced ischemia/ reperfusion (I/R) of lung tissues are also related. Necrosis of intestines or inflammation of pancreas also could induce systemic inflammatory responses, so that hemodynamic changes in the lung, pulmonary capillary permeability changes and lung edema ensured.
In the pilot study we proved that ischemia/ reperfusion of the lung tissues induced lung water increase (p < 0.01), lavage protein in pulmonary capillary bronchial alveolar lavage (PCBAL) increase (p < 0.001) and the permeability coefficient of pulmonary capillary increase (p < 0.05). After administration of water soluble oxygen radical scavenger, dimethylthiourea, the lung injury parameters attenuated significantly (PCBAL, p < 0.01; LW/BW, p < 0.05). However, big molecules of oxygen radical scavengers such as superoxide dismutase and catalase did not prove with protective effects (p > 0.05). These could be related to the big molecules of superoxide dismutase and catalase which canât penetrate into cells.
Further experiment, we demonstrated that ischemia/ reperfusion of pancreas induced inflammation of pancreas and the elevation of airways reactivity which could be related to the I/R of pancreas-induced increase of inflammation white cell, tumor necrotic factor, hydroxyl radicals, and nitrosative stress. After administration of curcumin (antioxidant), all these responses are attenuated. Since post I/R of pancreas induced increase of amylase, lipase, protease in the blood, it proved indirectly that pancreatic juice exudates into the blood (p < 0.01), so that intrapulmonary inflammation and lung edema.
In this study we set up the model by aerosolized of pancreatic juice and infused the aerosol directly into airway. We demonstrated that aerosolized of pancreatic juice into airway could induce obstructive and restrictive ventilatory insufficiency which are related to the juice-induced lung inflammation by which increase of blood concentration of white cells in pulmonary capillary. Decrease of RBC counts and increase of the concentrations of hemoglobin, hematocrit, mean corporal volume, and the mean weight of hemoglobin. Besides, platelet concentration also decreased. ▼ Damage to peripheral nerves following trauma or disease has a number of consequences including burning pain, muscle wasting, paralysis, or organ dysfunction. The most common form of neuropathy is that associated with metabolic abnormality, notably diabetes. Many diabetics, especially those with poor blood sugar control, ultimately develop a distal symmetrical and painful neuropathy that initially affects the longest peripheral axons, but with time spreads proximally. Deficiency in neurotrophic support has been proposed to contribute to the development of diabetic neuropathy. Recently, peripheral gene delivery of vascular endothelial growth factor (VEGF), neurotrophin-3 (NT-3), NGF, BDNF or hepatocyte growth factor (HGF) has been shown to facilitate the continuous production of neurotrophic factors and alleviate the diabetic neuropathy. The role of glial cell-derived neurotrophic factor (GDNF) in the pathogenesis and therapeutics of diabetic neuropathy is not well defined. The main objectives of this research sought to inspect the protective effect of GDNF peripheral gene delivery during hyperglycemia- or constriction- induced sciatic nerve injury in rats. In present proposal, we propose to investigate the change in organization and expressions of GDNF signaling complex in the sciatic nerve following injury in the initial stage. Subsequently, the recombinant adenovirus was used gene delivery system for GDNF to evaluate the potential of intramuscular administration of gene delivery for prevent nerve degeneration, and the molecular mechanism of GDNF to ameliorate neuropathy will be clarified. The above study would enable us to test the hypothesis that the topical gene delivery might be a suitable strategy for the treatment of diabetic neuropathy and other disorders in peripheral nerve. Furthermore, the results of animal studies might be extrapolated for future clinical application.

▼ Diabetes mellitus is characterized by elevated plasma glucose and increased rate of skin infection. Altered immune responses have been suggested to contribute to this prevalent complication. High glucose treatment reduced human β-defensin-3 (hBD3) expression of cultured keratinocytes. This pathogenic process involved inhibition of p38MAPK signaling, an event that resulted from increased formation of advanced glycation end product and showed worse anti-Staphylococcus aureus activity. In addition, high-glucose cultivated keratinocytes expressed reduced levels of human β-defensin-2 (hBD2) and pSTAT-1. Besides the impact on keratinocyte cultured under high glucose, the suboptimal interaction between keratinocytes and inflammatory cells also contributed to impaired diabetic wound healing. High-glucose environment enhanced interleukin (IL)-8 production via EGFR-ERK pathway in a ROS-dependent manner in keratinocytes. Treating diabetic rats with neutrophil inhibitor improved the healing. On the other hands, high glucose cultivated monocytes have significantly reduced production of IL-22, a molecule that is responsible for promoting keratinocyte migration. In summary, high glucose environment impared keratinocyte function directly or indirectly that contributed to impaired diabetic wound healing, and focusing on these defects will present a therapeutic approach to promote diabetic healing.
