1. Dimitrova, Maria. The effect of experimental diabetes on the cardiac oxytocin system.

Objective: To elucidate the effect of type I diabetes mellitus on the cardio-protective oxytocin (OT)-natriuretic peptides (NPs)-nitric oxide (NO) signaling cascade.

Methods: 200-250 grams Male Wistar rats injected intraperitoneally with 65 mg/kg streptozotocin (STZ) to induce experimental diabetes were sacrificed 5.5 and 10.5 weeks later. Gene and protein expression levels were determined by Real-Time PCR and Western blot, respectively.

Results: A significant increase in the gene expression of OT in the left ventricle (LV) was found after both 5.5 (640%) and 10.5-weeks (420%) of diabetes. Elevated gene expression was additionally found in the LV in both time frames for atrial natriuretic peptide (ANP) (370% and 160%, respectively) and brain natriuretic peptide (BNP) (210%, 190%).

Conclusion: Upregulation of the local OT-NPs system occurs in the LV of the diabetic rat after both 5.5 and 10.5 weeks of untreated diabetes, perhaps exemplifying an adaptive mechanism aimed at opposing the deleterious effects of hyperglycemia.

Objectif: Elucider l'effet du diabète mellitus type I sur la cascade signalisatrice de l'oxytocin (OT)- peptides natriurétiques (NPs)-oxyde nitrique (NO).

Méthode: Des rats Wistar masculin de 200-250 grammes ont été injectés intrapéritonéament avec 65 mg/kg de streptozotocin (STZ) pour produire un diabète expérimental. Ces derniers ont été sacrifiés 5.5 et 10.5 semaines plus tard. Les niveaux d'expressions des gênes et des protéines ont été déterminés en temps réel PCR et Western blot, respectivement.

Résultats: Une augmentation significative dans l'expression de d'OT dans le ventricule gauche à été trouvée dans les deux cas: après 5.5 (640%) et 10.5 semaines (420%) de diabète. Une augmentation élevée de l'expression des gênes à été observée dans le ventricule gauche dans les deux horizons temporels pour l'ANP (370% et 160%, respectivement) et pour le BNP (210%, 190%).

Conclusion: La régulation à la hausse du système OT-NPs se produit dans le ventricule gauche du rat diabétique après les deux périodes de 5.5 et 10.5 semaines de diabète laissé sans traitement, ce qui peut exemplifier un mécanisme adaptatif visant à opposer l'effet délétère de l'hyperglycémie.
